Description
Technical Field
The invention relates to a preparation method of a hand-use no-clean disinfectant, in particular to a preparation method of an alcohol-fibroin gel no-clean disinfectant.
Background
The hands usually contact the most bacteria and viruses in daily life, the bacteria and viruses can easily enter the body through food contacted by the hands or the hands touch the face to cause infection, and the bacteria and viruses infected on the hands can be further transmitted to other people through contacting common objects. Therefore, frequent hand washing is necessary to keep the hands clean, which is especially important before catering and when going home.
The hand is generally washed by the hand sanitizer and then is washed by water. But spraying a no-clean disinfectant on the hands is also a method in the case of no water around the body. The no-clean disinfectant with good sterilizing effect and safety has alcohol with the concentration of about 75 percent, but the alcohol also has the defect that the skin is irritated, and the skin on hands is easy to be rough and allergic after the no-clean disinfectant is frequently used. Therefore, some alcohol added with other skin care ingredients is taken as a no-clean disinfectant on the market, and various ingredients are often added into the no-clean disinfectant, and some added ingredients can be used for skin care, but are unsafe to be taken by the careless entrance. For example, in chinese patent publication No. CN101912358A, publication No. 2010, No. 12 and 15, triclosan is used as the main active ingredient, and ethanol, propylene glycol, glycerol, hydroxypropyl methylcellulose (HPMC), disodium ethylenediaminetetraacetate (EDTA-2Na), etc. are used as auxiliary ingredients, so as to provide a hand-washing-free disinfectant having an irritant effect on skin. Also, as disclosed in the chinese patent publication No. CN106265436A, publication No. 01/04 in 2017, a formulation of a leave-on disinfectant with skin care effect and a preparation method thereof are disclosed, wherein the leave-on disinfectant comprises a thickener, a disinfectant, a cosolvent, an encapsulating agent, a neutralizer, a humectant, a skin care agent, an essence and deionized water, and has complex components and poor skin care effect.

The invention aims to overcome the defects in the prior art and provides a preparation method of the alcohol-fibroin gel no-clean disinfectant which is safe to human bodies, convenient to use and capable of moistening skins.
The technical scheme adopted by the invention for solving the problems is as follows: the preparation method of the alcohol-fibroin gel washing-free disinfectant is characterized in that: the preparation method comprises the following steps: uniformly mixing 100ml of medical alcohol solution, 5-10ml of silk fibroin solution and 0-2ml of micromolecular sericin solution to prepare alcohol-fibroin gel washing-free disinfectant; the operation of homogeneous mixing is as follows: adding the silk fibroin solution into a container, then quickly adding the medical alcohol solution, stirring and mixing for 1-3 minutes, then slowly adding the small-molecular sericin solution, stirring for 2-3 minutes, and finishing mixing.
Preferably, the volume concentration of the medical alcohol solution is 30-70%.
Preferably, the concentration of the silk fibroin solution of the present invention is 0.5-8% w/v.

Although the alcohol-fibroin gel washing-free disinfectant prepared finally is not in a gel state any more after the treatment, but becomes a semitransparent thin emulsion state. The alcohol-fibroin gel washing-free disinfectant in the emulsion state has little influence on the skin care effect.
Preferably, the rotation speed of the stirring in the invention is 100-200 rpm.
Preferably, the operation of uniformly mixing according to the present invention is performed at a temperature of 0 to 30 ℃.
Preferably, the alcohol-fibroin gel washing-free disinfectant consists of 100ml of medical alcohol solution, 5-10ml of silk fibroin solution and 0-2ml of small molecular sericin solution.
Preferably, the preparation method of the silk fibroin solution provided by the invention comprises the following steps: applying 0.5% (w/v) Na on cocoon shell2CO2Degumming at bath ratio of 1:200 for 30min each time, washing with clear water, repeating the steps, and drying to obtain silk fibroin; with 50% (w/v) of CaC12The silk fibroin fiber is dissolved in the water solution at the temperature of 98 ℃ and the bath ratio of the silk fibroin fiber g to the CaC12The water solution ml is 1:25, and the time is 10 minutes; and filling the solution obtained after dissolution into a dialysis bag, intercepting the solution with the molecular weight of 12000-14000, dialyzing in running water for 3d, and adding water for dilution to obtain the silk fibroin solution.
Preferably, the preparation method of the silk fibroin solution provided by the invention comprises the following steps: applying 0.5% (w/v) Na on cocoon shell2CO2Degumming at bath ratio of 1:200 for 30min each time, washing with clear water, repeating the steps, and drying to obtain silk fibroin; dissolving the silk fibroin fibers by using a ternary solvent, wherein the bath ratio of the silk fibroin fibers to the ternary solution ml is 1:15, the dissolving temperature is 80 ℃, and the time is 10 minutes; and filling the solution obtained after dissolution into a dialysis bag, intercepting the molecular weight of 12000-14000, dialyzing in running water for 3d, and air-drying and concentrating to obtain the silk fibroin solution.
Preferably, the proportion of the ternary solvent is n (CaCl)2)∶n(H2O)∶n(C2H5OH)=1∶8∶2。
Compared with the prior art, the invention has the following advantages and effects: fibroin protein and sericin extracted from silk are collectively called as fibroin protein, are safe to human bodies, and have application in the aspects of food, medical materials and cosmetics. The silk fibroin and the sericin have various skin care effects of nourishing skin, cleaning, resisting oxidation, moisturizing and the like, and are ideal skin care additive components capable of being used as a washing-free disinfectant.
The alcohol-fibroin gel washing-free disinfectant with alcohol and fibroin in the invention does not contain other components except medical alcohol solution, fibroin solution and micromolecular sericin solution, and is very safe to use. The alcohol-fibroin gel washing-free disinfectant has a flowing gelatinous form, and is convenient to coat on hands; the volatilization of alcohol components can be delayed, and the sterilization effect is enhanced; in addition, it can moisten skin.
The principle of the invention is as follows:
the fibroin solution can generate structural change when meeting alcohol, and weak bonds such as hydrogen bonds and the like are generated between fibroin molecules for combination, because the dehydration effect of the alcohol changes the hydration state of the surfaces of the fibroin molecules in the solution. At alcohol concentrations below 70%, the weak bond binding and hydration state of fibroin reaches an equilibrium, forming a gel. Under the condition that the alcohol concentration is higher than 70%, the weak bond binding action of the fibroin molecules is enhanced, and the molecules are agglomerated and then separated out from the solution into flocculent or solid particulate. Therefore, the concentration of the alcohol solution added in the invention is 30-70%.
If the fibroin solution is added into the alcoholic solution, the fibroin which is firstly contacted with a large amount of alcoholic solution can be firstly gelled and agglomerated, and is not easy to mix uniformly. Therefore, the medical alcohol solution is quickly added into the fibroin solution and stirred and mixed.
When high concentration alcohol is added into the formed alcohol-fibroin gel, the fibroin still has the tendency of being precipitated from the solution, but because the fibroin is combined with weak bonds to form gel at the moment, the gel is in an irreversible state, and the precipitation of the fibroin is more precipitated in the state of gel particles. When the stirring is performed with strong force, the fine particles of the precipitates are relatively uniform and are not easily precipitated. Therefore, when the alcohol concentration of the alcohol-fibroin gel disposable disinfectant is adjusted to be more than 70%, high-concentration alcohol needs to be added on the basis of the prepared alcohol-fibroin gel disposable disinfectant.

As shown in FIG. 1, the evaporation rates of alcohol and the alcohol-fibroin gel disposable disinfectant of the present invention were compared, and "series 1" in FIG. 1 is the initial solution weight and "series 2" is the solution weight after two hours. Bar 1 in fig. 1 represents 5.5ml of alcohol with a concentration of 70% by volume; bar 2 represents 5ml of 70% alcohol by volume + 0.25ml of 4.8% w/v silk fibroin solution; bar 3 represents 5ml of 70% alcohol by volume + 0.4ml of 4.8% w/v silk fibroin solution; bar 4 represents 5ml of 70% alcohol by volume + 0.5ml of silk fibroin solution at 4.8% w/v. After the 4 solutions were added into glass dishes with a diameter of 3.7mm, the glass dishes were placed at a temperature of 20 ℃, RH 40% relative humidity, and volatilized indoors with a fan, comparative experiments were performed, and then the experimental results were prepared as shown in FIG. 1, and it can be seen from FIG. 1 that the fibroin solution can delay the volatilization of alcohol components.
The following experiment was performed by mixing 0.5ml of silk fibroin solutions with different concentrations with 10ml of alcohol with a volume percentage concentration of 60%, to obtain the time required for gelation after mixing the alcohol with the silk fibroin solutions with different concentrations, as shown in the following table.
Concentration% (w/v) of silk fibroin | 4.8 | 4 | 3 | 2 | 1 | 0.5 |
Gel time (minutes) | 5 | 8 | 15 | 30 | 60 | 90 |
Fluidity of gel | Flow of | Flow of | Flow of | Flow of | Flow of | Flow of |
From the above table, it can be seen that the concentration of the fibroin solution can be mixed with alcohol in a wide range to form a gel having fluidity, and the lower the concentration of the fibroin solution, the longer the time required for the gel. The lower the concentration of the fibroin solution, the thinner the gel formed, and the concentration of the fibroin solution can be selected within a certain range according to requirements in production.
An experiment was performed using 0.5ml of 4.8% (w/v) silk fibroin solution mixed with 10ml of different alcohol concentrations to obtain the time required for gelation after mixing the silk fibroin solution with the different alcohol concentrations, as shown in the following table.
Alcohol concentration% | 30 | 40 | 50 | 60 | 70 |
Gel time (minutes) | 120 | 30 | 15 | 5 | 3 |
Fluidity of gel | Flow of | Flow of | Flow of | Flow of | Flow of |
As can be seen from the above table, the alcohol with concentration range of 30-70% can be mixed with the fibroin solution to form a gel with fluidity. Different alcohol concentrations may be selected according to the use requirements.
The alcohol-fibroin gel washing-free disinfectant disclosed by the invention is not added with other components except for the medical alcohol solution, the fibroin solution and the micromolecular sericin solution, is very safe to use, and can moisten the skin. Has flowing gel form, is convenient for smearing on hands, can delay the volatilization of alcohol components, and enhances the sterilization effect.
Although the present invention has been described with reference to the above embodiments, it should be understood that the scope of the present invention is not limited thereto, and that various changes and modifications can be made by those skilled in the art without departing from the spirit and scope of the present invention.
