The invention also provides a method for preparing an algae sample by using the manufacturing device of the algae cured leaf sample in the embodiment 1 or 2.
1. In addition to the apparatus of example 1 or 2, the method also employed the following consumables:
absorbent paper: 42X 31cm, the specific number is according to the actual situation.
Gauze (2): the size is consistent with that of the absorbent paper.
Sample paper: 100-130 lbs. of ballast paper is commonly used in sizes of 37X 27cm, 27X 19.5cm, etc. according to the algae size.
And (3) table paper: the heavy ballast paper is 42X 30cm in size and is used for pasting finished cured wax leaf specimens.
Tweezers: and (5) adopting blunt forceps to tidy the specimen.
2. The method comprises the following steps:
(1) The collected algae are washed by seawater, sediment and attachments are removed, and then the collected algae are respectively placed in a basin for containing seawater for numbering, and the numbering on the basin is consistent with the numbering of the algae specimen when the algae specimen is collected in the field.
(2) And (5) taking sample paper with proper size, and writing numbers corresponding to the basins one by one.
(3) The specimen soaking solution is poured into the specimen tray 1.
The specimen soaking liquid for soaking the intolerant low-salt seaweed is all seawater; the specimen soaking liquid for soaking the intolerant low-salt seaweed is all fresh water; the specimen soaking liquid for soaking general seaweed consists of seawater: fresh water according to 1:1 by volume ratio.
(4) The support plate is placed in the sample tray 1, and then the sample paper with corresponding number is attached to the support plate, so that the sample soaking liquid in the sample tray 1 fully infiltrates the sample paper, and the sample paper is well attached to the upper surface of the support plate, so that no bubbles and wrinkles exist.
(5) The algae to be made is placed on specimen paper, proper positions of the algae are arranged by means of scales on a supporting plate, and the branches are stretched by forceps to enable the algae to be as close to a natural growth form as possible.
When the algae form is tidied, the algae is prevented from being folded, twisted and overturned as much as possible; if the algae body is provided with a fixer or a reproductive structure, the whole organ of the fixer or the reproductive structure is kept; if the algae branches are thicker, the algae branches can be properly evacuated; if the artistic creation of algae arrangement is needed, the algae arrangement should be treated in a way of easy and difficult.
(6) After finishing the algae, one end of the Z-shaped handle 3 is inserted at the lower side of the supporting plate, so that the supporting plate is slowly and lightly supported from the specimen tray 1, and redundant specimen soaking liquid flows out by the water permeable holes 205 of the supporting plate. In this process, the algae should be as gentle as possible to avoid the movement of the algae position or morphology by a large amount of movement.
If the algae is too thin and light, the algae is easy to shift in the lifting process, the gauze or the covering clip 4 can be covered above the algae, and the gauze or the covering clip 4 is clamped with the supporting plate below the algae by using the phoenix tail clip, so that the supporting plate, the specimen paper, the algae, the gauze or the covering clip 4 are fixed together, and then the supporting plate is gently supported by using the Z-shaped handle 3.
(7) Preparing a plurality of pieces of water absorbing paper, placing the supporting plate supported in the step (6) above the water absorbing paper, then gently withdrawing the supporting plate, and if the algae is covered with gauze or a covering clip 4, removing the gauze or the covering clip 4 to prevent water evaporation in the drying process, covering the gauze above the algae, and then pressing the plurality of pieces of water absorbing paper above the gauze.
(8) Stacking a plurality of algae sample units (including algae and water absorbing paper on two sides of the algae) treated in the step (7), tightly clamping and bundling the algae sample units by using a sample clamp 5, and adding a proper pressure to accelerate water absorption, wherein when the total thickness is insufficient due to insufficient algae sample quantity, more water absorbing paper is added for supplementing.
And then placed in a ventilated place or in the drying cabinet 6, waiting for the specimen to dry. If the amount of the collected specimen is small, the specimen can be dried in the drying oven 6. If the sample collection amount is large, the sample can be placed at a ventilation position for drying in the shade, so that the algae sample is prevented from being directly placed in the sun to be dried as much as possible, the algae is prevented from fading, and the number of the dried sample is uniform.
(9) The water absorbing paper is replaced twice a day 2-3 days before drying, then the water absorbing paper is replaced once a day, the algae specimens can be basically dried by using the natural air-dried algae specimens for 6-7 days, and the time for drying the algae specimens by using the drying box 6 can be shortened to 3-4 days.
The following problems should be noted during the drying process: incidentally, the algae forms are recorded during arrangement; the shape of the algae is easy to deform when water is discharged; the dehydration time is too long and is easily influenced by weather.
(10) And (3) sticking the dried sample in the step (9) on new table paper, and sticking a sample number and a label and then storing.
For the samples of which the algae bodies cannot be firmly adhered to the wax leaf specimens, the main parts can be adhered by using adhesive tapes and fixed on specimen paper so as to prevent the specimens from falling off. The identified specimens should be selected from each serial number, the best piece of specimens with the complete genital organs and the fixing device is attached to the table paper, the label is attached to the right lower part of the table paper, the serial numbers are registered, and the specimens are placed into a cured leaf specimen cabinet according to the system classification sequence.
The algae specimen should be stored with attention: after the algae wax leaf specimen is put into the cabinet, the cabinet door is closed, and the moisture resistance is noted; a small amount of camphor essence is put into the cabinet, so that the cabinet is mildew-proof and insect-proof, and the indoor is kept dry and clean; the specimen chamber needs to be provided with a thick dark curtain to avoid the fading and the color change of the specimen caused by direct sunlight.
The desiccant in the absorbent paper and the desiccant bag can be dried and reused. After the preparation of the specimen is completed, the supporting plate, the covering clamp 4, the Z-shaped handle 3, the specimen clamp 5, the binding belt and the specimen tray 1 are cleaned and dried, and then are put into a drying oven 6 for the next use.

Accordingly, such modifications or improvements may be made without departing from the spirit of the invention and are intended to be within the scope of the invention as claimed.
Claims (5)
1. The method for preparing the algae body specimen by utilizing the manufacturing device of the algae cured leaf specimen is characterized by comprising a supporting plate, a covering clamping piece, a Z-shaped handle, a specimen clamp, a binding belt, a specimen tray for containing specimen soaking liquid and a drying box for drying the specimen, wherein two adjacent edges on the supporting plate are respectively provided with a transverse scale and a longitudinal scale, the upper surface of the supporting plate is provided with scale grid lines, and a plurality of water permeable holes are also formed in the supporting plate; the supporting plate can be placed in a sample tray, sample paper for spreading algae is attached above the supporting plate, and the covering clamp sheet covers the upper part of the sample; each set of Z-shaped handles is two pairs, each Z-shaped handle comprises a connecting plate, a portable plate and a clamping groove clamp formed by two clamping plates, the portable plate is connected with the top end of the connecting plate and is away from the direction of the supporting plate, the clamping groove clamp is connected with the bottom end of the connecting plate, the opening of the clamping groove clamp faces the direction of the supporting plate, and the left end and the right end of the supporting plate are inserted into the clamping groove clamp when the Z-shaped handles are used; the two specimen clamps are respectively positioned at the upper side and the lower side of the algae body, the drying box comprises a box body with an open top and a box cover which is covered at the top of the box body, and a drying agent bag is arranged in the box body of the drying box; grid lines on the supporting plate are respectively latticed according to 1cm in the transverse direction and the longitudinal direction, and obvious marks are arranged every 10 cm; the supporting plate is a PVC plate, and a bendable and foldable interface is arranged at the position 5cm away from the top edge of the supporting plate, so that the whole supporting plate is conveniently soaked in a specimen tray and taken out later; the four sides of the specimen clamp are respectively provided with a plurality of insertion holes for tying the binding bands, and the plate body of the specimen clamp is provided with a plurality of strip-shaped holes; the surface of the Z-shaped handle is coated with an anti-corrosion layer; the drying agent bag is filled with copper sulfate or silica gel particles; the connecting plate, the portable plate and the two clamping plates forming the clamping groove clamp are all hard plastic plates, and the portable plate, the connecting plate and the clamping groove clamp are fixedly connected into a whole; the connecting plate and the portable plate are stainless steel plates, a clamping plate I positioned at the lower end of two clamping plates forming the clamping groove clamp is stainless steel plates, a clamping plate II positioned at the upper end of the clamping plate is a magnet block, the connecting plate, the portable plate and the clamping plate I are welded into a whole, and the clamping plate II is connected with the connecting plate through magnetic adsorption;
the method comprises the following steps:
(1) Washing the collected algae with seawater, removing sediment and attachments, and respectively numbering in a basin for containing seawater;
(2) Taking sample paper with proper size, and writing numbers corresponding to the basins one by one;
(3) Pouring a specimen soaking solution into a specimen tray;
(4) Placing the support plate into a sample tray, and attaching sample paper with corresponding number on the support plate to enable sample soaking liquid in the sample tray to fully infiltrate the sample paper, so that the sample paper is well attached to the upper surface of the support plate, and therefore no bubbles and wrinkles exist;
(5) Placing the algae to be made on specimen paper, arranging proper algae positions by means of a scale on a supporting plate, and stretching branches by forceps to enable the algae to be as close to a natural growth form as possible;
(6) After finishing the algae, inserting the bottom end of the Z-shaped handle at the lower side of the supporting plate so as to slowly and lightly support the supporting plate from the specimen tray, and flowing out redundant specimen soaking liquid by means of the water permeable holes of the supporting plate;
(7) Preparing a plurality of pieces of water absorbing paper, placing the supporting plate supported in the step (6) above the water absorbing paper, then gently drawing out the supporting plate, covering gauze above the algae, and pressing the plurality of pieces of water absorbing paper above the gauze;
(8) Stacking a plurality of algae samples processed in the step (7), tightly clamping the algae samples by using a sample clamp, and then placing the algae samples in a ventilation place or the drying box;
(9) The water absorbing paper is replaced twice a day in the first 2-3 days, then the water absorbing paper is replaced once a day, the algae specimens can be basically dried in 6-7 days by natural air drying, and the time for drying the algae specimens by a drying oven can be shortened to 3-4 days;
(10) And (3) sticking the dried sample in the step (9) on new table paper, and sticking a sample number and a label and then storing.

The method according to claim 1, characterized in that: in the step (1), the number on the basin is consistent with the number of the algae sample collected in the field; in the step (3), the specimen soaking liquid for soaking the intolerant low-salt seaweed is all seawater; the specimen soaking liquid for soaking the intolerant low-salt seaweed is all fresh water; the specimen soaking liquid for soaking general seaweed consists of seawater: fresh water according to 1:1 by volume ratio.

The method according to claim 1, characterized in that: in step (5), the forceps are blunt forceps; when the algae form is tidied, the algae is prevented from being folded, twisted and overturned as much as possible; when the algae body is provided with the sessile device or the reproductive structure, the whole organ of the sessile device or the reproductive structure is kept; when the algae branches are thicker, the algae branches can be properly evacuated; when the artistic creation of algae arrangement is needed, the algae arrangement should be treated in a way of first-and-last-difficulty.

The method according to claim 1, characterized in that: in the step (6), when the algae is too thin, covering gauze or covering the covering clamping piece above the algae, clamping the gauze or the covering clamping piece with a supporting plate below the algae by using a phoenix tail clamp, so that the supporting plate, sample paper, the algae, the gauze or the covering clamping piece are fixed together, and then slightly supporting the supporting plate by using a Z-shaped handle; in the step (8), when the total thickness is insufficient due to insufficient quantity of the algae body specimens, more quantity of absorbent paper is added for supplementation.

The method according to claim 1, characterized in that: in the step (10), the specimen is preserved without direct irradiation of sunlight; the drying agent in the water absorption paper and the drying agent bag can be dried and reused; after the preparation of the specimen is completed, the supporting plate, the covering clamping piece, the Z-shaped lifting handle, the specimen clamp, the binding belt and the specimen tray are cleaned and dried and then are put into a drying oven for the next use.
