As to the specific extraction method for the crude extraction of ganoderan in step (1), the present example provides the following three implementation manners, which are respectively:
the first implementation mode comprises the following steps:
(1.1) degreasing: weighing 150g of ganoderma lucidum fruiting body powder, and adding the ganoderma lucidum fruiting body powder into a mixture with a volume ratio of 2: 1, placing 550ml of chloroform-methanol mixed solution in a constant temperature water bath at 40 ℃, and condensing and refluxing for 5.5 h; so refluxing and degreasing for at least 3 times, filtering out the solvent, and drying filter residues in an oven at the temperature of 35 ℃;
(1.2) oligosaccharide removal: adding 550ml of 80% ethanol into the degreased filter residue according to the material-liquid ratio of 1:3, refluxing for 5.5h at 80 ℃, filtering out the solvent, repeating the process for at least 3 times, and drying and storing the filter residue;
(1.3) crude extraction of ganoderma lucidum polysaccharide: adding 950mL of distilled water into the filter residue obtained in the step (1.2), extracting at 95 ℃ for 5.5h, filtering out the solution, leaching for 4 times, and combining the filtrates; then, using a rotary evaporator to concentrate the filtrate to 450mL under the temperature condition of 35 ℃, and centrifuging for 18min at the rotating speed of 4500 r/min; then collecting the precipitate, centrifuging and washing the precipitate with absolute ethyl alcohol and diethyl ether in sequence, finally placing the precipitate in a vacuum drier, and vacuumizing and drying to obtain the crude extract of the ganoderan.
The second embodiment:
(1.1) degreasing: weighing 250g of ganoderma lucidum fruiting body powder, and adding the ganoderma lucidum fruiting body powder into a mixture with a volume ratio of 2: 1, putting 650ml of chloroform-methanol mixed solution in a thermostatic water bath at 50 ℃, and condensing and refluxing for 6.5 h; so refluxing and degreasing for at least 3 times, filtering out the solvent, and drying filter residues in an oven at the temperature of 45 ℃;
(1.2) oligosaccharide removal: adding 650ml of 80% ethanol into the degreased filter residue according to the material-liquid ratio of 1:3, refluxing for 6.5h at 90 ℃, filtering out the solvent, repeating the process for at least 3 times, and drying and storing the filter residue;
(1.3) crude extraction of ganoderma lucidum polysaccharide: adding 1050mL of distilled water into the filter residue obtained in the step (1.2), extracting at 100 ℃ for 6.5h, filtering out the solution, leaching for 5 times, and combining the filtrates; then, concentrating the filtrate to 550mL at the temperature of 45 ℃ by using a rotary evaporator, and centrifuging for 22min at the rotating speed of 5500 r/min; then collecting the precipitate, centrifuging and washing the precipitate with absolute ethyl alcohol and diethyl ether in sequence, finally placing the precipitate in a vacuum drier, and vacuumizing and drying to obtain the crude extract of the ganoderan.
The third embodiment is as follows:
(1.1) degreasing: weighing 200g of ganoderma lucidum fruiting body powder, and adding the mixture into a reaction kettle according to a volume ratio of 2: 1, placing 600ml of chloroform-methanol mixed solution in a constant temperature water bath at 45 ℃, and condensing and refluxing for 6.0 h; so refluxing and degreasing for at least 3 times, filtering out the solvent, and drying filter residues in an oven at the temperature of 40 ℃;
(1.2) oligosaccharide removal: adding 600ml of 80% ethanol into the degreased filter residue according to the material-liquid ratio of 1:3, refluxing at 85 ℃ for 6.0h, filtering out the solvent, repeating the process for at least 3 times, and drying and storing the filter residue;
(1.3) crude extraction of ganoderma lucidum polysaccharide: adding 1000mL of distilled water into the filter residue obtained in the step (1.2), extracting at the temperature of 97 ℃ for 6.0h, filtering out the solution, leaching for 4 times, and combining the filtrates; then, concentrating the filtrate to 500mL by using a rotary evaporator at the temperature of 40 ℃, and centrifuging for 20min at the rotating speed of 5000 r/min; then collecting the precipitate, centrifuging and washing the precipitate with absolute ethyl alcohol and diethyl ether in sequence, finally placing the precipitate in a vacuum drier, and vacuumizing and drying to obtain the crude extract of the ganoderan.
The ganoderma lucidum fruiting body powder adopted in the crude extraction mode of the three ganoderma lucidum polysaccharides is superfine crushed powder obtained by weighing, cleaning, drying and sterilizing the collected aquilaria sinensis ganoderma, crushing the crushed material into powder (more than 2000 meshes) and then extracting the powder by using water. The Aquilaria sinensis lucid ganoderma is lucid ganoderma cultured by taking Aquilaria sinensis wood chips as a novel high-quality basswood carrier, processing the high-quality basswood into a fungus bag after collection, crushing, high-temperature sterilization and disinfection and treatment, inoculating with the strains of the Ganoderma sinense in the fungus bag, and performing full-automatic intelligent three-dimensional plant factory culture.
Before the batching process in the step (2), the method further comprises a raw material processing step, and specifically comprises the following steps:
