Abstract

The invention discloses an initiator with disease resistance and insect resistance for small-sized seeds of non-heading Chinese cabbages and application thereof. The initiator has the advantages that the amylase activity of the small-sized seeds of the non-heading cabbages is improved through the matrine, the germination capacity of the small-sized seeds of the non-heading cabbages is improved, and the germination of the seeds of the non-heading cabbages is promoted. The small-sized seed of the non-heading Chinese cabbage has the disease-resistant and insect-resistant initiator matrine with insecticidal and bactericidal effects, and is more favorable for the growth of seedlings. The small-sized seed of the non-heading Chinese cabbage has the advantages of disease resistance and insect resistance, and the initiator is easy to operate when the seed is initiated. The small-sized seed of the non-heading Chinese cabbage has the advantages of natural property, environmental protection and easy degradation as well as disease and insect resistance.

Technical Field
The invention relates to the technical field of agriculture, in particular to an initiator for resisting diseases and insects for small-sized seeds of non-heading Chinese cabbages and application thereof.
Background
Seed priming (English name: Seed priming), also called "osmotic regulation", is the slow absorption of water by seeds under controlled conditions, so that the seeds undergo physiological and biochemical metabolism and repair actions of pregermination, promote the repair of cell membranes and organelles and the activation of enzymes, and are in a state ready for germination. The initiation technology is mainly applied to crops, flower plants, vegetable crops, medicinal plants and the like. The main priming methods are liquid (osmotic), water, solid matrix and biological priming.
Non-heading Chinese cabbage (Brassica rapa ssp. chinensis) belongs to Brassicaceae Brassica seed, commonly called green vegetable, pakchoi, rape, etc., and is an important vegetable crop native to China. The Chinese cabbage is fresh and tender, rich in nutrition, popular with people and plays an important role in the vegetable consumption in China. The non-heading Chinese cabbages are mostly cultivated in a direct seeding mode, but the seed particles are small, the practical production problems of large seeding quantity, low emergence rate, few strong seedlings and the like exist, and therefore, the improvement of the seed vitality and the seedling elements have direct influence on the yield and the quality of the non-heading Chinese cabbages. At present, most of initiators for non-heading Chinese cabbage seeds are potassium nitrate, sodium chloride, calcium chloride, polyethylene glycol 6000 and the like, however, the existing initiators for non-heading Chinese cabbage seeds are still not ideal in the aspects of improving the germination capacity of seeds and protecting the environment. Therefore, there is a need to provide an initiator with disease and insect resistance for small-sized seeds of non-heading Chinese cabbage and the application thereof to solve the above problems.
Disclosure of Invention
The invention provides an initiator with disease resistance and insect resistance for small-sized seeds of non-heading Chinese cabbages and application thereof, and aims to solve the problem that the existing initiator for the seeds of the non-heading Chinese cabbages still has poor effects on improving the germination capacity of the seeds and on the aspect of environmental protection.
In a first aspect, the invention provides an initiator for the disease and insect resistance of non-heading Chinese cabbage small-sized seeds, wherein the initiator is matrine.
Optionally, the concentration of matrine is 60-240 mg/L.
Optionally, the concentration of the matrine is 80 mg/L.
In a second aspect, the invention provides an application of any one of the above-mentioned initiators with disease and insect resistance in initiating small-sized seeds of non-heading Chinese cabbage.
In a third aspect, the invention provides a method for initiating small-particle seeds of non-heading Chinese cabbages, which comprises the following steps:
providing an initiator with a preset concentration, wherein the initiator is matrine;
and step two, placing the initiator and a specific number of the small-particle seeds of the non-heading Chinese cabbages in a beaker, and initiating for a preset time under the dark condition at room temperature.
Optionally, in the step one, the concentration of the initiator is 60-240 mg/L.
Optionally, in the step one, the concentration of the initiator is 80 mg/L.
Optionally, in the second step, the number of the non-heading Chinese cabbage small-sized seeds is 500, and the initiation treatment is carried out for 12 hours at room temperature in the dark.
The invention has the following beneficial effects: the initiator has the advantages that the amylase activity of the small-sized seeds of the non-heading cabbages is improved through the matrine, the germination capacity of the small-sized seeds of the non-heading cabbages is improved, and the germination of the seeds of the non-heading cabbages is promoted. The small-sized seed of the non-heading Chinese cabbage has the disease-resistant and insect-resistant initiator matrine with insecticidal and bactericidal effects, and is more favorable for the growth of seedlings. In order to make the objects, technical solutions and advantages of the present invention more apparent, the technical solutions of the present invention will be clearly and completely described below with reference to the specific embodiments of the present invention and the accompanying drawings. It is to be understood that the described embodiments are merely exemplary of the invention, and not restrictive of the full scope of the invention. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention. The technical solutions provided by the embodiments of the present invention are described in detail below with reference to the accompanying drawings.
The invention provides an initiator for non-heading Chinese cabbage small-sized seeds with disease resistance and insect resistance, wherein the initiator is matrine (English name: L-Ascorbic acid). The matrine is extracted from roots and stems of sophora plants such as sophora alopecuroides, sophora flavescens, sophora subprostratae and the like by using an organic solvent, and has wide medical and agricultural values. In the medical field, a plurality of modern pharmacological experiments prove that the matrine has the common functions of resisting virus, inflammation and arrhythmia, and the like, and also has the functions of regulating the central nervous system, protecting the liver, regulating the immunity, the cardiovascular system and the respiratory system and inhibiting the tumorigenesis.
In the aspect of agricultural value, the matrine has the functions of killing insects and bacteria and regulating plant growth. In the aspect of insect killing, research shows that the matrine can prevent and control vegetable pests such as cabbage caterpillar, cabbage aphid, melon aphid, stem borer, beet armyworm, diamond back moth, cabbage looper, yellow striped flea beetle, Chinese chive maggot and the like; fruit tree pests such as tenebrio molitor, moth, inchworm, red spider, and lecania; and grain pests such as armyworm, wheat midge, locust and the like. In the aspect of bacteriostasis, the matrine has certain bacteriostasis activity on pepper anthracnose pathogen, cucumber phytophthora, apple ring spot pathogen, wheat gibberellic disease, sclerotinia sclerotiorum and the like. In the aspect of plant growth regulation, research shows that exogenous matrine can promote the increase of fresh weight and dry weight of in-vitro cucumber cotyledons, the total chlorophyll content is increased, and the photosynthetic oxygen release amount is obviously increased; the matrine solution with proper concentration can effectively improve the germination rate, the germination vigor, the germination index, the vitality index and the amylase activity of wheat and mung bean seeds; the matrine with a certain concentration is externally applied, so that the growth of tomato plants can be promoted and the yield can be improved. However, the treatment effect of matrine on the non-heading Chinese cabbage seeds is not reported yet.
In this embodiment, the concentration of matrine is 60-240 mg/L. Preferably, the concentration of the matrine is 80 mg/L. The initiator with disease resistance and insect resistance can improve the germination vigor, the germination rate, the germination index and the vitality index of the non-heading Chinese cabbage seeds and obviously enhance the vitality of the seeds. The invention also provides application of any one of the above initiators with disease and insect resistance in initiating the non-heading Chinese cabbage small-sized seeds.
As shown in FIG. 1, the invention also provides a method for initiating small-particle seeds of non-heading Chinese cabbage, which comprises the following steps:
step one, providing an initiator with a preset concentration, wherein the initiator is matrine.
In this embodiment, the concentration of matrine is 60-240 mg/L. Preferably, the concentration of the matrine is 80 mg/L.
And step two, placing the initiator and a specific number of the small-particle seeds of the non-heading Chinese cabbages in a beaker, and initiating for a preset time under the dark condition at room temperature.
In this example, the number of the small-sized seeds of the non-heading Chinese cabbage is 500, and the priming treatment is carried out for 12 hours under the dark condition at room temperature.
The method for initiating the small-sized seeds of non-heading Chinese cabbage according to the present invention will be described in detail with reference to several embodiments.
Example 1
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 60 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 2
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 80 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 3
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 100 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 4
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 120 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 5
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 140 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 6
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 160 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 7
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the matrine is 180 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 8
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 200 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 9
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 220 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Example 10
The initiator for the small-sized seeds of the non-heading Chinese cabbage, which has disease resistance and insect resistance, is matrine, and the concentration of the initiator is 240 mg/L. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
Comparative example
The initiator of the small-grain seeds of the non-heading Chinese cabbage in the comparative example is distilled water. The seeds were selected to be full, uniform and placed in a 500ml beaker. 500 seeds were placed in each beaker and primed at room temperature for 12 h.
In the following germination tests were carried out on seeds obtained by comparative priming using the examples of the invention.
The test materials used in this experiment were the seeds of the non-heading cabbage variety Jiu tou bird and Huang jin Hua.
1. Seed germination test:
after the treatment according to the methods of examples 1 to 10 and comparative example, the seeds were left to air dry at room temperature. Then the seeds are moved into corresponding 90mm culture dishes, 50 grains are respectively placed in each group of treatment, six groups of parallel experiments are arranged in each group, and the seeds are cultured at room temperature. Water was replenished every 12 hours, keeping the filter paper wet.
2. Determination of germination index
4 germination indexes of germination vigor, germination rate, germination index and vitality index of the seeds are measured. The calculation method of each index is as follows:

The germination rate (%) of the seeds was 100% of the number of germinated seeds/test seeds at the final stage (day 7).

Wherein Gt is the number of germinated seeds in different germination days (counting 7d), Dt is the corresponding germination days, and S is the height of the bud seedling in the corresponding germination days.
3. Results and analysis
The seeds of the non-heading Chinese cabbage "jiugou" and "huanghua" treated according to the methods of examples 1 to 10 and comparative example were subjected to germination test according to the method mentioned in 1, and the test results were recorded as shown in tables 1 and 2.

As is clear from tables 1 and 2, the germination vigor, germination percentage, germination index and vigor index of the seeds of the "Jiuguang" and "yellow golden flower" of the other examples were slightly larger than those of the control group, except for examples 1 and 9. The germination vigor, the germination rate, the germination index and the vitality index of the seeds of the yellow golden flower are respectively improved by 1.33 percent, 2.91 percent and 41.47.
In conclusion, the initiator with disease resistance and insect resistance for the small-sized seeds of the non-heading Chinese cabbages and the application thereof provided by the invention have the advantages that the amylase activity of the small-sized seeds of the non-heading Chinese cabbages is improved through the matrine, the germination capacity of the small-sized seeds of the non-heading Chinese cabbages is favorably improved, and the germination of the seeds of the non-heading Chinese cabbages is promoted. The small-sized seed of the non-heading Chinese cabbage has the disease-resistant and insect-resistant initiator matrine with insecticidal and bactericidal effects, and is more favorable for the growth of seedlings. The small-sized seed of the non-heading Chinese cabbage has the advantages of disease resistance and insect resistance, and the initiator is easy to operate when the seed is initiated. The small-sized seed of the non-heading Chinese cabbage has the advantages of natural property, environmental protection and easy degradation as well as disease and insect resistance.
