(I) Experimental method
In order to verify the efficacy of the pure plant smoking cessation buccal tablets, 45 mice with the weight of 18-20g are selected and fed under the SPF-level condition; preparing 2mg/mL nicotine solution, and during the feeding period, taking 1mL solution every day to perfuse the stomach of the mouse, and continuously perfusing the stomach for 30 days to establish a mouse nicotine dependence model.
45 of the mice were randomly divided into experimental and control groups, experimental: dissolving 3 tablets of the smoking-quitting buccal tablets prepared in example 1 into 50mL of liquid medicine by water every day, performing intragastric administration on experimental mice, and treating for 40 d;
control group: dissolving 3 pure plant buccal tablets prepared in comparative example 1 into 50mL of liquid medicine with water every day, performing intragastric administration on experimental mice, and treating for 40 d;
blank group: mice in the experimental group were gavaged daily with an equal amount of physiological saline.
During the experiment, 1mL of feed containing the nicotine solution and common feed are simultaneously placed in front of an experimental group, a control group and a blank group of mice every day, and the selection of the mice on the feed is observed; in addition, the urine content of cotinine (a nicotine metabolite) in the urine of the mice in the experimental group, the control group and the blank group was measured periodically.
(II) results of the experiment
The experimental group mice had 1 mouse selected to eat common feed from day 5, 10 mice had 10 mice selected to eat common feed at day 10, and all the experimental group mice had the common feed after day 28, which shows that the pure plant buccal tablet for stopping smoking of the invention can eliminate the dependence on nicotine.
Only 2 mice in the control group selected to eat the common feed from the 10 th day, 10 mice in the 18 th day selected to eat the common feed, and the experimental group mice all selected to eat the common feed after 35 days, which shows that the smoking-quitting buccal tablets have better smoking-quitting effect compared with the smoking-quitting buccal tablets in the comparative example, namely, although the chrysanthemum has the similar efficacy with the dandelion, the smoking-quitting effect is obviously reduced after the chrysanthemum is used for replacing the dandelion, and the relationship that the pumpkin vine and the dandelion supplement each other is verified.
The control mice had been fed the nicotine supplemented diet for 40 days.
The structure of the content of cotinine in urine of the mice of the experimental group and the control group is shown in table 1.
TABLE 1 cotinine content in mouse urine (μmol/L)
As can be seen from the table 1, the cotinine content in the urine of the experimental mice is reduced all the time, while the cotinine content in the urine of the control mice is unchanged or even increased, which is related to the fact that the control mice use the feed added with nicotine all the time, and the pure plant smoking-quitting buccal tablets can eliminate the interest of users in smoking, can metabolize nicotine in the body and have better smoking-quitting effect compared with the control mice.
