Abstract

本发明涉及一种活性生物物质纯化分离介质的基体树脂,属于纯化分离介质技术领域。基体树脂包括油相:甲基丙烯酸缩水甘油酯、双甲基丙烯酸乙二醇酯、二乙烯基苯、二乙苯、2‑乙基己醇、奎醇、过氧化二苯甲酰、水相:去离子水、10%聚乙烯醇水液、生化级明胶、氯化钠、0.1%次甲基蓝水液。由此制备的生物活性大分子的分子分离纯化介质不仅具有天然、合成分离介质两者的优点,而且具有大孔结构、吸附速率高,比表面积大、吸附量大的蛋白质分离纯化介质,为蛋白质的分离纯化提高了一类性能优良的分离介质。

技术领域
本发明涉及一种纯化分离介质的基体树脂,更具体地说,本发明涉及一种活性生物物质纯化分离介质的基体树脂,属于纯化分离介质技术领域。
背景技术
随着生物工程技术的不断进步以及回归大自然的需求迅速提高,在保健食品、生物医学领域对生物活性制品需求的不断增加,对生物活性制品分离纯化技术的要求也越来越高。
而对作为分离纯化技术的核心组成部分——分离纯化介质的性能要求也越来苛刻。一般把分离纯化介质分为两大类:一是天然糖类分离介质,其主要特点是生物相容性较好,但基质较软、强度较差,在使用过程中容易破碎,柱操作压降也很大;第二类是高分子合成分离介质,其机械强度较佳,使用周期长。但对蛋白质的分离专一性较差,所得纯化产品纯度不高,产品质量较差。因此,如能制备出具有两者的优点的分离纯化介质就成为一项十分重要的研究工作。
发明内容
本发明旨在解决现有纯化分离介质的问题,提供一种活性生物物质纯化分离介质的基体树脂,由该基体树脂制得的纯化分离介质具备两种纯化分离介质的特定和优点。

过氧化二苯甲酰 3-5份
水相:
去离子水 1500-2500份
10%聚乙烯醇水液 5-10份
生化级明胶 45-75份
氯化钠 75-125份
0.1%次甲基蓝水液 15-25份。

本发明带来的有益技术效果:
本发明根据生物活性大分子的分子结构特点,即分子量较大、分子结构中一般都带有疏水链段和亲水链段等特性,在分离纯化介质的孔结构设计及介质分子化学组成等方面给予充分考虑。在合成制备过程中选择合适的聚会单体,使分离介质具有较好的亲水性,对生物活性大分子的分子具有较好的亲和性;通过选择适宜的致孔剂,使分离介质具有较佳的孔结构以利于生物活性大分子的分子的扩散。由此制备的生物活性大分子的分子分离纯化介质不仅具有天然、合成分离介质两者的优点,而且具有大孔结构、吸附速率高,比表面积大、吸附量大的蛋白质分离纯化介质,为蛋白质的分离纯化提高了一类性能优良的分离介质。
