Abstract
The invention provides application of a traditional Chinese medicine composition in preparation of a medicine for treating psoriasis, which is characterized by comprising the following raw material medicines in parts by weight: 20-200 parts of vinegar rhizoma corydalis, 10-160 parts of ligusticum wallichii, 10-160 parts of radix clematidis, 20-200 parts of divaricate saposhnikovia root, 20-200 parts of pepper, 10-160 parts of radix achyranthis bidentatae, 20-200 parts of lycopodium clavatum, 20-200 parts of northeast garden balsam stem, 10-160 parts of beautiful sweetgum fruit, 20-200 parts of erythrina bark, 1-10 parts of menthol and 2-15 parts of borneol. The invention relates to a guinea pig psoriasis model caused by propranolol, and researches the influence of the traditional Chinese medicine composition on the guinea pig psoriasis. The experimental result shows that the traditional Chinese medicine composition provided by the invention has a good protective effect on experimental psoriasis of guinea pigs.
Description
Technical Field
The invention belongs to the field of traditional Chinese medicines, and particularly relates to application of a traditional Chinese medicine composition in preparation of a medicine for treating psoriasis.
Background
Psoriasis is a common and recurrent chronic inflammatory skin disease, and the clinical manifestations are erythema, papule and scale. The prevalence rate in China is about 0.47%, and the prevalence rate in the world is 0.09% -11.43%, and the prevalence rate has a trend of rising year by year. Psoriasis is apt to develop or aggravate in spring and winter, and is relieved in summer and autumn. Has long course of disease and easy recurrence, and most patients often suffer from life-long disease, which causes great pain to the patients. The cause of psoriasis is not clear, and is thought to be mainly related to genetic, infectious, immune abnormality, endocrine, environmental factors and the like. Psoriasis can be classified into vulgaris, arthrosis, pustule and erythrodermia according to different clinical characteristics, wherein the vulgaris accounts for more than 99%, and other types are mostly converted from psoriasis vulgaris.
Western medicine treatment of psoriasis usually adopts oral medicines such as vitamins, antibiotics, anti-tumor medicines, heavy metals, immunomodulators or agonists, medicines acting on nervous systems and the like, or external medicines such as hormones, keratinocyte, cytotoxic agents and the like, and simultaneously combines ultraviolet irradiation, photochemical methods and the like to comprehensively treat the psoriasis. However, clinical practice proves that the medicament and the therapy have short remission period, more adverse reactions and high treatment cost.
Therefore, the development of a new medicine with remarkable treatment effect has important significance for treating psoriasis.

The invention aims to provide an application of a traditional Chinese medicine composition in preparing a medicine for treating psoriasis, wherein the traditional Chinese medicine composition comprises the following components: 20-200 parts of vinegar rhizoma corydalis, 10-160 parts of ligusticum wallichii, 10-160 parts of radix clematidis, 20-200 parts of divaricate saposhnikovia root, 20-200 parts of pepper, 10-160 parts of radix achyranthis bidentatae, 20-200 parts of lycopodium clavatum, 20-200 parts of northeast garden balsam stem, 10-160 parts of beautiful sweetgum fruit, 20-200 parts of erythrina bark, 1-10 parts of menthol and 2-15 parts of borneol.
Further, the traditional Chinese medicine composition comprises: 70-107.5 parts of vinegar rhizoma corydalis, 65-86 parts of ligusticum wallichii, 80-86 parts of radix clematidis, 107.5-120 parts of divaricate saposhnikovia root, 107.5-130 parts of pepper, 60-86 parts of achyranthes root, 75-107.5 parts of lycopodium clavatum, 107.5-110 parts of northeast speranskia herb, 40-86 parts of beautiful sweetgum fruit, 90-107.5 parts of erythrina bark, 3.6-7 parts of menthol and 7.2-10 parts of borneol
Preferably, the traditional Chinese medicine composition comprises the following raw material medicines in parts by weight: 107.5 parts of vinegar rhizoma corydalis, 86 parts of ligusticum wallichii, 86 parts of radix clematidis, 107.5 parts of divaricate saposhnikovia root, 107.5 parts of pepper, 86 parts of achyranthes root, 107.5 parts of lycopodium clavatum, 107.5 parts of northeast garden balsam stem, 86 parts of beautiful sweetgum fruit, 107.5 parts of erythrina bark, 3.6 parts of menthol and 7.2 parts of borneol.
The raw material medicines can be directly ground into powder, or can be extracts or other forms prepared by a conventional method. The composition of the present invention may also be directly ground into powder, or may be an extract prepared by a conventional method in the art, etc.
Specifically, the psoriasis treatment drug is selected from an oral administration dosage form, an injection administration dosage form or an external administration preparation.
Specifically, the medicine for treating psoriasis is selected from decoction, granules, capsules, tablets, oral liquid, pills, tinctures, syrups, suppositories, gels, sprays and injections.
Optionally, the preparation method of the traditional Chinese medicine composition comprises the following steps:
distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil, and collecting volatile oil; extracting the residue with 6-20 times of water, filtering the extractive solution, and collecting the filtrate; adding 50-90% ethanol 6-20 times the amount of rhizoma corydalis, herba Lycopodii, northeast herba speranskiae tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting, filtering the obtained extractive solution, recovering ethanol, mixing with the above water extract, and concentrating to obtain extract; mixing the volatile oil and the extract, and adding Mentholum and Borneolum Syntheticum.
Further, when the psoriasis treatment drug is selected from gels, the preparation method of the gels can be as follows:
distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil for 8 hr, and collecting volatile oil; extracting the residue with 15 times of water for 1.5 hr, filtering the extractive solution, and collecting the filtrate; adding 70% ethanol 12 times the amount of rhizoma corydalis, herba Lycopodii, northeast herba Speranskiae Tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting for 2 times, mixing extractive solutions, filtering, recovering ethanol, mixing with the above water extraction partial filtrate, and concentrating to obtain extract with relative density of 1.20-1.30;
dissolving carbomer in appropriate amount of distilled water, swelling completely, sequentially adding triethanolamine and glycerol, stirring, sequentially adding ground co-melted Mentholum and Borneolum Syntheticum, volatile oil, extract and appropriate amount of distilled water to 1000g, grinding, and packaging.
Further, when the psoriasis treatment drug is selected from gels, the preparation method of the gels can be as follows:
distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil for 6 hr, and collecting volatile oil; extracting the residue with 12 times of water for 2 hr, filtering the extractive solution, and collecting the filtrate; adding 60% ethanol 12 times the amount of rhizoma corydalis, herba Lycopodii, northeast herba Speranskiae Tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting for 2 times, mixing extractive solutions, filtering, recovering ethanol, mixing with the above water extraction partial filtrate, and concentrating to obtain extract with relative density of 1.20-1.25;
dissolving carbomer in appropriate amount of distilled water, swelling completely, sequentially adding triethanolamine and glycerol, stirring, sequentially adding ground co-melted Mentholum and Borneolum Syntheticum, volatile oil, extract and appropriate amount of distilled water to 1000g, grinding, and packaging.
Further, when the psoriasis treatment drug is selected from an ointment, the preparation method of the ointment can be as follows:
distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil for 8 hr, and collecting volatile oil; extracting the residue with 10 times of water for 1 hr, filtering the extractive solution, and collecting the filtrate; adding 70% ethanol 10 times the amount of rhizoma corydalis, herba Lycopodii, northeast herba Speranskiae Tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting for 2 times, mixing extractive solutions, filtering, recovering ethanol, mixing with the above water extraction partial filtrate, and concentrating to obtain extract with relative density of 1.20-1.25;
mixing the volatile oil and the extract uniformly, adding the menthol and borneol which are ground to be co-melted, a proper amount of glycerol, triethanolamine and liquid paraffin to 1000g, mixing uniformly and subpackaging to obtain the traditional Chinese medicine.
The invention relates to a guinea pig psoriasis model by using propranolol, and researches the influence of the traditional Chinese medicine composition on the guinea pig psoriasis. The experimental result shows that the traditional Chinese medicine composition provided by the invention has a good treatment effect on the psoriasis of the guinea pig.
Detailed Description
As mentioned above, the present invention discloses the application of a Chinese medicinal composition in the preparation of drugs for treating psoriasis, and the following will be described in detail with reference to the contents of the experimental examples.
It is specifically noted that all similar substitutes and modifications for the present invention which are obvious to those skilled in the art are deemed to be included in the present invention. While the methods and applications of this invention have been described in terms of preferred embodiments, it will be apparent to those of ordinary skill in the art that variations and modifications in the methods and applications described herein, as well as other suitable variations and combinations, may be made to implement and use the techniques of this invention without departing from the spirit and scope of the invention. It is to be understood that the described embodiments are merely exemplary of the invention, and not restrictive of the full scope of the invention.
If the specific conditions are not indicated, the method is carried out according to the conventional conditions or the conditions suggested by manufacturers, and the used raw material medicines or auxiliary materials and the used reagents or instruments are the conventional products which can be obtained commercially.
The invention is further illustrated by the following examples:
EXAMPLE 1 preparation of ointment of Chinese medicinal composition
The traditional Chinese medicine composition is prepared from the following raw material medicines:
107.5g of vinegar rhizoma corydalis, 86g of ligusticum wallichii, 86g of radix clematidis, 107.5g of divaricate saposhnikovia root, 107.5g of pepper, 86g of achyranthes root, 107.5g of lycopodium clavatum, 107.5g of northeast garden balsam stem, 86g of beautiful sweetgum fruit, 107.5g of erythrina bark, 3.6g of menthol, 7.2g of borneol, 16g of triethanolamine and 100g of glycerol.
Distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil for 8 hr, and collecting volatile oil; extracting the residue with 10 times of water for 1 hr, filtering the extractive solution, and collecting the filtrate; adding 70% ethanol 10 times the amount of rhizoma corydalis, herba Lycopodii, herba Speranskiae Tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting for 2 times (1 hr/time), mixing extractive solutions, filtering, recovering ethanol, mixing with the above water extraction partial filtrate, and concentrating to obtain extract with relative density of 1.20-1.25(50 deg.C);
mixing the volatile oil and the extract uniformly, adding the menthol and borneol which are ground to be co-melted, a proper amount of glycerol, triethanolamine and liquid paraffin to 1000g, mixing uniformly and subpackaging to obtain the traditional Chinese medicine.
EXAMPLE 2 preparation of gel of Chinese medicinal composition
The traditional Chinese medicine composition is prepared from the following raw material medicines: 107.5g of vinegar rhizoma corydalis, 86g of ligusticum wallichii, 86g of radix clematidis, 107.5g of divaricate saposhnikovia root, 107.5g of pepper, 86g of achyranthes root, 107.5g of lycopodium clavatum, 107.5g of northeast garden balsam stem, 86g of beautiful sweetgum fruit, 107.5g of erythrina bark, 3.6g of menthol, 7.2g of borneol, 20g of carbomer, 16g of triethanolamine and 100g of glycerol.
Distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil for 6 hr, and collecting volatile oil; extracting the residue with 12 times of water for 2 hr, filtering the extractive solution, and collecting the filtrate; adding 60% ethanol 12 times the amount of rhizoma corydalis, herba Lycopodii, northeast herba Speranskiae Tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting for 2 times (1.5 hr/time), mixing extractive solutions, filtering, recovering ethanol, mixing with the above water extraction partial filtrate, and concentrating to obtain extract with relative density of 1.20-1.25(50 deg.C);
dissolving carbomer in appropriate amount of distilled water, swelling completely, sequentially adding triethanolamine and glycerol, stirring, sequentially adding ground co-melted Mentholum and Borneolum Syntheticum, volatile oil, extract and appropriate amount of distilled water to 1000g, grinding, and packaging.
EXAMPLE 3 preparation of gel of Chinese medicinal composition
The traditional Chinese medicine composition is prepared from the following raw material medicines: 107.5g of vinegar rhizoma corydalis, 86g of ligusticum wallichii, 86g of radix clematidis, 107.5g of divaricate saposhnikovia root, 107.5g of pepper, 86g of achyranthes root, 107.5g of lycopodium clavatum, 107.5g of northeast garden balsam stem, 86g of beautiful sweetgum fruit, 107.5g of erythrina bark, 3.6g of menthol, 7.2g of borneol, 20g of carbomer, 16g of triethanolamine and 100g of glycerol.

dissolving carbomer in appropriate amount of distilled water, swelling completely, sequentially adding triethanolamine and glycerol, stirring, sequentially adding ground co-melted Mentholum and Borneolum Syntheticum, volatile oil, extract and appropriate amount of distilled water to 1000g, grinding, and packaging.
Experimental example 4 Effect of the Chinese medicinal composition of the present invention on the skin of the back of the ear of Guinea pig
1. Experimental Material
1.1 drugs and reagents
A Chinese medicinal composition prepared by the method of example 3, wherein the specification is 1g crude drug/g gel;
vaseline ointment, Shen Xin chemical Co., Ltd., specification 500 ml/bottle;
propranolol, institute for food and drug testing, specification 100 mg/bottle;
methotrexate for injection, qilu pharmaceuticals ltd, specification 0.1 g/bottle;
0.9% NaCl, specification 250m L: 2.25g, Shandong Kelun pharmaceutical Co., Ltd;
rat I L-2 (Interleukin 2) E L ISA Kit, Specification 96T, Elabscience;
rat I L-4 (Interleukin 4) E L ISA Kit, Specification 96T, Elabscience.
1.2 animals
Guinea pig, weight 220 ~ 250g, half female and half, supplied by Lefu farm in Pukou of Nanjing, quality certification number: SCXK (su) 2019-.

2. Experimental methods
2.1 selecting 60 guinea pigs, weighing 250-280g of guinea pigs in the experiment, randomly dividing the guinea pigs into 6 groups, namely a blank control group, a model group, low-traditional Chinese medicine composition, medium-high dose groups (3, 1.5 and 0.75g of crude drugs/kg) and positive drug methotrexate (1mg/kg), 10 blank control groups in each group, externally coating vaseline on the outer sides of two ears every day, uniformly coating 5% (mass percent of propranolol and vaseline) of propranolol ointment on the auricles on one side of the guinea pigs in the model group, coating vaseline ointment on the auricles on the other side of the auricles for 2 times every day, 21d, starting administration on the 22 th day of the experiment, administering matrix to the blank control group and the model group, coating the auricles of the drug groups every day for 2 times every day, continuously coating 7 days of the positive drug, injecting methotrexate every two days, measuring the thickness of the same auricles of the guinea pigs on the 1, 7, 14, 21 and 28 days, anesthetizing the guinea pigs with 10% of chloral, collecting main artery, fixing the pathological blood serum I, and observing the pathological change by using a standard formaldehyde staining method of measuring the skin, and measuring the HE, and measuring the pathological change the HE.

2.2 results of the experiment
2.2.1 general observations: after 5% propranolol cream is applied to guinea pigs for 1 week, auricle thickness is increased, hair of ears is completely shed, local skin is red and swollen, and skin temperature is high. After 2-3 weeks, the red swelling becomes worse, scales appear, and a little scales fall off to bleed in a punctate way. After treatment with the composition of the present invention, the thickness of the pinna of the guinea pig substantially returned to normal.
Measurement of auricle thickness: after 5% propranolol cream is applied for days 1, 7, 14, 21 and 28, the thickness of the auricle of the model group is obviously increased, and the auricle thickness is obviously different from that of the blank control group. After the composition is administered, the thickness of the auricle of the guinea pig is obviously reduced, and the difference between a medium-dose group and a high-dose group of the traditional Chinese medicine composition and a model group is more obvious.

2.2.2 histological Scoring of pathological sections
The blank control group auricle skin tissue morphology is normal; the model group can show wide hyperkeratosis, thick spinous layer, inflammatory cell infiltration in mastoid, thinning or disappearance of granular layer, extension and undulation of the skin process, telangiectasia and the like. After the composition is administered, parakeratosis is obviously reduced, inflammatory cells are reduced, and epidermal cells are thinned. The pathological scoring result shows that the pathological damage can be obviously improved by the low, medium and high dose groups of the traditional Chinese medicine composition.

2.2.3 results show that the level of I L-2 in the model group is increased, the level of I L-4 is reduced, after the composition is administered, the content of serum I L-2 in the traditional Chinese medicine composition and the high dose group is reduced, the content of I L-4 in the traditional Chinese medicine composition and the low, medium and high dose groups is increased, and the significant difference is realized compared with the model group.

The experiment conclusion is that the skin damage of guinea pigs caused by propranolol is adopted, extensive hyperkeratosis of an epidermal layer, thickening of a spinous layer, elongation of a skin process and wavy fluctuation can be seen under a model after model building, and vasodilatation congestion, edema and mild inflammatory cell infiltration in the dermal layer are achieved, compared with a blank control group, the pathological score has obvious difference, which indicates that the model building is successful.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.

7. The use according to any one of claims 1 to 3, wherein, when the psoriasis treatment agent is selected from a gel, the gel is prepared by the following method:
distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil for 8 hr, and collecting volatile oil; extracting the residue with 15 times of water for 1.5 hr, filtering the extractive solution, and collecting the filtrate; adding 70% ethanol 12 times the amount of rhizoma corydalis, herba Lycopodii, northeast herba Speranskiae Tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting for 2 times, mixing extractive solutions, filtering, recovering ethanol, mixing with the above water extraction partial filtrate, and concentrating to obtain extract with relative density of 1.20-1.30;
dissolving carbomer in appropriate amount of distilled water, swelling completely, sequentially adding triethanolamine and glycerol, stirring, sequentially adding Mentholum and Borneolum Syntheticum, volatile oil, extract and appropriate amount of distilled water, and grinding.
8. The use according to any one of claims 1 to 3, wherein, when the psoriasis treatment agent is selected from a gel, the gel is prepared by the following method:
distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil for 6 hr, and collecting volatile oil; extracting the residue with 12 times of water for 2 hr, filtering the extractive solution, and collecting the filtrate; adding 60% ethanol 12 times the amount of rhizoma corydalis, herba Lycopodii, northeast herba Speranskiae Tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting for 2 times, mixing extractive solutions, filtering, recovering ethanol, mixing with the above water extraction partial filtrate, and concentrating to obtain extract with relative density of 1.20-1.25;
dissolving carbomer in appropriate amount of distilled water, swelling completely, sequentially adding triethanolamine and glycerol, stirring, sequentially adding Mentholum and Borneolum Syntheticum, volatile oil, extract and appropriate amount of distilled water, and grinding.
9. The use according to any one of claims 1 to 3, wherein, when the psoriasis treatment agent is selected from an ointment, the ointment is prepared by the following method:
distilling fructus Zanthoxyli, rhizoma Ligustici Chuanxiong, and radix Saposhnikoviae with steam to obtain volatile oil for 8 hr, and collecting volatile oil; extracting the residue with 10 times of water for 1 hr, filtering the extractive solution, and collecting the filtrate; adding 70% ethanol 10 times the amount of rhizoma corydalis, herba Lycopodii, northeast herba Speranskiae Tuberculatae, fructus Lipuidambaris, cortex Erythrinae, Achyranthis radix, and radix Clematidis into vinegar, reflux-extracting for 2 times, mixing extractive solutions, filtering, recovering ethanol, mixing with the above water extraction partial filtrate, and concentrating to obtain extract with relative density of 1.20-1.25;
mixing the volatile oil and the extract, adding the co-melted menthol and borneol, proper amount of glycerol, triethanolamine and liquid paraffin into the mixed extract, and mixing.
