Test example 1 Effect on fat in obese mice
Taking 50 mice, each half of the male and the female, weighing 18-22g, randomly dividing into 12 groups: a blank control group, an obesity model group, an example 1 group, an example 2 group, an example 3 group, an example 4 group, an example 5 group, a comparative example 1 group, a comparative example 2 group, a comparative example 3 group, a comparative example 4 group, and a comparative example 5 group. Except for the blank control group, the rest groups all take the obesity feed, wherein the obesity feed is prepared by adding 10g of milk powder, 10g of lard oil, 1.5g of cholesterol and 0.3g of deoxycholic acid into every 100g of the basic feed, and the rest groups are the basic feed. Gavage is carried out for 1 time every day, gavage is carried out for 42 days continuously, the weight is weighed for 1 time every 7 days, the mice are sacrificed 24 hours after the last gavage, abdominal fat is taken and weighed as wet weight, and the fat index is calculated. The results of the experiment are shown in FIG. 1.
As can be seen from the results in FIG. 1, after the 42d intragastric administration, the groups of examples 4 to 5 have a very significant reduction effect on the increase of fat index of obese mice (P < 0.01) compared with the blank group and the model group; examples 1-3 groups significantly reduced the fat index (P < 0.05) in obese mice. Comparative examples 1 to 5 had only slight weight loss effect and the effect was not significant.
Test example 2 clinical test for weight-reducing effect
A study of a feeding trial was conducted on 180 subjects aged 20 to 55 years (average age: 35 years) in 180 adults by taking the weight-reducing oral liquids prepared in examples 1 to 5 and comparative examples 1 to 5 and the commercially available similar weight-reducing oral liquids for 60 days. Subject conditions: the BMI of the simple obese people, adult male and female, is more than or equal to 30, the thickness of abdominal skin folds is more than 30mm, the waist circumference is more than 101cm, and the weight exceeds the standard weight by 20 percent. Standard body weight (kg) ═ body height (cm) -100 × 0.9.
Subject exclusion: (1) patients with serious diseases of heart, liver, kidney and hemopoietic system, and psychosis are complicated. (2) Taking articles related to the tested function in a short time affects the judgment of the result.
The test method comprises the following steps: the subjects took the weight-reducing oral liquids prepared in examples 1 to 5 of the present invention and comparative examples 1 to 5, and the commercially available weight-reducing oral liquids, continuously for 60 days, 1 bottle per time, 3 times a day, and before or during each meal. The blank group was dosed with equal amount of water.
And (3) detecting an efficacy index: weight, height, waist circumference (periumbilical), hip circumference, thigh circumference, percent fat on weight, and Body Mass Index (BMI) calculated.
And (3) test results:
the results of the physical form index test of 60 days after the subjects took the oral liquid prepared by the invention examples 1-5 and the comparative examples 1-5 and the similar commercial weight-reducing oral liquid are shown in figure 2. Note that: capital letters are compared to the blank, P is less than 0.01; lower case letters are compared to the blank, P < 0.05.
FIG. 2 shows that the oral liquids prepared in examples 1 to 5 have significantly reduced efficacy indexes (except height index) and significantly different (P < 0.01) compared with the blank group, such as weight, height, waist circumference, hip circumference, thigh circumference, abdominal sebum thickness, and percentage of fat in the weight, when the subject takes the oral liquid for 60 days; compared with the traditional Chinese medicine in example 5, the traditional Chinese medicine in the comparative example 1 has different mixture ratio and does not have good weight-losing effect; comparative examples 2 to 5 show that the oral liquid prepared from the single raw material medicines in example 5 in the same amount has a slight weight-reducing effect and an unobvious effect, and thus the oral liquid prepared from the raw material medicines can obviously play a role in reducing weight and fat, and each group of raw material medicines has a synergistic effect.
Compared with the prior art, the invention further screens and obtains 4 Chinese medicinal material combinations with obvious fat-reducing and fat-reducing effects from 10 Chinese medicinal materials with homology of medicine and food, and the oral liquid raw material medicine is obtained, has obvious effect and wide raw material source;
according to the invention, water is decocted to extract water-soluble active ingredients in the raw material medicines, an ethanol solution is adopted to reflux and extract alcohol-soluble active ingredients in the raw material medicines at low temperature, part of the alcohol-soluble active ingredients are insoluble in water, a food-grade emulsifier is adopted to emulsify when the oral liquid is finally prepared, a homogeneous component is obtained, a stabilizer is added for stabilization, and the oral liquid with long shelf life, delicate taste and uniform solution is obtained after potassium sorbate is preserved;
the invention adopts pasteurization to sterilize oral liquid, heats the mixed raw materials to 68-70 ℃, and rapidly cools the mixed raw materials to 4-5 ℃ after keeping the temperature for 30 min. Because the common bacterial dead points are all at the temperature of 68 ℃ and the time of below 30min, the bacteria can be killed by sudden cooling and sharp changes of heat and cold after heating, and simultaneously the active ingredients in the oral liquid are kept from being damaged by high temperature, so that the weight-losing and lipid-lowering effects of the oral liquid are improved;
the composition is prepared by combining medicinal and edible raw materials, has the characteristics of pure nature, greenness and safety, does not need special diet when being taken, does not have any side effect or discomfort, is convenient to eat, has wide and easily-obtained medicinal sources and is low in price; the preparation process is simple, the operation is convenient, and the method is suitable for industrial production.
