1) respectively weighing the following raw materials in parts by weight: 20kg of astragalus membranaceus, 11kg of ligusticum wallichii, 12kg of angelica sinensis and 10kg of radix ophiopogonis, then mixing the astragalus membranaceus, the ligusticum wallichii, the angelica sinensis and the radix ophiopogonis, crushing the mixture, and sieving the crushed mixture through a 100-mesh sieve to obtain mixed powder;
2) adding 40% ethanol aqueous solution into the mixture powder, leaching for 48 hours, taking supernatant, filtering the supernatant, concentrating the supernatant into paste, and drying at 65 ℃ for 2 hours to obtain a leaching product; the adding amount of the ethanol water solution is 6 times of the total weight of the mixture powder, and the leaching temperature is 50 ℃;
3) dissolving the leaching product with water, adding β -xylosidase, alkaline phosphatase and pectin methylesterase for enzymolysis to obtain an enzymolysis solution, wherein the addition amount of the water is 6 times of the weight of the leaching product, the addition amount of the β -xylosidase is 0.4 percent of the weight of the leaching product, the addition amount of the alkaline phosphatase is 0.4 percent of the weight of the leaching product, the addition amount of the pectin methylesterase is 0.4 percent of the weight of the leaching product, the enzymolysis temperature is 45 ℃, and the enzymolysis time is 110 min;
4) filtering the enzymolysis liquid, concentrating the filtrate into paste, drying at 32 ℃ for 1.2 hours to obtain an enzymolysis product, adding a corn tassel extract, a corn root extract and white spirit into the enzymolysis product, standing and aging at 35 ℃ for 50 hours, and filtering to remove insoluble substances to obtain the astragalus membranaceus health-care wine; the addition amount of the corn tassel extract is 0.8 percent of the weight of the extraction product; the addition amount of the corn root extract is 0.4 percent of the weight of the extract; the adding amount of the white spirit is 50 times of the weight of the leaching product; the volume fraction of ethanol in the white spirit is 60%.

The preparation method of the corn root extract comprises the following steps:

Application test example
Effect of health care wine on experimental rat hyperlipidemia
Experimental animals: SD rats, SPF grade, male, body weight 200 + -20 g.
The experimental method comprises the following steps: after adaptive feeding for one week, dividing the rats into a normal control group and a modeling group randomly according to the weight, wherein 10 normal control groups are fed with basic feed; and (4) building 80 modules, after feeding with high-fat feed for 2 weeks, taking blood from eye sockets to measure blood fat, and selecting rats with remarkably increased blood fat as successful modeling of the hyperlipidemia.
The patients with blood TC higher than 2.5mmol/L were taken into the experiment, and the hyperlipidemic rats were divided into a hyperlipidemic model group, a health wine drug group prepared in examples 1-3 and comparative examples 1-3, and a positive drug simvastatin (5mg/kg body weight) group according to the blood lipid level, each group containing 10 rats. Normal control group and hyperlipemia model group were given physiological saline of equal volume, and experimental treatment: the rats in the normal control group and the model group are respectively administrated with physiological saline according to the intragastric administration of 20ml/kg body weight, the simvastatin in the positive control drug is administrated according to 5mg/kg body weight, the health care wine drug groups prepared in examples 1-3 and comparative examples 1-3 are administrated according to 20mg/kg body weight, and the normal control group and the model group are prepared into appropriate concentrations with the administration volume of 20ml/kg body weight for administration. The administration is performed by intragastric administration 1 time per day for 14 days, and 60 minutes after the last administration, blood is taken from ocular venous plexus for measurement and blood fat. The results are shown in Table 1.

Compared with the comparative example, the health-care wine obtained by the embodiment of the invention has obviously enhanced lipid metabolism regulation effect on rats, which shows that the health-care wine has obvious regulation effect on lipid metabolism abnormality of experimental rats caused by high-fat feeding.

Claims (10)

1. The preparation method of the astragalus membranaceus health-care wine is characterized by comprising the following steps of:
1) respectively weighing the following raw materials in parts by weight: 15-20 parts of astragalus membranaceus, 8-15 parts of ligusticum wallichii, 10-15 parts of angelica sinensis and 7-10 parts of radix ophiopogonis, and then the astragalus membranaceus, the ligusticum wallichii, the angelica sinensis and the radix ophiopogonis are mixed and crushed to obtain mixed powder;
2) adding 30-60% ethanol water solution into the mixture powder, leaching for 24-72 hr, collecting supernatant, filtering, concentrating, and drying to obtain extract;
3) dissolving the leaching product with water, and adding glycosidase, esterase and pectinase for enzymolysis to obtain an enzymatic hydrolysate;
4) filtering the enzymolysis liquid, concentrating and drying the filtrate to obtain an enzymolysis product, adding a corn tassel extract, a corn root extract and white spirit into the enzymolysis product, standing, aging and filtering to obtain the astragalus membranaceus health-care wine.
2. The preparation method of the astragalus membranaceus health-care wine as claimed in claim 1, wherein the corn tassel extract is added in an amount of 0.2-1% of the weight of the extracted product;
the addition amount of the corn root extract is 0.2-1% of the weight of the extract.
3. The preparation method of the astragalus membranaceus health-care wine as claimed in claim 2, wherein the preparation method of the corn tassel extract comprises the following steps:
taking the dried corn tassel, crushing, adding 30-60% ethanol water solution by volume fraction, leaching for 24-48 hours, taking supernatant, filtering the supernatant, concentrating and drying to obtain a corn tassel extract;
the preparation method of the corn root extract comprises the following steps:
pulverizing dried corn root, adding 30-60% ethanol water solution, leaching for 24-48 hr, collecting supernatant, filtering, concentrating, and drying to obtain corn root extract.
4. The preparation method of the astragalus membranaceus health-care wine as claimed in claim 3, wherein in the preparation method of the corn tassel extract, the adding amount of the ethanol water solution is 5-10 times of the weight of the dried corn tassel, and the extraction temperature is 50-60 ℃;
in the preparation method of the corn root extract, the adding amount of the ethanol water solution is 5-10 times of the weight of the dried corn root, and the extraction temperature is 50-60 ℃.
5. The preparation method of the astragalus membranaceus health-care wine according to any one of claims 1 to 4, wherein in the step 2), the adding amount of the ethanol aqueous solution is 4-8 times of the total weight of the mixture powder, and the extraction temperature is 40-60 ℃.

The preparation method of the astragalus membranaceus health-care wine as claimed in any one of claims 1 to 5, wherein in the step 3), the addition amount of the glycosidase is 0.2 to 0.5 percent of the weight of the extracted product; the addition amount of the esterase is 0.3 to 0.6 percent of the weight of the leaching product; the addition amount of the pectinase is 0.2-0.5% of the weight of the leaching product.
7. The method for preparing the astragalus membranaceus health-care wine as claimed in any one of claims 1 to 6, wherein in step 3), the enzymolysis temperature is 40-50 ℃, and the enzymolysis time is 100-120 min.
8. The preparation method of the astragalus membranaceus health-care wine according to any one of claims 1 to 7, wherein the adding amount of the white wine is 30-50 times of the weight of the extracted product;
the volume fraction of ethanol in the white spirit is 30-60%.
9. The method for preparing astragalus membranaceus health-care wine according to any one of claims 1 to 8, wherein in step 4), the standing and aging temperature is 30-40 ℃, and the standing and aging time is 48-72 hours.
