本发明公开了一种孔雀草精油及其应用,该孔雀草精油的制备方法是:将孔雀草的叶浸泡在水中,通过加热使浸泡液沸腾,当收集到第一滴挥发油时,持续保持沸腾2至5个小时,并持续收集剩余的挥发油,之后将收集到的挥发油进行脱水处理,得到孔雀草精油。应用时,将孔雀草精油作为抑菌活性成分用于制备抑菌性洗涤用品或皮肤护理产品,对埃希氏大肠杆菌、金黄色葡萄球菌、伤寒沙门氏菌具有明显的抑菌作用。本发明产品不仅具有明显的抑菌效果,而且扩大了孔雀草的应用范围,增加了孔雀草的应用价值。

技术领域
本发明涉及植物加工及其应用技术领域,尤其是一种孔雀草精油的制备及应用。
背景技术
孔雀草是一种菊科、万寿菊属一年生草本植物,一般植株高为30-100厘米,具有很强的环境适应性和抗逆性。由于外观较好,易于种植,目前该种植物常用于城市、道路、园林观赏种植,也可入中药作药用。由于应用范围窄,开发程度低,其经济价值低。因此,研究孔雀草深加工技术是提高孔雀草经济价值的重要途径。
发明内容
本发明所要解决的技术问题提供一种孔雀草精油及其应用,以解决现有孔雀草应用范围窄、价值低的问题,并且提供一种新的抑菌原料。
本发明以如下技术方案解决上述技术问题:
本发明孔雀草精油,它采用如下方法制取:
将孔雀草的叶浸泡在水中,通过加热使浸泡液沸腾,当收集到第一滴挥发油时,持续保持沸腾2至5个小时,并持续收集剩余的挥发油,之后将收集到的挥发油进行脱水处理,得到孔雀草精油。
本发明所述浸泡水的用量以浸没孔雀草的叶为宜。
本发明在脱水之前,将收集到的挥发油静置1h,待油水分离后,取上层液体即粗油,采用无水硫酸钠脱水,即得到孔雀草精油。
本发明孔雀草精油的应用:将孔雀草精油用于制备抑菌性洗涤用品、皮肤护理产品,即将孔雀草精油作为辅助原料加入抑菌性洗涤用品、皮肤护理产品中,使抑菌性洗涤用品、皮肤护理产品对埃希氏大肠杆菌、金黄色葡萄球菌、伤寒沙门氏菌具有明显的抑菌作用。
本发明具有如下有益效果:
通过本发明方法制备得到的孔雀草精油,可以作为抑菌活性成分用于制备抑菌性洗涤用品以及皮肤护理产品,不仅具有明显的抑菌效果,而且扩大了孔雀草的应用范围,增加了孔雀草的应用价值。
具体实施方式
以下结合具体实例对本发明的技术方案作进一步描述:
实施例1
孔雀草精油的制备方法如下:
采集新鲜的孔雀草的叶,不超过24h,先称取150g孔雀草叶,剪碎并置于1000mL圆底烧瓶中,加入500mL水浸没剪碎的孔雀草叶;之后在180℃下恒温加热,在第一滴挥发油滴出时开始计时,沸腾2h(根据挥发情况,也可以加热至5h)后,结束蒸馏,停止加热后,将收集到的挥发油静置1h,待油水分离后,取油水分离器中上层液体即粗油,用无水硫酸钠脱水,得到孔雀草精油。
孔雀草精油的性能测试:
抑菌活性测定,采用平板计数法:
在生物安全柜中通过接种环分别取埃希氏大肠杆菌(Escherichaia coli)、金黄色葡萄球菌(Staphyloccocus aureus)、伤寒沙门氏菌(Salmonella typhimurium)各一环,分别通过划线接种法接种在经灭菌的营养琼脂培养基斜面试管中,接种完成的试管用胶塞封口并用封口膜密封,于36℃生化培养箱中培养48h。分别配制BP培养基、MCA培养基、BS培养基并在超净工作台中以每个培养皿10~15mL的倾注量倾注于经过灭菌处理的培养皿中。
在生物安全柜中用接种环取每种菌各一环,分别转移至含有10mL灭菌生理盐水无菌试管中,混匀后梯度稀释至103、104、105倍,对抑菌限度的进行测定。
在冷却的培养基上加入100μL菌液与200μL、100μL、50μL、20μL为实施例,并通过无菌三角涂布棒进行涂布。103、104、105每个稀释度的菌液均与实施例进行一次如上涂布,并且每个稀释度设置一个100μL菌液与200μL无菌水的空白对照。
以上实验中,培养基与菌种的对应关系为:BS培养基接种伤寒沙门氏菌;BP培养基接种金黄色葡萄球菌;MCA培养基接种埃希氏大肠杆菌。将接种好的培养皿与没有进行接种作业的BS培养基、BP培养基、MCA培养基各一个放置于生化培养箱中36℃培养24h后,进行菌落计数。实验结果如表1所示。

由表1见,未进行涂布作业的样品均没有菌落生长,故可以认为培养基灭菌和培养条件没有向实验组引入杂菌。
在对伤寒沙门氏菌的实验结果中,三个稀释度空白组的沙门氏菌菌落计数均大于100,明显高于对薄荷、香茅草、艾草抑菌效果初筛实验中空白组生长量,这可能是由于在菌种稀释时用接种环挑取的菌量过大引起的。虽然空白组的稀释程度不足造成菌落数过多难以计数,但添加了孔雀草精油的实验组菌落计数均小于10,与空白组形成了明显的对比,足以说明孔雀草精油对伤寒沙门氏菌有显著的抑菌效果。降低精油用量后,可以看出来当浓度在50μL时,精油的抑菌效果基本与阳性对照抗菌素效果相当。用量加大后,对沙门氏菌的抑制作用非常明显。
在对金黄色葡萄球菌和埃希氏大肠杆菌的抑制实验中,在200μL和100μL条件下,抑菌效果最好,50μL时,大体与阳性对照效果持平,20μL也有一定的抑菌效果,说明孔雀草精油这两种菌的生长也同样有一定的抑制效果。
在对大肠杆菌的抑制实验中,可以得到同样的结论。
综上所述,孔雀草精油对伤寒沙门氏菌、金黄色葡萄球菌和埃希氏大肠杆菌均有抑制效果。
将实施例1中获得的精油取800μL作为辅料加入200g无抑菌效果洗手液中,用无抑菌效果洗手液作为空白实验对比(无抑菌效果洗手液在添加精油前经过121℃。15min灭菌处理,作为空白实验的洗手液亦经过灭菌),将加入精油的洗手液样品和空白对照样品置于敞开空气中,每120min取1~2g样品,按照上述金黄色葡萄球菌(平板计数法),以稀释度为104的菌液进行抑菌活性实验(即取200μL洗手液样品与100μL菌液混合涂布于培养基上,培养24h后进行菌落计数,平行进行三次实验取平均值),实验总时长为480min。实验结果如表2所示。

由表2可知:在置于敞开的空气中,添加了实施例1的精油的洗手液抑菌效果会随着时间的增加而下降,但整体抑菌效果明显强于未添加精油的洗手液样本,故可以看出在抑菌性洗涤用品中,孔雀草精油有着较好的抑菌效果,在洗涤用品中有应用价值。
将本发明孔雀草精油应用于制备其它皮肤护理产品也具有较好的抑菌效果。

1.孔雀草精油的应用,其特征在于,将孔雀草精油作为抑菌活性成分用于制备抑菌性洗涤用品或皮肤护理产品,即将孔雀草精油作为辅助原料加入抑菌性洗涤用品或皮肤护理产品中,使抑菌性洗涤用品或皮肤护理产品对埃希氏大肠杆菌、金黄色葡萄球菌、伤寒沙门氏菌具有明显的抑菌作用,
