Abstract
本发明涉及油脂合成领域,具体涉及一种保肝护眼中长链结构脂质组合物,由以下成分组成:中链脂肪酸甘油三酯、长链脂肪酸甘油三酯、抗氧化剂、固定化酶、游离脂肪酶、包埋材料、二氧化硅、辅料。将中链脂肪酸甘油三酯与长链脂肪酸甘油三酯于反应器中进行酯交换反应,加入固定化酶和游离脂肪酶进行反应,然后与抗氧化剂混合,反应后得到中长链脂肪酸甘油三酯;将中长链甘油三酯乳化均质,与包埋材料、二氧化硅和辅料混合,过筛,得到组合物。本发明得到的中长链结构脂质组合物对保护眼睛具有良好的功效且在减少肥胖、降低血脂方面具有良好功效,起到保护肝脏的作用。本发明的制备方法简单,易于操作,有利于实现工业化生产。

背景技术
我国脂肪肝患者1.2亿人,因为肝炎而造成的直接经济损失达3600亿元人民币;我国肥胖人口达到3.25亿,未来20年将会增加一倍,青少年、全国肥胖儿中脂肪肝发生率40-50%,Ⅱ型糖尿病发病率20年间增长了11-33倍;小学生近视率32.5%,初中59.4%,高中生77.3%,大学生80%。
现代社会经济快速发展,饮食结构发生巨大改变,全国肥胖症日益增多,其肥胖儿中脂肪肝发病率为40-50%。现代电子设备的频繁使用,使得人们比以往任何时候吸收更多的短波蓝光,而短波蓝光对眼睛的黄斑区危害极大。从日常生活中获得改善眼干症状、平均视敏度、炫光耐受度、提高眼球视力、缓解眼晕等眼部症状的目标成为青少年的苛求。基于以上现状我国青少年的健康面临着巨大挑战,而学习工作紧张,在日常生活实现集营养、治疗为一体的综合性解决方案成为行业研究的方向,填补临床空白,也是本发明的研究课题。
中链脂肪酸在转运时不需要与其他脂类物质形成乳糜微粒,也不易与蛋白质结合形成脂蛋白,因此不会增加血液中的胆固醇的含量,对降低血脂有一定的调节作用。由于中链脂肪不经过淋巴系统而直接经静脉进入肝脏,在肝脏内不合成脂类,故不易形成脂肪肝,但是由于天然的中链脂肪酸种类较少,市场上大多均为长链脂肪酸。而长链脂肪酸要经胆汁乳化,胰脂酶水解后,进入肠道后被酯化为甘油三酯,经淋巴系统进入血液,与脂蛋白结合后参与全身的循环,对人身体健康影响较大。所以,通过酯交换合成中长链脂肪酸酯可以克服现有困难,既可以保证脂肪酸快速被人体吸收,又可以实现来源广泛易得。
发明专利CN 101255380A公开了一种甘油三酯型鱼油及其制作方法,属于动物油精制技术领域,旨在解决现有乙酯型鱼油药用保健效果差的问题。它是以乙酯型鱼油、甘油为原料,以脂肪酶为催化剂,按重量配比为:乙酯型鱼油100、甘油8.0-12.0、脂肪酶5.0-10.0,通过物料准备、酯交换反应、后处理及检验包装组成的制作方法所制成的甘油三酯型鱼油。其制作工艺方法简便易行,所制得甘油三酯型鱼油的药用保健效果显著,便于后续产品生产,富含的ERA、DHA均为ω-3多不饱和脂肪酸,是人体自身不能合成但又不可缺少的重要营养素,适用于制造药品和各种保健食品、鱼油制剂等。此发明申请本质上即利用鱼油中的ω-3多不饱和脂肪酸来达到保健的效果。但是ω-3多不饱和脂肪酸作用有限,不能达到实现人体健康多个指标研究的目的,其起到的保健作用具有局限性。
发明专利CN 105412227A公开了一种不饱和脂肪酸膳食补充剂,主要由以下原料制备而成:磷虾油5-30份、鱼油30-100份、奇亚籽油5-20份。本发明提供的膳食补充剂的制备方法,依次包括胶液制备、压丸、定型、干燥,最终得成品。本发明提供的不饱和脂肪酸膳食补充剂,其中的三种有效成分相辅相成,不饱和脂肪酸的功效能够充分发挥并被人体很好地吸收,提高了药效。本发明所提供的制备方法,其操作简单、便捷、安全、高效、节能、适于工业化推广。但是由于其原料均为长链脂肪酸,人体对于其吸收性差,要经胆汁乳化,胰脂酶水解后,进入肠道后被酯化为甘油三酯,经淋巴系统进入血液,与脂蛋白结合后参与全身的循环,容易形成高血脂,所以应用前景不佳。
目前,现有技术中还没有一种中长链脂肪酸酯组合物的研究,既可以充分发挥中链脂肪酸的优势,又可以发挥长链脂肪酸的优势。同时,现有技术中还没有一种中长链脂肪酸酯组合物,可以同时达到减少肥胖、脂肪肝以及保护眼睛的效果。
发明内容
为了解决现有技术中缺乏具有减少肥胖、脂肪肝以及保护眼睛的效果的保肝护眼中长链脂肪酸酯的问题,本发明提供一种保肝护眼中长链结构脂质组合物及其制备方法和应用。
一种保肝护眼中长链结构脂质组合物由以下成分组成:中链脂肪酸甘油三酯、长链脂肪酸甘油三酯、抗氧化剂、固定化酶、游离脂肪酶、包埋材料、二氧化硅、辅料。
进一步地,按重量份计,由以下成分组成:50-80份中链脂肪酸甘油三酯、240-430份长链脂肪酸甘油三酯、5-15份抗氧化剂、5-8份固定化酶、20-30份游离脂肪酶、20-30份包埋材料、10-20份二氧化硅、5-10份辅料。
进一步地,所述中链脂肪酸甘油三酯为8-10碳的辛癸酸甘油三酯,来源于棕榈仁油或椰子油。
进一步地,所述长链脂肪酸甘油三酯为16-25碳的脂肪酸甘油三酯,来源于鱼油中的EPA、鱼油中的DHA、藻油中的EPA、藻油中的DHA、水飞蓟籽油、光皮梾棵油、玉米胚芽油、小麦胚芽油、鳄鱼油、娃娃鱼油、菜籽油、杏仁油、月见草油、琉璃苣油、共轭亚油酸、紫苏籽油、红花油、辣木籽油、辣木叶粉、光皮梾木果油、共轭亚油酸甘油酯、植物甾醇、植物甾醇酯的一种或几种。
进一步地,按重量计,中链脂肪酸甘油三酯:长链脂肪酸甘油三酯=(10-20):(65-75)。
进一步地,所述抗氧化剂为维生素E和抗坏血酸棕榈酸酯。
进一步地,按重量份计,维生素E和抗坏血酸棕榈酸酯的质量比为(5-8):1。
进一步地,所述固定化酶为435脂肪酶、胰脂肪酶、酵母菌中的一种或几种,载体为甲基丙烯酸、二乙烯苯、大孔径丙烯酸树脂、二氧化硅、硅胶中的一种或几种。
进一步地,所述游离酶为DF15游离脂肪酶。
进一步地,所述包埋材料为低聚异麦芽糖、低聚麦芽糖、葡萄糖浆、抗性糊精、酶解奶油、L-阿拉伯糖、乳化剂、磷酸氢二钾的一种或几种。
更进一步地,所述乳化剂为酪蛋白酸钠,单双甘油脂肪酸酯。
进一步地,所述辅料为液体或粉末蓝莓、叶黄素、虾青素、花青素、原花青素、玉米黄质、蔓越莓、山竹提取物、左旋肉碱、魔芋葡甘露聚糖、β-羟基-β-甲基丁酸钙和接骨木莓提取物。

本发明还提供一种上述保肝护眼的中长链结构脂质组合物的制备方法,包含以下步骤:
(1)将中链脂肪酸甘油三酯与长链脂肪酸甘油三酯于反应器混合,加入固定化酶和游离脂肪酶进行反应,经降温、脱色、脱臭、蒸馏、钝化酶活、精滤后得到混合物A;
(2)将混合物A与抗氧化剂混合,反应后得到中长链脂肪酸甘油三酯;
(3)将中长链甘油三酯与蒸馏水乳化均质,与包埋材料混合,喷雾干燥,得到混合物B;
(4)将混合物B与二氧化硅和辅料混合,过筛,得到组合物。
进一步地,所述步骤(1)中,反应温度为40-60℃,反应条件为在超声波振动下反应8-15h。
进一步地,所述步骤(1)中,降温温度为10-20℃。
进一步地,所述步骤(3)中,将中长链甘油三酯乳化均质,与包埋材料在低温辅助干燥塔中进行混合,进风温度185-195℃,一段干燥温度为85-95℃,二段干燥温度为45-55℃,三段干燥温度为25-45℃。
进一步地,所述步骤(4)中,将混合物B与二氧化硅和辅料混合后过筛14-100目。
本发明还提供一种上述保肝护眼的中长链结构脂质组合物的制备方法,包含以下步骤:将中链脂肪酸甘油三酯、长链脂肪酸甘油三酯、固定化酶、游离脂肪酶、抗氧化剂与包埋材料混合均质包埋,喷雾干燥,与二氧化硅和辅料混合均质后即得组合物。
进一步地,所述喷雾干燥条件为:送入干燥塔中,控制进风温度为185-195℃,一段干燥温度为85-95℃,二段干燥温度为45-55℃,三段干燥温度为25-45℃。
本发明还提供上述一种上述中长链结构脂质组合物或上述的制备方法得到的组合物在制备减少肥胖、脂肪肝、糖尿病同时保护视力的营养品中的应用。
本发明中药中的功能特性脂肪酸,包括菜籽油、水飞蓟籽油、鳄鱼油、娃娃鱼油、杏仁油、月见草油、紫苏籽油、琉璃苣油、鱼油藻油EPA/DHA、玉米胚芽油、小麦胚芽油、椰子油、棕榈仁油等与维生素E、醋酸酯结合形成结构脂,在人体形成梯度营养,又与葡萄糖、低聚异麦芽糖、抗性糊精、聚葡萄糖、异麦芽酮糖醇等制成速溶粉,经与蔓越莓、蓝莓、花青素、虾青素、叶黄素、花青素、玉米黄质等形成综合营养配方组合。
与现有技术对比,本发明的有益效果是:
1.本发明的原料易得,克服短链脂肪酸一过性,引起酮体中毒等并发症的问题以及长链脂肪酸酯不易吸收等问题,通过酶、抗氧化剂将两者进行酯交换得到中长链脂肪酸酯,易于人体吸收。
2.通过添加叶黄素、虾青素、花青素、原花青素、玉米黄质等辅料,使得组合物对保护眼睛具有良好的功效。
3.本发明合理控制各组分的含量,使得组合物在减少肥胖、降低血脂方面具有良好功效,起到保护肝脏的作用。
4.本发明的制备方法简单,易于操作,有利于实现工业化生产。

下面结合具体实施例来进一步描述本发明,本发明的优点和特点将会随着描述而更为清楚,但这些实施例仅是范例性的,并不对本发明的范围构成任何限制。本领域技术人员应该理解的是,在不偏离本发明的精神和范围下可以对本发明技术方案的细节和形式进行修改或替换,但这些修改和替换均落入本发明的保护范围内。
实施例1
(1)将椰子油提取的50份中链脂肪酸甘油三酯与光皮梾棵油提取的240份长链脂肪酸甘油三酯于反应器混合,加入8份435脂肪酶-二乙烯苯和20份DF15游离酶,在超声波振动、40℃条件下反应8h,降温至10℃、脱色、脱臭、蒸馏、钝化酶活、精滤后得到混合物A;
(2)将混合物A与5份抗氧化剂混合,反应后得到中长链脂肪酸甘油三酯;其中抗氧化剂为维生素E和抗坏血酸棕榈酸酯的混合物,按重量份计,维生素E:抗坏血酸棕榈酸酯=5:1
(3)将中长链甘油三酯与蒸馏水乳化均质,与20份低聚麦芽糖混合,喷雾干燥,得到混合物B;喷雾干燥条件为进风温度185℃,一段干燥温度为95℃,二段干燥温度为45℃,三段干燥温度为45℃;
(4)将混合物B与10份二氧化硅和10份辅料混合,过14目筛,得到组合物,其中,辅料为液体蓝莓、叶黄素、虾青素、花青素、原花青素、玉米黄质、蔓越莓、山竹提取物、左旋肉碱、魔芋葡甘露聚糖、β-羟基-β-甲基丁酸钙和接骨木莓提取物的混合物;按重量计,叶黄素占辅料总量20%,虾青素占2%,玉米黄质占4%,其它辅料余量。
实施例2
(1)将棕榈仁油提取的80份中链脂肪酸甘油三酯与鳄鱼油提取的430份长链脂肪酸甘油三酯于反应器混合,加入5份酵母菌-二氧化硅和30份DF15游离酶,在超声波振动、60℃条件下反应8h,降温至20℃、脱色、脱臭、蒸馏、钝化酶活、精滤后得到混合物A;
(2)将混合物A与15份抗氧化剂混合,反应后得到中长链脂肪酸甘油三酯;其中抗氧化剂为维生素E和抗坏血酸棕榈酸酯的混合物,按重量份计,维生素E:抗坏血酸棕榈酸酯=8:1
(3)将中长链甘油三酯与蒸馏水乳化均质,与30份抗性糊精混合,喷雾干燥,得到混合物B;喷雾干燥条件为进风温度195℃,一段干燥温度为85℃,二段干燥温度为55℃,三段干燥温度为25℃
(4)将混合物B与20份二氧化硅和5份辅料混合,过100目筛,得到组合物,其中,辅料为粉末蓝莓、叶黄素、虾青素、花青素、原花青素、玉米黄质、蔓越莓、山竹提取物、左旋肉碱、魔芋葡甘露聚糖、β-羟基-β-甲基丁酸钙和接骨木莓提取物的混合物;按重量计,叶黄素占辅料总量30%,虾青素占5%,玉米黄质占3%,其它辅料余量。
实施例3
(1)将椰子油提取的70份中链脂肪酸甘油三酯、杏仁油提取的350份长链脂肪酸甘油三酯、6份胰脂肪酶-硅胶、25份DF15游离酶、10份抗氧化剂与25份葡萄糖、低聚异麦芽糖、低聚麦芽糖、抗性糊精、变性淀粉、L-阿拉伯糖、酶解奶油、蒸馏水、单双甘油脂肪酸酯、磷酸氢二钾的混合物混合均质包埋,喷雾干燥,与15份二氧化硅和8份辅料混合均质,得到组合物。
其中抗氧化剂为维生素E和抗坏血酸棕榈酸酯的混合物,按重量份计,维生素E:抗坏血酸棕榈酸酯=7:1;辅料为液体蓝莓、叶黄素、虾青素、花青素、原花青素、玉米黄质、蔓越莓、山竹提取物、左旋肉碱、魔芋葡甘露聚糖、β-羟基-β-甲基丁酸钙和接骨木莓提取物的混合物;按重量计,叶黄素占辅料总量25%,虾青素占3%,玉米黄质占2%,其它辅料余量;喷雾干燥条件为进风温度185℃,一段干燥温度为95℃,二段干燥温度为45℃,三段干燥温度为45℃。
实施例4
(1)将棕榈仁油提取的80份中链脂肪酸甘油三酯、鳄鱼油提取的430份长链脂肪酸甘油三酯、5份酵母菌-二氧化硅、30份DF15游离酶、15份抗氧化剂与30份葡萄糖、低聚异麦芽糖、低聚麦芽糖、抗性糊精、变性淀粉、L-阿拉伯糖、酶解奶油、蒸馏水、酪蛋白酸钠、磷酸氢二钾的混合物混合均质包埋,喷雾干燥,与20份二氧化硅和5份辅料混合均质,得到组合物。
其中抗氧化剂为维生素E和抗坏血酸棕榈酸酯的混合物,按重量份计,维生素E:抗坏血酸棕榈酸酯=8:1;辅料为液体蓝莓、叶黄素、虾青素、花青素、原花青素、玉米黄质、蔓越莓、山竹提取物、左旋肉碱、魔芋葡甘露聚糖、β-羟基-β-甲基丁酸钙和接骨木莓提取物的混合物;按重量计,叶黄素占辅料总量30%,虾青素占5%,玉米黄质占3%,其它辅料余量;喷雾干燥条件为进风温度195℃,一段干燥温度为85℃,二段干燥温度为55℃,三段干燥温度为25℃。
对比例1
与实施例1相比,区别仅为中链脂肪酸甘油三酯与长链脂肪酸甘油三酯的配比不同。
对比例1中,原料为椰子油提取的中链脂肪酸甘油三酯10份,光皮梾棵油提取的长链脂肪酸甘油三酯220份,此时,中链脂肪酸甘油三酯与长链脂肪酸甘油三酯质量比为1:22。
对比例2
与实施例1相比,区别仅为以435脂肪酶代替435脂肪酶-二乙烯苯。
对比例3
与实施例1相比,区别仅为维生素E与抗坏血酸棕榈酸酯配比不同。
对比例3中,按重量份计,维生素E:抗坏血酸棕榈酸酯=1:1。
对比例4
与实施例1相比,制备方法不同。
反应原料为棕榈仁油提取的中链脂肪酸甘油三酯13份和大豆油提取的长链脂肪酸甘油三酯87份,加入435固定化脂肪酶,用量为原料质量的1%,在55℃温度下充氮气进行酯交换反应,反应时间15小时终止反应,得产物A。
将产物A放入脱酸容器中,加入产物A质量1.5%的氢氧化钾饱和水溶液,边搅拌边加入,搅拌速度为50r/min,温度50℃继续搅拌1小时后,取样检测,酸价是0.8mgKOH/g。酸价合格后,加入占油脂质量2.0%的硅胶,边搅拌边加入,搅拌速度为30r/min,搅拌开始后,油脂降温到40℃,搅拌40min;搅拌结束后,用过滤机进行分离;硅胶颗粒的直径为0.8mm(粒度20目),过滤后得清液B。
将清液B升温至160℃与蒸汽一起雾化,并喷射入闪蒸釜中进行脱臭和脱除中链甘油三酯残基,闪蒸釜的真空度50Pa,蒸汽温度180℃。真空回收的蒸发物由冷凝器收集并由分离器分馏,分馏物为产品中长链脂肪酸食用油脂。
实验方法:
实验动物及环境:SD大鼠,实验动物室温度:21-23℃,相对湿度:65-75%。
1.安全性试验
大鼠急性经口毒性试验:采用最大耐受试验法,选体重150g左右的SD大鼠20只,雌雄各半。试验前动物禁食16小时,不限饮水。称取50.0g实施例1得到的样品,加纯水至100mL,混匀、配成500mg/mL浓度溶液,然后给动物灌胃2次(间隔6h),每次灌胃量为0.4mL/kg体重。灌胃后观察、记录动物的中毒表现。每周称重一次,观察两周时间,试验结束解剖动物进行大体观察。按毒性分级标准评价受试物的急性毒性。实验结果如下:

结果可见,灌胃后,动物生长良好,未见体重收到影响。受试小鼠均未见中毒症状,观察14天无动物死亡。试验结束解剖动物、大体观察,肝、肾、脾、心、肺、胃、肠等主要脏器均未见明显异常改变。
2.功能性试验
选取SD大鼠80只,体重150g左右,平均随机分为8组,分别为高脂组、实施例1-3组、对比例1-4组。每组动物10只。实施例组用实施例中所记载得到的产品灌胃,对比例组用对比例中所记载得到的产品灌胃,高脂组用等量猪油灌胃,灌胃量为0.4mL/kg体重,每天三次,连续灌胃30天。实验结束后禁食16h,从大鼠股动脉取血测量TC、TG及HDL-C。

试验例2保护眼睛功效测定实验
参考《保健食品检验与评价技术规范》(2003年版)中缓解视疲劳功能检验方法对本发明进行功效检验。
受试者选择及分组:受试者为有视疲劳症状者,年龄分布为30-40岁人群,且三个月内未服用过缓解视疲劳的保健品或功能食品,共160人,将其随机分为8组,分别为空白组、
实施例1-3组、对比例1-4组。
受试者服用方案:每日三餐后,各组受试者服用0.1g/kg体重的产品,空白对照组不服用本发明中任何组合,仅服用等量的水。服用一个月后,观察各组人员眼痛、眼胀、畏光、视物模糊、眼干涩等症状,改善评价结果见下表:

综合测试结果可知,本申请具备以下优点:

Claims (10)
1.一种保肝护眼中长链结构脂质组合物,其特征在于,由以下成分组成:中链脂肪酸甘油三酯、长链脂肪酸甘油三酯、抗氧化剂、固定化酶、游离脂肪酶、包埋材料、二氧化硅、辅料。
2.根据权利要求1所述的组合物,其特征在于,按重量份计,50-80份中链脂肪酸甘油三酯、240-430份长链脂肪酸甘油三酯、5-15份抗氧化剂、5-8份固定化酶、20-30份游离脂肪酶、20-30份包埋材料、10-20份二氧化硅、5-10份辅料。
3.根据权利要求1所述的组合物,其特征在于,所述中链脂肪酸甘油三酯为8-10碳的辛癸酸甘油三酯,来源于棕榈仁油或椰子油;所述长链脂肪酸甘油三酯为16-24碳的脂肪酸甘油三酯,来源于鱼油中的EPA、鱼油中的DHA、藻油中的EPA、藻油中的DHA、水飞蓟籽油、光皮梾棵油、玉米胚芽油、小麦胚芽油、鳄鱼油、娃娃鱼油、菜籽油、杏仁油、月见草油、琉璃苣油、共轭亚油酸、紫苏籽油、红花油、辣木籽油、辣木叶粉、光皮梾木果油、共轭亚油酸甘油酯、植物甾醇、植物甾醇酯的一种或几种;按重量计,中链脂肪酸甘油三酯:长链脂肪酸甘油三酯=(10-20):(65-75)。
4.根据权利要求1所述的组合物,其特征在于,所述抗氧化剂为维生素E和抗坏血酸棕榈酸酯,按重量份计,维生素E:抗坏血酸棕榈酸酯=(5-8):1。
5.根据权利要求1所述的组合物,其特征在于,所述辅料为液体或粉末蓝莓、叶黄素、虾青素、花青素、原花青素、玉米黄质、蔓越莓、左旋肉碱、魔芋葡甘露聚糖、β-羟基-β-甲基丁酸钙、山竹提取物和接骨木莓提取物。
6.权利要求1-5任一所述的组合物的制备方法,其特征在于,包含以下步骤:
(1)将中链脂肪酸甘油三酯与长链脂肪酸甘油三酯于反应器混合,加入固定化酶和游离脂肪酶进行反应,经降温、脱色、脱臭、蒸馏、钝化酶活、精滤后得到混合物A;
(2)将混合物A与抗氧化剂混合,反应后得到中长链脂肪酸甘油三酯;将中长链甘油三酯与蒸馏水乳化均质,与包埋材料混合,喷雾干燥,得到混合物B;
(3)将混合物B与二氧化硅和辅料混合,过筛,得到组合物。
7.根据权利要求6所述的制备方法,其特征在于,所述步骤(1)中,反应温度为40-60℃,反应条件为在超声波振动下反应8-15h。
8.根据权利要求6所述的制备方法,其特征在于,所述步骤(4)中,过筛过14-100目。
9.权利要求1-5任一所述的组合物的制备方法,其特征在于,包含以下步骤:将中链脂肪酸甘油三酯、长链脂肪酸甘油三酯、固定化酶、游离脂肪酶、抗氧化剂与包埋材料混合均质包埋,喷雾干燥,与二氧化硅和辅料混合均质后即得组合物。
10.权利要求1-5任一所述的组合物或权利要求6-9任一所述的制备方法得到的组合物在制备减少肥胖、脂肪肝、糖尿病同时保护视力的营养品中的应用。
