Detailed Description
In order to well prevent and control the novel goose astrovirus and solve the hidden danger of antibiotic residue caused by the prevention and treatment of the antibiotic, the invention provides a compound traditional Chinese medicine injection for the goose gout caused by the novel goose astrovirus according to the traditional Chinese medicine theory, and the injection can effectively control the goose gout caused by the novel goose astrovirus. The present invention is further illustrated below with reference to examples to better understand the technical solutions of the present invention, and it should be noted that: in the following examples, 10g of each Chinese medicinal material is 1 part in selected amount.
Example 1: the embodiment provides a compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus, which comprises the following raw materials by weight, 60g of scutellaria baicalensis, 60g of artemisia capillaris, 200g of pyrrosia lingua, 160g of gardenia, 120g of honeysuckle, 400g of astragalus mongholicus, 100g of cockroach, 30g of sophora flavescens and 20g of polygonum aviculare.
The preparation method comprises the following steps:
step one, putting 120g of honeysuckle into a distillation retort, adding 1200mL of water into the retort, distilling, collecting distillate, adding 1.5 times of distilled water into the distillate for redistillation, collecting redistillate, refrigerating the redistillate at 0-4 ℃, and preserving distilled honeysuckle residues and aqueous solution for later use;
step two, respectively crushing cockroaches and polygonum aviculare, uniformly mixing, adding 90-95% ethanol with the weight of 4 times of that of the cockroaches and the polygonum aviculare, soaking for 0.5h, percolating after soaking, collecting percolate, and storing residues after percolating for later use;
putting artemisia capillaries, pyrrosia lingua, astragalus mongholicus, sophora flavescens, gardenia and scutellaria baicalensis into a multifunctional decoction pot, adding water with the weight 5 times that of the medicines into the pot, heating to 60 ℃, keeping the temperature for 2 hours at the temperature of 60 ℃, then filtering with gauze, collecting filtrate, and storing filtered residues for later use; carrying out reduced pressure distillation and concentration on the collected filtrate until half of the filtrate is concentrated to obtain a first concentrated solution;
step four, adding water in an amount which is 4 times that of the residue into the residue obtained in the step two and the residue obtained in the step three for soaking, combining the residue with the honeysuckle residue and the water solution obtained in the step one, boiling for 30min, filtering, and collecting the first filtrate; adding water 3 times the weight of the medicinal residue into the medicinal residue, boiling for 30min, filtering, collecting the second filtrate, mixing with the first filtrate, mixing, and concentrating under reduced pressure to obtain second concentrated solution;

step six, pouring the concentrated solution obtained in the step five into 95% ethanol which is 1.6-2 times of the concentrated solution, continuously stirring, adjusting the alcohol content to 60%, separating out flocculent precipitate, standing for 24 hours, performing suction filtration, collecting filtrate as second refined alcohol solution, recovering ethanol, and concentrating under reduced pressure to half amount;
and step seven, slowly pouring the concentrated solution obtained in the step six into 95% ethanol in an amount which is 3.2 times that of the concentrated solution, continuously stirring, adjusting the alcohol content to 75%, separating out a paste, standing for 24 hours, performing suction filtration to obtain a filtrate, namely a third refined alcohol solution, recovering ethanol, performing reduced pressure concentration to one third, volatilizing the ethanol completely, and cooling.
Step eight, adding 1 time of distilled water into the pure concentrated liquid medicine, uniformly stirring, adding 0.5% of active carbon, freezing and standing for 24 hours, performing suction filtration by using a No. 3 filter stick or filter paper pulp, and evaporating and concentrating the filtrate to one fifth of the original medicine of the preparation;
and step nine, adding honeysuckle distillate into the concentrated solution, adding 1N sodium hydroxide solution to adjust the pH to about 7, adding 0.1% bone charcoal, sealing, insulating water and heating for 2 hours, cooling, measuring the pH to about 5.5, adding 1N sodium hydroxide solution, adjusting the pH to 6.5-7.0, freezing and placing for 48 hours, performing suction filtration by using a No. 3 filter stick or filter paper pulp, adding 2% benzyl alcohol into the filtrate, uniformly stirring, freezing and placing for 24 hours, performing suction filtration by using a No. 4 filter stick, filling and sealing the filtrate, and sterilizing to obtain a finished product, wherein the finished product is a brown yellow clear solution, and the pH is 5.0-6.5.
Example 2: the embodiment provides a compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus, which comprises the following raw materials, by weight, 30g of scutellaria baicalensis, 100g of artemisia capillaries, 150g of pyrrosia lingua, 200g of gardenia, 100g of honeysuckle, 450g of astragalus mongholicus, 150g of cockroaches, 50g of sophora flavescens and 10g of polygonum aviculare.
The preparation method comprises the following steps:
step one, putting honeysuckle into a distillation tank, adding water which is 10 times of the weight of the honeysuckle into the distillation tank, distilling, collecting distillate, adding distilled water which is 1.5 times of the weight of the distillate into the distillate for redistillation, collecting redistillation, placing the redistillation in a refrigeration mode at 0-4 ℃, and preserving honeysuckle residues and aqueous solution after distillation for later use;
step two, respectively crushing cockroaches and polygonum aviculare, uniformly mixing, adding 90-95% ethanol with the weight of 4 times of that of the cockroaches and the polygonum aviculare, soaking for 0.5h, percolating after soaking, collecting percolate, and storing residues after percolating for later use;
putting artemisia capillaries, pyrrosia lingua, astragalus mongholicus, sophora flavescens, gardenia and scutellaria baicalensis into a multifunctional decoction pot, adding water with the weight 5 times that of the medicines into the pot, heating to 60 ℃, keeping the temperature for 2 hours at the temperature of 60 ℃, then filtering with gauze, collecting filtrate, and storing filtered residues for later use; carrying out reduced pressure distillation and concentration on the collected filtrate until half of the filtrate is concentrated to obtain a first concentrated solution;
step four, adding water in an amount which is 4 times of the total amount of the residues into the residues obtained in the step two and the step three, soaking, combining the residues with the honeysuckle residues and the water solution obtained in the step one, boiling for 30min, filtering, and collecting primary filtrate; adding water 3 times the weight of the medicinal residue into the medicinal residue, boiling for 30min, filtering, collecting the second filtrate, mixing with the first filtrate, mixing, and concentrating under reduced pressure to obtain second concentrated solution;

Example 3: the same parts of this embodiment as those of embodiment 1 are not repeated, except that the traditional Chinese medicine injection comprises the following raw materials, by weight, 100g of scutellaria baicalensis, 30g of artemisia capillaries, 250g of pyrrosia lingua, 100g of gardenia, 150g of honeysuckle, 350g of astragalus mongholicus, 50g of cockroach, 10g of sophora flavescens and 50g of polygonum aviculare.
Experimental example: in order to demonstrate the effectiveness of the injectable preparation of the invention, the applicant carried out the following tests:
selecting 20 diseased goslings and 20 diseased adult geese, randomly dividing the test geese into 2 groups, namely a test group and a control group, and ensuring that each group comprises 10 goslings and 10 adult geese.
The experimental group injects the injection according to the method of injecting 2ml per young goose and injecting 5ml per adult goose; the control group was injected with physiological saline according to the method of injecting 2 ml/young goose and 5 ml/adult goose. Blood is collected before and after administration, and the concentration of urate in goose serum is measured before and after administration, and the measurement results are shown in table 1.

As can be seen from Table 1, there was no significant difference in the concentrations of urate in the blood serum of the diseased goose in the test group and the control group before administration, and the concentrations of urate in the blood serum of the diseased goose were both high; after the injection is used, the concentration of the urate in the serum of the diseased goose in a test group is obviously reduced, a control group is not obviously changed, and the test group is obviously reduced compared with the control group (P is less than 0.05), which shows that the injection has good treatment effect on the goose gout.
Clinical application effect
Specific case 1: in 8 months and 10 days in 2017, 6000 laying geese exist in a certain goose farmer in Shangqiu, the geese individually suffer from lassitude, pale beak and pale skin, anemia, loose and fallen feathers, slow movement, lameness, swollen joints of legs and wings, pain feeling caused by touch, loose anus, weak contraction, white excrement discharge and pollution of feathers near the anus, and the goose gout is confirmed to be caused by the novel goose astrovirus through pathogen separation and identification.
The injection is used for intramuscular injection twice a day, 4 ml/time is used for 2 days, the disease condition is controlled on the 3 rd day, and the diseased goose is raised to be normal.
Specific case 2: 26 days in 2019, 4 months, 10000 geese of 6 days old in a certain goose farm in south Yang. On-site observation shows that the sick goose has poor growth, anorexia, lassitude, dyspnea, acceleration and deepening, shows systemic dystrophy and has weight lower than normal value; the anus is loose and weak in contraction, white semi-viscous water-like excrement is usually discharged, a large amount of off-white urate is contained, and white semi-liquid dilute excrement is arranged around the anus; the sick goose likes lying, does not want to launch and does not want to move; gradually emaciation and weakness, loose and disorderly feathers, loss of hair and lusterless hair, anemia. The goose gout caused by the novel goose astrovirus is confirmed and diagnosed through pathogen separation and identification.
The traditional Chinese medicine injection is used for intramuscular injection twice a day, 2 ml/injection is carried out each time, and the disease condition is controlled on the 3 rd day after the injection is used for continuous use for 2 days; after 5 days, the return visit is carried out, the symptoms disappear, and the goose group returns to normal.
Specific case 3: in 2019, 23 days after 6 months, 40000 geese of 65 days old in a certain goose farm at the mouth of the week are diseased, and the diseased geese are difficult to walk, limp and paralyzed in severe cases and cannot walk. Swelling joints, particularly leg joints, can gradually form hard and obviously contoured movable nodules due to different degrees of deposition of urate in joint cavities, and discharge gray-yellow cheese-like urate nodules and hemorrhagic ulcer after the nodules are broken; the movement is difficult, some goose wings and leg joints are deformed obviously and take the squatting or single-limb standing posture; the food intake is greatly reduced, the weight is reduced, and particularly the sick conical geese and the gosling grow slowly. The goose gout caused by the novel goose astrovirus is confirmed and diagnosed through pathogen separation and identification.
The traditional Chinese medicine injection is used for intramuscular injection twice a day, 5 ml/injection is carried out each time, and the disease condition is controlled on the 2 nd day after the injection is used for 2 days; after 4 days, the symptoms disappeared and the goose flocks recovered to normal.

A compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus is characterized by comprising the following traditional Chinese medicine raw materials in parts by weight: 3-10 parts of scutellaria baicalensis, 3-10 parts of artemisia capillaries, 15-25 parts of pyrrosia lingua, 10-20 parts of gardenia, 10-15 parts of honeysuckle, 35-45 parts of astragalus membranaceus, 5-15 parts of cockroaches, 1-5 parts of radix sophorae flavescentis and 1-5 parts of polygonum aviculare.

The compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus according to claim 1, which is characterized by comprising the following traditional Chinese medicine raw materials in parts by weight: 6 parts of scutellaria baicalensis, 6 parts of artemisia capillaries, 20 parts of pyrrosia lingua, 16 parts of cape jasmine, 12 parts of honeysuckle, 40 parts of astragalus mongholicus, 10 parts of cockroaches, 3 parts of sophora flavescens and 2 parts of polygonum aviculare.
3. The preparation method of the compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus as claimed in claim 1 or 2, which comprises the following steps:
step one, putting honeysuckle into a distillation tank, adding water which is 10 times of the weight of the honeysuckle into the distillation tank, distilling, collecting distillate, adding distilled water which is 1.5 times of the weight of the distillate into the distillate for redistillation, collecting redistillation, placing the redistillation in a refrigeration mode at 0-4 ℃, and preserving honeysuckle residues and aqueous solution after distillation for later use;
step two, respectively crushing cockroaches and polygonum aviculare, uniformly mixing, adding 90-95% ethanol with the weight of 4 times of that of the cockroaches and the polygonum aviculare, soaking for 0.5h, percolating after soaking, collecting percolate, and storing residues after percolating for later use;
putting artemisia capillaries, pyrrosia lingua, astragalus mongholicus, sophora flavescens, gardenia and scutellaria baicalensis into a multifunctional decoction pot, adding water which is 5 times of the total amount of the medicines into the pot, heating to 60 ℃, keeping the temperature for 2 hours at the temperature of 60 ℃, then filtering with gauze, collecting filtrate, and storing filtered residues for later use; carrying out reduced pressure distillation and concentration on the collected filtrate until half of the filtrate is concentrated to obtain a first concentrated solution;
step four, adding water in an amount which is 4 times that of the residues into the residues obtained in the step two and the step three for soaking, combining the residues with the honeysuckle residues and the water solution obtained in the step one, boiling for 0.5h, filtering, collecting the first filtrate, and keeping the filtered residues for later use; adding water 3 times the weight of the medicinal residue into the medicinal residue, boiling for 30min, filtering, collecting the second filtrate, mixing with the first filtrate, mixing, and concentrating under reduced pressure to obtain second concentrated solution;

(1) adding 95% ethanol into the mixed solution, stirring, adjusting ethanol content to 50%, standing for 12 hr after a large amount of flocculent precipitate is separated out, vacuum filtering, collecting filtrate as first refined ethanol solution, recovering ethanol, and concentrating under reduced pressure to half amount;
(2) pouring the concentrated solution obtained in the step (1) into 95% ethanol with the volume of 1.6-2 times of that of the concentrated solution, continuously stirring, adjusting the alcohol content to 60%, precipitating flocculent precipitate, standing for 24h, performing suction filtration, collecting filtrate as second refined alcohol solution, recovering ethanol, and concentrating under reduced pressure to half amount;
(3) slowly pouring the concentrated solution obtained in the step (2) into 95% ethanol with the alcohol content being 3.2 times that of the concentrated solution, continuously stirring, adjusting the alcohol content to 75%, separating out a paste, standing for 24h, performing suction filtration to obtain a filtrate, namely a third refined alcohol solution, recovering ethanol, performing reduced pressure concentration to one third, volatilizing the ethanol completely, and cooling.
5. The preparation method of the compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus according to claim 3, wherein a finished product prepared in the seventh step is a brownish yellow clear solution, and the pH value is 5.0-6.5.
