本发明提供了一种热稳定的甲肝疫苗可溶性微针贴,包括针体和背衬,所述的针体由甲肝疫苗、基质材料和疫苗稳定剂组成,其中,所述的疫苗稳定剂为聚乙烯吡咯烷酮和右旋糖酐的混合,所述的疫苗稳定剂与甲肝疫苗的含量比为(2:1)~(5:1)。本发明制备的甲肝疫苗可溶性微针贴,针体药物热稳定性好,机械强度适中,能有效刺入皮肤。

技术领域
本发明涉及疫苗的微针给药技术领域,提供了一种热稳定的甲肝疫苗可溶性微针贴及其制备方法。
背景技术
甲型病毒性肝炎,简称甲肝,是由甲型肝炎病毒引起的,以肝脏炎症病变为主的传染病,主要通过粪-口途径传播,任何年龄均可患本病,但主要为儿童和青少年,目前有效预防该病的措施是使用甲肝疫苗。目前的甲肝疫苗一般都是通过针头注射给药,患者顺应性差,因此亟待一种新型的疫苗给药途径。相比注射途径,口服依然存在着局限性,尤其是对于生物大分子类药物而言,容易受胃肠道的蛋白酶和肝脏的首过效应的影响导致生物利用度降低。除了注射和口服途径以外,透皮给药领域是目前热门的领域,而且皮肤作为人体最大的免疫器官,这使得疫苗与透皮给药领域的结合备受青睐。
阻碍透皮递送生物类大分子药物的关键因素是皮肤最外层的角质层障碍。微针是近十年发展迅速的物理促渗新技术,它可以打破皮肤的角质层障碍进入皮肤的表皮层而不触及真皮层的神经末梢,达到无痛微创给药,且给药效果突破了传统经皮给药制剂。根据不同的结构和功能特点,目前已有的微针可以分为以下四个类型即固体实心微针、中空微针、涂层微针以及可溶性微针。固体实心微针与中空微针虽然制备较为简单但是如果不小心断在皮内会造成有害残留;涂层微针载药量小且难以控制给药量;相比之下,可溶性微针具有载药量大,刺入皮内可以自行溶解且不造成有害残留等优点而被广泛研究。
公开号为KR20170032810A的韩国专利提供了一种将甲肝疫苗载入可溶性微针的技术方案,该专利虽然采用了一些水溶性聚合物来制备微针以及进行了药物释放行为的表征,但仅仅是简单罗列了可以采用哪些材料来制备可溶性微针的举例,并未针对解决甲肝疫苗可溶性微针给药的某一技术难题去筛选特定的处方与配比;而且,该微针的制备方法也是较为常规的一体化注模法。
此外,和甲肝疫苗类似的乙肝疫苗载入可溶性微针已有多篇文献进行报道。如:文献《Hepatitis B surface antigen incorporated in dissolvable microneedle arraypatch is antigenic and thermostable》(Poirier D,Renaud F,Dewar V,etal.Biomaterials,2017,145:256-265),文献《DNA-based vaccination againsthepatitis B virus using dissolving microneedle arrays adjuvanted by cationicliposomes and CpG ODN》(Qiu Y,Guo L,Zhang S,et al.Drug delivery,2016,23(7):2391-2398)等。众所周知,乙肝疫苗相比于甲肝疫苗活性成分发生了变化,则不同的基质材料与疫苗药物组合后,会导致疫苗可溶性微针的机械性能、存在效果差异,这也与文献《用于构建可溶性微针的基质材料及其复合材料》(章捷,马凤森,占浩慧等,材料导报,2017,31(19):129-134.)的结论一致。故,若借鉴乙肝疫苗可溶性微针处方来开发甲肝疫苗可溶性微针,仍存在诸多不确定性。
甲肝疫苗的本质是蛋白质,和一般的疫苗一样,属于温度敏感型。为了保护疫苗的活性成分不发生变性,往往需要加入适合的稳定剂,以保持疫苗在贮藏或干燥过程中的活性。根据文献(孙东坡,胡一桥.蛋白质冷冻干燥制品中的保护剂及其保护机制[J].药学进展,2003,27(4):201-205)和(李键.冻干保护剂在活疫苗生产中的应用进展[J].中国生物制品学杂志,2017,30(2):221-224)可知,不同的疫苗活性成分不同导致其理化性质不同,因此要根据具体的疫苗进行相应的处方筛选才能达到较好的保护效果;任何单一的保护剂都不可能具备所有的保护特性,因此合用保护剂的效果往往大于单一保护剂的效果;保护剂对某一疫苗的保护作用通常有一个最佳的使用浓度,且其对微针的刺入性能有影响,故筛选保护剂时还应考虑合理的使用量。
综上所述,如何开发出一种处方适宜、具有良好热稳定性、机械强度适中的甲肝疫苗可溶性微针贴,是本领域技术人员急需解决的技术难题。
发明内容
本发明目的在于提供了一种热稳定的甲肝疫苗可溶性微针贴,解决了上述现有技术存在的瓶颈。

一种热稳定的甲肝疫苗可溶性微针贴,包括针体和背衬,所述的针体由甲肝疫苗、基质材料和疫苗稳定剂组成,其中,所述的疫苗稳定剂为聚乙烯吡咯烷酮和右旋糖酐的混合,所述的疫苗稳定剂与甲肝疫苗的含量比为(2:1)~(5:1)。
优选的,所述的疫苗稳定剂中聚乙烯吡咯烷酮与右旋糖酐的含量比为(3:1)~(1:3)。
优选的,所述的基质材料为聚乙烯醇。
优选的,所述的疫苗稳定剂中聚乙烯吡咯烷酮与右旋糖酐的含量比为2:1。
优选的,所述的针体中各组分及其含量占比如下:30wt%聚乙烯醇,37.3wt%聚乙烯吡咯烷酮,18.7wt%右旋糖酐,14wt%甲肝疫苗。
本发明还提供了一种热稳定的甲肝疫苗可溶性微针贴的制备方法,包括如下步骤:
(1)微针的阴模制备
先采用光刻和蚀刻工艺制备阳模微针,再将聚硅氧烷与固化剂按质量比6:1混合后,浇注到放有单晶硅阳模微针的长方体容器中;将容器置于真空干燥箱,抽真空使得混合液中的气泡去除;再放入烘箱,干燥3h后取出,得到成型的聚硅氧烷模具;
(2)第一次真空入模
将处方量的甲肝疫苗、基质材料、疫苗稳定剂按比例混合后,加入伊红水溶液,用适量溶剂溶解成均一的针体液;取适量针体液涂覆于步骤1)制得的阴模上,放入真空干燥箱,抽真空使针体液进入阴模的孔洞;取出阴模,刮净孔洞外的溶液;并放入烘箱中干燥2h;
(3)第二次真空入模
将背衬材料用适量溶剂溶解成均一的背衬液,取适量背衬液涂覆于步骤(2)制得的阴模上,放入真空干燥箱抽真空;并放入烘箱中干燥4h,脱模,即得。
本发明提供的热稳定性甲肝疫苗可溶性微针贴片及其制备方法,与现有技术相比,具有如下有益效果:
①本发明选择特定的组分与含量比的疫苗稳定剂(PVP与右旋糖酐)与甲肝疫苗结合,使得可溶性微针中的疫苗可以在高的温度下保持稳定。
②本发明选择特定的含量比的基质材料(聚乙烯醇)与甲肝疫苗和疫苗稳定剂结合,使得可溶性微针机械强度适中,刺入皮肤较为有效。
总之,本发明制备的甲肝疫苗可溶性微针贴,针体药物热稳定性好,机械强度适中,能有效刺入皮肤。

具体实施方式
为了使本技术领域的人员更好的理解本发明方案,下面将结合本发明中的附图,对本实施例中的技术方案进行具体说明。必须说明的是,下述实施例仅用来解释本发明,而不是对发明进行限制,在本发明的精神和权利要求的保护范围内,对本发明作出的任何修改和改变,都落入到了本发明的保护范围。
实施例1甲肝疫苗可溶性微针阴模具的制备方法
如图1,本发明采用光刻和蚀刻工艺结合倒模法制备甲肝疫苗微针的阴模,步骤如下:
1)单晶硅阳模微针的制备
采用光刻和蚀刻工艺制备阳模微针,微针针列为5*5,每根微针的高度为400μm,针的底部直径为200μm,针尖直径为20μm。
2)制备聚硅氧烷模具
将聚硅氧烷与固化剂按质量比6:1混合后,浇注到放有单晶硅阳模微针的长方体容器中;将容器置于真空干燥箱,于真空度0.1MPa下抽真空3min,使得混合液中的气泡去除;再放入烘箱,于80℃下干燥3h后取出,得到成型的聚硅氧烷模具。
实施例2甲肝疫苗微针的制备
如图2,本发明采用二次真空入模法制备甲肝疫苗微针,步骤如下:
1)将基质材料、疫苗稳定剂以及甲肝疫苗按处方比例混合后,用适量溶剂(去离子水)溶解成均匀的混合液(即针体液),取适量针体液装入离心管,置于离心沉淀器,离心使得针体液中的气泡去除,静置备用。
2)取步骤1)离心后所得的针体液,浇注于本发明实施1制备的聚硅氧烷模具上,再将聚硅氧烷模具置于真空干燥箱,于真空度0.1MPa下3min,使得针体液注入模具的孔洞中;完毕后,取出模具,去除模具表面的溶液仅保留孔洞中的溶液;将上述装有溶液的模具置于37℃烘箱中干燥2h。
3)将背衬层材料用溶剂(去离子水)溶解成均匀的液体(即背衬液),取适量背衬液装入离心管,置于离心沉淀器,离心使得背衬液中的气泡去除,静置备用。
4)取步骤3)离心后所得的背衬液,浇注于步骤2)所得的聚硅氧烷模具上,再将聚硅氧烷模具置于真空干燥箱中,于真空度0.1MPa下3min;完毕后,取出模具,放入烘箱,于37℃下干燥4h后取出,脱模,即得成品甲肝疫苗微针贴片。
实施例3不同疫苗稳定剂对甲肝疫苗稳定性的影响
假定疫苗稳定剂在针体总质量中的占比为一定值,仅改变疫苗稳定剂的组分,考察如表1所示的单一或不同组合的疫苗稳定剂对微针中甲肝疫苗稳定性的影响,本实施例涉及的微针与阴模,其制备方法参照本发明的实施例1和实施例2。
微针中疫苗稳定性的表征方法:将按表1各处方制得的微针,置于10mL 37℃去离子水中溶解,用ELISA(酶联吸附免疫测定)法检测溶液中的疫苗抗原含量,记录为初始0月疫苗抗原含量(疫苗稳定性视为100%),另将前述同一批次制得的各种微针,放置在温度37℃、相对湿度30%的空气环境中保存3个月后,再用相同的ELISA法检测各微针疫苗的抗原含量相对于初始时的百分比,记为疫苗抗原含量百分比。
根据我们的大量预实验结果,当甲肝疫苗在温度37℃、相对湿度30%的空气环境中保存3个月后,疫苗抗原含量百分比依然高于75%则视为符合要求。

根据表2的甲肝疫苗活性数据可知:是否在可溶性微针中加入疫苗稳定剂对疫苗的稳定性有很大的影响;单一的耐热保护剂的效果逊色于复合耐热保护剂的效果,其中,右旋糖酐和PVP的组合(即处方6)的疫苗活性最高,表明其对疫苗的保护作用最好。分析其原因,作为疫苗稳定剂的原理,目前有“水替代”假说和“玻璃态”假说,处方6采用右旋糖酐和PVP的组合,由于右旋糖酐作为多羟基化合物会与甲肝疫苗以氢键形式结合形成“类水化膜”,而且PVP的加入提高了微针体系中的玻璃化转变温度,使得处方6的对疫苗的保护作用最佳。因此,本发明的稳定剂选择右旋糖酐和PVP的组合。
实施例4疫苗稳定剂比例对微针中的甲肝疫苗稳定性影响实验
在实施例3的基础上,进一步考察不同疫苗稳定剂比例对微针中的甲肝疫苗稳定性影响的差异。
假定疫苗稳定剂在针体总质量中的占比为一定值,且稳定剂的组分固定,仅改变疫苗稳定剂的比例,考察如表3所示的不同比例的稳定剂对微针性能的影响。
同理,本实施例涉及的微针与阴模,其制备方法参照本发明的实施例1和实施例2。对表3中各微针处方的疫苗抗原含量检测,测试步骤及考察标准参照本发明的实施例3。

结合表3和表4可知,
1)处方1和7由于疫苗抗原含量低于75%,故不符合要求。
2)PVP:右旋糖酐的含量为(3:1)~(1:3)时,均符合要求。
3)当疫苗稳定剂组分PVP与右旋糖酐比例在2:1时(即处方3),甲肝疫苗活性最高,对疫苗的保护作用最佳(将处方3与处方5进行配对t检验,p=0.007<0.05,具有显著性差异)。
实施例5复合疫苗稳定剂与疫苗不同比例对微针中疫苗稳定性的影响
假定基质材料的质量比例以及复合疫苗稳定剂的组分比例(PVP:右旋糖酐=2:1)为一定值,仅改变甲肝疫苗与疫苗稳定剂的比例,考察如表5所示的不同比例的复合疫苗稳定剂与疫苗对微针中疫苗稳定性的影响。
同理,本实施例涉及的微针与阴模,其制备方法参照本发明的实施例1和实施例2。对表5中各微针处方的疫苗抗原含量检测,测试步骤及考察指标参照本发明的实施例3。

根据参考文献(孙东坡,胡一桥.蛋白质冷冻干燥制品中的保护剂及其保护机制[J].药学进展,2003,27(4):201-205)可知,疫苗稳定剂的用量有一个最佳浓度,过高或过低都会导致疫苗稳定作用下降,处方1和6由于疫苗抗原含量低于75%,故不符合要求;处方2、3、4、5符合要求,即疫苗稳定剂与甲肝疫苗的含量比为(2:1)~(5:1)。
结合表5和表6可知,随着疫苗稳定剂与甲肝疫苗的比例的上升,甲肝疫苗的稳定性先上升后下降,当比例为(4:1)时,稳定剂保护作用最佳(将处方3和4进行配对t检验,p=0.008<0.05,具有显著性差异)。
实施例6基质材料质量占比对微针机械性能的影响
假定甲肝疫苗、复合疫苗稳定剂的质量比例为一定值,仅改变基质材料的占比,考察如表7所示的不同载药量的甲肝疫苗对微针性能的影响。
同理,本实施例涉及的微针与阴模,其制备方法参照本发明的实施例1和实施例2。
微针机械性能测试:由实施例2可知,在每个微针处方的基础上加入了伊红染料水溶液作为刺入指示剂。将每片制得的甲肝疫苗可溶性微针贴片利用给药器以10N的力刺入离体猪耳皮肤1min后,取下微针贴片观察离体皮肤的红点数量并计算其与微针针数的比例,比例越高则微针机械强度越好。
表7不同占比的基质材料构成的微针处方及对微针机械性能的影响
由文献《用于构建可溶性微针的基质材料及其复合材料》(章捷,马凤森,占浩慧等,材料导报,2017,31(19):129-134.)可知,前述实施例所筛选的PVP和右旋糖酐均可作为微针构筑的基质材料,并且具有一定的韧脆性。随着基质材料聚乙烯醇占比上下波动,微针的机械性能将会发生改变,以此确定基质材料的优选占比。结果如表7所示,当基质材料聚乙烯醇的占比为30wt%时,微针成功刺入百分比最高,表明微针机械强度最佳。
综上所述,甲肝疫苗可溶性微针的最优处方为:30wt%聚乙烯醇,37.3wt%PVP,18.7wt%右旋糖酐,14wt%甲肝疫苗。
Claims (4)
1.一种热稳定的甲肝疫苗可溶性微针贴,包括针体和背衬,其特征在于,所述的针体由甲肝疫苗、基质材料和疫苗稳定剂组成,其中,所述的基质材料为聚乙烯醇,所述的疫苗稳定剂为聚乙烯吡咯烷酮和右旋糖酐的混合,疫苗稳定剂中聚乙烯吡咯烷酮与右旋糖酐的含量比为(3:1)~(1:3);所述的疫苗稳定剂与甲肝疫苗的含量比为(2:1)~(5:1)。
2.根据权利要求1所述的疫苗稳定剂,其特征在于,聚乙烯吡咯烷酮与右旋糖酐的含量比为2:1。
3.根据权利要求1或2所述的热稳定的甲肝疫苗可溶性微针贴,其特征在于,所述的针体中各组分及其含量占比如下:30wt%聚乙烯醇,37.3wt%聚乙烯吡咯烷酮,18.7wt%右旋糖酐,14wt%甲肝疫苗。
4.如权利要求1所述的热稳定的甲肝疫苗可溶性微针贴的制备方法,其特征在于,包括如下步骤:
(1)微针的阴模制备
先采用光刻和蚀刻工艺制备阳模微针,再将聚硅氧烷与固化剂按质量比6:1混合后,浇注到放有单晶硅阳模微针的长方体容器中;将容器置于真空干燥箱,抽真空使得混合液中的气泡去除;再放入烘箱,干燥3h后取出,得到成型的聚硅氧烷模具;
(2)第一次真空入模
将处方量的甲肝疫苗、基质材料、疫苗稳定剂按比例混合后,加入少量的伊红水溶液,用适量溶剂溶解成均一的针体液;取适量针体液涂覆于步骤1)制得的阴模上,放入真空干燥箱,抽真空使针体液进入阴模的孔洞;取出阴模,刮净孔洞外的溶液;并放入烘箱中干燥2h;
(3)第二次真空入模
将背衬材料用适量溶剂溶解成均一的背衬液,取适量背衬液涂覆于步骤(2)制得的阴模上,放入真空干燥箱抽真空;并放入烘箱中干燥4h,脱模,即得。
