Abstract
The invention belongs to the technical field of veterinary drugs, and particularly relates to a compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus, which is an injection prepared by processing traditional Chinese medicines through processes of extraction, distillation, filtration, concentration, liquid preparation, sterilization and the like, wherein the raw materials of the traditional Chinese medicine injection comprise 3-6 parts of scutellaria baicalensis, 3-6 parts of artemisia capillaris, 10-20 parts of pyrrosia lingua, 8-16 parts of gardenia, 6-12 parts of honeysuckle, 20-40 parts of astragalus mongholicus, 5-10 parts of cockroaches, 1-3 parts of radix sophorae flavescentis and 1-2 parts of polygonum aviculare. The preparation is a pure traditional Chinese medicine preparation, has no antibiotic residue, and has a good treatment effect on goose gout caused by the novel goose astrovirus.

Technical Field
The invention belongs to the technical field of veterinary drugs, and particularly relates to a compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus and a preparation method thereof.
Background
The new goose astrovirus is a tiny and circular virus, and the diameter of the new goose astrovirus is 25-35 nm. Under an electron microscope, the virus has 5 or 6 star-shaped protrusions on the surface, so the virus is called as a star virus. The astrovirus is a non-capsular membrane positive strand RNA virus, can cause gosling gout, and is mainly characterized in that urate and urate crystals are deposited in organs such as viscera, ureter, kidney, joint cavity and the like; the initial diagnosis can be performed according to the age of the disease and clinical symptoms, and the molecular diagnosis is required for the accurate diagnosis.
At present, the disease is widely prevalent in the world, which causes great economic loss to the goose farming industry in the world, and the occurrence and prevalence of the disease become serious day by day. In recent years, new goose astrovirus has become an important disease affecting the development of the goose industry. After the disease occurs, the mortality of animals can be obviously reduced by adopting antibiotics for treatment, and the incidence of the disease can be reduced to a certain extent by properly adding the antibiotics in the production process; however, with the wide use of a large number of antibacterial drugs, particularly broad-spectrum antibacterial drugs, in the breeding industry, the problem of drug resistance of bacteria to antibacterial drugs is more and more prominent, and meanwhile, research data shows that antibiotics with sub-inhibitory concentration can influence the metabolic process of bacteria to change the toxicity of bacteria or the adaptability of bacteria to the environment and the like. There have been no effective vaccines, chemotherapeutic drugs or other methods reported to date to control and prevent astrovirus infection, which has been a problem that continues to plague the goose industry.
The early stage of the disease is effectively treated by antibiotics, the death can be reduced, common intramuscular or subcutaneous injection needs large dose repeated administration, but the drug resistance is easy to generate, serious drug residue is caused, and the food safety is influenced, so that a drug which can treat goose gout and does not generate antibiotic residue is needed.
Disclosure of Invention
Aiming at the problem that antibiotic residues in meat and eggs are easily caused by the fact that goose gout is treated by antibiotic, the invention provides a non-residue traditional Chinese medicine injection for treating goose gout and a preparation method thereof.
The technical scheme adopted by the invention for solving the technical problems is as follows: a compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus comprises the following traditional Chinese medicine raw materials in parts by weight: 3-10 parts of scutellaria baicalensis, 3-10 parts of artemisia capillaries, 15-25 parts of pyrrosia lingua, 10-20 parts of gardenia, 10-15 parts of honeysuckle, 35-45 parts of astragalus membranaceus, 5-15 parts of cockroaches, 1-5 parts of radix sophorae flavescentis and 1-5 parts of polygonum aviculare.
The compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus comprises the following traditional Chinese medicine raw materials in parts by weight: 6 parts of scutellaria baicalensis, 6 parts of artemisia capillaries, 20 parts of pyrrosia lingua, 16 parts of cape jasmine, 12 parts of honeysuckle, 40 parts of astragalus mongholicus, 10 parts of cockroaches, 3 parts of sophora flavescens and 2 parts of polygonum aviculare.
The invention also provides a preparation method of the compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus, which comprises the following steps:
step one, putting honeysuckle into a distillation tank, adding water which is 10 times of the weight of the honeysuckle into the distillation tank, distilling, collecting distillate, adding distilled water which is 1.5 times of the weight of the distillate into the distillate for redistillation, collecting redistillation, placing the redistillation in a refrigeration mode at 0-4 ℃, and preserving honeysuckle residues and aqueous solution after distillation for later use;
step two, respectively crushing cockroaches and polygonum aviculare, uniformly mixing, adding 90-95% ethanol with the weight of 4 times of that of the cockroaches and the polygonum aviculare, soaking for 0.5h, percolating after soaking, collecting percolate, and storing residues after percolating for later use;
putting artemisia capillaries, pyrrosia lingua, astragalus mongholicus, sophora flavescens, gardenia and scutellaria baicalensis into a multifunctional decoction pot, adding water which is 5 times of the total amount of the medicines into the pot, heating to 60 ℃, keeping the temperature for 2 hours at the temperature of 60 ℃, then filtering with gauze, collecting filtrate, and storing filtered residues for later use; carrying out reduced pressure distillation and concentration on the collected filtrate until half of the filtrate is concentrated to obtain a first concentrated solution;
step four, adding water in an amount which is 4 times that of the residues into the residues obtained in the step two and the step three for soaking, combining the residues with the honeysuckle residues and the water solution obtained in the step one, boiling for 0.5h, filtering, collecting the first filtrate, and keeping the filtered residues for later use; adding water 3 times the weight of the medicinal residue into the medicinal residue, boiling for 30min, filtering, collecting the second filtrate, mixing with the first filtrate, mixing, and concentrating under reduced pressure to obtain second concentrated solution;
step five, mixing the concentrated solution I and the concentrated solution II, concentrating the mixture into concentrated juice under reduced pressure, cooling the concentrated juice, pouring the concentrated juice into the cockroach percolate to obtain mixed solution, extracting the mixed solution with alcohol, and concentrating the mixed solution under reduced pressure to one third to obtain pure concentrated liquid medicine;
adding 1 time of distilled water into the pure concentrated liquid medicine, uniformly stirring, adding 0.5% of activated carbon, freezing and standing for 24 hours, performing suction filtration by using a No. 3 filter stick or filter paper pulp, and evaporating and concentrating the filtrate to one fifth;
and seventhly, adding honeysuckle distillate into the concentrated solution obtained in the sixth step, adding 1N sodium hydroxide solution to adjust the pH to about 7, adding 0.1% bone charcoal, sealing, insulating water and heating for 2 hours, adding 1N sodium hydroxide solution after cooling to adjust the pH to 6.5-7.0, freezing and placing for 48 hours, performing suction filtration by using a No. 3 filter stick or filter paper pulp, adding 2% benzyl alcohol into the filtrate obtained by suction filtration, uniformly stirring, freezing and placing for 24 hours, performing suction filtration by using a No. 4 filter stick, filling and sealing the filtrate, and sterilizing to obtain a finished product.
The preparation method of the compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus comprises the following steps:
(1) adding 95% ethanol into the mixed solution, stirring, adjusting ethanol content to 50%, standing for 12 hr after a large amount of flocculent precipitate is separated out, vacuum filtering, collecting filtrate as first refined ethanol solution, recovering ethanol, and concentrating under reduced pressure to half amount;
(2) pouring the concentrated solution obtained in the step (1) into 95% ethanol with the volume of 1.6-2 times of that of the concentrated solution, continuously stirring, adjusting the alcohol content to 60%, precipitating flocculent precipitate, standing for 24h, performing suction filtration, collecting filtrate as second refined alcohol solution, recovering ethanol, and concentrating under reduced pressure to half amount;
(3) slowly pouring the concentrated solution obtained in the step (2) into 95% ethanol with the alcohol content being 3.2 times that of the concentrated solution, continuously stirring, adjusting the alcohol content to 75%, separating out a paste, standing for 24h, performing suction filtration to obtain a filtrate, namely a third refined alcohol solution, recovering ethanol, performing reduced pressure concentration to one third, volatilizing the ethanol completely, and cooling.
According to the preparation method of the compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus, the finished product prepared in the seventh step is a brown yellow clear solution, and the pH value is 5.0-6.5.
The scutellaria in the formula is bitter in taste and cold in nature, is a monarch drug in the formula, has the effects of clearing heat, drying dampness, purging intense heat and detoxifying, and can be used for treating symptoms such as chest distress, nausea and vomiting, diarrhea, lung heat cough and the like.
Herba Artemisiae Scopariae, bitter and pungent in taste, slightly cold in nature, enter spleen, stomach, liver, gallbladder channel, and serve as the ministerial drugs of the formula together with folium Pyrrosiae and fructus Gardeniae, has effects of clearing heat and promoting diuresis, promoting bile flow and removing jaundice, and clearing heat and protecting liver, and can be used for treating rheumatism, cold and heat, jaundice, and protecting liver and gallbladder when being combined with fructus Gardeniae.
Pyrrosia lingua, sweet and bitter in taste and slightly cold in nature, enters lung and bladder channels, has the effects of promoting urination, clearing lung heat, relieving cough, cooling blood, stopping bleeding, relieving cough, eliminating phlegm, inhibiting bacteria and resisting inflammation.
Gardenia, bitter in taste and cold in nature, enters heart, lung and triple energizer channels, has the effects of clearing heat and promoting urination, cooling blood and dissipating blood stasis, and can purge fire and remove toxicity when being combined with radix scutellariae and honeysuckle.
The honeysuckle flower is sweet in taste and cold in nature, and has the effects of clearing away heat and toxic materials, diminishing inflammation and relieving swelling.
Astragalus root, radix astragali, sweet in taste and slightly warm in nature, enters spleen and lung channels, and is used together with cockroach in the formula to tonify qi, strengthen superficies, promote urination and promote tissue regeneration, and is used for treating symptoms such as qi deficiency, hypodynamia, chronic diarrhea and rectocele.
The cockroach is salty in taste and cold in nature, enters liver, spleen and kidney channels, and has the effects of promoting blood circulation to remove blood stasis, detoxifying and diminishing swelling, and resolving masses and removing food retention.
Kuh-seng, bitter in taste and cold in nature, enters heart, liver, stomach, large intestine and bladder meridians, and is used together with Biao knotgrass to clear heat and dry dampness, kill parasites and induce diuresis.
The herba polygoni avicularis is bitter in taste and cold in nature, enters bladder channels, and has the effects of promoting urination, treating stranguria, killing parasites and relieving itching.
The invention has the beneficial effects that: the invention is a pure Chinese medicinal preparation, has reasonable compatibility and obvious medicinal effect, and can be used as a special injection preparation for preventing and treating goose gout caused by novel goose astrovirus. The injection has effects of clearing heat, eliminating dampness, inducing diuresis and relieving edema, and has good therapeutic effect on goose gout caused by new goose astrovirus; particularly has obvious effects on the swelling, the broken walking and the difficult walking of the joint type goose toe and leg joint and the visceral gout. Compared with antibiotic medicines, the traditional Chinese medicine composition has the characteristics of pure nature, preventive treatment, no toxic or side effect, no residue, no drug resistance, complete cure and the like, is reasonable in compatibility, safe and effective, treats both principal and secondary aspect of disease, has no recurrence, avoids drug residue for prevention and treatment by antibiotics, ensures food safety, and is suitable for popularization in animal production. Has better curative effect on preventing and treating goose gout caused by the novel goose astrovirus, and has wide market prospect and larger economic benefit.
Detailed Description
In order to well prevent and control the novel goose astrovirus and solve the hidden danger of antibiotic residue caused by the prevention and treatment of the antibiotic, the invention provides a compound traditional Chinese medicine injection for the goose gout caused by the novel goose astrovirus according to the traditional Chinese medicine theory, and the injection can effectively control the goose gout caused by the novel goose astrovirus. The present invention is further illustrated below with reference to examples to better understand the technical solutions of the present invention, and it should be noted that: in the following examples, 10g of each Chinese medicinal material is 1 part in selected amount.
Example 1: the embodiment provides a compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus, which comprises the following raw materials by weight, 60g of scutellaria baicalensis, 60g of artemisia capillaris, 200g of pyrrosia lingua, 160g of gardenia, 120g of honeysuckle, 400g of astragalus mongholicus, 100g of cockroach, 30g of sophora flavescens and 20g of polygonum aviculare.
The preparation method comprises the following steps:
step one, putting 120g of honeysuckle into a distillation retort, adding 1200mL of water into the retort, distilling, collecting distillate, adding 1.5 times of distilled water into the distillate for redistillation, collecting redistillate, refrigerating the redistillate at 0-4 ℃, and preserving distilled honeysuckle residues and aqueous solution for later use;
step two, respectively crushing cockroaches and polygonum aviculare, uniformly mixing, adding 90-95% ethanol with the weight of 4 times of that of the cockroaches and the polygonum aviculare, soaking for 0.5h, percolating after soaking, collecting percolate, and storing residues after percolating for later use;

step four, adding water in an amount which is 4 times that of the residue into the residue obtained in the step two and the residue obtained in the step three for soaking, combining the residue with the honeysuckle residue and the water solution obtained in the step one, boiling for 30min, filtering, and collecting the first filtrate; adding water 3 times the weight of the medicinal residue into the medicinal residue, boiling for 30min, filtering, collecting the second filtrate, mixing with the first filtrate, mixing, and concentrating under reduced pressure to obtain second concentrated solution;
step five, combining the concentrated solution I and the concentrated solution II, concentrating the mixture into thick juice under reduced pressure, cooling the thick juice, pouring the mixture into cockroach percolate to obtain mixed solution, adding 95% ethanol into the mixed solution, continuously stirring, adjusting the ethanol content to be 50%, standing for 12h after a large amount of flocculent precipitates are separated out, performing suction filtration, collecting filtrate as first refined alcohol solution, recovering ethanol, and concentrating the filtrate under reduced pressure to half amount;
step six, pouring the concentrated solution obtained in the step five into 95% ethanol which is 1.6-2 times of the concentrated solution, continuously stirring, adjusting the alcohol content to 60%, separating out flocculent precipitate, standing for 24 hours, performing suction filtration, collecting filtrate as second refined alcohol solution, recovering ethanol, and concentrating under reduced pressure to half amount;
and step seven, slowly pouring the concentrated solution obtained in the step six into 95% ethanol in an amount which is 3.2 times that of the concentrated solution, continuously stirring, adjusting the alcohol content to 75%, separating out a paste, standing for 24 hours, performing suction filtration to obtain a filtrate, namely a third refined alcohol solution, recovering ethanol, performing reduced pressure concentration to one third, volatilizing the ethanol completely, and cooling.
Step eight, adding 1 time of distilled water into the pure concentrated liquid medicine, uniformly stirring, adding 0.5% of active carbon, freezing and standing for 24 hours, performing suction filtration by using a No. 3 filter stick or filter paper pulp, and evaporating and concentrating the filtrate to one fifth of the original medicine of the preparation;
and step nine, adding honeysuckle distillate into the concentrated solution, adding 1N sodium hydroxide solution to adjust the pH to about 7, adding 0.1% bone charcoal, sealing, insulating water and heating for 2 hours, cooling, measuring the pH to about 5.5, adding 1N sodium hydroxide solution, adjusting the pH to 6.5-7.0, freezing and placing for 48 hours, performing suction filtration by using a No. 3 filter stick or filter paper pulp, adding 2% benzyl alcohol into the filtrate, uniformly stirring, freezing and placing for 24 hours, performing suction filtration by using a No. 4 filter stick, filling and sealing the filtrate, and sterilizing to obtain a finished product, wherein the finished product is a brown yellow clear solution, and the pH is 5.0-6.5.
Example 2: the embodiment provides a compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus, which comprises the following raw materials, by weight, 30g of scutellaria baicalensis, 100g of artemisia capillaries, 150g of pyrrosia lingua, 200g of gardenia, 100g of honeysuckle, 450g of astragalus mongholicus, 150g of cockroaches, 50g of sophora flavescens and 10g of polygonum aviculare.
The preparation method comprises the following steps:
step one, putting honeysuckle into a distillation tank, adding water which is 10 times of the weight of the honeysuckle into the distillation tank, distilling, collecting distillate, adding distilled water which is 1.5 times of the weight of the distillate into the distillate for redistillation, collecting redistillation, placing the redistillation in a refrigeration mode at 0-4 ℃, and preserving honeysuckle residues and aqueous solution after distillation for later use;

step four, adding water in an amount which is 4 times of the total amount of the residues into the residues obtained in the step two and the step three, soaking, combining the residues with the honeysuckle residues and the water solution obtained in the step one, boiling for 30min, filtering, and collecting primary filtrate; adding water 3 times the weight of the medicinal residue into the medicinal residue, boiling for 30min, filtering, collecting the second filtrate, mixing with the first filtrate, mixing, and concentrating under reduced pressure to obtain second concentrated solution;

and step nine, adding honeysuckle distillate into the concentrated solution, adding 1N sodium hydroxide solution to adjust the pH to about 7, adding 0.1% bone charcoal, sealing, insulating water and heating for 2 hours, cooling, measuring the pH to about 5.5, adding 1N sodium hydroxide solution, adjusting the pH to 6.5-7.0, freezing and placing for 48 hours, performing suction filtration by using a No. 3 filter stick or filter paper pulp, adding 2% benzyl alcohol into the filtrate, uniformly stirring, freezing and placing for 24 hours, performing suction filtration by using a No. 4 filter stick, filling and sealing the filtrate, and sterilizing to obtain a finished product, wherein the finished product is a brown yellow clear solution, and the pH is 5.0-6.5.
Example 3: the same parts of this embodiment as those of embodiment 1 are not repeated, except that the traditional Chinese medicine injection comprises the following raw materials, by weight, 100g of scutellaria baicalensis, 30g of artemisia capillaries, 250g of pyrrosia lingua, 100g of gardenia, 150g of honeysuckle, 350g of astragalus mongholicus, 50g of cockroach, 10g of sophora flavescens and 50g of polygonum aviculare.
Experimental example: in order to demonstrate the effectiveness of the injectable preparation of the invention, the applicant carried out the following tests:
selecting 20 diseased goslings and 20 diseased adult geese, randomly dividing the test geese into 2 groups, namely a test group and a control group, and ensuring that each group comprises 10 goslings and 10 adult geese.
The experimental group injects the injection according to the method of injecting 2ml per young goose and injecting 5ml per adult goose; the control group was injected with physiological saline according to the method of injecting 2 ml/young goose and 5 ml/adult goose. Blood is collected before and after administration, and the concentration of urate in goose serum is measured before and after administration, and the measurement results are shown in table 1.
As can be seen from Table 1, there was no significant difference in the concentrations of urate in the blood serum of the diseased goose in the test group and the control group before administration, and the concentrations of urate in the blood serum of the diseased goose were both high; after the injection is used, the concentration of the urate in the serum of the diseased goose in a test group is obviously reduced, a control group is not obviously changed, and the test group is obviously reduced compared with the control group (P is less than 0.05), which shows that the injection has good treatment effect on the goose gout.

A compound traditional Chinese medicine injection for treating goose gout caused by novel goose astrovirus is characterized by comprising the following traditional Chinese medicine raw materials in parts by weight: 3-10 parts of scutellaria baicalensis, 3-10 parts of artemisia capillaries, 15-25 parts of pyrrosia lingua, 10-20 parts of gardenia, 10-15 parts of honeysuckle, 35-45 parts of astragalus membranaceus, 5-15 parts of cockroaches, 1-5 parts of radix sophorae flavescentis and 1-5 parts of polygonum aviculare.

The compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus according to claim 1, which is characterized by comprising the following traditional Chinese medicine raw materials in parts by weight: 6 parts of scutellaria baicalensis, 6 parts of artemisia capillaries, 20 parts of pyrrosia lingua, 16 parts of cape jasmine, 12 parts of honeysuckle, 40 parts of astragalus mongholicus, 10 parts of cockroaches, 3 parts of sophora flavescens and 2 parts of polygonum aviculare.
3. The preparation method of the compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus as claimed in claim 1 or 2, which comprises the following steps:
step one, putting honeysuckle into a distillation tank, adding water which is 10 times of the weight of the honeysuckle into the distillation tank, distilling, collecting distillate, adding distilled water which is 1.5 times of the weight of the distillate into the distillate for redistillation, collecting redistillation, placing the redistillation in a refrigeration mode at 0-4 ℃, and preserving honeysuckle residues and aqueous solution after distillation for later use;
step two, respectively crushing cockroaches and polygonum aviculare, uniformly mixing, adding 90-95% ethanol with the weight of 4 times of that of the cockroaches and the polygonum aviculare, soaking for 0.5h, percolating after soaking, collecting percolate, and storing residues after percolating for later use;
putting artemisia capillaries, pyrrosia lingua, astragalus mongholicus, sophora flavescens, gardenia and scutellaria baicalensis into a multifunctional decoction pot, adding water which is 5 times of the total amount of the medicines into the pot, heating to 60 ℃, keeping the temperature for 2 hours at the temperature of 60 ℃, then filtering with gauze, collecting filtrate, and storing filtered residues for later use; carrying out reduced pressure distillation and concentration on the collected filtrate until half of the filtrate is concentrated to obtain a first concentrated solution;
step four, adding water in an amount which is 4 times that of the residues into the residues obtained in the step two and the step three for soaking, combining the residues with the honeysuckle residues and the water solution obtained in the step one, boiling for 0.5h, filtering, collecting the first filtrate, and keeping the filtered residues for later use; adding water 3 times the weight of the medicinal residue into the medicinal residue, boiling for 30min, filtering, collecting the second filtrate, mixing with the first filtrate, mixing, and concentrating under reduced pressure to obtain second concentrated solution;
step five, mixing the concentrated solution I and the concentrated solution II, concentrating the mixture into concentrated juice under reduced pressure, cooling the concentrated juice, pouring the concentrated juice into the cockroach percolate to obtain mixed solution, extracting the mixed solution with alcohol, and concentrating the mixed solution under reduced pressure to one third to obtain pure concentrated liquid medicine;
adding 1 time of distilled water into the pure concentrated liquid medicine, uniformly stirring, adding 0.5% of activated carbon, freezing and standing for 24 hours, performing suction filtration by using a No. 3 filter stick or filter paper pulp, and evaporating and concentrating the filtrate to one fifth;
seventhly, adding honeysuckle flower distillate into the concentrated solution obtained in the sixth step, adding 1N sodium hydroxide solution to adjust the pH to about 7, adding 0.1% bone charcoal, sealing, heating in a water-proof manner for 2 hours, cooling, adding 1N sodium hydroxide solution, adjusting the pH to 6.5-7.0, standing for 48 hours in a freezing manner, performing suction filtration by using a No. 3 filter stick or filter paper pulp, adding 2% benzyl alcohol into the filtrate, stirring uniformly, standing for 24 hours in a freezing manner, performing suction filtration by using a No. four filter stick, filling and sealing the filtrate, and sterilizing to obtain a finished product.

(1) adding 95% ethanol into the mixed solution, stirring, adjusting ethanol content to 50%, standing for 12 hr after a large amount of flocculent precipitate is separated out, vacuum filtering, collecting filtrate as first refined ethanol solution, recovering ethanol, and concentrating under reduced pressure to half amount;
(2) pouring the concentrated solution obtained in the step (1) into 95% ethanol with the volume of 1.6-2 times of that of the concentrated solution, continuously stirring, adjusting the alcohol content to 60%, precipitating flocculent precipitate, standing for 24h, performing suction filtration, collecting filtrate as second refined alcohol solution, recovering ethanol, and concentrating under reduced pressure to half amount;
(3) slowly pouring the concentrated solution obtained in the step (2) into 95% ethanol with the alcohol content being 3.2 times that of the concentrated solution, continuously stirring, adjusting the alcohol content to 75%, separating out a paste, standing for 24h, performing suction filtration to obtain a filtrate, namely a third refined alcohol solution, recovering ethanol, performing reduced pressure concentration to one third, volatilizing the ethanol completely, and cooling.
5. The preparation method of the compound traditional Chinese medicine injection for treating goose gout caused by the novel goose astrovirus according to claim 3, wherein a finished product prepared in the seventh step is a brownish yellow clear solution, and the pH value is 5.0-6.5.
