Abstract
本发明涉及一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,涉及这类双模态造影剂的制备方法及其在肿瘤及转移淋巴结诊断方面的用途。基于荧光共振能量转移的多色荧光/超声双模态造影剂的结构示意图如图所示,其膜成份包括用于荧光共振能量转移的的一系列荧光染料、各种常规磷脂以及用于稳定的表面活性剂,内腔包载用于超声成像的惰性气体或液体,该双模态造影剂可对肿瘤进行荧光和超声成像。此外,利用荧光染料的荧光共振能量转移,在单一波长激发光的照射下,通过调控荧光染料的比例可以改变发射光的信号,对不同的淋巴结进行区分,可为临床提供更多的诊断信息。
Description
技术领域
本发明属于生物医用材料领域,具体涉及一类基于荧光共振能量转移的多色荧光/超声多模态造影剂,以及其在肿瘤和转移淋巴结诊断方面的用途。
背景技术
超声成像具有无创、便捷、实时成像等特点,是临床上使用最广泛的一类成像技术。超声造影剂能够显著增强超声成像信号,为医生提供更加清晰的组织和血池图像,在医学影像诊断方面具有很好的应用前景。理想的超声造影剂能够随血流分布到全身各处,反映器官的血流灌注情况,而且稳定时间长,造影效果好,显著提高了诊断的准确率。
荧光成像是分子生物学和医学研究中极为重要的手段。其中,近红外波段(600-900nm)是光学成像的“诊断窗”,其范围内的生物分子的自身荧光较弱,可避免背景干扰而获得较高的分析灵敏度,而且近红外荧光染料的穿透能力也明显强于可见光波段的荧光染料。
荧光共振能量转移是距离很近的两个荧光分子间产生的一种能量转移现象。当供体荧光分子的发射光谱与受体荧光分子的吸收光谱重叠,并且两个分子的距离在10nm范围以内时,就会发生非放射性的能量转移,使得供体的荧光强度比它单独存在时要低的多(荧光猝灭),而受体发射的荧光却大大增强(敏化荧光)。荧光共振能量转移可以增加荧光染料的斯托克斯位移,具有背景干扰低、信噪比高等优点。同时,荧光共振能量转移技术使得在单一波长激发光的照射下,对两种及以上不同发射波长的荧光染料的检测成为可能,大大降低操作成本,在多通道生物标记和荧光成像领域具有重要意义。
淋巴结的无创性检查一直备受临床关注。经皮下注射近红外荧光染料行活体荧光成像寻找前哨淋巴结在国内外已有应用,具有很高的灵敏度,但是受限于成像深度,影响了在深部淋巴结检测中的应用。而超声成像定位淋巴结具有较高的分辨率,但是灵敏度不够高,逐一寻找较为麻烦且容易遗漏。
综上所述,单一成像模式的造影剂已经不能满足临床诊断的需要。鉴于超声和荧光成像在成像灵敏度和图像分辨率方面具有互补优势,我们开发了一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,能够综合荧光和超声两种成像模式,精准定位肿瘤位置,达到两种成像模式优势互补的目的。此外,考虑到淋巴系统的复杂性,我们利用多种荧光染料的荧光共振能量转移,在单一波长激发光的照射下,通过调控染料的比例来改变发射光的信号,可以对不同的淋巴结进行区分,为临床提供了更多的关于淋巴结的诊断信息。
发明内容
本发明的目的是提供一类基于荧光共振能量转移的多色荧光/超声双模态造影剂以及该类造影剂的制备方法。
本发明的另一目的是提供上述基于荧光共振能量转移的多色荧光/超声双模态造影剂在肿瘤和转移淋巴结诊断中的应用。
本发明所述的基于荧光共振能量转移的多色荧光/超声双模态造影剂的结构如附图1所示。
本发明中双模态造影剂其特征在于该造影剂的壳层是由脂质单分子层构成,其组成同时包括:用于荧光共振能量转移的一系列荧光染料、各类常规磷脂以及用于稳定的表面活性剂,微泡内包载惰性气体或液体,荧光染料可以和常规磷脂以及表面活性剂在水溶液中共同自组装形成纳米尺寸的微泡。
其中所述的荧光染料首选生物相容性好、具有疏水长链或者双亲性、发射光谱在近红外波段的染料。一般其结构如下:
其中R1,R2=C6~18烷基,R3,R4=H或SO3H-;n=2或3; X=H,CH3,CH3O,对位吡啶环,吡嗪等;Y=Cl-,Br-,I-等;例如 1,1-dioctadecyl-3,3,3,3-tetramethylindotricarbocyanine iodide(DiR)、吲哚菁绿(ICG)及其衍生物等。所述的荧光染料与常规磷脂自组装形成微泡,荧光染料和磷脂通过静电作用力或范德华力结合。
上述荧光染料,其特征在于所述的一系列荧光染料可以进行荧光共振能量转移,即前一种荧光染料的发射光谱与后一种荧光染料的吸收光谱有重叠,如附图2所示。
所述一系列染料的种类为二到四种,例如附图2中染料1、2、 3即为一系列荧光染料,首选染料1为1,1′-dioctadecyl-3,3,3,3- tetramethylindocarbocyanineperchlorate(DiI),染料2为1,1′- dioctadecyl-3,3,3,3-tetramethylindodicarbocyanine perchlorate(DiD),染料3为1,1-dioctadecyl-3,3,3,3-tetramethylindotricarbocyanine iodide (DiR)。
本发明所述的一种基于荧光共振能量转移的多色荧光/超声双模态造影剂的制备方法,包括以下步骤:
1)采用薄膜水化法,将一定比例的磷脂和多种荧光染料于乙醇中溶解混匀(近红外染料总比例0~20%),旋转蒸发除去溶剂,形成均匀的薄膜。
2)加入含有表面活性剂及甘油的生理盐水,水化30分钟。
3)将上述所得体系转移至西林瓶中,混匀。
4)往西林瓶中填充惰性内包物质,封口后,使用银汞调和器剧烈震荡45s,分离提纯后得到基于荧光共振能量转移的多色荧光/超声双模态造影剂。
在步骤1)中所述的磷脂包含12~24个碳的碳链长度以及包括磷脂酰胆碱、磷脂酰基乙醇胺、磷脂酸和磷脂酰基甘油,优选1,2-二硬脂酰基-sn-甘油基-3-磷酸胆碱(DSPC)、1,2-二棕榈酰基-sn-甘油基-3-磷脂酰胆碱(DPPC)、1,2-二棕榈酰基-sn-甘油基-3-磷脂酸(DPPA)等。
在步骤2)中所述的表面活性剂的分子结构具有两亲性:一端为亲水基团,另一端为疏水基团,包括但不限于泊洛沙姆 (Poloxamer)、聚山梨酯-80(Tween 80)、聚乙二醇4000(PEG 4000) 等。
在步骤4)中所述的惰性内包物质包含空气、氮气、二氧化碳、氟碳烃气体,液体选自C5-C12氟碳烃。
本发明中基于荧光共振能量转移的多色荧光/超声双模态造影剂,其一系列荧光染料和磷脂、表面活性剂共成膜,能够综合荧光和超声两种成像模式,精准定位肿瘤位置。此外,一系列荧光染料可以进行荧光共振能量转移,通过调控荧光染料的比例可以改变发射光的信号,在单一波长激发光的照射下,利用比例不同的造影剂对不同的淋巴结进行区分,为临床提供了更多的诊断信息。

具体实施方式
通过以下具体实施方式将有助于理解本发明,但并不限制本发明的内容。
实施例1
将二棕榈酰基磷脂酰胆碱(DPPC)、二棕榈酰基磷脂酰乙醇胺 (DPPE)和二棕榈酰基磷脂酸(DPPA)按照5∶2∶1的质量比用乙醇配成8mg/mL的母液。加入按照一定摩尔比混合(1∶1)的荧光染料DiI、DiD,旋转蒸发除去溶剂。加入1mL 0.6mg/mL的pluronic L61溶液(含有50uL的甘油,63.1mg)68℃水化30min。将制备好的混合溶液转移到西林瓶中,充气,机械震荡,从而得到负载两种染料的超声造影剂。粒径分布如附图3所示。
实施例2
将二棕榈酰基磷脂酰胆碱(DPPC)、二棕榈酰基磷脂酰乙醇胺 (DPPE)和二棕榈酰基磷脂酸(DPPA)按照5∶2∶1的质量比用乙醇配成8mg/mL的母液。加入按照一定摩尔比混合(1∶1∶1)的荧光染料DiI、DiD、DiR,旋转蒸发除去溶剂。加入1mL 0.6mg/mL的 pluronic L61溶液(含有50uL的甘油,63.1mg)68℃水化30min。将制备好的混合溶液转移到西林瓶中,充气,机械震荡,从而得到负载三种染料的超声造影剂。
实施例3

实施例4
将二棕榈酰基磷脂酰胆碱(DPPC)、二棕榈酰基磷脂酰乙醇胺 (DPPE)和二棕榈酰基磷脂酸(DPPA)按照5∶2∶1的质量比用乙醇配成8mg/mL的母液。加入按照一定摩尔比混合(1∶2∶0.5)的荧光染料DiI、DiD、DiR,旋转蒸发除去溶剂。加入1mL 0.6mg/mL 的pluronicL61溶液(含有50uL的甘油,63.1mg)68℃水化30 min。将制备好的混合溶液转移到西林瓶中,充气,机械震荡,从而得到负载三种染料的超声造影剂。
实施例5
将实施例1-4中获得的双模态造影剂与生理盐水按体积比1∶3配置后注入乳胶管中,在水槽中放入500ml超声脱气水,将上述乳胶管置于液体中部,使用超声诊断仪contrast模式,MI:0.04(机械指数),探头频率:3-12MHz,观察体外超声造影效果。体外超声造影图像见附图4所示,由于硅胶管壁本身较高的密度,导致其在超声场里呈现较高的回声效果,而硅胶管内腔在没有造影剂存在的时候,呈现无回声状态。当注射之后,在硅胶管的内腔中观察到显著的回声信号增强,从而证实了具有很好的体外超声显影增强的能力。
实施例6
为了评估实施例1-4中获得的双模态造影剂在体内超声显影增强性能,对新西兰大白兔进行了腘窝淋巴结超声成像。微泡的浓度为108/mL,注射50μL到大白兔右腿足垫内,轻揉按摩十分钟。使用超声诊断仪contrast模式,MI:0.04(机械指数),探头频率:3-12MHz。观察腘窝淋巴结的超声成像情况,见附图5所示。
实施例7
为了评估实施例1-4中获得的双模态造影剂在体内对不同淋巴结进行荧光成像的能力,对小白鼠头颈部淋巴结进行荧光成像。微泡的浓度为108/mL,按照10μL的剂量分别在左右耳以及左右前肢足垫处注射实施例1-4中获得的造影剂,注射后轻揉按摩十分钟。随后对小鼠头颈部淋巴结进行荧光成像,激发光设置在543nm,接受的发射光分别为570nm、670nm以及780nm。体内活体荧光成像图像如附图6示。
Claims (8)
1.一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,其特征在于该造影剂的壳层是由脂质单分子层构成,其组成同时包括:用于荧光共振能量转移的一系列荧光染料、各类常规磷脂以及用于稳定的表面活性剂,荧光染料可以和常规磷脂以及表面活性剂在水溶液中共同自组装形成纳米尺寸的微泡,微泡内包载惰性气体或液体;
其中一系列荧光染料的特征在于可以进行荧光共振能量转移,即前一种荧光染料的发射光谱与后一种荧光染料的吸收光谱有重叠;所述一系列染料为三种染料,染料1为 DiI(1,1'-dioctadecyl-3,3,3,3-tetramethylindocarbocyanine perchlorate);染料2为DiD(1,1'-dioctadecyl-3,3,3,3-tetramethylindodicarbocyanine perchlorate);染料3为DiR(1,1-dioctadecyl-3,3,3,3-tetramethylindotricarbocyanine iodide)。
2.根据权利要求1所述的一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,其制备方法包括以下步骤:
1) 采用薄膜水化法,将一定比例的磷脂和一系列荧光染料于乙醇中溶解混匀,旋转蒸发除去溶剂,形成均匀的薄膜;
2) 加入含有表面活性剂及甘油的生理盐水,水化30分钟;
3) 将上述所得体系转移至西林瓶中,混匀;
4) 往西林瓶中填充惰性内包物质,封口后,使用银汞调和器剧烈震荡45 s,分离提纯后得到基于荧光共振能量转移的多色荧光/超声双模态造影剂。
3.根据权利要求1所述的一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,其特征在于所述的超声造影剂由成膜材料包裹气体或液体构成,所述造影剂的粒径范围为100 nm-8 μm。
4.根据权利要求1所述的一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,所用磷脂的特征在于包含12~24个碳的碳链长度以及包括磷脂酰胆碱、磷脂酰基乙醇胺、磷脂酸和磷脂酰基甘油。
5.根据权利要求4所述的一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,所用磷脂材料包括1,2-二硬脂酰基-sn-甘油基-3-磷酸胆碱(DSPC)、1,2- 二棕榈酰基-sn-甘油基-3-磷脂酰胆碱(DPPC)、1,2- 二棕榈酰基-sn-甘油基-3-磷脂酸(DPPA)。
6.根据权利要求1所述的一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,所用的表面活性剂的分子结构具有两亲性:一端为亲水基团,另一端为疏水基团。
7.根据权利要求6所述的一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,所用的表面活性剂包括泊洛沙姆(Poloxamer)、聚山梨酯-80(Tween 80)、聚乙二醇4000(PEG 4000)。
8.根据权利要求1所述的一种基于荧光共振能量转移的多色荧光/超声双模态造影剂,所用的惰性气体包含空气、氮气、二氧化碳、氟碳烃气体,液体选自C5-C12氟碳烃。
