五梨跤提取物的用途
技术领域
本发明涉及一种五梨跤提取物的用途。
背景技术
夏天的时候,人们着装较少,而太阳强烈,为了减轻长时间在户外活动被阳光灼伤肌肤的刺痛感,需要有防晒措施。从古至今,人们最常用的防晒工具是伞。然而,伞虽然能够防止大部分的太阳直射,但仍难以避免走路晃动或是其它方向反射、折射的阳光。且对于小孩,运动时不喜欢打伞。因此,防晒霜起到了很重要的作用。
紫外线依波长可分为UVA、UVB及UVC三种,而UVC波长最短(200nm~280nm)能量最强,但皆被大气层的臭氧层所吸收,因此对皮肤之影响不大。UVB(中波紫外线,280nm~320nm)的波长比UVA(320nm~400nm)的波长短,且能量较大。UVB会有立即性之伤害、红斑、起水泡,且无法穿透玻璃,作用至皮肤表皮,造成麦拉宁色素增加,甚至可能引起自由基物质之发生,而破坏皮肤细胞,造成皮肤癌。现今讲求以“天然”为目标,利用天然植物提取物做为防制皮肤光损伤之研究,天然物对皮肤具有温和、不刺激、安全性高等之特点,因此,发现新的天然防晒组分具有重要价值。
发明内容
本发明的主要目的,在于提供五梨跤提取物作为抗紫外组分的用途。
优选地,是在抗紫外或防晒产品中作为抗紫外组分的用途。
优选地,所述的产品包括天然防晒护肤品。
优选地,所述的五梨跤提取物为五梨跤低极性或非极性有机溶剂提取物。所述的低极性或非极性有机溶剂,包括乙醚、异丙醚、二氯甲烷、氯仿、环己烷、正己烷中的至少一种。
优选地,所述的五梨跤提取物为五梨跤正己烷提取物。
优选地,所述的五梨跤提取物提取方法包括,将新鲜五梨跤用低极性和/或非极性有机溶剂进行冷浸提取12-60h小时,过滤之后,去除溶剂,冷冻干燥,所得产物即为五梨跤提取物。
本发明所述的五梨跤,其拉丁文学名为Rhizophora mucronata,所述的五梨跤提取部位,优选为叶。其生长于滨海地区。本发明确认五梨跤低极性或非极性有机溶剂提取物尤其是五梨跤正己烷提取物,具有吸收紫外线的能力,进而达到防晒保护的效能。

具体实施方式
滨海植物五梨跤(Rhizophora mucronata)的萃取
(a)将采集回来的新鲜五梨跤叶片,称重并记录重量,再以粉碎机打碎,分别以纯水进行微波萃取(30分钟),以及正己烷溶剂进行冷浸提取48小时。
(b)过滤提取液在真空旋转浓缩机将溶剂抽干,再予以冷冻干燥6小时。
(c)所获得干燥产物,称重后放置-20℃冰箱冰存备用。
紫外线吸收能力之测定
将各个五梨跤水提与正己烷提取物与阳性对照标准品配制成0.40mg/ml之溶液,装入1ml之石英槽,以紫外/可见光分光光谱仪(Hitachi U-2900)测其吸光值,需先将空白样品放入归零,再放入样品测定其200nm~600nm之吸光值,并套入公式计算出消光系数。
提取物毒性评估
采用MTT方法检测皮肤角质细胞毒性。利用活细胞粒线体(mitochondria)内之脱氢酶(dehydrogenase)将MTT之四唑环(tetrazolium环)切断,形成甲臜(formazan)的蓝色沉淀物,使颜色由淡黄色转为深蓝色,而死细胞不含脱氢酶,所以MTT颜色并不会变化。在去除培养液后,加入DMSO(二甲基亚砜)溶出此蓝色沉淀物,即可用亮度计(multiscanreader)读取波长540nm之吸收度,可计算出细胞的存活或死亡率。实际应用系在96孔培养盘上做连续浓度稀释的实验组及不含化合物的控制组,加入一定浓度的细胞,培养三后,以MTT法呈色,扣除控制组结果,以同一样品在不同浓度下所得之百分比与浓度的对数值作线性回归,推算在百分之五十时的浓度值作为之毒性指标。
(4)pBR322质体DNA进行修复性试验
利用芬頓反应(Fenton reaction)原理产生自由基攻击DNA,若样品具有清除自由基能力,将可保护DNA不会断裂,经过电泳分析可验证其保护結果。质体pBR322质体DNA为超螺旋型型态(Supercoiled,SC),受氢氧自由基攻击后会使链结断裂,而造成损伤,并转变成开环(Open circular,OC)或线性(Linear)的形式。DNA受损的程度由SC和OC的比例变化表现,SC比例越大,DNA损伤越轻,OC比例越大则损伤越严重。实验方法:将定量之pBR322质体DNA取2μl于1.5ml离心管中,其次每管各加入3μl之1X PBS以及已配制成不同浓度之待测样品,最后分别各加入5μl之0.3%H2O2待测(反应条件为37℃,10分钟)。从每一离心管将已配制好之样品浓度分别取出置于石英玻片上。将石英玻片置于UV箱上,照射UV 30mJ/cm2。照射完,再分别各加入3μl之6X loading dye,混合均匀,再取出已制好的电泳胶片,分别把不同浓度的样品加到电泳胶片进行电泳。待电泳跑完后,戴上手套取出胶片移置已加入适量溴化乙锭(EtBr)的去离子水槽中,进行染色约30分钟。待染色完成,戴上手套取出胶片,以电泳胶片影像撷取分析系统拍照存盘。
(5)SPF之测定
SPF值(Sun Protection Factor)为防晒效的指标,亦称之为“防晒系數”,为物质防御紫外线的防御能力。(仪器名称:防晒系数测定仪Labsphere UV-1000S)
实验方法:将机器预先暖机20分钟,于石英玻片上黏上3M Tape,于280~400nm下进行背景空白校正,戴上指套将待测样品涂抹至3M Tape,每次涂抹标准品与各个萃取物2mg/cm2,于室温静置20分钟,于280~400nm下进行紫外线穿透率之测定,重复测定后即为平均之SPF值。
(五)结果
(1)提取率

(2)紫外线吸收能力之测定
市售防晒剂标准品之UV吸收值(0.4mg/mL)
五梨跤水萃取物之UV吸收值(0.40mg/mL)
五梨跤正己烷萃取物之UV吸收值(0.40mg/mL)
结果显示:以紫外/可见光分光光谱仪(Hitachi U-2900)测定,取最大吸收波长代入消光系数之公式计算,得知0.4mg/mL浓度的五梨跤提取物于UVB吸收范围确实有优异的吸收能力。
(3)细胞毒性评估
样品(100μg/mL、培养24小时) 类皮肤角质株化细胞株(HaCaT)存活度(%)
正己烷为溶剂以浸提法提取物 82.8±1.6%
结果显示:本实验针对类皮肤角质株化细胞株(HaCaT)观察24小时,经过24小时后,五梨跤正己烷提取物细胞存活率82.8%,显示其对HaCaT的毒性低。
(4)pBR322质体DNA进行修复性试验
结果显示:在未经处理的pBR322质体DNA(呈超螺旋型型态)经由芬頓反应后会产生氢氧自由基,导致DNA断裂。而作用于添加浓度0.40mg/mL五梨跤正己烷提取物后发现DNA的断裂现象减少,且皆维持超螺旋型型态,代表五梨跤正己烷提取物具有保护质体DNA的能力。
结论
1.在紫外线吸收能力的测定结果发现,虽然五梨跤萃取物的消光系数较标准品(Parsol MCX、Parsol 1789)低,但以正己烷萃取的五梨跤消光系数也高达5以上,说明五梨跤能抵抗紫外线的成分确实为低极性的部分。
2.在保护pBR322质体DNA的能力试验中,五梨跤正己烷提取物发现DNA的断裂逐渐减少,且质体DNA都维持超螺旋型型态,抑制芬頓反应所产生的氢氧自由基所引起的DNA断裂效果最好。
3.从紫外线光谱图可得之滨海植物的吸收范围大部分皆在UVB的范围内,可防止皮肤晒伤,可作为天然植物防晒原料。
以上所述,仅为本发明较佳实施例而已,故不能依此限定本发明实施的范围,即依本发明专利范围及说明书内容所作的等效变化与修饰,皆应仍属本发明涵盖的范围内。

3.根据权利要求2所述的五梨跤提取物的用途,其特征在于:所述的抗紫外产品或防晒产品包括防晒霜。
4.根据权利要求1所述的五梨跤提取物的用途,其特征在于:所述的五梨跤提取物为五梨跤低极性或非极性有机溶剂提取物。
5.根据权利要求4所述的五梨跤提取物的用途,其特征在于:所述的五梨跤提取物为五梨跤正己烷提取物。
6.根据权利要求1所述的五梨跤提取物的用途,其特征在于:所述的五梨跤提取物的提取方法包括,将新鲜五梨跤以低极性和/或非极性有机溶剂进行冷浸提取12-60h小时,过滤之后,去除溶剂,冷冻干燥,所得产物即为五梨跤提取物。
7.根据权利要求6所述的五梨跤提取物的用途,其特征在于:所述的新鲜五梨跤,为新鲜五梨跤叶片。
