Abstract
An application of lentinan in preventing and treating fungal diseases of tea trees belongs to the technical field of agriculture. The application of lentinan in preventing and treating tea tree fungus diseases, wherein the tea tree fungus refers to tea anthracnose pathogen and tea leaf spot pathogen. The activity of the lentinan for inhibiting the fungal pathogens of tea trees is researched, and the result shows that the lentinan has good bacteriostatic activity on two main fungal diseases of the tea trees, namely anthracnose and alternaria alternate, and can be applied to control the fungal diseases of tea. At present, the prevention and the treatment of the fungal diseases of tea trees mainly adopt chemical drug prevention and treatment, pathogenic bacteria drug resistance is easy to cause due to long-term single use of similar medicaments, and meanwhile, chemical pesticide residues threaten the food safety of human beings and restrict the export of tea leaves. The lentinan is a novel natural functional polysaccharide separated and extracted from lentinus edodes sporocarp, is green and environment-friendly, can not cause pesticide residue while ensuring the quality of tea leaves, and has great potential for preventing and treating tea tree fungal diseases.
Description
Technical Field
The invention belongs to the technical field of agriculture, and particularly relates to a new application of lentinan in preventing and treating tea tree fungal diseases.
Background
Tea trees are important economic crops in China, but because the tea trees mainly grow in warm and humid environments, diseases of the tea trees are bred. In recent years, with the increase of tea demand, the area of a tea garden is continuously enlarged, and the tea tree diseases are increasingly aggravated due to the fact that tea trees in a part of tea areas are single in variety and extensive in management. Tea tree fungi are mainly controlled by chemical methods at present, but biological control methods are still very limited. The long-term single use of the same medicament can easily cause the increasing of the problems of pathogenic bacteria drug resistance and food and ecological safety caused by pesticide residue, so the environment-friendly biological pesticide has good application prospect.
Lentinan (LNT for short) is a novel natural functional polysaccharide separated and extracted from lentinus edodes sporocarp, mainly contains glucose and a small amount of mannose, fucose, galactose, xylose, arabinose and the like; the peptide chain is composed of 18 amino acids such as aspartic acid, histidine, serine, lysine, glutamic acid and the like.
Chinese patent CN101869111B discloses a plant virus disease resistant composition containing lentinan and ribavirin, which takes the lentinan and the ribavirin as effective components, wherein the content of the lentinan is 0.5-5 wt%, the content of the ribavirin is 0.5-5 wt%, and the composition is applied to the prevention and treatment of tomato virus diseases or tobacco virus diseases.
Therefore, lentinan is a biological regulator with the functions of inhibiting tumor growth and improving immunity in animals, has the functions of stimulating the response of a plant immune system, improving the disease resistance of plants and regulating the growth of plants in plants, and has a good control effect on plant virus diseases and bacterial diseases, but the application of lentinan in the control of tea tree fungal diseases is not reported yet.

Detailed Description
The effects of the present invention will be further described below by way of specific embodiments.
Example 1: inhibition of tea tree pathogenic fungi by lentinan
The pathogenic fungi used in the invention refer to tea anthracnose pathogen and tea alternaria, and are tea plant pathogenic fungi. And performing activated culture (at 25 ℃ and in dark) on the test strain on a PDA (personal digital assistant) plate for 5-10 d, and performing subculture for 1-2 times to obtain the strain for experiment. The lentinan used as an experimental agent was purchased from Shanghai Hui union biological medicine (Xinyi) GmbH.
Inhibition of hyphal growth by hyphal growth rate assay:
measuring the test reagent solution with different concentrations by using a pipette, adding a sterilized PDA culture medium cooled to about 60 ℃, uniformly mixing, pouring into a culture dish, and preparing into the culture medium containing the lentinan with different concentrations (the concentration gradient is 2.5 mu g/mL, 5 mu g/mL, 10 mu g/mL, 20 mu g/mL, 40 mu g/mL, 80 mu g/mL and 160 mu g/mL). Transferring the stored pathogenic bacteria into PDA plate without adding medicament, activating at 26 deg.C for 96 hr, preparing bacterial cake with diameter of 7mm near the colony edge with a puncher, and transferring to the center of PDA plate containing different concentrations of medicament. At the same time, the treatment was repeated 3 times with the medium without the agent as a control, all the operations being carried out on a sterile operating table.
Placing the inoculated culture dish in a constant-temperature incubator at 26 ℃ for inverted culture for 5d, observing the growth condition of each treated colony, measuring the diameter of the colony by a cross method, calculating the mean value of the diameters of the colonies, and calculating the average inhibition rate of the growth of hyphae according to the following formula. The average inhibition rate of hypha growth is (pure growth amount of negative control-pure growth amount of treatment)/pure growth amount of negative control x 100%; pure growth amount-average diameter of colony-cake diameter. The virulence regression equation, the correlation coefficient (r) between the inhibition probability value (y) and the log of concentration (x) was found using Microsoft Excel.
The inhibition effect of lentinan with different concentrations on the growth of hyphae of tea anthracnose pathogen and tea round spot pathogen is shown in table 1, the lentinan has a good inhibition effect on the tea anthracnose pathogen and the tea round spot pathogen, and the inhibition effect on the two pathogenic fungi reaches nearly 80% when the concentration is 160 mug/mL; the toxicity of lentinan on tea plant fungi is calculated, and the inhibition medium concentrations EC50 of lentinan on tea anthracnose and tea round spot pathogenic fungi are respectively 10.55 mug/mL and 2.68 mug/mL, so that the results show that the lentinan has obvious inhibition effect on the growth of the tea anthracnose and tea round spot pathogenic fungi, and the lentinan has great potential in the prevention and treatment of tea plant fungal diseases.

The invention discloses a new application of lentinan in preventing and treating tea tree fungal diseases, and proves that the lentinan has good inhibitory activity on tea anthracnose and tea round spot pathogenic fungi through indoor toxicity measurement. Lentinan belongs to natural functional polysaccharide, is green and environment-friendly, ensures the quality of tea leaves, does not cause pesticide residue, and has great potential in the prevention and control of tea tree fungal diseases.
