Abstract

背景技术
基质辅助激光解吸/电离飞行时间质谱(“MALDI-TOF-MS”)、电喷雾电离(“ESI”)以及联合利用电喷雾电离和低能量碰撞诱导解离(“CID”)的串联质谱(“MS-MS”)方法均可用于产生碎片离子以调查聚糖的序列和键,并且可在糖基化的表征中起着关键作用。
肽图分析为药物物质或产物选择性碎片化为离散肽的方法,这些离散肽可用于确认所需的产物结构。这些碎片的分离和鉴定以可重现的方式进行,使得深入分析可鉴定蛋白质一级结构中的细微(甚至同质异位)差异,诸如互补DNA的转录错误、点突变以及翻译后修饰(诸如糖基化、置换和截短)。由于肽图分析的复杂性和固有变异性,因此进行比较性程序,其中将测试样品的肽图与并列实验中制备的参照物质的肽图进行比较。

发明内容

具体实施方式
本文描述了聚糖氨基酸复合物及用于制备和分析所述复合物的方法。更具体地讲,聚糖(即,O-连接的聚糖或N-连接的聚糖)分别缀合到天冬酰胺、丝氨酸、苏氨酸、酪氨酸或羟赖氨酸残基以形成聚糖氨基酸复合物。聚糖氨基酸复合物可包含单个氨基酸或包含二至四个共价连接的氨基酸的肽。聚糖氨基酸复合物可为具有与聚糖部分组合的短肽的复合物。例如,在一个实施方案中,使用一种或多种酶将连接到肽或蛋白质的O-连接的聚糖分子从蛋白质或肽裂解或以其他方式释放。蛋白质或肽被消化,从而产生聚糖氨基酸复合物。任选地,可将聚糖氨基酸复合物从样品中存在的其他组分分离和/或纯化。然后,用衍生化试剂对聚糖氨基酸复合物加标签(标记)以检测和分析样品中存在的聚糖。

适用的消化技术和方案包括但不限于使用单种蛋白酶的消化及下面确定的方案、或使用蛋白酶组合的混合物消化及相关联的方案。例如,如表1中指出,存在通过蛋白酶与蛋白质的比率、温度和时间进行溶液中消化的一般性指南。

可用于本发明方法的附加的蛋白酶包括Arg-C(梭菌蛋白酶),即,在精氨酸残基的C末端处(包括脯氨酸旁边的位点)裂解的内肽酶。在赖氨酸残基处也可发生裂解。该测序级酶可单独使用或结合其他蛋白酶以用于通过质谱及其他应用的蛋白质分析。Arg-C活性在7.6-7.9的pH范围内最佳。另外,如下所述,可使用链霉蛋白酶裂解蛋白质或肽并且生成聚糖氨基酸复合物。

在一个实施方案中,消化方案可包括作为蛋白水解酶的胃蛋白酶。可使用游离(溶液中)或固定化的胃蛋白酶。胃蛋白酶可按照甘油、乙酸钠及作为防腐剂的叠氮化钠中的浆液形式供应。为了制备供使用的浆液,可将浆液转移到抗体或树脂制备物,并且在消化缓冲液(20mM乙酸钠,pH4.5)中洗涤或重悬。可将蛋白质溶解和/或悬浮于消化缓冲液中,并且添加到胃蛋白酶中,温育并混合,然后离心以使树脂沉淀并收集上清液。
蛋白水解酶也可为胰蛋白酶或胰凝乳蛋白酶。在一个实施方案中,固定化胰凝乳蛋白酶消化方案可包括在37℃下介于pH 7.5至9.0之间的反应条件。可通过增加酶与蛋白质底物的比率和温育温度来增加反应速率。例如,可使用大约1:25酶与蛋白质底物的比率来实现4小时至过夜的典型消化。为了实现加速的胰凝乳蛋白酶消化(大约0.5至1.0小时),可使用1:10酶比蛋白质底物。消化缓冲液可为诸如0.08Tris-HCL,0.1M氯化钙,pH7.8的缓冲液,或诸如1.0M碳酸氢铵,0.01M氯化钙,pH 8.0的另一种缓冲液。可将蛋白质溶解于消化缓冲液中。可用消化缓冲液洗涤固定化胰凝乳蛋白酶,并且在每次洗涤之后通过离心从缓冲液分离凝胶。可重悬固定化胰凝乳蛋白酶凝胶并且将其添加到样品中。可在37℃下的水浴中在快速搅拌下温育酶底物混合物2至18小时。如上所指出的那样从消化混合物分离固定化胰凝乳蛋白酶凝胶。
实施例I
基于胰蛋白酶的预示性方案
为了使蛋白质样品变性,添加50mM NH4HCO3或50mM Tris-HCl(pH7.8)、1mM CaCl2,直到盐酸胍或脲浓度小于1M。为了消化天然蛋白质,将蛋白质溶解于pH介于7和9之间的50mM NH4HCO3或Tris-HCl缓冲液中。然后添加胰蛋白酶,达到1:20(w/w)的最终蛋白酶:蛋白质比率。优选的是,蛋白质或肽浓度为至少0.1mg/ml并且在37℃下过夜温育至少4小时。可使该胰蛋白酶消化物经受附加的蛋白酶或化学消化技术以便产生聚糖氨基酸复合物。
实施例II
链霉蛋白酶方案

实施例III
固定化链霉蛋白酶预示性方案
可通过将链霉蛋白酶(53702 EMD MILLIPORE)偶联到亚乙基桥杂化(BEH)有机二氧化硅颗粒上来构造原型固定化链霉蛋白酶树脂。使用三乙氧基甲硅烷基丁醛,可使蛋白质经由其伯胺来官能化并且还原胺化以表现出硅烷官能度。通过随后的硅烷键合,可然后将蛋白质键合到树脂或颗粒上。该反应机制也可相反地进行,其中首先使颗粒与醛部分键合,并且使用还原胺化反应将蛋白质偶联到该醛。此外,还可通过其他化学方法(包括但不限于基于环氧化物或碳二亚胺的化学)完成酶与树脂的缀合。
简而言之,可在1mL至3mL的pH 5.0下的50mM柠檬酸三钠中制备含有50mg至100mg的链霉蛋白酶的溶液。然后将一mL至二mL的含有1M氰基硼氢化钠和50mg至200mg三乙氧基甲硅烷基丁醛的溶液添加到链霉蛋白酶溶液中,并且允许还原胺化反应在10℃至50℃下进行1至4小时。然后将0.1g至1.0g的BEH颗粒添加到该溶液中,并且允许硅烷键合进行0.5小时至12小时。然后经由添加pH 5.0下的50mM柠檬酸三钠中的1mL至4mL的2M硫酸钠而使该混合物经受盐析条件,并且在室温下旋转过夜。最后,通过添加0.5mL至2mL的碱性溶液(诸如1M乙醇胺)来淬灭固定化反应,并且在室温下旋转2小时。最终,使用50mM柠檬酸三钠和1M氯化钠的重复洗涤来洗涤链霉蛋白酶固定化BEH颗粒。参见Ahn,J.et al.,PepsinImmobilized on High-Strength Hybrid Particles for Continuous Flow OnlineDigestion at 10,000psi,Anal.Chem 84(16):7256-7262(Ahn,J.等人,在高强度杂化颗粒上固定化的胃蛋白酶用于10,000psi下的连续流在线消化,《分析化学》,第84卷,第16期,第7256-7262页),该文献以引用方式并入。

步骤2产生聚糖氨基酸复合物
如上所指出,消化步骤可涉及将多种蛋白酶(本文称为蛋白酶的混合物)与缓冲液浓缩物一起混合,以获得适当的pH及酶与蛋白质的比率。各种蛋白酶包括但不限于胰蛋白酶、赖氨酰内肽酶(“Lys C”)、胃蛋白酶、肾素、胰凝乳蛋白酶、弹性蛋白酶、羧肽酶、氨肽酶、三肽酶、N-氨肽酶、谷氨酰内肽酶、肽酰-ASP金属内肽酶、梭菌蛋白酶和二肽酶的组合。
在本主题方法中,在消化蛋白质或肽后,直接从蛋白质或肽产生聚糖氨基酸复合物。如下表2中所示,每种蛋白酶或酶在不同位点处裂解。例如,胰蛋白酶对Arg和Lys的C端侧具有特异性,并且胰凝乳蛋白酶对疏水性残基(例如,Leu、Met、Ala和芳族化合物)的C端侧具有特异性。因此,基于该混合物,可以这样一种方式细调该裂解,使得聚糖与其对应氨基酸残基一起被裂解。蛋白酶的混合物还可包含已知能消化蛋白质的某些化学物质,包括但不限于溴化氰、2硝基-5-硫代-氰基苯甲酸、O-亚碘酰苯甲酸、稀酸、BNPS-粪臭素。

步骤3聚糖氨基酸复合物的纯化或分离
可通过利用亲水作用色谱固相萃取(“HILIC SPE”)来纯化聚糖氨基酸复合物。蛋白质样品通常来源于复杂基质。在变性步骤期间存在的污染物和非蛋白质材料可包括DNA、RNA和完整细胞器。此类污染物可影响聚糖氨基酸复合物的检测并损害柱效率,以及可增加LC系统中的背压。另外,复杂样品基质可含有干扰下游标记反应的化合物。
在一个实施方案中,为了分离和纯化聚糖氨基酸复合物,可用200μL的水调节μElution板形式的SepPak氨丙基SPE吸附剂。可用200μL的15:85水/乙腈使孔平衡。然后可加载经乙腈稀释的样品。可用两(2)个600μL体积的1:9:90(v/v/v)甲酸/水/乙腈来洗涤孔。然后可用三(3)个30μL体积的SPE洗脱缓冲液(5%ACN中的200mM乙酸铵)来洗脱聚糖氨基酸复合物。另选地,二醇键合的或亲水-亲脂平衡的吸附剂(含有充分亲水部分)可用于完成该步骤。
步骤4聚糖氨基酸复合物的标记
可使用不同标记技术将标记引入到聚糖氨基酸复合物,当分析员采用光学检测(即,荧光)或质谱时,该标记能够改善这些复合物的可检测性。
在一个实施方案中,可使用快速加标签试剂来快速产生分析即用型O-连接的聚糖复合物或分析即用型N-连接的聚糖复合物。“分析即用型O-连接的聚糖复合物”意指在用最少处理或不做任何附加的处理的情况下即可用于LC或MS分析的O-连接的聚糖复合物。
虽然并非所有基团都具有质子亲和力或荷电标签基团,但可用的步骤包括用标记试剂(也称为加标签试剂)对聚糖氨基酸复合物标记(也称为“加标签”),该标记试剂由荧光部分、质子亲和力/荷电标签基团和N-羟基琥珀酰亚胺酯或氨基甲酸酯反应性基团构成,如下所示:

标记反应的条件(包括温度、有机溶剂组成、有机溶剂浓度、缓冲液组成、pH、离子强度、试剂的摩尔过量和时间)被选择和控制为使得实现在伯胺与羟基基团之间的所需的反应选择性。继而,优化标记的反应产物的收率,并且最小化“过度标记的”聚糖(由>1个标记修饰的聚糖或葡基胺)的生成。换句话讲,优化反应产物,并且最小化由大于伯胺/仲胺数量的任何数量的标记修饰的过度标记的聚糖氨基酸复合物的生成。然而,有时过度标记无法避免。
基于对提供快速标记动力学、高荧光量子产率和显著增强的MS可检测性的合理的设计考虑,来合成促进分析的快速加标签试剂。除了用NHS氨基甲酸酯或HNS酯活化试剂的快速加标签之外,样品制备还可取决于还原胺化。在这个过程中,聚糖氨基酸复合物在无水条件下被还原胺化以便最小化去唾液酸作用。样品制备从水性条件转变为无水条件。在这个反应中,含有醛的加标签试剂与聚糖氨基酸复合物的胺进行缩合反应,导致亚胺或席夫碱,还亚胺或席夫碱被还原剂还原以产生仲胺。该反应通常在含有乙酸的二甲基亚砜中进行,但是已经描述了使用四氢呋喃和甲醇的替代方法。另一方面,通过利用快速加标签试剂,可消除还原胺化并且可进行水性反应。
在实验室中可采取使用GLYCOWORKSTMRAPIFLUOR-MSTMN-聚糖试剂盒的组分对聚糖氨基酸复合物快速加标签。同样,这些方法也可用于使专门为分析即用型标记的聚糖氨基酸复合物的制备而优化的新型试剂盒商业化。如本文所提供,用于制备分析即用型标记的聚糖氨基酸复合物的优化聚糖样品制备工作流程需要三个步骤:(1)从蛋白质产生聚糖氨基酸复合物;(2)进行标记以向聚糖氨基酸复合物赋予可检测的化学实体;以及(3)净化步骤以从样品去除潜在的干扰。可将聚糖氨基酸复合物与一种或多种快速加标签试剂快速反应,并且用由高效荧光团和强碱性叔胺构成的标签对其进行标记,从而对荧光和MS检测产生增强的敏感度。下面紧接着示出了快速标记的聚糖氨基酸复合物的结构的描绘。

值得注意的是,快速标记的聚糖具有一个特别独特的键部分,其不同于来自还原胺化反应的仲胺键。快速标记的聚糖氨基酸复合物含有中性(非酸性)的脲键。这种结构可影响快速标记的聚糖的物理化学特性和/或性质,包括它们的色谱保留及其荧光性质和其他物理化学性质诸如等电点(“pI”)、酸性、碱性、疏水性、亲水性、螯合金属的能力、UV吸光度、荧光、在可见光谱中的吸光度、比色变化、分子大小、与结合伴侣相互作用的亲和力(即生物素酰化残基与亲和素或链霉亲和素之间,表位与对位之间)、还原/氧化电位、交联倾向、可裂解性(化学和热),以及不同长度的聚合取代基(即4个重复的聚乙二醇(PEG),40个重复的PEG)。

步骤5标记的聚糖氨基酸复合物的纯化或分离
在本发明方法中,在LC、MS或LC-MS分析之前,并且如果需要,可使标记的聚糖氨基酸复合物经受许多纯化类型处理之一,包括但不限于透析、使用反相(RP)的固相萃取样品净化、亲水作用色谱、或表面活性剂沉淀。Ying-Qing Yu,出处同上。值得注意的是,净化方法可能很耗时,在一些情况下仅部分有效,并且在没有适当方案的情况下可导致样品回收率降低。同上。
在一个实施方案中,为了从标记混合物分离和纯化标记的聚糖氨基酸复合物,可用200μL的水调节μElution板形式的SepPak氨丙基SPE吸附剂。可用200μL的15:85水/乙腈使孔平衡。然后可加载经乙腈稀释的样品。可用两(2)个600μL体积的1:9:90(v/v/v)甲酸/水/乙腈来洗涤孔。然后可用三(3)个30μL体积的SPE洗脱缓冲液(5%ACN中的200mM乙酸铵)来洗脱聚糖氨基酸复合物。另选地,二醇键合的或亲水-亲脂平衡的吸附剂(含有充分亲水部分)可用于完成该步骤。
步骤6糖基化分析
本文所述标记的聚糖氨基酸复合物可使用各种技术来分析,包括但不限于不同类型的分离和检测方法。分离技术可包括但不限于亲水作用色谱(“HILIC”)、固相萃取(“SPE”)、毛细管电泳、高pH阴离子交换色谱或反相液相色谱。
此外,色谱装置(诸如美国专利公布No.2013/03199086中描述的那些)提供了多峰色谱介质(保留机制)和聚糖分析方法,这提供了在不同生物大分子之间的高分辨率,基于大小、组成、结构(例如,异构、键合)和/或电荷的独特的选择性。这些类型的柱允许通过标准方法(例如,质谱、荧光检测)检测从介质中洗脱的大分子,而无需或很少需要净化或纯化后分析和预检测(例如,荧光加标签)。此外,为了实现足以完全分离聚糖(特别是N-连接的聚糖)的敏感度和选择性,可使用具有包含色谱表面的高纯度色谱材料(“HPCM”)的色谱装置。此处,色谱表面(保留机制)具有疏水性表面基团以及一种或多种可电离改性剂。此类HPCM在2016年4月24日提交的未公布的美国专利申请号62/326,783第5和6页中有所描述,该申请以引用方式并入本文。

与通过这种新的样品制备方法提供的效率和敏感度增益以及相关联的方法同等重要的是,其稳健性以及其产生与历史聚糖特征谱分析一致的结果的能力。此外,从聚糖氨基酸复合物生成的聚糖数据可用于查询聚糖数据库,但首先需要转换成被称为葡萄糖单元(“GU”)值的标准化值。从而,实施校准基准的另一个益处就是能够将现有聚糖数据转换成搜索可能的聚糖数据库的格式。转换的数据可用于正在研究未知聚糖组成的样品的发现过程。一旦聚糖被转换成GU值,用户就能够查询在线数据库以深入了解其样品中可能存在的潜在聚糖结构,潜在地降低了进行典型表征需要的时间。在液相色谱中,校准进行得非常频繁,有时频繁到在每次样品分析之后都要进行一次。
也可直接对所获得的衍生化混合物进行色谱分析,并且这种分析包括但不限于高压液相色谱(“HPLC”)、超高效液相色谱(UHPLC)、质谱、超临界流体色谱、紫外(“UV”)和/或荧光(“FLR”)检测。然而,分离任选地包括如本文所述的SPE离线和SPE在线技术。
为了检测荧光(“FLR”)标记的聚糖氨基酸复合物,可将亲水作用液相色谱(“HILIC”)与荧光检测偶联。对于分离处理和与某些常规的高效液相色谱(“HPLC”)方法相比,在HILIC模式下操作的装填有酰胺键合的亚乙基桥杂化颗粒的柱(下文称为“BEH聚糖柱”或“BEH柱”)通过其分离中性和酸性标记的聚糖氨基酸复合物两者的能力可提供峰值分辨率的改善。BEH聚糖柱实现并产生可重现的分离数据,并在花费更少的时间优化方法。
BEH柱利用杂化二氧化硅BEH、桥连技术颗粒,所述颗粒用稳定的含酰胺的物质官能化。BEH技术产生了直径范围为1.7μm至5μm的多种粒度的固定相,在HPLC与超高效液相色谱(“UPLC”)技术平台之间提供桥梁。BEH颗粒提供碱性分析物的峰形和效率、合理的一系列色谱选择性,以及在流动相极限下(特别是在升高的pH下)化学稳定性的改善。BEH聚糖/BEH酰胺柱的分辨能力部分为由于具有用于相关联的应用的最佳酰胺配体浓度的多孔颗粒。可对该柱进行优化以与UPLC或HPLC系统一起使用。
为了充分利用BEH聚糖柱,或简单地利用标记的聚糖氨基酸复合物的任何基于HILIC的特征谱分析,可使用葡聚糖校准梯(在下文中有时称为“葡聚糖梯”或“葡聚糖梯标准物”)。从HILIC/FLR系统获得的聚糖特征谱可针对葡聚糖梯进行校准,并分配葡萄糖单元(GU)值。例如,一个已知的此类梯,即2AB标记的葡聚糖校准梯(沃特世公司,产品编号186006841),不同于其他市售产品。该葡萄糖均聚物的平均分子量较高(约4,500道尔顿),因此“可用”的GU值范围为其他葡聚糖梯标准物的两倍;观察到的GU为2升到30。因此,大分子量聚糖的保留时间分配得到改善。葡聚糖梯的纯度和结构完整性可通过HILIC和质谱两者(“MS”)进行评估。
聚糖(如由标记的聚糖氨基酸复合物表示)的洗脱时间可参照葡聚糖梯,用葡萄糖单元(“GU”)表示。每个单独的聚糖结构具有与其键和组成单糖直接相关的GU值。由于对于给定的聚糖,特定键中的每个单糖均以特定的方式贡献GU值,所以GU值可用于预测结构。因此,本文提供的葡聚糖梯可用于校准LC的运行,防止其因不同时日或不同系统而改变。GU值可通过将五阶多项式分布曲线或三次样条拟合曲线拟合到葡聚糖梯来计算,然后使用该曲线根据保留时间来分配GU值。只要在柱之间的标准偏差小于0.3,聚糖的GU值可为非常可重现的。这就允许与在一段时间内从一系列仪器中收集到的数据库值进行直接比较。
具有GU值,就能够查询以GU值储存的聚糖的数据库,从而有助于阐明聚糖群内存在的潜在聚糖结构。葡聚糖梯也提供经质量控制的标准物,其可用于校准在不同实验室用不同仪器获得的色谱图。使用HILIC-FLR仪器采集的保留时间将因仪器与仪器和实验室与实验室而异。通过将保留时间转换为GU值,所得的数据可用于现场和非现场比较在不同地方之间的信息。虽然GU值的使用在此只是作为一个示例,但其他色谱方法可从使用葡聚糖校准物中获益,所述方法包括但不限于反相色谱以及反相与HILIC两者的混合型迭代。
聚糖氨基酸复合物的快速标记可简化用于分析的聚糖样品的制备,特别是在应用于O-糖基化分析时。但是,产生适于与这些类型的标记的化合物一起使用的葡聚糖梯并非一项无足轻重的任务,因为将需要该葡聚糖梯生成所谓的葡萄糖单元值。如作为WO 2016/077548公布的PCT申请No.PCT/US2015/060326中所述,用于还原性聚糖的加标签的两步法可调节色谱响应和化学性质诸如荧光和MS活性,以及调节具有不同检测性质的多个标签。每个步骤在与还原性聚糖连接的性质上有所不同。术语“还原性聚糖”是指还原糖、还原性糖、还原性多糖(杂多糖不同的糖单元,或不同的单糖,或均多糖),并且包括任何醛封端的糖诸如壳三糖、壳二糖、半乳糖-β-(1-3)-GalNAc-聚糖、甘露三糖-二-(N-乙酰基-D-葡糖胺)和麦芽糖。还原性聚糖或还原糖为能够充当还原剂的任何糖,因为其具有游离的醛基团。关于聚糖氨基酸复合物,有可能上述技术可用于制备化学上类似的葡聚糖梯。在一个具体实施方案中,葡聚糖可用丝氨酸还原胺化,并且然后用RapiFluor-MS衍生化而产生下面紧接着描绘的结构。
Claims (35)
1.一种表征蛋白质或肽的糖基化的方法,包括以下步骤:
提供多种糖基化蛋白质和/或肽;
用变性溶液使所述多种糖基化蛋白质和/或肽变性以产生变性的混合物,其中所述变性溶液包含MS相容的表面活性剂、有机溶剂、脲和/或胍;
将所述变性的混合物与多种蛋白酶组合以产生聚糖氨基酸复合物,其中所述多种蛋白酶裂解所述蛋白质或所述肽;
用HILIC SPE纯化所述聚糖氨基酸复合物,其中所述聚糖氨基酸复合物被分离;以及
用加标签试剂对所述聚糖氨基酸复合物加标签以产生标记的聚糖氨基酸复合物;
检测所述标记的聚糖氨基酸复合物;以及
通过所述标记的聚糖氨基酸复合物的检测来表征所述蛋白质或肽的糖基化。
2.根据权利要求1所述的方法,其中所述变性溶液包含3-[(2-甲基-2-十一基-1,3-二氧戊环-4-基)甲氧基]-1-丙烷磺酸钠。
3.根据权利要求1所述的方法,其中所述蛋白质或所述肽使用溶液中胃蛋白酶消化。
4.根据权利要求1所述的方法,其中所述蛋白质或所述肽用固定化胃蛋白酶消化。
5.根据权利要求1所述的方法,其中所述蛋白质或所述肽用溶液中链霉蛋白酶消化。
6.根据权利要求1所述的方法,其中所述蛋白质或所述肽用固定化链霉蛋白酶消化。
7.根据权利要求1所述的方法,其中所述蛋白质或所述肽用溶液中胰蛋白酶消化。
8.根据权利要求1所述的方法,其中所述蛋白质或所述肽用固定化胰蛋白酶消化。
9.根据权利要求1所述的方法,其中所述蛋白质或所述肽用pH为7.5至9.0的溶液中胰凝乳蛋白酶消化。
10.根据权利要求1所述的方法,其中使所述蛋白质或所述肽在约50℃至90℃之间的温度下变性。
11.根据权利要求1所述的方法,其中固定化内肽酶和外肽酶同时用于消化所述蛋白质或所述肽。
12.根据权利要求1所述的方法,其中所述加标签试剂为具有大于约的非极性表面区域和pKa大于约7的碱性残基的两亲化合物,并且形成所述两亲化合物和所述聚糖氨基酸复合物的缀合物。
13.根据权利要求12所述的方法,其中所述非极性表面区域介于约和约之间。
14.根据权利要求12所述的方法,其中所述非极性表面区域介于约和约之间。
15.根据权利要求1所述的方法,其中所述蛋白质或所述肽具有连接到聚糖的丝氨酸、苏氨酸、酪氨酸或羟赖氨酸。
16.根据权利要求1所述的方法,其中通过液相色谱、质谱、荧光和/或紫外检测来检测所述标记的聚糖氨基酸复合物。
17.一种表征蛋白质或肽的糖基化的方法,包括以下步骤:
提供多种糖基化蛋白质和/或肽;
将所述多种糖基化蛋白质和/或肽与多种蛋白酶和消化缓冲液混合,其中所述多种蛋白酶裂解具有丝氨酸、苏氨酸、酪氨酸或羟赖氨酸的所述蛋白质和/或所述肽以产生聚糖氨基酸复合物;以及
用加标签试剂对所述聚糖氨基酸复合物加标签以产生标记的聚糖氨基酸复合物;
检测所述标记的聚糖氨基酸复合物;以及
通过所述标记的聚糖氨基酸复合物的LC、MS或LC/MS分析和检测来表征所述蛋白质和/或所述肽的糖基化。
18.根据权利要求17所述的方法,其中所述消化缓冲液为MS相容的。
19.根据权利要求17所述的方法,其中所述消化缓冲液为MS不相容的。
20.根据权利要求17所述的方法,其中所述蛋白酶为固定化的。
21.根据权利要求17所述的方法,其中所述蛋白酶为未固定化的。
22.根据权利要求17所述的方法,其中所述蛋白酶为固定化蛋白酶和非固定化蛋白酶的混合物。
23.根据权利要求17所述的方法,其中所述蛋白酶单独混合或与结合其他类型的蛋白酶混合。
24.根据权利要求23所述的方法,其中所述蛋白酶依次与所述消化缓冲液混合。
25.根据权利要求17所述的方法,其中所述蛋白质被部分消化。
26.根据权利要求17所述的方法,还包括用HILIC SPE纯化所述聚糖氨基酸复合物并且从基质分离所述聚糖氨基酸复合物的步骤。
27.根据权利要求17所述的方法,还包括纯化所述标记的聚糖氨基酸复合物并且分离所述标记的聚糖氨基酸复合物的步骤。
28.一种制备标记的聚糖氨基酸复合物用于糖基化分析的方法,包括以下步骤:
从蛋白质或肽释放聚糖氨基酸复合物;以及
用加标签试剂标记所述聚糖氨基酸复合物以产生分析即用型聚糖氨基酸复合物。
29.根据权利要求28所述的制备标记的聚糖氨基酸复合物的方法,还包括使所述蛋白质或所述肽变性的步骤。
30.根据权利要求29所述的方法,其中用变性溶液使所述肽或所述蛋白质变性以产生变性的混合物,所述变性溶液包含MS相容的表面活性剂、有机溶剂、脲或奎尼丁。
31.根据权利要求28所述的方法,还包括纯化所述聚糖氨基酸复合物的步骤。
32.根据权利要求28所述的方法,还包括纯化所述加标签的聚糖氨基酸复合物的步骤。
33.根据权利要求28所述的方法,其中所述加标签试剂为快速加标签试剂。
34.根据权利要求1所述的方法,其中所述多种蛋白酶包括一种酶或多种不同酶。
35.根据权利要求35所述的方法,其中将所述多种不同酶中的每种酶一次性或依次添加到所述变性的混合物中。
