Abstract

本发明涉及一种四氢姜黄素前体脂质体的制备方法,其特征在于采用以下步骤:A、按质量比称取四氢姜黄素0.5‑4%,复配剂1‑15%,膜材1‑10%,支撑剂0‑15%,抗氧化剂1‑10%,增溶剂1‑8%,冻干保护剂1%,余量无水乙醇;B、将所述四氢姜黄素、膜材、支撑剂、复配剂混合均匀,得到混合溶液I;C、将所述抗氧化剂和增溶剂加热至70℃,搅拌均匀,得混合溶液II;D、将所述混合溶液I加入到所述混合溶液II中,混合均匀,得复合脂质体混悬溶液,真空条件下除去溶液中的无水乙醇,得初乳液;E、将所述冻干保护剂加入步骤D所得初乳液中,冷冻干燥得四氢姜黄素前体脂质体。本发明具有水溶性好、稳定性好、药物包封率高、四氢姜黄素的生物利用度高等有益效果。

Description

一种四氢姜黄素前体脂质体的制备方法
技术领域
本发明涉及化妆品制备技术中的载体技术,特别涉及一种四氢姜黄素前体脂质体的制备方法。
背景技术
四氢姜黄素是由姜科植物姜黄(curcuma longa)根茎中分离出的姜黄素(curcumin)氢化而来。分子式为C21H26O6,分子量为372.2,密度1.222,熔点95℃-97℃,具有抑制酪氨酸酶的强效活性。四氢姜黄素的美白效果优于熊果苷;能有效抑制氧自由基(oxygen free radicals)的生成并能清除已经形成的自由基,具有明显的抗氧化作用,可用于抗人体皮肤衰老、修复、淡化色素、祛斑等,目前已作为天然功能性美白原料用于化妆品的研究开发。但其存在溶解性差的问题,难溶于水,所以生物利用率低。
脂质体是脂类分子(类脂)的自组装体,是一种由一个或多个脂质双层中间包覆微水相的结构。在脂质体的形成过程中,亲水的头部形成膜的内外表面层,而亲油性的尾部处于膜的中间,膜壁厚度约为5-7nm,而囊的直径一般在25-500nm之间。脂质体的这种结构使其能够携带各种亲水的、疏水的或两亲性的物质,这些物质被包入脂质体内部水相,或插入类脂双分子层,或吸附连结在脂质体的表面。脂质体的亲水性囊泡特殊结构,能够提高被包覆成分的稳定性、促进活性成分的透皮吸收、延长作用时间、对局部部位靶向作用、降低毒副作用等。现有技术中有采用脂质体包裹四氢姜黄素的,比如中国发明专利申请“负载四氢姜黄素的纳米载体及其制备方法”,专利申请号“201310733658.9”,中国发明专利“一种复合美白脂质纳米粒乳液的制备方法”,专利号“201210295096.X”。但脂质体混悬液在贮存期间易发生聚集、融合及药物渗漏,尤其水溶性药物的渗漏更显著;且磷脂易氧化、水解,难以满足药物制剂稳定性的要求。
发明内容
本发明要解决的技术问题是提供一种水溶性好、稳定性好、药物包封率高、生物利用度高的四氢姜黄素前体脂质体。
为解决上述技术问题,本发明采用以下技术方案:一种四氢姜黄素前体脂质体的制备方法,其特征在于采用以下步骤:
A、按如下质量比称取四氢姜黄素0.5-4%,复配剂1-15%,膜材1-10%,支撑剂0-15%,抗氧化剂1-10%,增溶剂1-8%,冻干保护剂1%,余量无水乙醇;所述复配剂为单硬脂酸甘油酯,所述膜材为大豆卵磷脂,所述支撑剂为胆固醇,所述抗氧化剂为维生素C,所述增溶剂为聚山梨醇酯20,所述冻干保护剂为甘露醇。
B、将步骤A中称取的四氢姜黄素、膜材、支撑剂、复配剂混合均匀,溶于所述无水乙醇,加热至60℃,搅拌至完全溶解,得到混合溶液I;
C、将步骤A中称取的抗氧化剂和增溶剂加热至70℃,搅拌均匀,得混合溶液II;
D、将步骤B所得混合溶液 I 加入到步骤 C所得混合溶液 II中,混合均匀,得复合脂质体混悬溶液,真空条件下,除去溶液中的无水乙醇,得初乳液;
E、将步骤A所述冻干保护剂加入步骤D所得初乳液中,在-20℃下预冻3-12 h;然后在真空环境下,-20℃冷冻至水印消失;然后升温至20-30℃,持续干燥4-12h,得四氢姜黄素前体脂质体。
本发明制备的四氢姜黄素前体脂质体,相对于现有四氢姜黄素载体技术,具有包封率高的优点,包封率>98%,经有效包覆的四氢姜黄素使用过程中损失小,稳定性好。本发明制备的四氢姜黄素前体脂质体经水合后为纳米级,水分散性好,能够广泛应用于各类化妆品的制备中。
本发明将四氢姜黄素制备成前体脂质体,使用时,将其溶于水或缓冲液中,即可形成脂质体。由于所得四氢姜黄素前体脂质体为固体制剂,克服了普通脂质体存在的聚集、沉降、药物渗漏及磷脂氧化等技术问题,相对于现有的普通脂质体具有稳定性好的优点,另外还有便于贮存和运输的优点。
本发明采用单硬脂酸甘油酯、大豆卵磷脂、胆固醇、维生素C、聚山梨醇酯20等进行包裹,采用玻璃化温度合适的甘露醇作为冻干保护剂,制得的四氢姜黄素前体脂质体疏松、水分散性好,溶于水后脂质体粒径分布均匀 ,药物包封率高,四氢姜黄素的包封率>98%。
综上所述,本发明制备的四氢姜黄素前体脂质体具有水溶性好、稳定性好、药物包封率高、四氢姜黄素的生物利用度高等有益效果,便于将四氢姜黄素加入到各类化妆品中使用。

具体实施方式:
以下结合具体实施例对上述方案作进一步说明。以下所述仅为体现本发明的目的、效果和技术特征的较佳实施例,并不因此而限定本发明的保护范围。
实施例1:
一种四氢姜黄素前体脂质体的制备方法,采用以下步骤:称取四氢姜黄素1%、大豆卵磷脂5%、胆固醇3%、单硬脂酸甘油酯10%,搅拌使之混合均匀,溶于无水乙醇70%,加热至60℃,充分搅拌至完全溶解,得到混合溶液I;称取维生素C 8%、聚山梨醇酯20 3%,加热至70℃,充分搅拌搅拌至完全均匀,得到混合溶液II;将得到的混合溶液I倒入混合溶液II,搅拌使之混合均匀,即得到复合脂质体混悬溶液,然后在真空条件下,除去溶液中的无水乙醇,制成初乳液;在初乳液中加入10%甘露醇溶液,在-20℃下预冻12 h。冷冻干燥过程:先降低冷阱温度,放入预冻好的样品,抽真空,待水印消失后,温度升至20℃,继续干燥4h,取出,得到的冻干样品密封、避光,常温保存于干燥容器中,即得所述四氢姜黄素前体脂质体。如图1所示,该四氢姜黄素前体脂质体经激光粒度仪测定其平均粒径约为37.3nm。如图2所示,该四氢姜黄素前体脂质体分别在25℃、40℃和光照条件下存储28天,其平均粒径(Averagesize)无明显变化,说明稳定性很好。如图3所示,该四氢姜黄素前体脂质体在不同稀释度的情况下,平均粒径(Average size)和聚合物分散性指数(PDI)无明显变化,说明水分散性很好。
实施例2:
一种四氢姜黄素前体脂质体的制备方法,采用以下步骤:称取四氢姜黄素4%、大豆卵磷脂8%、胆固醇5%、单硬脂酸甘油酯7%,搅拌使之混合均匀,溶于无水乙醇40%,加热至60℃,充分搅拌至完全溶解,得到混合溶液I;称取维生素C 6%、聚山梨醇酯20 5%和无水乙醇25%,加热至70℃,充分搅拌搅拌至完全均匀,得到混合溶液II;将得到的混合溶液I倒入混合溶液II,搅拌使之混合均匀,即得到复合脂质体混悬溶液,然后在真空条件下,除去溶液中的无水乙醇,制成初乳液;在初乳液中加入10%甘露醇溶液,在-20℃下预冻8h。冷冻干燥过程:先降低冷阱温度,放入预冻好的样品,抽真空,待水印消失后,温度升至25℃,继续干燥6h,取出,得到的冻干样品密封、避光,常温保存于干燥容器中,即得所述四氢姜黄素前体脂质体,该四氢姜黄素前体脂质体经激光粒度仪测定其平均粒径约为42nm。
实施例3:
一种四氢姜黄素前体脂质体的制备方法,采用以下步骤:称取四氢姜黄素2.5%、大豆卵磷脂3%、胆固醇5%、单硬脂酸甘油酯和芦巴胶油12%,搅拌使之混合均匀,溶于无水乙醇35%,加热至60℃,充分搅拌至完全溶解,得到混合溶液I;称取维生素C 5%、聚山梨醇酯20 6%和无水乙醇31.5%,加热至70℃,充分搅拌搅拌至完全均匀,得到混合溶液II;将得到的混合溶液I倒入混合溶液II,搅拌使之混合均匀,即得到复合脂质体混悬溶液,然后在真空条件下,除去溶液中的无水乙醇,制成初乳液;在初乳液中加入15%甘露醇溶液,在-20℃下预冻7h。冷冻干燥过程:先降低冷阱温度,放入预冻好的样品,抽真空,待水印消失后,温度升至22℃,继续干燥12h,取出,得到的冻干样品密封、避光,常温保存于干燥容器中,即得所述四氢姜黄素前体脂质体,该四氢姜黄素前体脂质体经激光粒度仪测定其平均粒径约为65nm。

Claims (1)

1.一种四氢姜黄素前体脂质体的制备方法,其特征在于采用以下步骤:
A、按如下质量比称取四氢姜黄素0.5-4%,复配剂1-15%,膜材1-10%,支撑剂0-15%,抗氧化剂1-10%,增溶剂1-8%,冻干保护剂1%,余量无水乙醇;所述复配剂为单硬脂酸甘油酯,所述膜材为大豆卵磷脂,所述支撑剂为胆固醇,所述抗氧化剂为维生素C,所述增溶剂为聚山梨醇酯20,所述冻干保护剂为甘露醇;
B、将步骤A中称取的四氢姜黄素、膜材、支撑剂、复配剂混合均匀,溶于所述无水乙醇,加热至60℃,搅拌至完全溶解,得到混合溶液I;
C、将步骤A中称取的抗氧化剂和增溶剂加热至70℃,搅拌均匀,得混合溶液II;
D、将步骤B所得混合溶液 I 加入到步骤 C所得混合溶液 II中,混合均匀,得复合脂质体混悬溶液,真空条件下,除去溶液中的无水乙醇,得初乳液;
E、将步骤A所述冻干保护剂加入步骤D所得初乳液中,在-20℃下预冻3-12 h;然后在真空环境下,-20℃冷冻至水印消失;然后升温至20-30℃,持续干燥4-12h,得四氢姜黄素前体脂质体。
