Description

Traditional Chinese medicine composition for preventing or treating sub-health, nano-medicine and preparation method thereof
Technical Field
The invention relates to the technical field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition for preventing or treating sub-health, a nano-medicine and a preparation method thereof.
Background
The sub-health is often firstly shown on the body in real life, namely the body is sub-health, the main symptoms comprise fatigue, aching pain, insomnia, haggard, function reduction, dysfunction and the like, people in the sub-health state often feel poor physical strength, fatigue and sleepiness, lack of vitality and facing qi, ① cardiovascular symptoms comprise chest distress, palpitation, shortness of breath, breath holding and the like, ② digestive system symptoms comprise poor appetite, anorexia (the vegetables are seen to have no appetite and sometimes feel hungry but do not want to eat), ③ osteoarticular symptoms comprise waist soreness and backache, joint discomfort or general discomfort, ④ nervous system symptoms comprise frequent head opening, or oppression, fullness, dizziness, poor memory, general weakness, sleepiness, and fatigue, ⑤ sleep symptoms comprise easier waking up in the morning, easier dreaminess, easier urinary dream, less urinary urgency and lower reproductive function, ⑥ urinary urgency and urinary urgency.
The sub-health hazard can cause great influence on daily life all the time, and is mainly characterized in that ① influences work efficiency, life and learning quality, ② influences sleeping quality, aggravates physical and mental fatigue and causes chronic fatigue syndrome, ③ causes vicious circle and causes mental or body diseases, ④ is susceptible to most chronic diseases, such as most malignant tumors, cardiovascular and cerebrovascular diseases and diabetes, specifically comprises hyperlipidemia, fatty liver, hypertension, prostate diseases, abnormal liver functions, gynecological diseases, coronary heart diseases, diabetes, cataract, gallstone and cancer prevention general survey abnormality, ⑤ influences health life, causes premature senility and even causes early death (overwork death) due to sudden emergency.
In view of this, the combination of medicines for conditioning or treatment is required, wherein western medicines are mostly required for symptomatic treatment, the curative effect is single, and the conditioning and treatment effects are limited. Compared with western medicines, the traditional Chinese medicine has more advantages in conditioning and treating sub-health, but the traditional Chinese medicine is poor in relative curative effect and not obvious in effect at present, needs to be taken for a long time, is taken for a long time, increases the burden of a patient, and easily causes adverse symptoms such as diarrhea.
The evaluation of the traditional Chinese medicine on the sub-health is carried out by students of traditional Chinese medicine on the sub-health according to the traditional Chinese medicine theory, and the evaluation method comprises the following 10 standards of ① patients with uneasiness, palpitation and insomnia, ② patients with body fluid secretion caused by sweating and frequent colds, ③ patients with red tongue and tongue fur, bitter taste and dry stool, ④ patients with poor complexion, dark gray and dark surrounding eyes, ⑤ patients with limb expansion, silkworm lying below eyes, ⑥ nail imaging and abnormal change, ⑦ chest swelling before tide, breast nodules, ⑧ vomit adhesive, hiccup fullness, ⑨ abnormal body temperature, lassitude and weakness, ⑩ patients with blurred vision and headache.
Assessment of "sub-health" by modern medicine: modern medicine's assessment of sub-health includes clinical and laboratory examinations; the former includes electrocardiogram, electromyogram, electroencephalogram, ultrasonic examination, X-ray examination, CT examination, nuclear magnetic examination, etc., and the latter includes routine examination (blood, urine, stool, sputum), body fluid examination (gastric juice, cerebrospinal fluid, blood, puncture fluid), bone marrow, liver function, kidney function, blood lipid, blood glucose, tumor marker, etc. The evaluation indexes of sub-health are summarized into 30 items including: mental stress, anxiety and restlessness; autistic, bitter and stuffy; distraction, shallow thinking skin; easy excitement and no trouble; memory decline, the acquaintance forgets the name; the interest becomes light and the desire declines; lazy to contact, low mood; the eyes are easy to feel tired and lack of strength; decreased energy and bradykinesia; dizziness and brain distension, and difficult recovery; dizziness and blurred vision when standing for a long time; the limbs are limp and weak, and the force is not strong; weight loss, weakness; the sleep is not easy to fall, and dreaminess is easy to wake up; morning unwilling and daily dozing; local numbness, easy cooling of hands and feet; sweats in the palms and axilla, dry tongue and dry mouth; spontaneous low fever, night sweat; soreness of the waist and back pain, which causes petechia; the tongue has white fur and the halitosis is self-generated; mouth and tongue ulcers, which occur repeatedly; the sense of taste is not flexible, and the appetite is not good; acid eructation and dyspepsia; defecating and constipation, abdominal distention; susceptible to cold, herpes labialis; nasal obstruction, nasal discharge, sore throat; suffocating and shortness of breath, and urgent respiration; chest pain, chest distress, feeling of pressure in the heart; palpitation, and irregular heart rhythm; tinnitus, ear-back and easy carsickness.
The important idea of preventing and treating diseases in the traditional Chinese medicine is that the traditional Chinese medicine is used for preventing and treating diseases, and the method comprises the following four aspects: the disease is not cured and is kept in good health, and the disease is prevented in the first place; to be rescued, the patient is protected from mild disease; the disease is cured early, and the transmission and the transformation of the disease are prevented; recuperating after illness to prevent recurrence.
The traditional Chinese medicine is a traditional Chinese medicine for tonifying qi and nourishing blood with medicines, and common formulas in daily life are red date, donkey-hide gelatin, astragalus, polygonum multiflorum, American ginseng and the like, but sub-health people are not enough for simply tonifying blood and qi due to the comprehensive imbalance of qi, blood and yin. Taking a formula for tonifying spirit, strengthening qi and essence and blood (trade name: jianyanling capsules) as an example, the formula takes polygonum multiflorum and rhizoma polygonati which are monarch drugs for tonifying liver and kidney and benefiting essence and blood; the prepared rehmannia root, human placenta and other blood nourishing and essence replenishing assistant monarch drugs are used for synergism; the black soya bean is used for promoting blood circulation and diuresis, and the effect of promoting menstruation is matched with monarch and minister tonifying medicines, the seventeen medicines are strictly matched and are complementary to each other, the tonic is supplemented with the product for clearing away the heat in the tonifying process, the tonic is not greasy, and the yin and yang are adjusted, so that the effect that the viscera are strong and the diseases are not disturbed when qi and blood are sufficient in the traditional Chinese medicine is realized, but the effect is still to be improved.

The nanometer powder prepared from the traditional Chinese medicine composition can improve bioavailability, increase targeting property, reduce toxic and side effects, show new drug effect, broaden the indications of the original drug, enrich the dosage form selection of the traditional Chinese medicine, reduce the dosage and save the traditional Chinese medicine resources. More specifically, the following advantages are provided:
(1) improve or increase the solubility and bioavailability of Chinese medicinal materials. Traditional Chinese medicines are usually decoction, but some components in the traditional Chinese medicines are low in solubility in water (or other solvents), so that the traditional Chinese medicines are not suitable for being prepared into various dosage forms for clinical use. The prepared nano powder can well solve the problem of solubility of the components, is convenient for preparation and improves the effectiveness of the medicament.
(2) Is convenient for preparing sustained release preparation or controlled release preparation. The half-life period of part of the traditional Chinese medicine components is short, the slow release or controlled release purpose cannot be achieved, the traditional Chinese medicine composition is similar to western medicines with short half-life period, the effective blood concentration in vivo can be maintained only by frequent administration, and the problem can be solved by adopting the nanometer traditional Chinese medicine composition.
(3) Targeted drug delivery. The prepared nano traditional Chinese medicine composition can adopt different high molecular materials or carry out surface modification according to the difference of physiological enzyme and bacteria distribution, cell distribution, reticular structure, lymph distribution and the like of different parts in vivo, so as to prepare the required traditional Chinese medicine nano particles which are conveyed to a specified target area; the magnetic nano-particles can also be prepared by adopting a method of combining in vivo and in vitro to achieve the aim of targeted drug delivery.
(4) Smart drug delivery systems (feedback regulated release of drug). The nanometer Chinese medicine composition may be prepared into nanometer particle, liposome, etc. and through proper surface modification or grafting with polymer material sensitive to some enzyme, pH value, protein and other change in physiological environment, the medicine release of the Chinese medicine preparation may be controlled based on some change information.
(5) Reduce the stimulation of some traditional Chinese medicines to the stomach and intestine. The stimulation of the oral traditional Chinese medicine to the stomach and intestine is related to the granularity and the release speed of the medicine, the medicine with the stimulation to the stomach and intestine is prepared into nano particles, nano emulsion and the like, and then the proper administration form is adopted to reduce the local 'accumulation' release of the medicine in the stomach and avoid or reduce the stimulation of the medicine to the stomach and intestine.

The herba Dendrobii powder is prepared by pulverizing herba Dendrobii into powder.
The invention also provides a preparation method of the traditional Chinese medicine composition, which comprises the following steps:
mixing the components of the Chinese medicinal composition to obtain the Chinese medicinal composition.
Further, the step of mixing the components of the Chinese medicinal composition may comprise the steps of:
s101: pulverizing the above materials to obtain pulverized product.
It is understood that step S101 may further include a step of cleaning and drying the components, for example, a high-speed centrifuge is used to clean and separate the Chinese herbal medicines, such as cordyceps sinensis, panax notoginseng, panax quinquefolius, astragalus membranaceus, ganoderma lucidum spores, bear gall and dendrobium nobile, so as to remove silt and various impurities of the Chinese herbal medicines. The Chinese herbal medicines can be fresh medicinal materials or medicinal materials after treatment (such as sun drying and the like), and are not particularly limited.
In step S101, the related components may be pulverized by using an existing pulverization method, or related pulverized materials may be directly purchased, and the selection of the pulverization method may be selected according to the properties of the medicinal materials.
Preferably, the cordyceps, the panax notoginseng, the American ginseng, the ganoderma robe, the bear gall and the dendrobium are respectively crushed by adopting an ultramicro crushing method; drying and pulverizing radix astragali at low temperature. Wherein, the superfine grinding method has the grinding principle that: the material gets into crushing room by the loading hopper, smashes through the blade of high-speed rotation, adjusts the distance that grading wheel and classifying disc reach required material fineness, and the material that reaches the requirement is introduced into the collection box sack from crushing room to the high-speed rotatory fan blade, and the material that does not reach the requirement continues to smash in crushing room, and the dust that collection box sack produced is collected by the suction box and is smashed to required particle size distribution. In one embodiment, the particle size distribution after pulverization is 5 μm to 10 μm. In one embodiment, the minimum temperature used in the low-temperature drying and pulverizing method is-50 ℃ to-20 ℃.
And S102, performing nanocrystallization treatment on the crushed materials of the components to prepare nano powder.
Further, step S102 includes the steps of:
s1021: carrying out micronization treatment on the crushed materials of the components by adopting a ball mill;
in one embodiment, the micronization process is carried out by a high-energy horizontal ball mill, the ball powder ratio of the high-energy horizontal ball mill is kept in a certain proportion by high temperature and high pressure, vacuum or inert gas atmosphere is kept in a tank, the rotating speed and time of the high-energy ball mill are controlled, and the micronization process of the traditional Chinese medicine can be completed by controlling the temperature (the control range is 50-100 ℃). In one embodiment, the particle size of the pulverized material of each component is less than 10 microns after micronization.
After micronization, a vibration mill (preferably an eccentric vibration mill) can be used for breaking the wall to ensure that the particle size of the powder is less than 10 microns, which is beneficial to the subsequent steps.
S1022: the pulverized material subjected to the micronization treatment was sequentially extracted.
Further, step S1022 includes the steps of:
(a) mixing the micronized powder with water, treating for 3-24h at 90-110 deg.C (preferably 100 deg.C), filtering, collecting filtrate, and concentrating the filtrate to obtain concentrated solution with solid content of 30% -50% (preferably 40%);
(b) mixing the concentrated solution with ethanol at volume ratio of (0.9-1.2):1, precipitating, filtering, collecting precipitate, and drying to obtain nanopowder.
By adopting the method to prepare the nano powder, the uniformity of the particle size of the nano powder can be effectively improved.
S103: mixing the nanometer powders of the above materials, and making into Chinese medicinal composition.
In step S103, mixing may be carried out by using a mixer, and preferably, mixing may be carried out by using a twin-screw high-speed mixer.
Compared with the method that the raw materials are directly mixed and then subjected to nano treatment, the method has the advantages that appropriate parameters can be conveniently selected according to the properties of the medicinal materials of all the components, so that the uniformity of the particle size of all the components in a final product is ensured, and the preparation is convenient.
