Abstract

背景技术

如本文所用,术语“抗体”具有其在本领域中的一般含义,并且是指具有抗原结合区的任何抗体样分子,该术语包括包含抗原结合结构域的抗体片段,例如Fab'、Fab、F(ab')2和单结构域抗体(DAB)。在天然抗体中,两条重链通过二硫键相互连接,并且每条重链通过二硫键与轻链连接。有两种轻链,lambda(1)和kappa(k)。有五种主要的重链类别(或同种型),其决定抗体分子的功能活性:IgM、IgD、IgG、IgA和IgE。每条链含有不同的序列结构域。轻链包括两个结构域,一个可变结构域(VL)和一个恒定结构域(CL)。重链包括四个结构域,一个可变结构域(VH)和三个恒定结构域(CH1、CH2和CH3,统称为CH)。轻链(VL)和重链(VH)的可变区决定了对抗原的结合识别和特异性。轻链的恒定区结构域(CL)和重链的恒定区结构域(CH)赋予重要的生物学特性,例如抗体链结合、分泌、经胎盘移动、补体结合和与Fc受体(FcR)的结合。Fv片段是免疫球蛋白的Fab片段的N-末端部分,并且由一条轻链和一条重链的可变部分组成。抗体的特异性在于抗体结合位点和抗原决定簇之间的结构互补性。抗体结合位点由主要来自高变区或互补决定区(CDR)的残基组成。偶尔,来自非高变区或框架区(FR)的残基影响整个结构域的结构,从而影响结合位点。互补决定区或CDR是指一起定义天然免疫球蛋白结合位点的天然Fv区的结合亲和力和特异性的氨基酸序列。免疫球蛋白的轻链和重链各自具有三个CDR,分别称为L-CDR1、L-CDR2、L-CDR3和H-CDR1、H-CDR2、H-CDR3。因此,抗原结合位点包括六个CDR,其包含来自各重链和轻链V区中的CDR组。框架区(FR)是指插入CDR之间的氨基酸序列。

在一些实施方案中,抗体是多克隆抗体或单克隆抗体。可以使用Kohler和Milstein的方法(Nature,256:495,1975)产生单克隆抗体。为制备可用于本发明的单克隆抗体,以适当的间隔(例如,每周两次、每周一次、每月两次或每月一次)用相关的病毒抗原形式免疫接种小鼠或其他合适的宿主动物(例如小鼠、山羊、骆驼科动物......)。例如,病毒抗原形式通常根据以下方法制备:可以根据实施例的培养系统制备一定量的病毒颗粒,然后在碘克沙醇垫上纯化,最后用一定量的胰蛋白酶和脱氧胆酸钠(DOCA)处理。或者,抗原形式直接由衍生自本发明多肽的肽组成。可以在处死后一周内向动物施用最终“加强”的抗原形式。通常需要在免疫接种期间使用免疫佐剂。合适的免疫佐剂包括弗氏完全佐剂、弗氏不完全佐剂、明矾、Ribi佐剂、Hunter's Titermax、皂苷佐剂如QS21或Quil A,或含CpG的免疫刺激性寡核苷酸。其他合适的佐剂是本领域众所周知的。可以通过皮下、腹膜内、肌肉内、静脉内、鼻内或其他途径免疫接种动物。在免疫接种方案后,从动物的脾、淋巴结或其他器官分离淋巴细胞,并使用诸如聚乙二醇的试剂与合适的骨髓瘤细胞系融合以形成杂交瘤。融合后,如所述使用标准方法将细胞置于允许杂交瘤生长但不允许融合伴侣生长的培养基中(Coding,Monoclonal Antibodies:Principles and Practice:Production andApplication of Monoclonal Antibodies in Cell Biology,Biochemistry andImmunology,3rd edition,Academic Press,New York,1996)。培养杂交瘤后,分析细胞上清液中所需特异性抗体(即,选择性结合抗原)的存在。合适的分析技术包括ELISA、免疫荧光、流式细胞术、免疫沉淀和蛋白质印迹。其他筛选技术是本领域众所周知的。优选的技术是确认抗体与构象完整的天然折叠抗原结合的技术,例如非变性ELISA、流式细胞术和免疫沉淀。
在进一步的方面,在样品中检测本发明的ORF2i或ORF2g多肽对于诊断目的可能特别有意义。特别地,检测本发明的ORF2i或ORF2g多肽适用于测定样品中戊型肝炎病毒感染性颗粒的存在。更具体地,检测本发明的ORF2i或ORF2g多肽适用于诊断受试者的戊型肝炎病毒感染。
如本文所用,术语“样品”包括任何固体或液体样品,其易于含有戊型肝炎病毒的感染性颗粒。在一些实施方案中,样品选自腹水、尿、唾液、汗液、乳汁、滑液、腹膜液、羊水、percerebrospinal fluid、淋巴液、肺栓塞、脑脊液和心包液。在一些实施方案中,样品是粪便样品。在一些实施方案中,样品是尿液样品。在一些实施方案中,样品是血液样品。如本文所用,术语“血液样品”是指源自受试者的任何血液样品。血液样品的收集可以通过本领域技术人员熟知的方法进行。例如,受试者的血液可以由受过训练的医务人员直接抽取到抗凝血剂如柠檬酸盐和EDTA中。通过在3500×G下冷冻离心2分钟,可以将全血分离成血浆部分、细胞和血小板部分。离心后,上清液为血浆。
本发明的抗体特别适用于检测样品中本发明的ORF2i或ORF2g多肽的存在。
因此,本发明的另一个目的涉及用于检测样品中本发明的ORF2i或ORF2g多肽的存在的方法,包括在允许多肽和抗体形成免疫复合物的条件下,将样品与本发明的抗体接触,其中检测到免疫复合物表明样品中存在多肽。
更具体地,本发明的另一个目的涉及用于检测样品中戊型肝炎病毒的感染性颗粒的存在的方法,包括在允许抗体和感染性颗粒形成免疫复合物的条件下,将样品与对ORF2i多肽具有特异性的抗体接触,其中检测到免疫复合物表明样品中存在感染性颗粒。
在一些实施方案中,本发明的检测方法可以进一步包括将样品与对ORF2c多肽(即,ORF2c多肽的质量为约75kDa)具有特异性的抗体接触。
本发明的检测方法特别适用于诊断急性HEV感染、近期HEV感染、慢性HEV感染或弱活性HEV感染。本发明的检测方法也适用于诊断清除的HEV感染,其特征在于检测到ORF2g多肽(连同检测到ORF2c多肽)但未检测到ORF2i多肽。
用于检测免疫复合物的测定和条件是本领域技术人员已知的。此类测定包括例如竞争测定、直接反应测定、夹心型测定和免疫测定(例如ELISA)。测定可以是定量的或定性的。存在许多不同的用于检测包含本发明多肽的抗体-肽复合物的形成的常规测定法。例如,检测步骤可包括进行ELISA测定、进行侧流免疫测定、进行凝集测定、分析分析转子中的样品,或用电化学、光学或光电传感器分析样品。这些不同的测定法是本领域技术人员所熟知的。

具体实施方式
实施例1:
材料和方法
化学品和细胞培养。在补充有10%灭活胎牛血清(DMEM/FCS)的Dulbecco改良的Eagle培养基(DMEM)中,于37℃生长PLC/PRF/5(CRL-8024)、PLC1、PLC3和A549(CCL-185)细胞。在32℃下,在含有DMEM/M199(1v:1v)、1mg/ml富含脂质的白蛋白(Albumax ITM)和40nMNa2SeO3的培养基中维持转染的细胞。
质粒和转染。表达适应细胞培养的gt3Kernow C-1株的质粒(HEV-p6,GenBank登录号JQ679013)或表达高斯荧光素酶基因的复制子(HEV-p6GLuc)由SU Emerson提供6。通过将GDD基序突变到RNA依赖性RNA聚合酶基因中来产生复制缺陷型复制子HEV-p6GLucGAD7。用mMESSAGE试剂盒(Ambion)产生加帽的RNA转录物。使用Gene PulserXcellTM装置(Bio-Rad),通过电穿孔将加帽的RNA递送至细胞。
动力学实验和病毒生产。
用加帽的HEV-p6RNA(20μg/4×106个细胞)电穿孔PLC1和PLC3细胞。对于动力学实验,每两天收集上清液,并在含有50mM TrisHCl(pH 7.5)、150mM NaCl、5mM EDTA、0.5%(v/v)NP40、0.05%脱氧胆酸钠、1mM PMSF和蛋白酶抑制剂鸡尾酒(Complete;Roche)的缓冲液中裂解细胞。对于病毒生产,细胞每10天分离一次。收集上清液并储存在-80℃。
密度梯度。用Vivaspin超滤旋转柱(Sartorius)浓缩PLC3/HEV-p6上清液。将浓缩的上清液或患者血清(500μl)置于7.5-40%碘克沙醇梯度上,将其在4℃以160,000g离心16小时。收集12个级分1ml,通过折射法测量其密度。通过RT-qPCR测定HEV RNA滴度。用每个级分感染A549细胞。通过间接免疫荧光、免疫印迹以及RNA和病毒滴定测定感染性。对于患者样品,将级分用于ORF2探针检测、GNA pull-down和RNA定量。
在碘克沙醇垫上纯化感染性颗粒。通过以160,000g在4℃下离心4小时通过20%碘克沙醇垫,浓缩PLC3/HEV-p6的上清液。将垫重悬于PBS中,第二次超速离心,并将沉淀重悬于PBS中。
感染人源化小鼠。如前所述,将原代人肝细胞移植到纯合的uPA+/+-SCID小鼠中8。通过脾内途径,用以下接种人源化小鼠:在碘克沙醇梯度上纯化的HEV-p6(级分6,F6p6,8.7x107IU/小鼠),或相同体积的来自用未转染PLC3细胞的浓缩上清液制备的对照梯度的级分6(F6对照)。用这些制剂接种的未移植小鼠作为阴性对照。用gt1(Sar55,2.8×105IU/小鼠)粪便悬浮液接种的嵌合小鼠用作阳性对照9。每周收集粪便和血浆样品。如所述,使用RT-qPCR检测并定量小鼠血浆和粪便中的病毒RNA9。如所述进行S17区的测序9。
透射电子显微镜(TEM)。将formvar-碳TEM网格(S162,Oxford Instruments)用0.01%聚-L-赖氨酸在室温(RT)下孵育30分钟,或在RT下用聚-L-赖氨酸孵育,然后用抗ORF3(Bioss抗体)、抗-ORF2(1E6)或同种型匹配的抗体(20μg/ml)孵育1小时。将网格在PBS中洗涤,并在RT下与病毒样品一起孵育2小时。将TEM网格在PBS中洗涤,并在4%多聚甲醛和1%戊二醛的0.1M磷酸盐缓冲液(pH7.2)中孵育20分钟。用0.5%乙酸铀酰染色在网格上捕获的颗粒,然后在JEOL-1230TEM下检查。对于脱脂,在37℃下将颗粒用1%脱氧胆酸钠、0.1%胰蛋白酶处理1小时,然后加工用于TEM,或在相同条件下处理,然后在碘克沙醇梯度上分层。在测定密度和RNA水平后,将级分11加工用于TEM。
糖苷酶消化。在95℃下,将蛋白质样品在糖蛋白变性缓冲液(New EnglandBiolabs)中变性10分钟。在1%NP40和制造商(NEB)提供的缓冲液存在下,在37℃下进行糖苷酶消化4小时。在相同条件下(但不含糖苷酶)制备的样品用作对照。
GNA pull-down。在室温下,将琼脂糖偶联的GNA珠与50-500μl患者血清或100μlPLC3/HEV-p6上清液一起孵育2小时。用PBS 0.5%NP40洗涤珠子8次。将蛋白质在Laemmli缓冲液中洗脱,并通过SDS-PAGE分析。未偶联的琼脂糖珠用作阴性对照。
患者样品。患者样品于2012年至2017年间在法国收集。这是一项非介入性研究。仅根据医生的指示(无补充或修改取样)通过标准病毒诊断获得样品。匿名分析数据。法国公共卫生法(CSP Art L1121-1.1)不要求患者签署此类方案的书面知情同意书。
结果
建立有效的HEV细胞培养系统
为了建立HEV的细胞培养系统,我们首先分析了PLC/PRF/5细胞系的两个亚克隆(即PLC1和PLC3细胞)中的HEV复制。我们使用名称为HEV-p6GLuc的表达高斯荧光素酶(GLuc)的HEV gt3复制子6,其中ORF2编码序列被分泌的GLuc序列替换。因此,分泌的GLuc的量与病毒RNA合成成比例,并因此与HEV复制成比例。用HEV-p6GLuc RNA或非复制型HEV-p6GLuc基因组(HEV-p6GLucGAD)电穿孔PLC1、PLC3和亲本PLC/PRF/5细胞。在这三种细胞系中HEV-p6GLuc的复制水平随时间稳定增加(数据未显示),但与PLC/PRF/5细胞相比,PLC1和PLC3细胞显示出更高的HEV复制倍数增加。之后使用PLC1和PLC3细胞进行实验。
我们在动力学实验中评估了PLC1和PLC3细胞表达病毒蛋白和产生感染性颗粒的能力。用HEV-p6RNA菌株电穿孔PLC1和PLC3细胞6。用抗ORF2和抗ORF3抗体的免疫荧光显示,超过80%的细胞表达病毒蛋白(数据未显示),表明PLC1和PLC3细胞是高度可转染的,并且可能在这些细胞中发生病毒基因组的强烈复制和表达。必须注意的是,在Sofosbuvir(SFV)存在下,ORF2表达被抑制(数据未显示),证实了信号的特异性。
我们接下来通过蛋白质印迹(WB)在不同时间点分析了HEV-p6电穿孔的PLC1和PLC3细胞的细胞裂解物和上清液中ORF2和ORF3蛋白的表达。重要的是,早在电穿孔后(p.e.)第2天和第4天,就在PLC1细胞的细胞裂解物和上清液中检测到ORF2蛋白(数据未显示)。在HEV-p6电穿孔的PLC3细胞中,ORF2蛋白的表达略微延迟。必须注意的是,PLC3细胞比PLC1细胞生长更慢,这可以解释观察到的差异。除主要产物ORF2外,还在细胞和上清液中检测到具有较低分子量的ORF2相关蛋白,表明ORF2衣壳蛋白可能经历翻译后修饰。也早在p.e.的第2天和第4天,就分别在PLC1和PLC3细胞裂解物中检测到ORF3蛋白(数据未显示)。相反,在电穿孔细胞的上清液中微弱地检测到ORF3蛋白,表明该蛋白主要在细胞内表达。
上清液中的主要ORF2产物(数据未显示)显示出比细胞内形式更高的表观分子量(数据未显示),表明高度分泌的ORF2蛋白可能经历翻译后修饰。细胞内和细胞外ORF3蛋白的比较显示,分泌的ORF3蛋白比细胞内蛋白迁移略快(数据未显示),可能反映蛋白磷酸化的差异10或未描述的ORF3蛋白修饰。
还监测了在p.e.不同天数收集的转染细胞的上清液中的病毒RNA和感染性颗粒。通过RT-qPCR测定HEV RNA水平。早在p.e.第2天检测到高RNA水平(数据未显示)。RNA滴度逐渐增加,并且在第10天对于PLC1和PLC3细胞分别达到1.1×108和3.3×107拷贝/ml(数据未显示)。同时,测定感染性病毒滴度,其对于PLC1和PLC3细胞分别达到7×103和1.5×103ffu/ml。这些结果表明,感染性病毒颗粒的组装在非常早期发生,并且在HEV-p6电穿孔的PLC1和PLC3细胞中同样发生。然而,PLC1细胞的HEV感染性为1.5x104RNA拷贝/ffu,PLC3细胞的HEV感染性为2.2x104RNA拷贝/ffu,表明感染性HEV颗粒的组装可能是低效的过程。然而,我们不能排除病毒或细胞抑制剂阻断HEV感染并使感染性滴度偏差的可能性。使用PLC3细胞进行进一步的实验。
ORF2衣壳蛋白大量产生,但只有一小部分组装成感染性颗粒。
为了产生大量的感染性上清液,我们将HEV-p6转染的PLC3细胞培养了47天。合并上清液,浓缩,并在碘克沙醇梯度上分级。分析每个级分的ORF2蛋白、ORF3蛋白、RNA和感染性病毒颗粒的分布(数据未显示)。从级分2-7中检测到ORF2蛋白,但在级分3-5中检测到更多(数据未显示)。如在转染细胞的上清液中观察到的(图1C),检测到级分2-5中的ORF2蛋白为两种产物,一种主要产物为约90kDa(ORF2g),较小产物为约75kDa(ORF2c)。在级分2-5的梯度上层也检测到具有较低分子量的ORF2相关蛋白,这可能对应于蛋白质的其他加工形式。相反,在级分6和7中检测到的ORF2蛋白主要是80kDa产物(ORF2i),其对应于细胞内ORF2的大小(图1C)。ORF3蛋白仅在级分5-7中检测到,但在级分6中最丰富(数据未显示)。有趣的是,在级分6中仅检测到一个主要的RNA峰,密度为1.10g/ml(数据未显示)。因此,大量的衣壳蛋白(级分2-4)与病毒RNA无关,而ORF3蛋白则相关。
通过感染A549细胞分析每个级分的感染性。在感染后5天通过WB(数据未显示)和间接免疫荧光(数据未显示)分析ORF2和ORF3蛋白的表达。在用级分5、6和7感染的细胞的裂解物中检测到ORF2i和ORF3蛋白(数据未显示),表明感染性颗粒与这些级分相关。实际上,用级分5、6和7接种的A549细胞的免疫荧光染色对于ORF2蛋白表达是阳性的,而用级分1-4和级分8-12接种的细胞是阴性的(数据未显示)。级分6显示出最高的感染滴度(5×106ffu/ml)。虽然在用级分3和4接种的细胞中检测到ORF2g和ORF2c蛋白(数据未显示),但通过免疫荧光未观察到这些级分的特异性感染(数据未显示),表明ORF2g和ORF2c可能是结合细胞表面的非常稳定的蛋白质,其可在孵育数天后检测到。
必须注意的是,在感染性部分中检测到外泌体CD81四跨膜蛋白(数据未显示),支持了HEV颗粒可能利用外泌体分泌途径进行出口这一假设11。
总之,我们的结果表明,在HEV生命周期中,ORF2衣壳蛋白大量产生,但只有一小部分(ORF2i)组装成通过外泌体途径分泌的感染性颗粒。
用HEVcc颗粒感染人肝嵌合小鼠
近来,人肝嵌合小鼠已被描述为用于研究体内慢性HEV感染和评估抗病毒分子的有价值的模型9,12-14。在接种gt1或gt3病毒粒子后,在嵌合小鼠中建立HEV感染。然而,用源自未处理的细胞培养物上清液的颗粒接种小鼠不会导致强烈的感染。因此,我们接下来评估我们的HEVcc颗粒是否能够感染原代人肝细胞(PHH)移植的小鼠9。通过脾内途径用以下接种嵌合小鼠:在碘克沙醇梯度上纯化的HEV-p6的级分6(F6p6)(图2),或用未转染的PLC3细胞上清液制备的对照梯度的级分6(F6对照,neg 1)(数据未显示)。将用gt1粪便悬浮液(Sar55)接种的人嵌合小鼠用作阳性对照9。用F6p6接种的非移植小鼠作为第二阴性对照(neg 2)。虽然程度低于感染gt1的小鼠,但接种F6p6的嵌合小鼠仍然显示出活动性感染的体征(数据未显示)。实际上,在第2周至第10周,在F6p6接种的嵌合小鼠的粪便中检测到HEVRNA,并且从第8周到第10周显著增加(数据未显示)。还在小鼠肝脏(数据未显示)和小肠内容物(数据未显示)中检测到基因组HEV RNA。如先前所述,与接种gt1的小鼠相比,接种F6p6的小鼠中的RNA载量增加得更慢9。但是我们首次证明了用来自细胞培养物上清液的HEV颗粒接种的嵌合小鼠的强烈感染。
由于HEV-p6菌株含有赋予生长优势的人S17核糖体蛋白片段6,15,我们接下来对来自F6p6感染的小鼠的HEV-p6区域进行测序(粪便,p.i.第9周和一个肝片),并将其与接种物比较。我们未发现S17区域中任何适应性突变的逆转(数据未显示)。
HEV颗粒的超微结构。
最近,基于丙型肝炎病毒(HCV)颗粒的在透射电子显微镜(TEM)网格上的直接特异性免疫捕获(IC)的新策略首次导致其超微结构的精确描述16。为了定义HEVcc颗粒的形态(其从未被精确描述),我们对分离的感染性颗粒使用相同的方法。我们首先分析了在涂有聚-L赖氨酸(一种聚阳离子附着因子)的网格上捕获的材料。我们观察到相当均匀的大小为40-70nm的颗粒群(数据未显示)。我们接下来用ORF3蛋白特异性抗体进行IC,该抗体已被描述为暴露于脂质相关的HEV颗粒上。我们观察到颗粒具有明显的内部二十面体亚结构,其可能对应于衣壳(数据未显示)。大小分布的计算表明,粒径范围为40nm至70nm。相反,当使用抗ORF2(1E6)(数据未显示)或同种型对照抗体(数据未显示)时,未观察到颗粒。然而,当在使用抗ORF2抗体进行IC前,先通过脱氧胆酸钠和胰蛋白酶(DT)处理将病毒制剂部分脱脂时,免疫捕获了两个颗粒群:主要群体由大小为30-50nm的颗粒组成,更加异质的群体由90-140nm的颗粒组成(数据未显示)。因此,DT处理揭露了病毒颗粒上的1E6表位,并导致可能对应于裸病毒粒子的小HEV颗粒的IC。然而,DT处理没有完全去除ORF3和相关脂质,因为一些颗粒仍然被抗ORF3抗体捕获(数据未显示)。这些颗粒的较大尺寸可能是由于它们的脂质包层的分离。相反,当在IC之前在密度梯度上纯化DT处理的颗粒时(数据未显示),用1E6免疫捕获了高度均质的约32nm的颗粒群,而IC抗ORF3没有捕获颗粒。这些颗粒的密度为1.18g/ml,显示出可能与裸衣壳相对应的二十面体结构。
我们接下来分析了来自三位HEV感染患者的血清(HEVser)的真实颗粒的超微结构。对于HEVcc,IC抗ORF2或用无关抗体的IC没有观察到颗粒(数据未显示)。相反,IC抗ORF3导致多形性群体的有效分离。HEVser1和HEVserST-1颗粒具有与HEVcc颗粒相似的形态,而HEVser2颗粒显示出可能对应于脂质的厚外层(数据未显示)。免疫捕获颗粒的大小分布的计算显示,HEVser颗粒的平均直径和中值直径通常大于HEVcc颗粒,并且患者之间有所不同(数据未显示)。来自HEVser2的颗粒最大,平均直径为120nm。这些结果表明,颗粒的脂质含量可能决定颗粒大小,如对于HCV颗粒所述16。
最后,如上所述,我们分析了来自HEV感染患者的粪便的颗粒的超微结构。与HEVcc和HEVser相反,粪便颗粒被IC抗ORF2捕获,而IC抗ORF3没有捕获颗粒(数据未显示)。尽管颗粒似乎缠绕在杂质中,限制了大小分布计算,但HEV粪便颗粒的平均直径为28nm(n=34,平均值=28nm,中值=28nm,SD=4nm),这与先前的观察一致17。对于脱脂的HEVcc,粪便颗粒显示出可能对应于裸衣壳的二十面体结构。
表征ORF2蛋白的不同形式
我们开发了一种使用20%碘克沙醇垫的纯化系统,可以容易地分离感染性颗粒(ORF2i,图1A,沉淀)和与感染性颗粒无关的ORF2g/ORF2c蛋白(图1A,上层)。
HEV产生大量分泌的ORF2g/ORF2c蛋白,其可能是ORF2蛋白的糖基化/加工形式。相反,通过碘克沙醇梯度(F6p6)或垫(沉淀/垫)纯化的感染性颗粒相关ORF2蛋白(ORF2i)显示出与细胞内ORF2蛋白(细胞)相同的大小(图1B),表明ORF2i可能不经历翻译后修饰。
由于ORF2蛋白质序列含有三个可能的N-连接糖基化位点和多个O-连接糖基化位点,我们接下来分析了ORF2蛋白质对不同糖苷酶处理的敏感性(图1C)。内切糖苷酶H(EndoH)在来自N-糖蛋白的高甘露糖和一些杂合寡糖的壳二糖核心内切割。肽-N-糖苷酶F(PNGaseF)在来自N-糖蛋白的高甘露糖、杂合和复合寡糖的最内部N-乙酰基葡糖胺和天冬酰胺残基之间切割。O-糖苷酶(O-Gly)催化一些O-连接的二糖的去除。神经氨酸酶(Neura)催化N-和O-糖蛋白的唾液酸残基的水解。应注意,末端唾液酸阻断O-Gly的作用。从细胞裂解物或纯化的HEV颗粒表达的ORF2i蛋白对糖苷酶消化具有抗性(图1C),表明该蛋白不是N-或O-糖基化的。相反,如通过处理后的迁移变化证明,分泌的ORF2g/ORF2c蛋白对EndoH和PNGaseF糖苷酶消化敏感,表明这些蛋白是N-糖蛋白。ORF2g/ORF2c蛋白对O-Gly处理具有抗性,但对Neura处理敏感,对Neura和O-Gly的组合更敏感。这些结果表明ORF2g/ORF2c蛋白是唾液酸化的O-糖基化蛋白。重要的是,ORF2g/ORF2c蛋白通过雪花莲凝集素(GNA)pull-down沉淀,所述GNA是与高甘露糖型聚糖相互作用的凝集素,证实了ORF2g/ORF2c N-聚糖的寡甘露糖苷性质(图1D)并且允许ORF2g/ORF2c蛋白被容易地特异性沉淀。
我们证明了分泌的ORF2g/ORF2c是糖基化蛋白质,表明这些蛋白质通过分泌途径。然而,在细胞内水平未检测到ORF2g/ORF2c蛋白。因此,我们假设ORF2g/ORF2c蛋白可能快速通过分泌途径,并在没有细胞内积累的情况下快速分泌。因此,我们用布雷菲德菌素A(BFA)(一种蛋白质分泌抑制剂18)处理表达HEV-p6的PLC3细胞(图1E)。在BFA处理后,ORF2g/ORF2c蛋白的分泌被阻断。有趣的是,在经BFA处理的细胞的裂解物中检测到可能对应于ORF2g的扩散条带和可能对应于ORF2c的微弱条带,表明在阻断蛋白质分泌后,ORF2g/ORF2c蛋白在细胞中积累,从而证实了我们的假设。
我们接下来通过质谱法分析了ORF2蛋白的序列。通过SDS-PAGE解析在碘克沙醇垫上纯化的病毒颗粒和用抗ORF2抗体(4B2)免疫沉淀的ORF2g/ORF2c蛋白。用胰蛋白酶或AspN在凝胶中消化WB2中对应于ORF2i、ORF2g和ORF2c的胶体蓝染条带(图2),然后通过nanoLC-MS/MS分析。图2示出了鉴定的肽和序列覆盖。对于这三种蛋白质,C-末端完全被覆盖,证明在它们的C-末端没有加工。对于ORF2i蛋白,意外地鉴定了覆盖一半信号肽(SP)的半胰蛋白酶肽(图2A,虚线),表明ORF2i的SP可能没有被切割。对于ORF2g,鉴定了以Ser34开始的胰蛋白酶和半AspN肽,表明ORF2g蛋白的第一个aa可能对应于Ser34(图2B)。对于ORF2c,鉴定了以Iso234开始的胰蛋白酶肽,表明ORF2c蛋白的第一个aa可能对应于与Iso234接近的aa之一(图2C)。由于在蛋白质组学分析中半胰蛋白酶和半AspN肽对应于天然蛋白质加工或非特异性蛋白水解事件,我们进一步用N-琥珀酰亚胺基氧基羰基甲基三(2,4,6-三甲氧基苯基)溴化鏻(TMPP-Ac-OSu,其特异性结合完整蛋白质的N-末端19)进行标记。用TMPP-Ac-OSu修饰鉴定的肽证实了ORF2i和ORF2g的第一个aa分别对应于Leu14和Ser34(图1F,2)。相反,由于TMPP-Ac-OSu标记未鉴定ORF2c的第一个aa,因此需要进一步研究以阐明该观察结果。总之,这些数据表明,ORF2g蛋白质失去其SP,并且可能由分泌途径蛋白酶加工。相反,ORF2i蛋白不通过信号肽酶加工,因此可能不会转移到ER腔中。
ORF2g/ORF2c蛋白是HEV感染患者血清中的主要抗原
由于我们证明了在细胞培养中,HEV主要产生与感染性颗粒无关的糖基化形式的ORF2蛋白,我们接下来试图确定在感染患者中是否发生相同的情况。如上所述,来自17名感染HEV患者和来自5名HEV阴性患者的血清通过GNA pull-down沉淀并用探针检测ORF2蛋白(图3A)。PLC3/HEV-p6上清液的GNA pull-down用作阳性对照。异乎寻常地,17个HEV阳性血清中的13个和7个分别显示大量的ORF2g和ORF2c蛋白。在HEV阴性血清中未检测到ORF2蛋白。此外,ORF2g/ORF2c蛋白的检测既不依赖于患者的HEV毒株也不依赖于血清病毒载量(表1)。重要的是,在碘克沙醇梯度上对患者血清(P6患者)进行分级,然后对每个级分进行GNApull-down(图3B)表明,如在细胞培养中一样,非常大量的ORF2g/ORF2c蛋白在轻度级分(级分4和5)中被分离,并通过GNA拉下来。然而,这些级分与感染性物质无关,因为它们与病毒基因组无关(图3C)。应当注意,由于存在大量人白蛋白,存在于级分4中的ORF2蛋白可能不会通过GNA珠沉淀。我们的结果表明,ORF2g/ORF2c蛋白可能是HEV感染患者血清中的主要抗原。因此,我们使用HEV抗原ELISAPlus测定法对梯度的每个级分中的ORF2蛋白进行定量(图3D)。有趣的是,在含有ORF2g/ORF2c蛋白的级分4中检测到最高量的抗原,而在其他级分中,尤其是级分7(感染性级分)中检测到较低量的抗原。总之,我们的结果表明,在感染患者中,HEV产生大量与感染性颗粒无关的糖基化抗原蛋白,并且可能导致活动性HEV感染诊断的偏差。

讨论
通过高度复制且细胞培养适应的p6菌株6和高度可转染的PLC/PRF/5细胞亚克隆的组合,我们开发了一种新的细胞培养系统,在转染后很早期就检测到病毒复制和蛋白质表达。时间过程实验表明,如最近报道的20,21,ORF2蛋白早期大量分泌到转染细胞的上清液中。有趣的是,当在细胞裂解物或上清液中检测到时,ORF2和ORF3蛋白的迁移不同,表明这些蛋白质在其分泌过程中可能经历翻译后修饰。需要进一步的实验来鉴定ORF3蛋白中的这种修饰。ORF2蛋白含有三个可能的N-连接糖基化位点和多个O-连接糖基化位点。使用表达载体,先前已经显示ORF2蛋白被糖基化并在细胞表面上表达22-24,但不清楚糖基化的ORF2蛋白是否是病毒粒子的天然形式。最近,有人提出HEV病毒粒子衣壳可能是糖基化的20,尽管Graff等显示相反结果25。我们的研究表明,HEV产生大量名称为ORF2g和ORF2c的ORF2蛋白,其是分泌的、唾液酸化的、N-和O-糖基化的,但与感染性病毒粒子无关。大部分ORF2蛋白可能转位到ER腔中,在那里它们被N-糖基化并且可能通过蛋白酶加工以产生ORF2g和ORF2c蛋白。这两种蛋白质迅速通过其中它们被O-糖基化且唾液酸化的分泌途径,然后迅速分泌。有趣的是,在ORF2g N-末端上游存在的RGRR残基表明,弗林蛋白酶样蛋白酶可能参与其成熟。需要进一步的实验来表征ORF2序列中的糖基化和加工位点,特别是导致产生ORF2c蛋白的机制。相反,在细胞内水平和病毒粒子中观察到的ORF2i蛋白可能不会转位到ER腔中,并停留在细胞质区室中。我们的结果表明,存在两条生产HEV衣壳蛋白的途径:(i)主要的非生产途径,其中ORF2蛋白被递送至分泌途径,在那里它们被加工并快速分泌。(ii)将细胞质ORF2蛋白递送至病毒粒子装配位点的生产途径。需要进一步的研究来深入研究这些途径。
与先前的研究9,13相反,我们成功地用gt3HEVcc颗粒感染了嵌合小鼠。通过密度梯度从ORF2g/ORF2c蛋白中分离感染性颗粒可以解释我们的成功,因为这些蛋白质使用待阐明的机制干扰病毒粒子感染靶细胞的能力。
我们发现,在其生命周期中,HEV高度分泌ORF2蛋白的糖基化形式,其在感染患者中循环并且是患者血清中的主要抗原。定义哪种形式的ORF2蛋白被已解决其感染的患者的抗体识别将是有趣的。HEV可以产生ORF2g/ORF2c蛋白作为免疫诱饵。有趣的是,使用TEM分析,我们发现,与其中表面抗原形成亚病毒颗粒的其他病毒(如乙型肝炎病毒)相反,ORF2g/ORF2c蛋白不形成颗粒物质(数据未显示)。
我们的TEM分析显示,HEVcc是大小为40-70nm的颗粒,在其表面显示内部结构和ORF3蛋白。DT处理和超速离心暴露了类似于在患者粪便中发现的那些的小二十面体衣壳(17和我们的研究),表明HEVcc与其中嵌入ORF3的脂质相关。我们的分析表明,尽管HEVser颗粒大得多,但其形态与HEVcc颗粒相似。HEVser病毒粒子在其表面显示ORF3蛋白,并且可能与脂质高度相关。由于HEVser直径在患者之间存在差异,因此确定观察到的大小变化是否与脂质含量相关将是有趣的,如最近对HCV颗粒所证实的16。
最近,HEV-Ag-ELISA测定上市销售用于诊断HEV感染。该测定的比较研究显示高水平的特异性和敏感性26-29。然而,在一些样品中发现抗原检测和RNA定量之间存在一些差异,其中在没有可检测的RNA的情况下检测到HEV抗原27-29。在我们手中,抗原测定也非常敏感。然而,我们发现,与感染性病毒粒子无关的ORF2g/ORF2c蛋白是该试剂盒识别的主要抗原。总之,这些数据表明必须小心解释用HEV-Ag-ELISA测定获得的结果。实际上,这种测定可能不一定能检测到活动性感染,因为ORF2g/ORF2c蛋白在细胞培养中似乎非常稳定,但也可能在患者血清中非常稳定28,这表明即使HEV在肝脏中不再复制,这些蛋白也可能保留在患者血清中。
实施例2
制备抗ORF2i蛋白的特异性抗体。合成衍生自本发明多肽(ORF2i)的肽(P1),并将其与KLH偶联。用P1肽以3周间隔免疫接种5只小鼠三次。在免疫接种期间使用弗氏完全佐剂和不完全佐剂。通过皮下和腹膜内途径免疫接种动物。第三次免疫后10天,使小鼠出血并测试其血清的免疫反应性。首先通过间接ELISA在涂有P1肽的平板上测定血清(数据未显示)。接下来在蛋白质印迹实验中用ORF2蛋白的混合物(ORF2i和ORF2g/ORF2c)作为抗原分析它们的特异性(图4)。与识别三种形式的1E6抗体相比,五种血清中的两种(P1S2和P1S5)显示出对ORF2i蛋白的高度特异性识别,其中与ORF2g/c蛋白没有交叉反应(图4)。单克隆抗体的克隆和生产正在进行中。
这些结果表明,可以产生特异性抗ORF2i多肽的抗体。这种抗体非常适合于测定样品中戊型肝炎病毒的感染性颗粒的存在。更具体地,本发明的ORF2i或ORF2g多肽的检测适用于诊断受试者的戊型肝炎病毒感染。

Claims (18)
1.分离的戊型肝炎病毒ORF2衣壳(ORF2i)多肽,其由与SEQ ID NO:1所示氨基酸序列具有至少90%同一性的氨基酸序列组成。
2.权利要求1所述的分离的戊型肝炎病毒ORF2衣壳(ORF2i)多肽,其未被糖基化。
3.分离的戊型肝炎病毒ORF2衣壳(ORF2g)多肽,其由与SEQ ID NO:2所示氨基酸序列具有至少90%同一性的氨基酸序列组成。
4.权利要求3所述的分离的戊型肝炎病毒ORF2衣壳(ORF2g)多肽,其是糖基化的。
5.编码权利要求1或3所述的多肽的核酸分子。
6.对权利要求1或3所述的多肽具有特异性的抗体。
7.权利要求6所述的抗体,其是单克隆抗体或多克隆抗体。
8.用于检测样品中戊型肝炎病毒的感染性颗粒的存在的方法,包括在允许抗体和感染性颗粒形成免疫复合物的条件下,将样品与对权利要求1所述的ORF2i多肽具有特异性的权利要求6所述的抗体接触,其中检测到免疫复合物表明样品中存在感染性颗粒。
9.权利要求8所述的方法,其中所述样品选自血液、腹水、尿、唾液、汗液、乳汁、滑液、腹膜液、羊水、percerebrospinal fluid、淋巴液、肺栓塞、脑脊液、心包液和粪便。
10.权利要求8所述的方法,进一步包括将样品与对权利要求3所述的ORF2g多肽具有特异性的权利要求6所述的抗体接触。
11.权利要求8所述的方法用于诊断戊型肝炎病毒(HEV)感染的用途。
12.权利要求11所述的用途,其中所述HEV感染是急性HEV感染、近期HEV感染、慢性HEV感染或弱活性HEV感染。
13.权利要求8所述的方法用于诊断清除的HEV感染的用途,其特征在于检测到ORF2g多肽,任选地一起检测到ORF2c多肽,但没有检测到ORF2i多肽。
14.包含至少一种如权利要求6所述的抗体的试剂盒或装置。
15.权利要求14所述的试剂盒或装置,其包含对权利要求1所述的ORF2i多肽具有特异性的抗体和对权利要求3所述的ORF2g多肽具有特异性的抗体。
16.权利要求14所述的试剂盒或装置,其是ELISA测定试剂盒。
17.权利要求14所述的试剂盒或装置,其中所述抗体固定在固体支持物上。
18.权利要求14所述的试剂盒或装置,其包含产生可检测信号的第二个权利要求6所述的抗体。
