技术领域
本发明属于医药技术领域,具体涉及一种天然抗菌抗病毒制剂、制备方法及其用途。
背景技术
梨火疫病病菌是一种具有毁灭性的植物病原细菌,寄主范围很广,能危害梨、苹果、山楂、木旬子、李等40多个属220多种植物,造成严重的经济损失。感染后的主要症状为:花器呈萎蔫状,深褐色,并向下蔓延至花柄,使花柄呈水渍状。叶片发病,先从叶缘开始变黑色,然后沿叶脉发展,最终全叶变黑、凋萎。病果初生水渍状斑,后变暗褐色,并有黄色粘液溢出,最后病果变黑而干缩。枝干被害,初呈水渍状,有明显边缘,后病部凹陷出现溃疡状呈褐色至黑色。梨火疫病病菌被视为最难控制的细菌性病害之一,严重感染可能会杀死一个季节内的全部树木。
目前,梨火疫病的防治主要是采用化学药剂链霉素,化学防治尽管能快速消除病原菌,但易造成植株耐药性、污染环境、破坏生态平衡和威胁人畜的健康和安全。因此,寻求绿色、环保的火疫病防治方法是现代农业发展中的一项重要任务。生物防治作为一种绿色安全的防治方法,受到越来越多的关注。
经检索,现有技术已有相关的申请案公开,如中国专利申请号为2019102668000,公开日期为2019年6月4日的申请案公开了一种利迪链霉菌在防治植物火疫病中的应用,该菌株对火疫病病原菌解淀粉欧文氏菌具有显著的拮抗效果,对蔷薇科植物尤其是梨、苹果和海棠等经济作物火疫病具有良好的防治效果,该申请案以生物菌作为药剂,较化学农药对植物病害的防治相比,效果显著,对环境无污染,有利于绿色、无公害产品的生产,可用于制备防治火疫病的功能性生防菌剂。
再如中国专利申请号为2014107962089,公开日期为2015年4月1日的申请案公开了一株侧孢短芽孢杆菌及其应用,并公开了该菌株具有如下特征:对多种植物病原真菌(番茄枯萎病菌、辣椒根腐茄柄镰刀菌、番茄灰霉病菌、水稻纹枯病菌、甘蓝黑斑病菌、梨炭疽病菌和梨轮纹病菌)及植物病原细菌(番茄青枯病菌、梨火疫病菌和水稻细条斑病菌)均具有较强的室内平板抑制作用;是微生物农药研发的有用资源,具有潜在的应用前景。
上述申请案的方法采用生物菌作为防治药物,相比化学类药物虽然能够避免环境污染,然而其需要的投入量一般较大,而且由于大量菌种的投加同样会有引发菌体泛滥的情况,具有破坏生态平衡的风险。
基于现有技术的缺陷,亟需发明一种既是绿色、无公害的,又可以避免菌体泛滥的防治火疫病的制剂。
发明内容
1.要解决的问题
针对现有技术中防治梨火疫病的方法通常采用化学药剂,易造成植株耐药性、污染环境、破坏生态平衡的缺陷,采用生物菌防治方法容易导致菌体泛滥,同样具有破坏生态平衡的风险,本发明提供了含有柚子类种籽提取液、柠檬酸、柠檬酸钠、甘油和复合生物酶的制剂,利用各组分协同,首先快速溶解其荚膜,再进一步促进细胞溶解,有效抑制梨火疫病病菌,对环境无污染,有利于绿色、无公害产品的生产。
进一步的,本发明的发明目的在于提供一种广谱性的抗菌制剂,不仅能够快速针对特殊细菌的防治,同时对于普通细菌抑制也更加快速有效。
2.技术方案
为了解决上述问题,本发明所采用的技术方案如下:
本发明提供了一种天然抗菌抗病毒制剂,所述制剂包括柚子类种籽提取液、柠檬酸、柠檬酸钠、甘油和复合生物酶。
作为本发明更进一步的改进,所述柠檬酸的质量占制剂总质量的0.1%~5%,柠檬酸钠的质量占制剂总质量的0.05%~1%,所述复合生物酶的质量占制剂总质量的1~30%,所述柚子类种籽提取液的质量占制剂总质量的0.1%~1%,所述甘油的质量占制剂总质量的0.2%~2%。
作为本发明更进一步的改进,所述的天然抗菌抗病毒制剂的制备方法,所述方法包括以下步骤:
1)筛选柚子类种籽,粉碎至一定粒度、采用乙醇为提取剂通过搅拌的方式提取,得到粗提液,将所述粗提液过滤得到滤液;再通过回收乙醇,得到提取液;
2)向步骤1)中的提取液中加入甘油,再添加水稀释一定倍数,得到稀释液,向所述稀释液中加入柠檬酸、柠檬酸钠和复合生物酶,得到混合溶液。
优选的,如果步骤2)中得到的混合溶液pH值较高,可以使用碱性水调节混合溶液pH到5.5~8的范围内。
上述混合溶液经过高温灭菌处理,制备成为抗菌产品。可直接以液体形式使用,可通过干燥处理制备成干燥制品,使用时进行加水稀释。
作为本发明更进一步的改进,所述复合生物酶是将猪粪尿原液进行发酵得到发酵液,再将发酵液过滤得到,所述的复合生物酶中含有溶菌酶。
作为本发明更进一步的改进,所述复合生物酶提取方法包括以下步骤:
步骤a)将猪粪尿原液投入多个反应器中,依次按照好氧-好氧-好氧-兼氧-厌氧-好氧-兼氧-厌氧-好氧-兼氧-厌氧-好氧工艺进行处理,经复合发酵后得到其代谢产物的液体制剂;前三个反应器内分别设有微纳米曝气装置;
步骤b)将得到的液体制剂进行过滤,取上清液作为复合生物酶使用,所述复合生物酶中微生物含量低于1%。
作为本发明更进一步的改进,所述种籽在粉碎前需将其在-25℃~-80℃下进行快速冷冻处理5小时以上。

3.有益效果
相比于现有技术,本发明的有益效果为:
(1)本发明的天然抗菌抗病毒制剂,制备过程中,首先从柚子类种籽中提取得到主要成分为脂肪酸类黄酮的提取液,然后在提取液中加入以利于主成分(脂肪酸类黄酮)稳定的甘油,最后加入柠檬酸、柠檬酸钠和复合生物酶(含有溶菌酶)进行复配制备而成,本发明制剂中各成分进行组合,能够协同作用于梨火疫病致病菌的外层荚膜,首先快速溶解致病菌的荚膜,再进一步促进细胞溶解,有效抑制梨火疫病病菌。本发明的制剂对环境无污染,有利于绿色、无公害产品的生产,可用于制备防治火疫病的功能性制剂。
(2)本发明的天然抗菌抗病毒制剂,具有广谱性的抗菌功能,其主要作用原理如下:1)该制剂含有能够抑制一般细菌的复合生物酶(其含有溶菌酶和多种复合酶),通过溶菌酶可以破坏一般细菌的细胞壁中的N-乙酰胞壁酸和N-乙酰氨基葡萄糖之间的β-1,4糖苷键,使细胞壁破裂、细菌溶解,因此复合生物酶针对普通细菌本身即可起到有效抑制作用,而在脂肪酸类黄酮与柠檬酸、柠檬酸钠的协同作用下,能够加速细菌细胞壁的溶解,使其快速被分解,缩短作用起效时间;2)对于自然界还存在的特殊细菌(如梨火疫病致病菌),其细胞壁被特殊结构-荚膜保护,而荚膜成分因不同菌种而异(主要是由葡萄糖与葡萄糖醛酸组成的聚合物,也有含多肽与脂质)。这成为该类病原菌难以通过溶解细胞壁来抑制的原因之一,本发明的制剂通过从柚子类种籽中提取得到的脂肪酸类黄酮与柠檬酸、柠檬酸钠组合,促进该类病原菌的荚膜被快速破坏,再通过脂肪酸类黄酮进一步灭活外层荚膜被去掉的病原菌,同时制剂中含有的复合生物酶也在该阶段发挥了强化杀菌的功能。因此,本发明的制剂不仅是针对普通的细菌还是外层具有荚膜的特殊细菌均可以发挥有效的杀菌作用,发挥作用快速有效。
(3)本发明的天然抗菌抗病毒制剂,在提取液中加入甘油成分以保证提取得到的主成分脂肪酸类黄酮能够稳定存在于该提取液体系,且使提取液不易挥发,以在最终的提取液中获得更多的有效成分,持续生效时间更长,更有利于杀菌杀毒作用的发挥。另外,本发明的方法在种籽粉碎前需要进行将其在-25℃~-80℃下进行冷冻处理,可以使大量的脂肪酸类黄酮物质聚集于种籽的核心部分,并且在冷冻状态下进行粉碎,易于更多的脂肪酸类黄酮物质乙醇方式提取,保留更多的有效成分。

具体实施方式
下面结合具体实施例对本发明进一步进行描述。
实施例1
本发明的天然抗菌抗病毒制剂,制备方法包括如下步骤:
1)筛选葡萄柚种籽、清洗,晒干后先在-25℃~-80℃下对种籽进行冷冻5小时,再进行粉碎,粉碎粒度小于5mm,将粉碎好的种籽装入提取罐中,加入70%乙醇作为提取剂,25℃搅拌条件下提取,保持料液比在1:(10~60)之间,提取120分钟后得到粗提液,将所述粗提液过滤得到滤液;再通过回收乙醇,得到提取液;
2)向所述提取液中加入甘油,再添加水进行稀释,得到稀释液,在所述稀释液中加入柠檬酸、柠檬酸钠和复合生物酶,得到混合溶液。
所述复合生物酶为酶液形式,具体制备过程如下:采用多个工程塑料或玻璃钢制的大反应桶,多个反应桶依次按照好氧-好氧-好氧-兼氧-厌氧-好氧-兼氧-厌氧-好氧-兼氧-厌氧-好氧(O/O/O-A/A/O-A/A/O-A/A/O)间歇设置并连接,前三个反应桶中均设有微纳米曝气装置,控制反应桶中溶解氧在2mg/L以上;每天从第一个反应桶中导入猪粪尿原液,以自然溢流方式依次进入下一反应桶中,三个月后从最后的反应桶中最终溢流出生物酶液,再经沉淀过滤桶过滤纯化后制得复合生物酶,经检验,上述制备的复合生物酶中含有溶菌酶和多种复合酶,微生物含量低于1%。
依据提取液含量不同,以制备不同浓度的制剂样品进行抑菌实验,本实施例中分别记为制剂样品1~4,所述制剂样品1中,柚子类种籽提取液的质量占制剂总质量的0.8~1%,所述甘油的质量占制剂总质量的1.6%~2%。所述柠檬酸的质量占制剂总质量的0.1%~5%,柠檬酸钠的质量占制剂总质量的0.05%~1%,所述复合生物酶的质量占制剂总质量的1~30%。
所述制剂样品2中,柚子类种籽提取液的质量占制剂总质量的0.5~0.8%,所述甘油的质量占制剂总质量的1%~1.6%,所述柠檬酸的质量占制剂总质量的0.1%~5%,柠檬酸钠的质量占制剂总质量的0.05%~1%,所述复合生物酶的质量占制剂总质量的1~30%。
所述制剂样品3中,柚子类种籽提取液的质量占制剂总质量的0.1~0.5%,所述甘油的质量占制剂总质量的0.2%~1%,所述柠檬酸的质量占制剂总质量的0.1%~5%,柠檬酸钠的质量占制剂总质量的0.05%~1%,所述复合生物酶的质量占制剂总质量的1~30%。
所述制剂样品4中,柚子类种籽提取液的质量占制剂总质量的0.02~0.05%,所述甘油的质量占制剂总质量的0.04%~0.1%,所述柠檬酸的质量占制剂总质量的0.1%~5%,柠檬酸钠的质量占制剂总质量的0.05%~1%,所述复合生物酶的质量占制剂总质量的1~30%。
抑菌实验
通过“滤纸片法”测定复合生物酶以及不同浓度的制剂样品对梨火疫病病菌的生长抑制。采用复合生物酶作为对比实验,目的在于:所述的复合生物酶中含有溶菌酶,可通过溶解大部分的细菌细胞壁使细菌被破坏,针对不同种类的细菌,通常具有抑菌效果。
供试菌株
梨火疫病病菌Erwiniaamylovora,从新疆库尔勒市的香梨产区正在受梨火疫病病菌侵害的山楂样本上分离得到菌株XJSZ0102,经准确鉴定和致病力测定后超低温保存,将其作为实验对象。
供试药剂:制剂样品1,制剂样品2,制剂样品3、制剂样品4,复合生物酶。
试验方法
分别取本发明的制剂1~4与复合生物酶制剂进行抑菌试验。吸取1ml(108CFU/ml)菌悬液在直径为90mm的NA平板培养基上均匀涂开,吸出多余菌液。晾干后,将直径为6mm的灭菌滤纸片分别充分浸泡上述药液,滴落多余药液后置于上述涂菌平板培养基上,每皿均匀放置3片,重复3次,以无菌水作为对照。置于28℃生化培养箱中培养24h后,采用十字交叉法测量抑菌圈直径。
结果与分析
试验结果表明,复合生物酶对梨火疫病病菌的药效与阴性对照无差异,对病原菌的生长无抑制作用。虽然复合生物酶中含有的溶菌酶可以通过破坏一般细菌的细胞壁中的N-乙酰胞壁酸和N-乙酰氨基葡萄糖之间的β-1,4糖苷键,使细胞壁破裂、细菌溶解,针对普通细菌可起到有效抑制作用,然而针对含有荚膜的特殊细菌(梨火疫病病菌)则难以发挥作用,具体见图2。本发明的制剂1~3均对梨火疫病病菌的生长有较好的抑制作用,具体见图1。
表1为不同制剂样品对梨火疫病病菌的抑菌圈直径统计,根据表1可知,制剂样品1的平均抑菌圈直径约为17mm,制剂样品2的平均抑菌圈直径约为9.2mm,制剂样品3的平均抑菌圈直径约为7.8mm。制剂样品4未体现出抑菌作用。
由上述结果可知,柚子类种籽提取液对梨火疫病病菌的荚膜破坏起到重要作用,其浓度越高,抑菌效果越好,加入含量至0.05%以下时无法发挥抑菌作用,本发明通过从柚子类种籽中提取得到的脂肪酸类黄酮与柠檬酸、柠檬酸钠组合,促进该类病原菌的荚膜被快速破坏,再通过脂肪酸类黄酮进一步灭活外层荚膜被去掉的病原菌,同时制剂中含有的复合生物酶也在该阶段发挥了强化杀菌的功能。本发明的制剂不仅是针对普通的细菌还是外层具有荚膜的特殊细菌均可以发挥有效的杀菌作用,发挥作用快速有效。

实施例2
按照实施例1的制备方法制备本发明的制剂,不同之处在于:所述柚子类种籽提取液的质量占制剂总质量的0.2%,所述甘油的质量占制剂总质量的0.4%。所述柠檬酸的质量占制剂总质量的0.5%,柠檬酸钠的质量占制剂总质量的0.1%,所述复合生物酶的质量占制剂总质量的5%。针对普通的细菌,复合生物酶在脂肪酸类黄酮与柠檬酸、柠檬酸钠的配合下,更能够加速细菌细胞壁的溶解,使其快速被分解。
将本实施例的制剂制备成喷雾,使用普通喷雾器对餐厅厨房(面积约50平方米)进行灭菌处理:喷雾数量每平方米30mL,总用量为1500mL。
检测部位为:食材储藏间、砧板周边区域、菜品制作台;测量仪器:LumitesterPD30;开始时检测值6350RLU,喷雾60秒停止5分钟后降到865RLU,停止10分钟后降至230RLU(1000RLU以下符合日本食品加工及餐饮业的卫生标准中一般杂菌标准)。之后持续观测一周,均可保持在500RLU以下。经过三个月以后,每天打烊前喷雾每平米15mL,持续监测三个月,一般杂菌值一直保持在300RLU以下。
实施例3
本实施例为采用本发明的产品进行家居室内消毒的实施例:
按照实施例1的制备方法制备本发明的制剂,不同之处在于:所述柚子类种籽提取液的质量占制剂总质量的0.1%,所述甘油的质量占制剂总质量的0.2%,所述甘油的质量占制剂总质量的0.4%。所述柠檬酸的质量占制剂总质量的0.5%,柠檬酸钠的质量占制剂总质量的0.1%,所述复合生物酶的质量占制剂总质量的5%。
制备成为普通喷雾,用于家居室内消毒:使用普通喷雾器对家居室内(面积约120平方米)进行灭菌处理:喷雾数量每平方米10mL升,总用量为1200mL。检测部位为:厨房,卫生间,起居室,卧室,储藏间等;测量仪器:Lumitester PD30;检测值开始时检测值最高3560RLU,喷雾60秒停止5分钟后降到751RLU,停止10分钟后降至246RLU(1000RLU以下符合日本食品加工及餐饮业的卫生标准)。之后持续观测一周,均可保持在300RLU以下。三个月之后,每3天喷雾一次每平米10毫升,持续监测一个月,一般杂菌值一直保持在210RLU以下。

1.一种天然抗梨火疫病病菌制剂,其特征在于:所述制剂包括葡萄柚种籽乙醇提取液、柠檬酸、柠檬酸钠、甘油和复合生物酶,所述柠檬酸的质量占制剂总质量的0.1%~5%,柠檬酸钠的质量占制剂总质量的0.05%~1%,所述复合生物酶的质量占制剂总质量的1~30%,所述葡萄柚类种籽提取液的质量占制剂总质量的0.1%~1%,所述甘油的质量占制剂总质量的0.2%~2%,复合生物酶具体制备过程如下:采用多个工程塑料或玻璃钢制的大反应桶,多个反应桶依次按照好氧-好氧-好氧-兼氧-厌氧-好氧-兼氧-厌氧-好氧-兼氧-厌氧-好氧间歇设置并连接,前三个反应桶中均设有微纳米曝气装置,控制反应桶中溶解氧在2mg/L以上,每天从第一个反应桶中导入猪粪尿原液,以自然溢流方式依次进入下一反应桶中,三个月后从最后的反应桶中最终溢流出生物酶液,再经沉淀过滤桶过滤纯化后制得复合生物酶。
2.权利要求1所述的天然抗梨火疫病病菌制剂的制备方法,其特征在于:所述方法包括以下步骤:
1)筛选葡萄柚类种籽,粉碎至一定粒度、采用乙醇为提取剂通过搅拌的方式提取,得到粗提液,将所述粗提液过滤得到滤液,再通过回收乙醇,得到葡萄柚类种籽的提取液;
2)向所述提取液中加入甘油,再添加水稀释一定倍数,得到稀释液,向所述稀释液中加入柠檬酸、柠檬酸钠和复合生物酶,得到混合溶液。
3.根据权利要求2所述的天然抗梨火疫病病菌制剂的制备方法,其特征在于:所述种籽在粉碎前需将其在-25℃~ -80℃下快速冷冻处理5小时以上。
4.根据权利要求2所述的天然抗梨火疫病病菌制剂的制备方法,其特征在于:所述步骤1)中以乙醇为提取剂时保持料液比为1:(10~60)。
5.根据权利要求2所述的天然抗梨火疫病病菌制剂的制备方法,其特征在于:所述步骤1)中的粉碎粒度小于5mm。
6.权利要求1所述的天然抗梨火疫病病菌制剂在抗梨火疫病病菌中的用途。
