技术领域
本发明涉及细胞分裂和细胞膨胀协同调控领域,特别涉及具有正向调控植株侧生器官大小的INP1基因及其同源基因。
背景技术
细胞分裂和细胞膨胀协同调控多细胞生物的生长及最终器官大小。例如在拟南芥中,其叶片发育受到五个相互交错的细胞增殖生长活动时期:起始期、初级细胞分裂期、转换期、拟分生组织细胞分裂期和细胞膨胀期的严格时空调控。截至目前,在拟南芥及相关的模式植物中已发现多个调控以上发育时期的调节因子,并解析了多条以此相关的调节通路。泛素-蛋白酶体途径广泛参与生物体生长发育的调控过程,其中包括细胞周期的调控。例如,在拟南芥叶片发育早期,F-box蛋白AtFBX92通过影响细胞分裂而抑制叶片生长;又如拟南芥F-box蛋白FBL17通过形成SCFFBL17E3泛素连接酶并降解KRP2,从而促进细胞增殖和核内复制。SAP-PPD-KIX-TPL和SAP-PPD-NINJA-TPL通路对于拟南芥最终器官大小和叶片形态的调控起着十分重要的作用。拟南芥F-box蛋白SAP形成SCFSAPE3泛素连接酶,一方面通过降解PPD2-KIX8/9抑制复合物来正调控器官大小,另一方面则通过降解PPD2-NINJA抑制复合物来调节叶片的形态。PPD1/2及其同源基因的抑制或敲除、KIX8/9基因的突变均可导致植物叶片器官变大并呈现半球状,而SAP及其同源基因的突变则导致器官变小。泛素受体DA1与E3泛素连接酶DA2或EOD1/BB协同作用来调控拟南芥细胞增殖和最终器官大小。
发明内容
为解决上述现有技术存在的问题,本发明的目的在于提供具有正向调控植株侧生器官大小的INP1基因及其同源基因。
为达到上述目的,本发明的技术方案为:
具有正向调控植株侧生器官大小的INP1基因及其同源基因,所述INP1基因及其同源基因序列如SEQ ID:1-14所示。
进一步的,所述INP1基因及其同源基因分别属于:蒺藜苜蓿、大豆、花生、鹰嘴豆、狭叶羽扇豆、番茄、马铃薯、烟草。
所述的INP1基因及其同源基因,在直接调控植物侧生器官的大小方面的应用。
INP1基因及其同源基因的突变导致侧生器官变小,过表达导致侧生器官变大。
相对于现有技术,本发明的有益效果为:
本专利申请中所涉及到的INP1基因为首次在蒺藜苜蓿中被克隆及功能报道,之前无任何相关研究和报道;专利申请中的INP1在相关物种中的同源基因也无任何功能报道,均为首次被分析。

下面结合附图和具体实施方式对本发明技术方案做进一步详细描述:

蒺藜苜蓿INP1基因及相关物种中同源基因,
我们从Noble Research Institute蒺藜苜蓿(Medicago truncatula)的烟草逆转录转座子Tnt1插入突变体库中筛选到6个表型一致且侧生器官变小的独立突变体株系,经等位性实验验证该6个突变体为等位突变体(同一个基因不同位置发生了插入突变),分别命名为inp1(incomplete pulvinus1)-1、inp1-2、inp1-3、inp1-4、inp1-5和inp1-6。该突变体有以下几个特征:①细胞大小增大:相对于野生型(WT)蒺藜苜蓿,inp1突变体细胞增大;对叶表皮细胞大小的追踪测量发现,在P5(Plastochron 5)期之前inp1突变体与野生型无明显区别,但在P6期及之后时期将超过野生型,且在后期差异会进一步扩大。②细胞数目减少:计算结果表明,inp1突变体每个小叶的表皮细胞总数少于野生型的50%;③叶表皮细胞的排列模式及气孔指数(Stomata Index)不变:inp1突变体的叶表皮细胞形态和气孔指数没有发生变化,表明在叶片发育过程中,inp1突变体中很可能初级细胞分裂受到抑制,而次级细胞分裂没有受到影响。综上所述,这些数据表明inp1突变体叶片大小的改变主要原因是由于初级细胞分裂受到抑制而导致细胞数量减少所引起的。
(1)蒺藜苜蓿inp1突变体的分离与特征描述
我们从蒺藜苜蓿(Medicago truncatula)的烟草逆转录转座子Tnt1插入突变体库中筛选到6个表型一致且侧生器官变小的独立突变体株系,分别为命名为inp1-1(incomplete pulvinus1-1)、inp1-2、inp1-3、inp1-4、inp1-5和inp1-6。该突变体有以下几个特征:①细胞大小增大:相对于野生型(WT),inp1突变体细胞增大;对叶表皮细胞大小的追踪测量发现,在P5期之前inp1突变体与野生型无明显区别,但在P6期及之后超过野生型,且在后期差异会进一步扩大。②细胞数目减少:计算结果表明,inp1突变体每个小叶的表皮细胞总数少于野生型的50%;③叶表皮细胞的排列模式及气孔指数(Stomata Index)不变:inp1突变体的叶表皮细胞形态和气孔指数没有发生变化,表明在叶片发育过程中,inp1突变体很可能初级细胞分裂受到抑制,而次级细胞分裂没有受到影响(Ge等,2016)。综上所述,这些数据表明inp1突变体叶片大小的改变主要原因是由于初级细胞分裂受到抑制而导致细胞数量减少所引起的。
(2)INP1基因克隆及蛋白结构特征描述
①材料收集与测序:将杂合的inp1-1、inp1-2和inp1-3突变体分别与野生型植株(R108)进行回交(inp1纯合突变体不育,所以只能用杂合突变体与野生型进行回交);分别收集inp1-1、inp1-2和inp1-3回交群体中的纯合突变体样品(50个茎尖),提取基因组DNA;利用提取的基因组DNA进行二代全基因组重测序。
②重测序数据分析:通过使用ITIS(转座子插入位点识别)工具对inp1-1、inp1-2和inp1-3基因组重测序数据进行比对分析,确定Tnt1在基因组中的插入位点(Chuan Jiang等,2015);因突变体基因组中有多个Tnt1插入位点,通过对插入位点的分析确认inp1-1、inp1-2和inp1-3在Medtr5g097060基因位点都有Tnt1的纯合插入,初步确认inp1突变体的表型是由于Medtr5g097060位点的插入突变所引起的;我们将Medtr5g097060命名为INP1(INCOMPLETEPULVINUS1)。
③连锁验证:利用PCR验证inp1-1、inp1-2和inp1-3突变体无法扩增Medtr5g097060基因序列,而能扩增Tnt1序列,进一步验证Medtr5g097060对应INP1基因。
④反向验证及遗传互补验证:反向在Tnt1插入突变体库中筛选到3个Medtr5g097060位点有Tnt1插入的突变体株系,表型与inp1-1、inp1-2和inp1-3相似,都呈现出侧生器官变小的表型,分别命名为inp1-4、inp1-5和inp1-6;然后,我们将INP1自身启动子驱动的INP1编码序列(CDS)通过农杆菌介导的方法转入inp1突变体中,其突变表型被互补。这些结果证实了Medtr5g097060位点与INP1基因相对应,INP1的突变是导致inp1突变体突变表型的原因。
⑤蛋白结构特征描述:通过进化分析和氨基酸序列比对分析表明INP1编码一个F-box蛋白,其蛋白N端具有高度保守的F-box结构域,C端具有WD40 Repeat结构域,说明INP1蛋白可通过F-box结构域与其他相关蛋白相互作用,形成SCF E3泛素连接酶,并通过其C端结构域识别特异底物并将其泛素化降解。
(3)INP1表达模式与蛋白亚细胞定位
qRT-PCR结果显示,INP1在叶枕、叶片、托叶、花器官和未成熟种子中均有高表达。RNA原位杂交结果显示,INP1的转录产物在茎尖分生组织(SAM)、早期叶原基(P0-P2)的外围区积累。4周龄GUS转基因幼苗的成熟叶片维管组织中GUS报告基因表达量较高。这些结果表明,INP1在蒺藜苜蓿叶片发育过程中起着重要的作用。
在花椰菜花叶病毒35S启动子(CaMV35S)的控制下,我们在烟草的叶中瞬时表达INP1与绿色荧光蛋白的融合蛋白(INP1-GFP或GFP-INP1),以确定INP1亚细胞定位。INP1-GFP和GFP-INP1融合蛋白均定位于烟草表皮细胞细胞核。
(4)INP1的过表达促进蒺藜苜蓿初级细胞分裂及器官大小
INP1突变导致蒺藜苜蓿的器官变小。为了进一步研究INP1基因在器官大小调控中的功能,我们将35S::INP1或35S::INP1-GFP载体转入野生型中,获得INP1过表达的植株。过表达植株呈现器官增大、叶片呈半球状的表型,该表型与已报道的bs1突变体表型相似(Ge等,2016)。与野生型相比,35S::INP1和35S::INP1-GFP转基因株系的INP1表达量分别增加了12倍和1000倍以上。与inp1突变体相反,INP1过表达植株的细胞体积相对于野生型减小。后续研究表明,类似于inp1突变体,INP1过表达植株与野生型叶片表皮细胞大小的分化可以追溯到P6期,后期这种差异逐渐增大。INP1过表达植株每片小叶的表皮细胞总数均为野生型的两倍以上。和inp1突变体相似,INP1过表达植株表皮细胞的模式和气孔指数与野生型相似,表明在INP1过表达的植株中,初级细胞分裂增强。叶片发育过程的分析表明,与inp1突变体相似,过表达的植株的小叶长、宽、面积到达稳定期的时间点较野生型有延迟,但小叶生长的持续时间无明显差异,进一步说明了在INP1过表达和inp1突变植株中拟分生组织细胞分裂不受影响。综上所述,在蒺藜苜蓿中INP1可能通过促进初级细胞分裂而正向调节器官大小。
(5)蒺藜苜蓿中F-box蛋白INP1调控BS1蛋白的稳定性
蒺藜苜蓿中BS1基因的突变会导致类似INP1基因过表达的器官增大、叶片呈现半球状的表型(Ge等,2016)。之前的研究报道的线索也表明F-box蛋白INP1很可能通过与其他相关蛋白结合形成SCF E3泛素连接酶,通过泛素-蛋白酶体途径特异识别并降解初级细胞分裂及侧生器官生长的正向调控因子BS1,从而促进细胞分裂及植物生长。酵母双杂交和双分子荧光互补实验表明INP1可以直接与MtASK结合,很可能形成SCF E3泛素连接酶。同样,通过酵母双杂交和双分子荧光互补实验证明INP1能与BS1直接相互作用。酵母双杂交实验结果表明,INP1通过其WD40 Repeat重复结构域与BS1相互作用,体外下拉实验也证实了这一点。此外,蛋白质泛素化降解实验也证明INP1可以直接降解BS1。综上所述,表明在蒺藜苜蓿中INP1可以形成SCF E3泛素连接酶,并通过泛素-蛋白酶体途径降解BS1并正向调控器官大小。
(6)蒺藜苜蓿中INP1影响细胞周期调控基因的表达
植物最终的器官大小被细胞分裂及细胞膨胀两个过程协同调控(Nathalie等,2012)。细胞周期基因在植物基因组中广泛分布,在植物发育过程中起着调节细胞分裂和生长的主要作用(Klaas等,2002;Janice等,2009)。为了揭示inp1突变体中核心细胞周期调控基因表达与细胞分裂及器官大小变化之间的关系,我们用qRT-PCR检测了inp1突变体和野生型植株中MtCYCD3;2,MtCDKB1;1,MtE2Fb和MtKRP3等细胞周期Marker基因的表达。结果显示,与野生型相比,inp1突变体中细胞分裂的正向调控因子MtCYCD3;2,MtCDKB1;1和MtE2Fb的表达降低,特别是在幼叶中。相反,在具有高细胞分裂活性的P4和P5期,inp1突变体中细胞分裂抑制因子MtKRP3的表达增加。这些结果表明,在蒺藜苜蓿中INP1会影响核心细胞周期调控基因的表达,进而调控细胞分裂和器官大小。
3运用价值
农作物侧生器官大小直接决定着其最终的产量。该研究结合之前的研究报道,说明INP1及其同源基因在正向调控植物器官大小方面具有较高的保守性。通过进化分析表明,INP1在其他重要的豆科作物,如大豆(Glycine max)、花生(Arachis duranensis及Arachis ipaensis)、鹰嘴豆(Cicer arietinum)、羽扇豆(Lupinus angustifolius),及其他经济、农业作物,如番茄(Solanumly copersicum)、马铃薯(Solanum tuberosum)、烟草(Nicotiana tabacum)中具有保守的同源基因。INP1基因直接调控植物侧生器官的大小,所以其与同源基因在分子育种方面具有较大的运用潜力及价值。

INP1及其相应同源蛋白在不同物种中参与器官大小调控的证据有以下几点:
如图1所示,对INP1蛋白及相应物种中同源蛋白进行进化分析,从进化树可以看出所有蛋白具有同源性,聚集在相应的分枝上,这从一方面说明其功能的保守性;
如图1所示,对INP1蛋白及相应物种中同源蛋白进行蛋白保守结构域分析,分析结果显示:INP1及其相应的同源蛋白都具有保守的F-box结构域(黑色方框)及WD40 repeat结构域(灰色方框);功能保守的特定家族的蛋白一般依赖于其特定的保守结构域来发挥功能,根据已有的研究报道,F-box蛋白中的F-box结构域一般通过结合ASK/SKP蛋白及Cullin蛋白从而形成SCF E3泛素连接酶复合物参与特定底物蛋白的泛素化,而F-box蛋白中的WD40结构域则是参与识别特定的被泛素化的底物,在生物体内,被泛素化的蛋白会被蛋白酶体(Proteasome)所降解,该蛋白调控机制在生物的生长发育过程中广泛参与各种生物学过程,发挥着十分重要的功能。所以从INP1及其同源蛋白的蛋白结构保守性,以及泛素化调控功能的保守性可以说明这些基因/蛋白器官大小调控功能的保守性;
INP1基因在蒺藜苜蓿中突变造成侧生器官变小,而过表达则导致变大;同样,根据已有的报道,INP1在拟南芥中的同源基因突变造成叶片和,果荚和花器官等变小,而过表达也导致变大;INP1在黄瓜中的同源基因突变后,叶片,花瓣和果实变小。由此可见,在多个物种中INP1或是其同源基因对侧生器官具有正调控功能,这是INP1及其同源基因功能保守性最直接的证据。
以上所述,仅为本发明的具体实施方式,但本发明的保护范围并不局限于此,任何不经过创造性劳动想到的变化或替换,都应涵盖在本发明的保护范围之内。因此,本发明的保护范围应该以权利要求书所限定的保护范围为准。
