Description
本发明涉及水痘带状疱疹病毒(VZV)前早蛋白质175和其衍生物以及包括这些蛋白质的疫苗在预防和治疗VZV感染中的应用。
水痘带状疱疹病毒(VZV)是人疱疹病毒,它是水痘和带状疱疹的病原体。初期感染或原发感染导致水痘,通常在幼年时感染上,此时为相对良性的。然而,对于幼年时未接触过水痘的成年人,以及被免疫包容的个体有时可以是致命的。同样,VZV感染对于新生儿可以是致命的,因为该病毒能穿越胎盘。人们知道,水痘是极易通过直接接触而传染的疾病。
与大多数疱疹病毒一样,VZV趋向于感染某些病毒发育在其中受到抑制的细胞。一段不同的潜伏期之后,水痘带状疱疹(VZ)病毒可被释放,开始感染其它细胞。这种VZ病毒的重新激活每年大约引起5百万例带状疱疹(Plotkin et al.Postgrad.Med.J.61:155-63(1985))。带状疱疹特征是大脑神经节和末梢神经发炎并伴有剧痛。目前,重新激活该病毒的因素尚不清楚。
现已表明,用VZV减毒品系接种的人从VZV感染获得了保护性免疫力(Arbeter et al.,J.pediatr 100,886-93(1982)and Brunell et al.,Lancet ü:1069-72(1982))。这种方法虽是有效的,但由于繁殖水痘带状疱疹病毒有困难而受到限制。在鉴定VZ病毒的抗原成分上人们做了一定的努力。为了开发改良的VZ疫苗特别是亚单位疫苗,分离VZV被膜蛋白是很重要的。Forghani等人(J.Virol.52:55-62(1984)).OKuno等人(Virol.129:357-68(1983))和Keller等人(J.Virol.52:293-7(1984))已从VZV感染的细胞和VZ病毒粒子中鉴定出许多病毒特异性糖蛋白。
迄今,人们生产抗VZV的重组亚单位疫苗的努力集中在作为潜在糖蛋白的外被膜糖蛋白上。本发明明显不同于这种方法,而是涉及采用VZV的前早非结构蛋白来提供对抗继后之VZV攻击的保护作用。
由于胞毒性淋巴细胞CTL的抗原识别机理涉及将天然抗原分解成肽,使蛋白质水解片段结合到MHC分子上并将复合物输送到分子表面,故任何病毒编码的多肽,而不是那些象糖蛋白一样的整合膜蛋白,都可以是T细胞介导的反应的潜在靶体。然而,由于VZV基因组除编码外糖蛋白外,还编码几种非结构蛋白质和内病毒粒子蛋白质,这样就导致有大量的潜在CTL靶体,而不知哪种蛋白质是最相关的。
VZV感染的特征是只有极微量的游离病毒存在。在潜伏期和重新激活期间,病毒主要是细胞内的。由此,复发疾病即使是用高水平中和抗体也不能被阻止,而且病毒控制依赖于细胞介导的免疫性。因此,为了用免疫接种法获得保护,人们不仅希望诱导出抗体反应,而且还希望诱导出CTL反应。有效的疫苗应当是只要潜伏病毒重激活信号一出现就能尽早发挥作用的预激态CTL。
为了鉴定用于预防或治疗目的之疫苗的最重要的CTL靶抗原,本发明人考虑了VZV复制循环。包含病毒基因组之细胞内的病毒蛋白质合成开始时将产生病毒蛋白质片段,该片段由MHC分子呈现于细胞表面上,使其成为适当特异性的CTL的靶。VZV的复制循环持续约24小时并涉及α或前早(IE)β和γ或晚期(L)基因产物的有序表达。因此早期CTL攻击和随后的先于晚结构基因表达的感染细胞的溶胞能阻止新的病毒粒子产生,并由此防止病毒扩散到邻近细胞。为了更为有用,CTL应测检到感染和重新激活后出现在细胞内的最早的病毒蛋白质。
IE蛋白质IEP 175是由开放读码设计的ORF62编码的,该蛋白质本身有时称作IE62。
该蛋白质作为相对分子量为175 KDa的磷蛋白出现(Kinchinton et al J.of Virology Vol 66(1)(1992)P.359-366)),但推测的分子量为140KDa。它是由人T细胞识别的(Bergen et al Viral.Immunology 4(3)1991 P151))并已提示是为重要的免疫靶体(Bergen et al J.Infectious Diseases(1990)162 P.1049))。
有许多系统可供本行业熟练技术人员采用重组DNA技术生产蛋白质,然而,其中大部分已被证实不能成功地生产全长度的IEP175。例如该蛋白在昆虫细胞中被降解。
然而,本发明人已发现在CHO细胞中表达可生产出天然IEP175的功能等同物和结构等同物(即正确的大小)。
因此,在本发明的实施方案中提供了无VZV污染物的功能上等同于其天然蛋白质的IEP175。
本发明还扩展到IEP175的生理功能衍生物。
本发明的一个方面是提供没有VZV污染物的具有与附图1中描述的顺序基本上同源之氨基酸顺序的IEP175蛋白质。
基本上同源是指本发明提供的功能等同物IEP 175蛋白质至少有75%同源于图1中的氨基酸顺序,优选80%,更优选至少90%,且最优选至少95%同源。
优选的IEP175衍生物是在大肠杆菌或CHO细胞中表达后得以分泌该蛋白质的那种。特别是提供了其中氨基酸226至257和氨基酸648至733已缺失的可分泌衍生物。
典型的其它免疫原衍生物为含有附加顺序的融合多肽。该附加顺序可以携带一或多个抗原决定基,其来自其它VZV蛋白质如VZV糖蛋白,例如gpⅠ,gpⅡ,gpⅢ,gpⅣ或gpⅤ(有时称为gpⅠ,gcⅡ,gcⅢ等)、其它VZV抗原、甚或其它非VZV抗原如乙肝表面或核心抗原。此外,本发明的免疫原衍生物可融合于具有免疫刺激特性的载体多肽如佐剂上,或提高对VZV蛋白质的免疫反应,或可用于表达、纯化或配制VZV蛋白质。
优选的融合蛋白质包括融合于上述可分泌形式之IEP175的“无锚地”gpⅡ。
另一方面,本发明提供在调节顺序控制下可表达DNA分子编码的能在异种宿主中发挥功能的IEP 175或其衍生物。特别是提供了基本上同源于附图1中所示DNA顺序的DNA分子。基本上同源是指DNA顺序至于75%同源于图1所示的DNA顺序,优选至少85%,更为优选90%且最优选至少95%。
可采用本行业熟知的方法,通过加入、缺失、取代或重排碱基来制备编码IEP175或衍生物的DNA顺序。图1中,开头的ATG编码N末端蛋氨酸,且最后的TGA为转译终止(即停止)信号。
本发明的再一个方面是提供编码水痘带状疱疹病毒IEP175或衍生物的重组DNA分子或包括DNA顺序的媒体,它可操作地连于在真核宿主细胞中更好是在CHO细胞中起作用的调节区域。
本发明的另一方面是提供制备水痘带状疱疹病毒IEP175或其衍生物的方法,该方法包括在宿主细胞中表达所述的DNA顺序并回收蛋白质产物。
在相关的方面,本发明提供了用重组DNA分子转化的重组CHO细胞系。
在另一个方面,本发明提供了根据本发明制备VZV IEP175蛋白质或衍生物的方法,该方法包括在重组宿主细胞中表达编码所述蛋白质或衍生物的DNA顺序并回收所得的蛋白质产物。
本发明的方法可采用常规重组技术如Maniatis等人在Molecular Cloning-A Laboratory Manual;Cold Spring Harbor,1982和DNA Cloning Vol Ⅰ,Ⅱ和Ⅲ卷(DM,Glover ed,IRL Press Ltd)中描述的技术来完成。
包含有这种编码顺序的DNA分子可以采用已知技术(例如由mRNA模板制造互补物或CDNAs)或从VZV基因组DNA分离得到。参见Ecker et al.Proc.Natl.Acad.Sci.USA 79:156-160(1982),Strans等,Proc.Natl.Acad.Sci USA.79:993-7(1982).Strans et al.,J.Gen.Virol.64:1031-41(1983)和Davison et al.,J.Gen.Virol.64:1811-1814(1983))。此外,可用标准DNA合成技术合成编码gpⅠ、gpⅡ和gpⅢ的DNA分子。
由此本发明还提供了通过缩合适当的一、二或寡聚核苷酸单位来制备DNA顺序。
制备可以化学方法、酶方法、或上述两种方法的组合在体外或适合时在体内进行。由此DNA顺序可以采用常规方法如DM Roberts等人在Biochemistry 1985.24,5090-5098中所描述的方法,用酶连接适合的DNA片段来制备。
经用适当的限制性内切酶消化含有所需核苷酸顺序的DNA、化学合成、酶促聚合,或这些方法的组合,可以获得该DNA片段。
用限制性内切酶消化可在适合的缓冲液中在20℃-70℃温度下,通常在含有0.1-10μg DNA的50μl或更小的体积中进行。
DNA的酶促聚合可在体外进行,采用DNA聚合酶如DNA聚合酶Ⅰ(Klenow片段)在含有所需要之核苷三磷酸dATP、dCTP、dGTP和dTTP的适当缓冲液中,在10-37℃下,通常在50μl或更小的体积中进行。
DNA片段的酶促连接可采用DNA连接酶如T4DNA连接酶在适合的缓冲液中,4℃至室温下,通常为50μl或更小的体积中进行。

该DNA顺序优选由两个或更多个DNA分子连接而制得,这些DNA分子合在一起包含编码该蛋白质的DNA顺序。
DNA分子可以经用适合的限制性内切酶消化携带所需编码顺序的载体而获得。
DNA分子的准确结构以及获得它们的途径依据所需蛋白质产物的结构而定。构建编码蛋白质之DNA分子的适宜策略的设计对本专业熟练技术人员来说是易于作到的。
裂解与宿主细胞相容的载体以提供线性DNA片段,并将所述的线性片段与一个或多个(与所述线性片段一起在连接条件下编码IE175蛋白质或衍生物的)DNA分子结合而制得本发明的表达载体。线性片段与一个以上DNA分子的连接可同时或按所需顺序进行。因此,该DNA顺序可以按需要在构建载体期间或之前制成。
载体的选择将部分地取决于宿主。更具体地说,本发明优选的宿主细胞为CHO细胞。本发明宿主细胞的适合载体包括质粒和装配型质粒。
为制备IE175表达载体,可采用适合的酶,按照例如上面提到的Maniatis等描述的步骤经限制、聚合和连接DNA而常规完成。聚合和连接可以按上述制备DNA聚合体的方法进行。用限制性内切酶消化可在适当的缓冲液中于20-70℃下,通常在含有0.1-10μgDNA的50μl或更小体积中进行。
根据本发明,重组宿主细胞通过在转化条件下转化带有本发明表达载体的宿主细胞制备。适宜的转化条件为常规的且描述于例如上面提到的Maniatis等人的文献中,或“DNA Cloning”VolⅡ,D.M.Glover ed.,IRL Press Ltd,1985中。
培养物中的哺乳动物细胞可以通过将载体DNA钙共沉淀到细胞上或经电穿孔而被转化。
在允许表达DNA顺序的条件下培养转化的宿主细胞是按照例如描述于上面提到的Maniatis等人的文献和“DNA Cloning”中的常规技术进行的。因此,细胞最好补充营养素并在45℃以下培养。
根据宿主细胞和产物是否为被分泌的或是否为经化学或酶释放并由所得溶胞物中分离的蛋白质产物,而用常规方法回收该VZV175蛋白质表达产物。如当产物是可分泌的,则产物通常可从营养培养基中分离。
可将DNA顺序组装到设计来分离稳定转化之哺乳动物细胞系表达IEP175蛋白质的载体中;例如牛乳头状瘤病毒或在中国仓鼠细胞中扩增的载体(DNA Cloning Vol.Ⅱ.D.M.ed.IRL Press 1985;Kamfman,R.J.et al,Molecular and Cellular Biology 5,1750-1759,1985;Pavlakis G.N.and Hamer,D.H.,Proceedings of the National Academy of Sciences(USA)80,397-401,1983;Goddel,D.V.et al.European Patent Application No.0093619,1983)。
在本发明的一个具体实施方案中,VZV IEP175蛋白质在CHO细胞中表达。表达IEP175蛋白质,优选采用Tad表达质粒。在此系统中,表达暗盒(包含VZV蛋白质编码顺序)被可操作地连接于劳氏肉瘤病毒(RSV)启动子上。该载体含有足够量的细菌DNA,得以在大肠杆菌或其它适合的原核宿主中繁殖。这种穿梭载体还含有足够的真核DNA侧接于VZV编码顺序上,以便可重组到真核宿主的基因组内并使用氨甲蝶呤作选择剂扩增整合的DNA。
优选RSV启动子,因为与其它启动子相比,它在启动转录上是有很高效率的。
优选CHO DHFR细胞,因其对氨甲蝶呤敏感。
通过常规蛋白分离技术,例如选择性沉淀、吸收光谱、和亲和层析包括单克隆抗体亲和柱层析将VZV IEP175蛋白质或衍生物从细胞培养物中纯化出来。
本发明还涉及含有免疫保护量之本发明VXV IEP 175蛋白质的疫苗。术语“免疫保护”是指足够量的VZV IEP175蛋白质,当施用于人时,足以避免或减轻疾病的产生保护性抗体或抗继后VZV感染的免疫反应。
因此,本发明提供了包含与药用载体、赋形剂或稀释剂混合之VZV IEP175或其衍生物的疫苗制剂。
本发明的疫苗另外还含有其它抗原成分诸如VZV gpⅠ、gpⅡ、gpⅢ、gpⅣ或gpⅤ或其截短的衍生物。特别是含有欧洲专利申请公开EP-A-0405867号中公开的截短之gpⅠ,gpⅡ或gpⅢ。本发明的最佳方案提供了含有与IEP175或其衍生物结合的无锚地gpⅡ组合物。EP-A-0405867号中公开的截短之gpⅡ的疫苗组合物。
术语“无锚地”是指VZV糖蛋白衍生物基本上没有所有的C末端锚地区域,且当在哺乳动物细胞中表达时可以被分泌出来。这些蛋白描述在EP-A-0405867中。
在另一实施方案中,提供了包含融合蛋白质的疫苗,该融合蛋白质含有包容IEP 175或衍生物的氨基酸顺序,以及包容gpⅠ、gpⅡ、gpⅢ、gpⅣ或gpⅤ或其衍生物之一的氨基酸顺序。
再一个实施方案中,涉及到VZV IEP 175或其衍生物在制造用于治疗或预防VZV感染之疫苗中的应用。
本发明还提供了用于医药的VZV IEP 175或其衍生物。本发明的再一个方面提供了治疗对VZV感染敏感或患有VZV感染之病人的方法。它包括使用安全且有效量的根据本发明的疫苗。
各疫苗剂量中蛋白质的量是作为能在典型接种者中诱导无明显不利副作用之免疫保护性反应的量而选择的。此量将根据所使用的特定免疫原和其怎么提供而有所不同。通常,期望各剂量含1-1000μg蛋白质,优选2-100μg,最优选4-40μg。特定疫苗的最佳量可由涉及在被试对象身上观察到的适当免疫反应的标准来确定。初始接种后,被试者可接受一次或几次有足够间隔的加强免疫接种。
为防止或显著降低带状疱疹的复发、发作次数、严重程度或持续时间,除了给对VZV感染敏感的人进行免疫接种外,本发明的药用组合物还可用于处置、免疫治疗患有VZV感染的病人。
本发明的疫苗中可直接使用VZV IEP 175蛋白质的水溶液。另外,可将冻干或未预先冻干的VZV IEP 175蛋白质混合在一起或与任何各种已知佐剂相混合。这些佐剂包括但不只限于氢氧化铝,胞壁酰二肽和皂甙如OuiaA,特别是QS21或3脱酰化-磷酰酸酯A(3D-MPL)。作为另一可替代的例子,可将该蛋白质包裹于微颗粒如脂质体中。在另一标准替代中,VZV IEP175蛋白质可被结合在免疫刺激性大分子上,如杀死的博德特氏杆菌或破伤风类病毒上。
疫苗制备一般地描述于New Trends and Developments in Vaccines.Voller et al.(eds),University Park Press,Baltimore,Maryland,1978中。在脂质体中胶囊包裹由Fullerton在美国专利4,235,877中作了描述。将蛋白质结合到大分子上公开于,例如,Likhite的美国专利4,372,954和Armor等的美国专利4,474,757中。Quil A的应用已由Dalsgaard等人在Acta Vet Scand,18:349(1977)中公开。3D-MPL可从Ribi immunochem,USA得到,并公开于英国专利申请2,220211号和美国专利4912094号中。QS21公开于美国专利5,057,540号中。
下列实施例说明但不是限定本发明。限制性内切酶或其它试剂基本上按照Vendors的说明书使用的。
实施例1
在用牛痘病毒重组体感染的细胞中表达IEP 175
VZV基因组DNA是从回收于水痘病人体内的病毒中提取的(材料由Dr.Rentier(Institut de Pathologie,Univesité de Liege Sart-Tilman,Liege,Belgium)提供)。
用EcoRl消化病毒DNA并分离约16.6Kb片段,其相当于碱基100441至117034(Davison et al.J.Gen.Virology,67,1759-1816,1986),然后克隆到质粒pUC9(一种标准的大肠杆菌克隆载体)的EcoRI位点中。由此质粒分离~7Kb SspI片段(102241至109293)并克隆到pUS19的incil位点中以产生质粒PNIV2017。该质粒编码完整的IEP 175蛋白质加上5′和3′未转译DNA。
然后用一套限制性内切酶消化质粒pNIV2017。产生三个片段:编码蛋白质N末端部分的有2199个碱基对的BstXI-BamHI片段、编码蛋白质C末端部分的1002个碱基对的BamHI-PpumⅠ片段和728个碱基对的PpumⅠ-MaeⅢ片段。将合成寡核苷酸加到这些片段上以产生由独特限制性位点侧接的编码暗盒。将该编码暗盒引入pUC19中储存(质粒pNIV2020)。进一步确定寡核苷酸及其相邻部分的顺序。从pNIV2020中回收IEP175编码暗盒,然后插入转移载体pULB5213(一种由Chakrabarti等描述的质粒pSC11的衍生物(Molecular and Cel-lular Biology 5.3403.3409,1985))中。最终构建体pNIV2026示于图2中。
将重组转移质粒pNIV2026转染到牛痘病毒感染的CV-1细胞中并在Bromo-Uridine筛选后分离重组病毒,并根据其在有X-gal存在时呈现的兰色进行噬斑纯化。将其称为VV 2026。对RAIZ工细胞的噬斑分析最好使用人H143成纤维细胞TK-细胞株。用于感染细胞的牛痘病毒为WR型(来源自Borgsiewicz L.K.)。步骤与前述获得牛痘病毒重组体的步骤相同(Mackett,M.and Smith,G.L.,J.Gen.Virology 67.2067-2082,1986;Mackett,M.,Smith,G.L.and Moss.B.,J.Virology 49,857-864,1984)。
用重组牛痘病毒VV2026,以1(Moil)感染多重性感染培养物中的CV-1细胞。感染后16和17小时收集感染的细胞(约3×105个/每次测定)和使用过的培养基(约2ml)。经Western吸印试验鉴定IEP 175蛋白质的存在。在12%SDS-聚丙烯酰胺凝胶上分辨蛋白质,转移到硝酸纤维素滤膜上并用抗IEP 175氨基酸顺序衍生(αα 1299至1310)之合成肽的小鼠血清探查之。按照标准方法,使用结合到碱性磷酸酶上的羊抗小鼠IgG和适当的生色底物检测复合物。
结果表明VV2026感染的细胞能有效地在细胞质中积聚IEP175,但不能将它输出到培养基中。重组蛋白质大小约为150K。

实施例2
在重组杆状病毒感染的昆虫细胞中表达IEP 175
鉴于生产出大量的重组IEP175蛋白质,我们采用了以杆状病毒和在培养基中的昆虫细胞为基础的表达系统。
从质粒pVIV2020开始,用EcoRI和XbaI消化以回收编码IEP 175的盒暗,并以钝端插入杆状病毒转移质粒pAcYMI中,质粒pAcYMI预先用BanHI切成平头。因此所得的重组质粒pNIV2038,在多角体启动子的控制下并以正确的方向载带编码IEP 175的顺序(图3)。
质粒pAcYMI是含有来自AcMNPY基因组(它包括多角体基因启动子,但不包括多角体基因)的顺序和来自高拷贝细菌质粒pUC8的顺序的杆状病毒穿梭载体。参见Matauura et al,J.Gen.Virol.68.1233-1250(1987)。
按公开的程序(Summer et al,TAES Bulletin NR 1555,May 1987;Texas Agricultural Experimental Station),与野生型DNA杆状病毒共转染,将重组杆状病毒转移质粒pNIV2038引入Spodoptera frugiperda(Sf9)昆虫细胞中,各自的比例为50比1μg。Spodoptera frugiperda细胞(Sf 9)可从ATCC(Rockville,Md,USA)得到。
收集上清液得到所产生的病毒颗粒。然后在噬斑检测中,用含该病毒的培养基感染Sf9细胞。分离并纯化出几种重组杆状病毒。然后用它们感染培养基中的Sf9细胞。感染后的不同时间内回收被感染细胞的总蛋白质,采用对IEP 175具特异性的小鼠抗肽血清(参见上文),用Western吸印法检测IEP 175的存在。在所有情况下,我们都不能证明在此系统中有完整IEP 175的表达。但我们观察到多种截短形式之蛋白质的表达,表明在细胞内发生广泛的蛋白水解降解作用。
重组杆状病毒转染质粒pNIV2038描述于图3中。
实施例3
在CHO细胞中表达IEP 175
为了试验附加的表达系统以获得IEP 175的大量表达,我们转向CHO系统。
从质粒pNIV2020开始,分离PstⅠ(已切成平头)-PvuⅡ 3946bp片段并插入质粒TDN的BgⅢ(已切成平头)和EcoRV位点之间,制成pNIV2042。质粒TDN描述于Connors等人的DNA,7,651-661(1988)一文中。它携带RSV LTR启动子、G418选择标志和DHFR扩增暗盒。pNVI12042编码组织血纤维蛋白溶酶原激活因子(tPA)的信号肽,接着是相应于成熟tPA之N末端氨基酸残基的4个氨基酸残基,然后接着IEP 175天然的起始蛋氨酸和该蛋白质的完整顺序(图1)。
通过电穿孔将质粒pNIV2042引入CHO dhfr-细胞。用geneticin(G418)选择重组细胞系并用氨甲蝶呤进行扩增。所有的步骤均按Moguilevsky等人(Eur.J.Biochem 197,605-614,1991)描述的步骤进行。
采用上述系统获得G418R克隆并检测全长度IEP175蛋白质的产生。显示出产生重组蛋白质的克隆用不同浓度(5至50nM)的氨甲蝶呤扩增并重新检测蛋白质产率。结果表明IEP可有效地在CHO细胞中产生,即它可在细胞质中积聚且其表观分子量为大约175千道尔顿。与昆虫细胞系统中观察到的结果相反,在CHO表达系统中没有蛋白水解降解作用。用50nM氨甲蝶呤扩增后获得最佳生产克隆18.5.22。用涉及两种对蛋白质(肽1299至1310和肽175-436)具有特异性的小鼠抗肽血清监测IEP 175的产生。如上所述用Western吸印分析法确定重组IEP 175的结构完整性。出乎预料的是,尽管IEP 175的DNA上存在信号肽顺序,但重组IEP 175并没有被分泌到CHO细胞培养基中。
CHO细胞的表达质粒pNIV2042描述于图4中。
为了证实该重组IEP蛋白质不仅结构上适合,而且还显示有天然蛋白质的已知调节功能,我们进行了如下试验(参见Jackers et al.1992;Liny et al,1992)。
用一套携带各种VZV启动子DNA顺序上游至报导基因氯霉素酰基转移酶(CAT)之编码顺序的质粒经电穿孔转染表达IEP175蛋白质的CHO细胞(克隆18-5-22)和对照组CHO细胞。(上述质粒可得自Professeus B.Rentier.Istitut de Pathologie,Université de Liège,Sart-Tilman,Liège,Belgium)。然后检测所得被转染细胞系的酶促活性(CAT),该酶促活性检测法用于检测IEP 175对VZV启动子功能的潜在激活效果。
表1概括了这些试验的结果。由表中可见,CHO细胞中产生的IEP 175蛋白质,在几种情况下能刺激启动子活性。这表明重组IEP 175蛋白质在此方面与其天然等同物的行为一样。

实施例4
构建编码缺乏亲核特型之可分泌性IEP 175蛋白质的DNA
4a.质粒pNIV2020(见实施例1)携带编码完整IEP 175蛋白质的顺序,并包括两个亲核特型。它们具有下列氨基酸顺序,特型1包含在氨基酸残基226和254之间且含有亲核氨基酸节段KSPKKKTLKVK;特型2包含在氨基酸残基648和733之间且含有亲核氨基酸节段PRKRKS。
用a)AatⅡ和SphⅠ,b)PpumⅠ和BstEⅡ消化pNIV2020,适当地修成平头,连在一起以重新构建缺乏含亲核特型之DNA顺序的质粒(图5)。
4b.回收编码所得截短之IEP 175的暗盒并插入质粒下游至指定tPA信号的顺序。方式如实施例3所述。
用所得质粒转化CHO细胞,可使IEP 175以可分泌的形式表达。
实施例5
IEP 175 gcⅡ融合体的构建
将4b中所述的片段作为融合体下游至编码gcⅡ的顺序(EP-A-405867)插入质粒。用所得的质粒转化CHO细胞,使之能够表达IEP 175截短gcⅡ的截短融合体蛋白质。
Claims (12)
1、功能上等同于天然VZV IEP 175的无VZV污染物的VZV IEP 175及其生理学功能衍生物。
2、根据权利要求1的VZV IEP 175,其具有与图1中描述的顺序基本相同的氨基酸顺序。
3、根据权利要求1的VZV IEP 175的生理学功能衍生物,它在表达后是可从适合的宿主中分泌的。
4、根据权利要求3的衍生物,其中缺失了氨基酸226至257和648至733。
5、一种融合蛋白质,其中该融合蛋白质的一部分包含VZV IEP 175或根据权利要求1-4的生理学功能衍生物。
6、根据权利要求5的融合蛋白质,其一部分包含来自VZV的无锚地gpⅡ蛋白质衍生物。
7、包含编码根据权利要求1-6中任一项之蛋白质并可操作地连接于在真核宿主细胞中起作用之调节区上的DNA顺序的载体。
8、一种生产根据权利要求1-6之任一项的蛋白质的方法,包括用根据权利要求7的载体转化宿主,并收回所得蛋白质。
9、含有与医药学上可接受的稀释剂、赋形剂或载体混合之根据权利要求1-6之任一项的蛋白质的疫苗组合物。
10、根据权利要求1-6之任一项的VZV IEP 175或其生理学功能衍生物在医药中的应用。
11、根据权利要求1-6之任一项的VZV IEP175或其生理学功能衍生物用于制造预防和治疗对VZV感染敏感之病人的疫苗。
12、一种预防性治疗对VZV感染敏感之病人的方法,包括混合无毒性有效剂量的根据权利要求1-6的蛋白质。
