Abstract
本发明公开了一种抗菌中草药复方制剂和制备方法及洗手液,洗手液包括有抗菌中草药复方制剂,该抗菌中草药复方制剂包括如下重量份数配比:荆芥穗15‑35份、连翘2‑10份、桔梗5‑10份、牛蒡子10‑20份、甘草8‑15份,并通过提取纯化的制备方法得到,以中草药荆芥穗、连翘、桔梗、牛蒡子、甘草植物萃取液和表面活性剂、增效剂等混合的抗菌洗手液,使中草药和其它活性成分优化组合,利于其协同效应抗菌,使得洗手液不但对普通致病菌金黄色葡萄球菌和大肠杆菌具有较好杀灭效果,而且该洗手液对耐药菌耐甲氧西林葡萄球菌具有较好的抗菌活性,且香气淡雅,无需再添加香精,降低了成本。

技术领域
本发明涉及日用品洗手液领域,尤其涉及一种抗菌中草药复方制剂和制备方法及洗手液。
背景技术
日常生活中,特别是春季和夏季,随着环境湿度的增大,细菌和病毒容易滋生,并通过各种传播渠道传播至人体皮肤上,如果不对其进行及时处理,会引发皮肤类疾病,而科学研究表明大部分细菌病毒感染都是通过手部接触或手部与口,鼻接触而发生,随着人们对健康意识的提高,手部卫生也得到了越来越多的关注,顺应这一趋势,各种抑菌洗手液已经成为市场的主流产品,大部分的抑菌洗手液中的抗菌剂还是来自于化学原料,例如三氯生,三氯卡班,对氯间二甲苯酚等。这些化学杀菌剂虽然杀菌力较好,但是也具有一些隐患,比如对皮肤的刺激,以及清洗不彻底而进入口部的风险。所以,迫切需要寻找一种安全无毒的抗菌护肤的洗手液。
现有技术中的中草药抗菌洗手液,用到的中药成分较单一,且制备得到的中草药抗菌洗手液具有杀菌效果及去污、滋润、保湿和护手的作用,但是洗手液杀菌功能主要限于普通致病菌,并没有显示对耐药菌比如耐甲氧西林葡萄球菌的杀菌效果,而像耐甲氧西林葡萄球菌是一种超级细菌,从传染病防控角度来看,像户外可能是超级细菌和其他耐药菌的传染源之一,而一旦被感染,对免疫力较差的人威胁比较大,且目前对耐甲氧西林葡萄球菌的治疗非常麻烦,因此,需要从防止感染方面去防止这种情况的发生,进而对洗手液应当具有特殊的要求。
发明内容
针对上述技术中存在的不足之处,本发明提供一种抗菌中草药复方制剂和制备方法及洗手液,不但对普通病菌有较好杀灭效果,而且对耐药球菌也具有很好的抗菌效果。
为实现上述目的,本发明提供一种抗菌中草药复方制剂,包括如下重量份数配比:荆芥穗15-35份、连翘2-10份、桔梗5-10份、牛蒡子10-20份、甘草8-15份。
其中,包括如下重量份数配比:荆芥穗20份、连翘8份、桔梗8份、牛蒡子16份、甘草8份。
本发明还提供一种抗菌中草药复方制剂的制备方法,用于制备上面所述的抗菌中草药复方制剂,包括以下步骤:
(1)将荆芥穗和连翘混合粉碎,采用水蒸气蒸馏法提取荆芥穗和连翘混合油;
(2)将桔梗,牛蒡子,甘草药材混合粉碎,加入6-9体积倍水浸泡,热回流提取2次,过滤后合并两次滤液,浓缩到提取液体积的十分之一,进行醇沉、离心后得到上清液,将上清液浓缩至比重为1.1-1.5,通过大孔树脂柱和乙醇洗脱,得到洗脱液,减压回收乙醇,浓缩至比重1.1-1.5,得到桔梗、牛蒡子和甘草混合提取物;
(3)将步骤(1)所得荆芥穗和连翘混合油,加入等体积的RH60和丙二醇,搅拌均匀,与步骤(2)所得的桔梗、牛蒡子和甘草混合提取物混合均匀,即得抗菌复方制剂。
其中,在步骤(1)中的荆芥穗和连翘粉碎的粒径和步骤(2)中的桔梗、牛蒡子、甘草药材混合粉碎的粒径均为60-200目。
其中,在步骤(1)中,所述水蒸气蒸馏法的工艺条件为:料液比1:10-1:20,提取温度为85-100℃,提取时间为1-5h。
其中,在步骤(2)中,热回流2次的工艺条件为:热回流的温度为85-100℃,每次回流时间为每次1.5小时。
其中,步骤(2)中醇沉,离心的条件是加入1-5倍体积量的95%乙醇,醇沉,离心。
其中,步骤(2)中,通过大孔树脂柱和乙醇洗脱,得到洗脱液的具体过程为:通过处理好的大孔树脂柱,分别以三个柱体积蒸馏水和20%乙醇洗脱,弃去,然后以50%-70%乙醇洗脱,得到洗脱液。
本发明还公开一种洗手液,包括上面所述的抗菌中草药复方制剂。

本发明的有益效果是:与现有技术相比,本发明提供的抗菌中草药复方制剂和制备方法及洗手液,以中草药荆芥穗、连翘、桔梗、牛蒡子、甘草植物萃取液按一定重量份数比制得的抗菌的复方制剂,且将抗菌复方制剂和表面活性剂、增效剂等混合的抗菌洗手液,使中草药和其它活性成分优化组合,利于其协同效应抗菌,使之在用量极低的情况下,在满足消费者实际洗涤条件的同时,达到去污、消毒的效果,不但对普通致病菌金黄色葡萄球菌和大肠杆菌具有较好杀灭效果,而且对耐药菌耐甲氧西林葡萄球菌也具有很好的抗菌效果。
具体实施方式
为了更清楚地表述本发明,本发明由多种具有抗菌功效的活性物质的天然中草药配伍制成,绿色健康,药理明确,安全无毒副作用,下面结合具体实施例来说明对本发明抗菌的效果。
本发明提供一种抗菌中草药复方制剂,该抗菌中草药复方制剂包括如下重量份数配比:荆芥穗15-35份、连翘2-10份、桔梗5-10份、牛蒡子10-20份、甘草8-15份,经过提取纯化的制备方法得到。
本发明实施例提供的抗菌中草药复方制剂,经过提取纯化而成。这五种药材是许多传统治疗呼吸道感染的药方的组成成分,方中荆芥穗,连翘芳香解表,牛蒡子和桔梗清热解毒,甘草调和诸药。现代药理研究证实荆芥穗中的胡薄荷酮,连翘中的连翘苷,牛蒡子中的牛蒡苷,桔梗中的桔梗皂苷,甘草中的甘草次酸都具有一定的抗菌消炎作用,进一步说明了本方得合理性。因为该复方制剂中荆芥穗油和连翘油是主要的抗菌成分和香气基础,因此该复方制剂在制备过程中采取先单独收集荆芥穗油和连翘油的方法,避免有效成分在后续提取纯化中损失破坏。
其中,荆芥(Schizonepeta tenuifolia Briq.),唇形科荆芥属多年生草本植物,荆芥穗为荆芥的花穗,始载于《神农本草经》,民间用于治疗祛风、解表、透疹和止血。近年来研究表明,荆芥穗具有解热降温、镇静、镇痛、抗炎、止血、抑制心肌收缩、祛痰平喘、抗氧化等药理作用。荆芥穗中含有黄酮类、酚酸类及挥发油等活性成分,挥发油中主要含有单萜类成分,以胡薄荷酮和薄荷酮化合物,这些化合物均具有特殊的香气。
连翘为木樨科连翘属植物[Forsythia suspensa(Thunb.)Vahl],连翘的干燥果实作为中药连翘(Forsythiae Fructus)使用,其主要功效为清热,解毒,散结,消肿等。治温热,丹毒,斑疹,痈疡肿毒,瘰疬,小便淋闭等。连翘挥发油是其主要抗菌功效成分。
牛蒡子(Fructus Arctii)为菊科(Compositae)植物牛蒡的干燥成熟果实。中医学将牛蒡种子称“牛蒡子”或“大力子”。牛蒡子属常用中药,具有疏散风热、宣肺透疹、解毒利咽的功能,用于风热感冒,咳嗽痰多,麻疹,风疹,咽喉肿痛,作腮丹毒,痈肿疮毒。
中药桔梗为桔梗科多年生草本植物桔梗(Platycodon grandiflorus)的根;苦、辛,平,微寒。归肺经。有止咳祛痰、宣肺、排脓等作用。为药食两用品种,在中国东北地区常被腌制为咸菜,在朝鲜半岛被用来制作泡菜。现代药理研究表明桔梗中的桔梗皂苷和黄酮类具有较好的抗菌作用。
甘草(Glycyrrhiza uralensis Fisch),豆科多年生草本植物。性平,味甘,为脾胃之良药,生用偏凉,能清热解毒,炙用性温,能益气补虚,其甘缓之性,又能缓急止痛,故可用治脾胃气虚、中气不足、气虚血少、心中动悸、痰嗽咳喘、腹痛挛急以及疮疡肿毒诸证。由于又能调和药性,解百药之毒,故为中医上使用最广的一种药物。
作为优选地,本发明通过采用如下提取方法,获取抗菌中草药复方制剂,包括以下步骤:
(1)将荆芥穗和连翘混合粉碎,采用水蒸气蒸馏法提取荆芥穗和连翘混合油;
(2)将桔梗,牛蒡子,甘草药材混合粉碎,加入6-9体积倍水浸泡,热回流提取2次,过滤后合并两次滤液,浓缩到提取液体积的十分之一,进行醇沉、离心后得到上清液,将上清液浓缩至比重为1.1-1.5,通过大孔树脂柱和乙醇洗脱,得到洗脱液,减压回收乙醇,浓缩至比重1.1-1.5,得到桔梗、牛蒡子和甘草混合提取物;
(3)将步骤(1)所得荆芥穗和连翘混合油,加入等体积的RH60和丙二醇,搅拌均匀,与步骤(2)所得的桔梗、牛蒡子和甘草混合提取物混合均匀,即得抗菌复方制剂。
其中,在步骤(1)中的荆芥穗和连翘粉碎的粒径和步骤(2)中的桔梗、牛蒡子、甘草药材混合粉碎的粒径均为60-200目;在步骤(1)中,所述水蒸气蒸馏法的工艺条件为:料液比1:10-1:20,提取温度为85-100℃,提取时间为1-5h;其中,在步骤(2)中,热回流2次的工艺条件为:热回流的温度为85-100℃,每次回流时间为每次1.5小时;其中,步骤(2)中醇沉,离心的条件是加入1-5倍体积量的95%乙醇,醇沉,离心;其中,步骤(2)中,通过大孔树脂柱和乙醇洗脱,得到洗脱液的具体过程为:通过处理好的大孔树脂柱,分别以三个柱体积蒸馏水和20%乙醇洗脱,弃去,然后以50%-70%乙醇洗脱,得到洗脱液。将荆芥穗和连翘的挥发油与其它成分分开收集,避免了挥发油长时间提取被损失和破坏,另外用大孔树脂柱纯化条件比较温和,不需要高温也不需要使用有毒试剂,制备方法简单,且使提取的有效成分达到最优。
下面,通过具体实施例说明本发明的抗菌中草药复方制剂在洗手液中的有益作用:
实施例1
(1)称取5重量份荆芥穗和1重量份连翘粉碎至150目,水蒸气蒸馏法在料液比1:15,提取温度为90℃,提取4h,得到荆芥穗油和连翘油;
(2)称取2重量份桔梗,4重量份牛蒡子,2重量份甘草药材粉碎至150目,加入6-9倍量水浸泡1-1.5小时后,在90℃热回流提取2次,每次1.5小时,滤过,合并两次滤液,浓缩到提取液体积的十分之一,加入3倍体积量的95%乙醇,醇沉,离心,上清液浓缩至比重1.3,通过处理好的大孔树脂柱,分别以三个柱体积蒸馏水和20%乙醇洗脱,弃去,然后以50%-70%乙醇洗脱,收集洗脱液,减压回收乙醇,浓缩至比重为1.2,得到桔梗、牛蒡子和甘草的混合提取物;
(3)将步骤(1)所得荆芥穗油和连翘油,加入等体积的RH60和丙二醇,搅拌均匀,与步骤(2)制得的桔梗、牛蒡子和甘草的混合提取物混合均匀,即得抗菌复方制剂1。
实施例2
(1)称取5重量份荆芥穗和2重量份连翘粉碎至150目,水蒸气蒸馏法在料液比1:15,提取温度为90℃,提取4h,得到荆芥穗油和连翘油;
(2)称取2重量份桔梗,4重量份牛蒡子,2重量份甘草药材粉碎至150目,加入6-9倍量水浸泡1-1.5小时后,在90℃热回流提取2次,每次1.5小时,滤过,合并两次滤液,浓缩到提取液体积的十分之一,加入3倍体积量的95%乙醇,醇沉,离心,上清液浓缩至比重1.3,通过处理好的大孔树脂柱,分别以三个柱体积蒸馏水和20%乙醇洗脱,弃去,然后以50%-70%乙醇洗脱,收集洗脱液,减压回收乙醇,浓缩至比重为1.2,得到桔梗、牛蒡子和甘草的混合提取物;
(3)将步骤(1)所得荆芥穗油和连翘油,加入等体积的RH60和丙二醇,搅拌均匀,与步骤(2)制得的桔梗、牛蒡子和甘草的混合提取物混合均匀,即得抗菌复方制剂2。
实施例3
(1)称取5重量份荆芥穗和3重量份连翘粉碎至150目,水蒸气蒸馏法在料液比1:15,提取温度为90℃,提取4h,得到荆芥穗油和连翘油;
(2)称取2重量份桔梗,4重量份牛蒡子,2重量份甘草药材粉碎至150目,加入6-9倍量水浸泡1-1.5小时后,在90℃热回流提取2次,每次1.5小时,滤过,合并两次滤液,浓缩到提取液体积的十分之一,加入3倍体积量的95%乙醇,醇沉,离心,上清液浓缩至比重1.3,通过处理好的大孔树脂柱,分别以三个柱体积蒸馏水和20%乙醇洗脱,弃去,然后以50%-70%乙醇洗脱,收集洗脱液,减压回收乙醇,浓缩至比重为1.2,得到桔梗、牛蒡子和甘草的混合提取物;
(3)将步骤(1)所得荆芥穗油和连翘油,加入等体积的RH60和丙二醇,搅拌均匀,与步骤(2)得到桔梗、牛蒡子和甘草的混合提取物混合均匀,即得抗菌复方制剂3。
实施例4
(1)称取1重量份荆芥穗和3重量份连翘粉碎至150目,水蒸气蒸馏法在料液比1:15,提取温度为90℃,提取4h,得到荆芥穗油和连翘油;
(2)称取2重量份桔梗,4重量份牛蒡子,2重量份甘草药材粉碎至150目,加入6-9倍量水浸泡1-1.5小时后,在90℃热回流提取2次,每次1.5小时,滤过,合并两次滤液,浓缩到提取液体积的十分之一,加入3倍体积量的95%乙醇,醇沉,离心,上清液浓缩至比重1.3,通过处理好的大孔树脂柱,分别以三个柱体积蒸馏水和20%乙醇洗脱,弃去,然后以50%-70%乙醇洗脱,收集洗脱液,减压回收乙醇,浓缩至比重为1.2,得到桔梗、牛蒡子和甘草的混合提取物;
(3)将步骤(1)所得荆芥穗油和连翘油,加入等体积的RH60和丙二醇,搅拌均匀,与步骤(2)得到桔梗、牛蒡子和甘草的混合提取物混合均匀,即得抗菌复方制剂4。
另外,本发明提供了抗菌的洗手液,包括上述制备的抗菌的复方制剂,具体的,该洗手液包括以下重量份的组分:脂肪醇聚氧乙烯醚硫酸钠(AES)(70%)10份、脂肪酸甲酯磺酸盐(MES)3份、乙二胺四乙酸二钠(EDTA-2Na)0.05、烷基葡萄糖苷(APG-2000)2份、CAB-354份、甘油2份、氯化钠0.9份、中草药抗菌复方制剂1份、柠檬酸0.04份和去离子水77.01份,其中,以上所述的各种原料均为工业级,按此配比的方式通过具体实施例4-6具体体现其抗菌的作用,其中RH60是氢化蓖麻油聚乙二醇醚,CAB是椰油酰胺丙基甜菜碱。
实施例5
(1)在化料釜加入40-45%70±5℃去离子水;
(2)搅拌下依次加入上述的中草药抗菌洗手液的配方中所述的AES、MES、CAB-35、APG-2000、甘油、EDTA-2Na、NaCl充分搅拌均匀溶解;
(3)温度降至40~45℃时,加入实施例1制得的中草药抗菌复方制剂,加入余下去离子水;
(4)用上述的中草药抗菌洗手液的配方中所述的柠檬酸调原液pH值至6.0~6.5,用200目尼龙筛网过滤形成洗手液1。
本实施例产品性能:
杀菌力:(QBT 2738-2012日化产品抗菌抑菌效果的评价方法,洗手液用量0.5g,作用时间3min)金黄色葡萄球菌99.62%,大肠杆菌82.05%,耐甲氧西林葡萄球菌93.15%。
下表是实验室条件下的检测结果:

(2)搅拌下依次加入上述的中草药抗菌洗手液的配方中所述的AES、MES、CAB-35、APG-2000、甘油、EDTA-2Na、NaCl充分搅拌均匀溶解;
(3)温度降至40~45℃时,加入实施例2制得的中草药抗菌复方制剂,加入余下去离子水;
(4)用上述的中草药抗菌洗手液的配方中所述的柠檬酸调原液pH值至6.0~6.5,用200目尼龙筛网过滤形成洗手液2。
实施例7
(1)在化料釜加入40-45%70±5℃去离子水;
(2)搅拌下依次加入上述的中草药抗菌洗手液的配方中所述的AES、MES、CAB-35、APG-2000、甘油、EDTA-2Na、NaCl充分搅拌均匀溶解;
(3)温度降至40~45℃时,加入实施例3制得的中草药抗菌复方制剂,加入余下去离子水;
(4)用上述的中草药抗菌洗手液的配方中所述的柠檬酸调原液pH值至6.0~6.5,用200目尼龙筛网过滤形成洗手液3。
实施例8
(1)在化料釜加入40-45%70±5℃去离子水;
(2)搅拌下依次加入上述的中草药抗菌洗手液的配方中所述的AES、MES、CAB-35、APG-2000、甘油、EDTA-2Na、NaCl充分搅拌均匀溶解;
(3)温度降至40~45℃时,加入实施例4制得的中草药抗菌复方制剂,加入余下去离子水;
(4)用上述的中草药抗菌洗手液的配方中所述的柠檬酸调原液pH值至6.0~6.5,用200目尼龙筛网过滤形成洗手液4。
实施例9
(1)在化料釜加入40-45%70±5℃去离子水;
(2)搅拌下依次加入上述的中草药抗菌洗手液的配方中所述的AES、MES、CAB-35、APG-2000、甘油、EDTA-2Na、NaCl充分搅拌均匀溶解;
(3)温度降至40~45℃时,加入余下去离子水;
(4)用上述的中草药抗菌洗手液的配方中所述的柠檬酸调原液pH值至6.0~6.5,用200目尼龙筛网过滤形成洗手液5。
本实施例产品性能:
杀菌力:(QBT 2738-2012日化产品抗菌抑菌效果的评价方法,洗手液用量0.5g,作用时间3min金黄色葡萄球菌1.25%,大肠杆菌3.17%,耐甲氧西林葡萄球菌1.56%。
下表是实验室条件下的检测结果:
其中,根据实际需要,制备好的洗手液进行检测、灌装、包装检测装箱即可。
总结
本发明的实施例5-9的测试结果表明,实施例5-8是加了抗菌复方制剂的洗手液,实施例5-8具有良好的抗菌效果,其中,在作用时间为3min时,实施例3对耐药菌耐甲氧西林葡萄球菌的杀灭性最好,达到了98.56%,而实施例2对金黄色葡萄球菌的杀菌率达到95.26%,对大肠杆菌80.05%,且对耐药菌耐甲氧西林葡萄球菌的杀菌率达到94.35%,无论是普通病菌还是耐药菌,都有良好的杀灭效果,是最优的实施例,再通过实施例3和4,实施例3和4主要是是荆芥穗和连翘占比的不同,当荆芥穗的重量份比大于连翘的重量份比时,效果更优,实施例9是不加抗菌复方制剂的洗手液,实验结果显示,其没有抗菌的效果,因此,综合而言本发明包含有抗菌复方制剂的洗手液不仅对普通的致病菌金黄色葡萄球菌和大肠杆菌具有较好杀灭效果,而且对耐药菌耐甲氧西林葡萄球菌也具有很好的抗菌效果。

Claims (10)
1.一种抗菌中草药复方制剂,其特征在于,包括如下重量份数配比:荆芥穗15-35份、连翘2-10份、桔梗5-10份、牛蒡子10-20份、甘草8-15份。
2.根据权利要求1所述的抗菌中草药复方制剂,其特征在于,包括如下重量份数配比:荆芥穗20份、连翘8份、桔梗8份、牛蒡子16份、甘草8份。
3.一种抗菌中草药复方制剂的制备方法,用于制备权利要求1或所述的抗菌中草药复方制剂,其特征在于,包括以下步骤:
(1)将荆芥穗和连翘混合粉碎,采用水蒸气蒸馏法提取荆芥穗和连翘混合油;
(2)将桔梗,牛蒡子,甘草药材混合粉碎,加入6-9体积倍水浸泡,热回流提取2次,过滤后合并两次滤液,浓缩到提取液体积的十分之一,进行醇沉、离心后得到上清液,将上清液浓缩至比重为1.1-1.5,通过大孔树脂柱和乙醇洗脱,得到洗脱液,减压回收乙醇,浓缩至比重1.1-1.5,得到桔梗、牛蒡子和甘草混合提取物;
(3)将步骤(1)所得荆芥穗和连翘混合油,加入等体积的RH60和丙二醇,搅拌均匀,与步骤(2)所得的桔梗、牛蒡子和甘草混合提取物混合均匀,即得到抗菌复方制剂。
4.根据权利要求3所述的抗菌中草药复方制剂的制备方法,其特征在于,在步骤(1)中的荆芥穗和连翘粉碎的粒径和步骤(2)中的桔梗、牛蒡子、甘草药材混合粉碎的粒径均为60-200目。
5.根据权利要求3所述的抗菌中草药复方制剂的制备方法,其特征在于,在步骤(1)中,所述水蒸气蒸馏法的工艺条件为:料液比1:10-1:20,提取温度为85-100℃,提取时间为1-5h。
6.根据权利要求3所述的抗菌中草药复方制剂的制备方法,其特征在于,在步骤(2)中,热回流2次的工艺条件为:热回流的温度为85-100℃,每次回流时间为每次1.5小时。
7.根据权利要求3所述的抗菌中草药复方制剂的制备方法,其特征在于,步骤(2)中醇沉,离心的条件是加入1-5倍体积量的95%乙醇,醇沉,离心。
8.根据权利要求3所述的抗菌中草药复方制剂的制备方法,其特征在于,步骤(2)中,通过大孔树脂柱和乙醇洗脱,得到洗脱液的具体过程为:通过处理好的大孔树脂柱,分别以三个柱体积蒸馏水和20%乙醇洗脱,弃去,然后以50%-70%乙醇洗脱,得到洗脱液。
9.一种洗手液,包括权利要求3-8任一项所述的抗菌中草药复方制剂。
