Abstract
A topical Chinese medicinal composition for treating acne and its preparation method, the raw material composition is 10-50 parts of radix Sophorae Flavescentis, 10-50 parts of cortex Dictamni Radicis, 5-30 parts of radix Stemonae, 5-15 parts of cortex Phellodendri, 1-8 parts of radix et rhizoma Rhei, and 1-10 parts of caulis Sargentodoxae. The method comprises the following steps: (1) preparing raw materials; (2) soaking radix Sophorae Flavescentis, cortex Dictamni Radicis, radix Stemonae and cortex Phellodendri in water, and extracting to obtain radix Sophorae Flavescentis extractive solution, cortex Dictamni Radicis extractive solution, radix Stemonae extractive solution and cortex Phellodendri extractive solution; (3) mixing radix Sophorae Flavescentis extractive solution, cortex Dictamni Radicis extractive solution, radix Stemonae extractive solution and cortex Phellodendri extractive solution, and filtering to obtain purified filtrate; (4) evaporating and concentrating; (5) drying radix et rhizoma Rhei and caulis Sargentodoxae, grinding, and micronizing; (6) putting the mixed powder into the concentrated solution, and adding a preservative and a humectant; and dispersing, homogenizing, shearing and refining the mixed materials by a homogenizing emulsifying machine to prepare the paste. The method can effectively improve the production efficiency, reduce the loss of the effective components of the medicine and enhance the medicine effect; the product is convenient to carry and use and has better market prospect.

Technical Field
The invention belongs to the technical field of traditional Chinese medicine, and relates to an external traditional Chinese medicine composition for treating acne and a preparation method thereof.
Background
Acne is a common chronic inflammatory skin disease which is frequently found in facial follicular sebum units of teenagers, has certain damage capacity, is better found in young men and women of 15-30 years old, and is clinically mainly manifested by polymorphic skin lesions such as facial acne, papules, pustules, nodules and the like, and is frequently found in parts rich in sebaceous glands such as the face, the back, the chest and the like. Acne occurs mainly in association with factors such as hyperseborrhea, blockage of pilosebaceous ducts, bacterial infection and inflammatory reaction; after adolescence, the androgen level in the human body is increased, sebaceous glands are developed vigorously and a large amount of sebum is produced. Abnormal keratinization of the pilosebaceous canal can cause blockage of the canal, obstruction of sebum discharge, and formation of keratotic plugs, i.e., comedones. A number of microorganisms in the hair follicle (especially propionibacterium acnes) multiply, producing lipases, decomposing sebum, producing free fatty acids, which in turn chemotact inflammatory cells and mediators, inducing and exacerbating the inflammatory response. Because acne is well developed in adolescence, the personal image is seriously influenced, and psychological problems such as anxiety, depression and the like can be brought to patients, particularly teenagers.
At present, modern medicine is mainly applied to the treatment of medicines such as vitamin A acid, antibiotics, hormones and the like, but certain drug resistance and adverse drug reactions exist, and the effective rate is not ideal. At present, a plurality of traditional Chinese medicine external treatment products for treating acne exist in the market, and in the preparation process, most products adopt a single extraction mode or a mode of simply crushing and adding a solvent for mixed use, so that the content of the effective ingredients of the medicines in the products is low, or the water-insoluble ingredients are lost due to the limitation of the extraction process of the effective ingredients; or due to the volume and dosage of the raw medicinal materials, the product contains more lignin and cellulose, so that the content of the effective components of the medicine in unit volume is low, thereby affecting the curative effect and being inconvenient to use.

The invention aims to provide an external traditional Chinese medicine composition for treating acne and a preparation method thereof.
The external traditional Chinese medicine composition for treating acne comprises 10-50 parts of radix sophorae flavescentis, 10-50 parts of cortex dictamni, 5-30 parts of radix stemonae, 5-15 parts of cortex phellodendri, 1-8 parts of rheum officinale and 1-10 parts of sargentgloryvine stem.
The external traditional Chinese medicine composition for treating acne comprises 20-40 parts of radix sophorae flavescentis, 20-40 parts of cortex dictamni, 10-20 parts of radix stemonae, 6-12 parts of cortex phellodendri, 2-5 parts of rheum officinale and 3-8 parts of sargentgloryvine stem.
The raw material components of the external traditional Chinese medicine composition for treating acne are 30 parts of radix sophorae flavescentis, 30 parts of cortex dictamni, 15 parts of radix stemonae, 10 parts of cortex phellodendri, 3 parts of rheum officinale and 6 parts of sargentgloryvine stem.
The preparation method of the external traditional Chinese medicine composition for treating acne comprises the following steps:
1. preparing raw materials according to the raw material component proportion;
2. soaking radix Sophorae Flavescentis, cortex Dictamni Radicis, radix Stemonae and cortex Phellodendri in water for at least 2 hr, and respectively extracting with water under normal pressure, negative pressure or ultrasonic wave to obtain radix Sophorae Flavescentis extractive solution, cortex Dictamni Radicis extractive solution, radix Stemonae extractive solution and cortex Phellodendri extractive solution;
3. mixing radix Sophorae Flavescentis extractive solution, cortex Dictamni Radicis extractive solution, radix Stemonae extractive solution and cortex Phellodendri extractive solution, and filtering to remove residue, ash and macromolecular compounds to obtain purified filtrate;
4. heating the purified filtrate, evaporating and concentrating to obtain a concentrate;
5. drying the rhubarb and the sargentgloryvine stem in the raw materials to remove water, then crushing, and carrying out ultrafine crushing by using an ultrafine crusher until the particle size is less than or equal to 40 microns to prepare ultrafine mixed powder;
6. putting the mixed powder into the concentrated solution, and adding a preservative and a humectant to obtain a mixed material; dispersing and homogenizing the mixed materials by adopting a homogenizing emulsifying machine at a stirring speed of 2000-4000 revolutions per minute, and then shearing and refining the mixed materials by adopting a stirring speed of 6000-9000 revolutions per minute to prepare paste so as to obtain the external traditional Chinese medicine composition for treating acne.
In the step 2, when the normal pressure extraction, the negative pressure extraction or the ultrasonic extraction is performed, the mass ratio of the kuh-seng, the cortex dictamni, the radix stemonae or the cortex phellodendri to the water is 1: (8-10), extracting for 2 times, and mixing the liquid obtained after the two-time extraction to obtain a radix sophorae flavescentis extracting solution, a cortex dictamni extracting solution, a radix stemonae extracting solution and a cortex phellodendri extracting solution; wherein, the normal pressure extraction adopts traditional Chinese medicine normal pressure reflux extraction equipment, and the extraction time is 1.5-2.0 hours each time; the negative pressure extraction adopts traditional Chinese medicine vacuum extraction equipment, the extraction time is 1.0-1.5 hours each time, and the working pressure during extraction is-0.06-0.08 MPa; traditional Chinese medicine ultrasonic extraction equipment is adopted for ultrasonic extraction, and the extraction time is 25-35 minutes each time.
In the step 2, when the normal pressure extraction or the negative pressure extraction is carried out, the radix sophorae flavescentis, the cortex dictamni, the radix stemonae and the cortex phellodendri are in a sheet shape; when ultrasonic extraction is carried out, radix Sophorae Flavescentis, cortex Dictamni Radicis, radix Stemonae and cortex Phellodendri are firstly pulverized to particle size of below 5 mm.
In the step 3, the filtration is sequentially performed by adopting a centrifugal filter for rough filtration, ceramic membrane ultrafiltration and hollow fiber membrane nanofiltration; the molecular weight of the substance in the obtained purified filtrate is less than 10000.
In the step 4, a steam boiler, a single-effect evaporator or a multi-effect evaporator is adopted for evaporation concentration, and the mass of the obtained concentrate is 20-25% of the total mass of the purified filtrate.
In the step 5, the dried material is firstly crushed, then the part below 60 meshes is screened out to be used as primary powder, the material on the screen is continuously ground to be below 80 meshes to be used as secondary powder, and the primary powder and the secondary powder are mixed for superfine grinding.
In the step 6, the preservative is ethylparaben, and the addition amount of the preservative is less than 0.02 percent of the total mass of the mixed materials; the humectant is sodium carboxymethylcellulose, and the addition amount of the humectant is 2-3% of the total mass of the mixed material.
In the step 6, the external traditional Chinese medicine composition for treating acne is filled by a full-automatic filling machine, and is quantitatively packaged by a medical aluminum ointment tube, a single paper small box of a hose is used for packaging, and the product is cased and sterilized.

the preparation method is the same as example 1, and is different from the following steps:
(1) respectively carrying out negative pressure extraction, wherein the mass ratio of the kuh-seng, the cortex dictamni, the radix stemonae or the cortex phellodendri to the water is 1: 9; the extraction time is 1.0-1.5 hours each time, and the working pressure during extraction is-0.06-0.08 MPa;
(2) the mass of the concentrate is 22% of the total mass of the purified filtrate;
(3) the addition amount of the preservative is 0.01 percent of the total mass of the mixed materials; the adding amount of the humectant accounts for 3 percent of the total mass of the mixed materials; firstly, dispersing and homogenizing at a stirring speed of 3000 r/min, and then shearing and thinning at a stirring speed of 7000 r/min to prepare the paste.
Example 3
The external traditional Chinese medicine composition for treating acne comprises the raw material components of 40 parts of radix sophorae flavescentis, 20 parts of cortex dictamni, 20 parts of radix stemonae, 6 parts of cortex phellodendri, 5 parts of rheum officinale and 3 parts of sargentgloryvine stem;
the preparation method is the same as example 1, and is different from the following steps:
(1) respectively carrying out ultrasonic extraction, wherein the mass ratio of the kuh-seng, the cortex dictamni, the radix stemonae or the cortex phellodendri to the water is 1: 9, extracting for 25-35 minutes each time; extracting radix Sophorae Flavescentis, cortex Dictamni Radicis, radix Stemonae and cortex Phellodendri, pulverizing to particle size below 5 mm;
(2) the mass of the concentrate is 24% of the total mass of the purified filtrate;
(3) the addition amount of the preservative is 0.01 percent of the total mass of the mixed materials; the adding amount of the humectant accounts for 3 percent of the total mass of the mixed materials; firstly, the mixture is dispersed and homogenized at the stirring speed of 4000 revolutions per minute, and then the mixture is sheared and refined at the stirring speed of 8000 revolutions per minute to prepare the paste.

(2) the mass of the concentrate is 25% of the total mass of the purified filtrate;
(3) the addition amount of the preservative is 0.01 percent of the total mass of the mixed materials; the adding amount of the humectant accounts for 2 percent of the total mass of the mixed materials; firstly, the mixture is dispersed and homogenized at the stirring speed of 4000 revolutions per minute, and then the mixture is sheared and refined at the stirring speed of 8000 revolutions per minute to prepare the paste.
Example 5
The external traditional Chinese medicine composition for treating acne comprises the raw material components of 20 parts of radix sophorae flavescentis, 20 parts of cortex dictamni, 10 parts of radix stemonae, 12 parts of cortex phellodendri, 5 parts of rheum officinale and 8 parts of sargentgloryvine stem;
the preparation method is the same as example 1, and is different from the following steps:
(1) respectively extracting the sophora flavescens, the cortex dictamni, the stemona or the phellodendron bark by negative pressure, wherein the mass ratio of the water to the sophora flavescens, the cortex dictamni, the stemona or the phellodendron bark is 1: 10, the extraction time is 1.0 to 1.5 hours each time, and the working pressure during extraction is-0.06 to-0.08 MPa;
(2) the mass of the concentrate is 25% of the total mass of the purified filtrate;
(3) the addition amount of the preservative is 0.01 percent of the total mass of the mixed materials; the adding amount of the humectant accounts for 3 percent of the total mass of the mixed materials; firstly, the mixture is dispersed and homogenized at the stirring speed of 4000 revolutions per minute, and then the mixture is sheared and refined at the stirring speed of 9000 revolutions per minute to prepare the paste.
Claims (10)
1. A topical Chinese medicinal composition for treating acne is characterized by comprising 10-50 parts of radix sophorae flavescentis, 10-50 parts of cortex dictamni, 5-30 parts of radix stemonae, 5-15 parts of cortex phellodendri, 1-8 parts of rheum officinale and 1-10 parts of sargentgloryvine stem.

The external traditional Chinese medicine composition for treating acne according to claim 1, which is characterized by comprising 20-40 parts of radix sophorae flavescentis, 20-40 parts of cortex dictamni, 10-20 parts of radix stemonae, 6-12 parts of cortex phellodendri, 2-5 parts of rheum officinale and 3-8 parts of sargentgloryvine stem.

The external traditional Chinese medicine composition for treating acne according to claim 1, which is characterized in that the raw material components comprise 30 parts of radix sophorae flavescentis, 30 parts of cortex dictamni, 15 parts of radix stemonae, 10 parts of cortex phellodendri, 3 parts of rheum officinale and 6 parts of sargentgloryvine stem.
4. A method for preparing the topical Chinese medicinal composition for treating acne as claimed in claim 1, which is characterized by comprising the following steps:
(1) preparing raw materials according to the raw material component proportion;
(2) soaking radix Sophorae Flavescentis, cortex Dictamni Radicis, radix Stemonae and cortex Phellodendri in water for at least 2 hr, and respectively extracting with water under normal pressure, negative pressure or ultrasonic wave to obtain radix Sophorae Flavescentis extractive solution, cortex Dictamni Radicis extractive solution, radix Stemonae extractive solution and cortex Phellodendri extractive solution;
(3) mixing radix Sophorae Flavescentis extractive solution, cortex Dictamni Radicis extractive solution, radix Stemonae extractive solution and cortex Phellodendri extractive solution, and filtering to remove residue, ash and macromolecular compounds to obtain purified filtrate;
(4) heating the purified filtrate, evaporating and concentrating to obtain a concentrate;
(5) drying the rhubarb and the sargentgloryvine stem in the raw materials to remove water, then crushing, and carrying out ultrafine crushing by using an ultrafine crusher until the particle size is less than or equal to 40 microns to prepare ultrafine mixed powder;
(6) putting the mixed powder into the concentrated solution, and adding a preservative and a humectant to obtain a mixed material; dispersing and homogenizing the mixed materials by adopting a homogenizing emulsifying machine at a stirring speed of 2000-4000 revolutions per minute, and then shearing and refining the mixed materials by adopting a stirring speed of 6000-9000 revolutions per minute to prepare paste so as to obtain the external traditional Chinese medicine composition for treating acne.

The method for preparing the external traditional Chinese medicine composition for treating acne according to claim 4, characterized in that in the step (2), when normal pressure extraction, negative pressure extraction or ultrasonic extraction is carried out, the mass ratio of the lightyellow sophora root, the dittany bark, the sessile stemona root or the amur corktree bark to water is 1 (8-10), 2 times of extraction are carried out, and the liquid obtained after the two times of extraction is mixed to be used as lightyellow sophora root extracting solution, dittany bark extracting solution, sessile stemona root extracting solution and amur corktree bark extracting solution; wherein, the normal pressure extraction adopts traditional Chinese medicine normal pressure reflux extraction equipment, and the extraction time is 1.5-2.0 hours each time; the negative pressure extraction adopts traditional Chinese medicine vacuum extraction equipment, the extraction time is 1.0-1.5 hours each time, and the working pressure during extraction is-0.06-0.08 MPa; traditional Chinese medicine ultrasonic extraction equipment is adopted for ultrasonic extraction, and the extraction time is 25-35 minutes each time.

The method for preparing a topical Chinese medicinal composition for the treatment of acne according to claim 4, characterized in that in step (3), the filtration is performed by rough filtration with centrifugal filter, ultrafiltration with ceramic membrane and nanofiltration with hollow fiber column; the molecular weight of the substance in the obtained purified filtrate is less than 10000.

The method for preparing a topical Chinese medicinal composition for the treatment of acne according to claim 4, characterized in that in step (4), the evaporation concentration is performed by using a steam boiler, a single-effect evaporator or a multiple-effect evaporator, and the mass of the obtained concentrate is 20-25% of the total mass of the purified filtrate.

The method for preparing a topical Chinese medicinal composition for treating acne as claimed in claim 4, wherein in step (5), the dried material is pulverized, then the part below 60 mesh is sieved to obtain a primary powder, the sieved material is further pulverized to below 80 mesh to obtain a secondary powder, and the primary powder and the secondary powder are mixed and subjected to micronization.
