Abstract

The invention belongs to the field of biotechnology, and particularly relates to preparation of Geranium wilfordii (Geranium wilfordii Maxim.) polysaccharide and application of Geranium wilfordii polysaccharide in development of liver-protecting medicines. The invention adopts an immune liver injury model induced by concanavalin A and investigates the liver protection activity of the immune liver injury model. The experimental result shows that the geranium polysaccharide can obviously reduce the liver function index of mice with liver injury, and can be used for researching and developing liver-protecting medicaments, in particular medicaments for treating immunological liver injury. The invention has important significance for further development and utilization of the geranium wilfordii.

Research shows that the polysaccharide component has various pharmacological activities, such as immunity regulation, anti-inflammation, antioxidation, antitumor and the like, and has a plurality of researches and applications in the fields of medicines and health-care foods.
Geranium wilfordii (Geranium wilfordii Maxim.) is a plant of Geranium of Geraniaceae, is one of the common Chinese medicines, and has the effects of dispelling wind-damp, dredging channels and collaterals, and the like. The dose of Geranium sibiricum polysaccharide is 2-3 times per day, and 0.4-1.8g each time.
The dosage of Geranium polysaccharide is 1g for 3 times per day.
Adding pharmaceutically acceptable adjuvants into the Geranium polysaccharide, and making into tablet, hard capsule, soft capsule, powder, pill, and granule.

Example 1
Preparing the geranium crude polysaccharide: pulverizing herba Erodii seu Geranii, and defatting with methanol at 60 deg.C for 3 times. Decocting the defatted herba Erodii seu Geranii with hot water at 80 deg.C for 3 hr for 2 times, mixing extractive solutions, and concentrating under reduced pressure. Precipitating the concentrated extract with 80% ethanol, centrifuging, and removing supernatant to obtain crude polysaccharide of herba Erodii seu Geranii.
Example 2
And (3) refining geranium polysaccharide: re-dissolving the obtained geranium polysaccharide with distilled water, removing protein by a sevege method, centrifuging, discarding the precipitate for 5 times, and centrifuging until no precipitate exists. And finally, decompressing and concentrating the polysaccharide after protein removal, freezing and drying, and detecting the content of the polysaccharide by a sulfuric acid-phenol method to finally obtain the refined geranium polysaccharide.
Example 3
Liver-protecting efficacy research of geranium polysaccharide
1. Animal grouping, modeling and administration method
Geranium polysaccharide was prepared from example 2.
ICR mice were randomly divided into a normal control group, a model group, a positive group (bifendate 150 mg/kg), a geranium polysaccharide low dose group (200 mg/kg), and a geranium polysaccharide high dose group (600 mg/kg), each group containing 10 mice. The administration was performed by gavage, and the normal group and the model group were administered with physiological saline, and the other groups were administered with the corresponding dose of the drug 1 time for 1 day for 7 consecutive days. After 30min of the last administration, 12mg/kgConA is intravenously injected to the model group and each administration group to prepare an acute immune liver injury model, and normal groups are intravenously injected with normal saline with the same volume. After fasting for 7h without water deprivation, the animals were sacrificed by orbital bleeding and liver collection.
2. Serum liver function index detection
Blood is taken from eyeballs in a 1.5mL centrifuge tube, and the eyeballs are centrifuged at 4500r/min at normal temperature for 10min, and supernatant is left. Processing according to the procedures of alanine Aminotransferase (ALT) and aspartate Aminotransferase (AST) kits, and measuring the absorbance values (OD values) of ALT and AST in serum by using a microplate reader. And calculating the corresponding enzyme activity value according to the OD value by searching a standard curve.
The experimental results are shown in fig. 1, compared with the normal group, the AST and ALT values of the model group are obviously increased, and compared with the model group, the AST and ALT values of the positive drug and the administration groups with different doses are obviously reduced, and the dose dependence is shown. The results show that the geranium polysaccharide has a protective effect on immune liver injury mice.
In the mouse acute immunological liver injury test, the dosage of the geranium polysaccharide is 200-600 mg/kg/d. The dosage is 1.2-3.6g/d converted into human (calculated by adult weight 60 kg). In consideration of the difference in body weight of human and various other pathological conditions, the recommended dose is 2-3 times daily, 0.4-1.8g each time. The most common dosage is 1g each time 3 times daily.
In conclusion, the geranium polysaccharide has the characteristics of obvious liver protection activity and small toxic and side effects, and has wide application in the development of preparing liver protection medicines, particularly in the treatment of immunological liver injury diseases. Has important significance for further development and utilization of the geranium.
