The invention relates to a device for rapidly extracting hydrogen sulfide from cerebrospinal fluid and serum samples and a using method thereof, wherein the device for rapidly extracting hydrogen sulfide from cerebrospinal fluid and serum samples comprises a20 mL closed injector containing a phosphoric acid solution, a centrifugal tube arranged at the needle head of the injector and a micro-sampling needle inserted into a rubber plug of the injector, wherein the centrifugal tube contains a sodium hydroxide solution, and the micro-sampling needle is used for absorbing a sample to be extracted; the method has the characteristics of simple operation, quick treatment, low cost and the like, and can quickly extract the hydrogen sulfide in the cerebrospinal fluid and the serum sample.

Technical Field

Background
Hydrogen sulfide (H)2S) is colorless, inflammable and extremely toxic gas with the odor of rotten eggs, and mainly comes from factories, mining industry and petrochemical industry. If the waste water containing hydrogen sulfide is not treated or is discharged beyond the standard, serious damage can be caused to the ecological environment, and even the health of human beings is seriously threatened. Recent studies have demonstrated that hydrogen sulfide is the third gas signaling molecule found following Nitric Oxide (NO) and carbon monoxide (CO), playing an important role in physiological processes such as nerve conduction, inflammation, vasodilation, apoptosis, cell growth regulation, and cardiovascular protection. The physiological level of hydrogen sulfide in mammalian serum is in the range of about 30-100. mu.M, while the level of hydrogen sulfide in brain can be as high as 160. mu.M. In addition, abnormal levels of hydrogen sulfide in vivo have been associated with a number of diseases such as down syndrome, alzheimer's disease, diabetes and cirrhosis of the liver. However, the cerebrospinal fluid and the serum contain a large amount of interferents which can affect the detection of the hydrogen sulfide, so that the selective extraction of the hydrogen sulfide in the serum and the brain tissue can avoid the influence of interfering species during detection, and the method has important significance for deeply understanding the physiological function of the serum and even for early diagnosis of diseases and cancers.
Disclosure of Invention
Aiming at a series of problems of complicated pretreatment, long time consumption, complex equipment operation, high cost and the like in the prior detection of hydrogen sulfide in complex environments of serum and cerebrospinal fluid, the invention provides a device for rapidly extracting the content of hydrogen sulfide in cerebrospinal fluid and serum samples and a using method thereof.
In order to achieve the purpose, the invention adopts the following technical scheme:

1) adding 5 mL of 1 mol/L phosphoric acid solution into a20 mL closed syringe;
2) a micro sample injection needle sucks 100 mu L of sample to be extracted, and a rubber plug is inserted into the lower end of the injector;
3) inserting the needle head of the syringe needle into a centrifugal tube;
4) 1 mL of 0.1M NaOH solution is added into the centrifugal tube;
5) and slowly pushing the sample in the micro sample injection needle into the injector to enable the sample to fully react with the phosphoric acid, slowly pushing the piston of the injector upwards until the upper liquid level of the phosphoric acid solution is contacted with the top in the injector without air when the phosphoric acid solution does not generate bubbles any more, and collecting the reacted sodium hydroxide solution in the centrifugal tube by using a pipette.
The device for rapidly extracting the hydrogen sulfide content in the cerebrospinal fluid and the serum sample is constructed by utilizing the simple injector and the sample injection needle, has low cost and convenient operation, and is suitable for rapidly extracting the hydrogen sulfide in the cerebrospinal fluid and the serum sample on site so as to be used for next detection.
