Abstract
The invention discloses a culture dish sealing method, which is used for sealing a culture dish of 6-12 cm by using a winding film and comprises the following steps: (1) cutting the length direction of a winding shaft of the winding film by using a modified LZ-manual cutting machine, cutting into an annular ring with the width of 3cm, and placing on a super-clean working table for sterilization; (2) and (3) taking a 6-12 cm glass culture dish, sealing the cover opening by using the sterilized winding film in the step (1), closely pressing the winding film at the joint of the bottom of the culture dish and the cover opening during sealing, rotating the culture dish for 1.5-2 weeks, and then slightly pulling the winding film to break the winding film to finish sealing. The invention uses the 'hand stretching and winding film' to replace the traditional sealing film and preservative film, can reduce the limitation of sealing, can seal glass culture dishes and plastic culture dishes of 6cm, 9cm and 12cm, does not need to be cut, is not easy to break and age, saves time and labor and greatly reduces the cost.

Technical Field
The invention relates to a sealing method of a culture dish.
Background
At present, a microorganism laboratory generally uses a culture dish to culture fungi, bacteria, actinomycetes, and the like. At present, some laboratories do not seal culture dishes, and although time and labor are saved, the culture dishes are easy to pollute. Some laboratories use a sealing film imported from the united states (Parafilm M) for sealing, but have the disadvantages of time consuming, difficult cleaning, high cost, etc. Therefore, a sealing film which is time-saving, labor-saving, excellent in quality and low in cost is urgently needed to replace the sealing film.
Currently, most laboratories use an inlet sealing membrane to seal the petri dish (attachable photos). The sealing film is in a reel shape and has the size of 4 inches × 125 feet (namely 10 cm × 3810 cm), the sealing film is cut into strips of 10 cm × 3cm, and the sealing film is stretched to seal the culture dish interface, so that the sealing film is suitable for sealing culture dishes with the diameter of 9cm and below.
The sealing film has the following defects: 1. it takes time. The sealing film needs to be cut before sealing, which takes a lot of time. 2, not easy to clean. The sealing film is easy to age, the microorganism is cultured for more than 15 days, the sealing film is easy to age and break, the sealing film is not easy to tear off during cleaning, the cleaning difficulty is increased, and the cleaning time is prolonged. 3. Has limitations. A culture dish with a diameter of 120mm cannot be sealed. 4. The cost is high. The sealing film is in a reel shape, the size of the sealing film is 4 inches × 125 feet (namely 10 centimeters × 3810 centimeters), the sealing film needs to be cut into strips of 10 centimeters × 3 centimeters when in use, 1270 strips can be cut, and the price of one reel of the sealing film is 150 yuan, so that each strip is 0.1181 yuan, and the cost of each sealed 100 culture dishes is 11.81 yuan.
Sealing a 6-9 cm glass culture dish by using a preservative film, wherein the operation steps are as follows: (a) cutting the common preservative film roll into annular rings with the width of 2-3 cm by using a scalpel, and placing the annular rings on an ultra-clean working table for disinfection and sterilization; (b) and (3) taking a 6-9 cm glass culture dish, sealing the cover opening by using the sterilized common preservative film in the step (a), tightly attaching the preservative film to the cover opening to be sealed at the bottom of the glass culture dish during sealing, rotating the culture dish for 2 circles, and slightly pulling the preservative film to tear off the preservative film to finish sealing [1 ].
The preservative film has the following defects: 1. thinner, easy to break and high in rework rate in use. 2. Disposable plastic petri dishes cannot be sealed. 4. 3, high cost. Taking a commercial preservative film skilful as an example, a roll of 26 yuan, which is 20cm × 30m in specification, is cut into annular rings with the width of 3cm according to requirements, and 7 rolls are used, the total length is about 30m × 7 to 210m, taking a culture dish with the diameter of 9cm as an example, two circles are needed for wrapping one culture dish, so the length is 3.14 × 9 × 2 to 57cm, the preservative film can wrap 368 culture dishes according to the specification, and the cost for sealing 100 culture dishes is 7 yuan.

The invention discloses a culture dish sealing method, which utilizes a winding film to seal a culture dish of 6-12 cm and comprises the following steps: the method comprises the following steps of sealing a 9cm glass culture dish by using a winding film: (1) cutting the winding film reel in length direction with a modified LZ-manual cutting machine into annular ring with width of 3cm, and sterilizing with ultraviolet rays for 30min on an ultraclean workbench; (b) and (3) taking a 9cm glass culture dish, sealing the cover opening by using the sterilized winding film in the step (1), closely pressing the winding film at the joint of the bottom edge of the culture dish and the cover opening during sealing, rotating the culture dish for 1.5-2 circles, and slightly pulling the winding film to break the winding film to finish sealing.
In the invention, the hand stretch wrapping film (also called PE stretch film) has higher tensile strength and tear strength, and good self-adhesion, and the adhesion is divided into single-sided adhesion and double-sided adhesion, so that an object can be wrapped into a whole. Meanwhile, the film has excellent transparency, and does not influence the growth of microorganisms needing illumination culture. Meanwhile, the composite material has good retractive force, the prestretching multiplying power is 400 percent, and the common specifications are as follows: the width is 30cm,35cm,45cm,50cm, and the thickness is 15um-50 um. According to the requirements of the invention, the steel plate needs to be cut into a width of 3cm by self.
The wound film is 50cm multiplied by 500m in price of 58 yuan, cut into annular rings with the width of 3cm according to requirements, and wound into 17 rolls, the total length is about 500m multiplied by 17 to 8500m, taking a culture dish with the diameter of 9cm as an example, 2 circles (or 1.5 circles) are needed for wrapping one culture dish, so the length is 3.14 multiplied by 9 multiplied by 2 to 57cm, the wound film can wrap 14912 culture dishes according to the specification, and the cost of each sealed 100 culture dishes is 0.39 yuan.

(2) and (3) taking a 6-12 cm glass culture dish, sealing the cover opening by using the sterilized winding film in the step (1), closely pressing the winding film at the joint of the bottom of the culture dish and the cover opening during sealing, rotating the culture dish for 1.5-2 weeks, and then slightly pulling the winding film to break the winding film to finish sealing.
2. A method of closing a culture dish according to claim 1, wherein the culture dish is a glass or plastic culture dish.
3. The method of claim 1, wherein the wrapping film is a PE stretch film.
