Abstract
本发明公开了一种具有专一性抗痤疮丙酸杆菌作用及抗炎作用的短脂肽,属于生物技术领域。所述短脂肽是由C6~C20脂肪酸与通式为KWKW的氨基酸序列组成。本发明的短脂肽能够专一性抑制痤疮丙酸杆菌生长,减少毛囊周围组织的炎症反应,体内、体外实验表明本发明短脂肽具有较高的抗菌、抗炎活性,较低毒性。此外,该短脂肽具有与传统抗生素不同的作用机制,短脂肽极难产生耐药性,尤其是对现有抗生素已经产生耐药的菌株有效,因而能够达到防治痤疮,提高生活质量的根本目的。本发明的短脂肽是含有脂质与氨基酸的化合物或复合体,可以添加于各种洗护用品或者化妆品中,安全环保,具有广阔的市场前景。

本发明属于生物技术领域,具体涉及一种具有专一性抗痤疮丙酸杆菌作用及抗炎作用的短脂肽。
背景技术
痤疮是一种青少年常见的毛囊皮脂腺的慢性炎症性皮肤病,多发于头面部,颈部等皮脂腺丰富的部位,对人体主要危害表现在:(1)引起继发性感染,影响皮肤的正常功能,严重的还可形成皮肤癌;(2)可能导致形成痘斑、痘坑、痘疤痕等,影响青年择业、择偶、交际,引发青春期忧郁症,性格变得孤僻,自卑,影响患者的心理健康。引起痤疮的主要致病因素之一是痤疮丙酸杆菌,目前对于中度到重度炎症性损害一般通过全身或局部抗菌药物治疗。但传统的抗生素易使细菌产生耐药性,导致疗效下降,因此迫切需要研制新型的药物。
痤疮的发病机制主要与下列4个因素有关:皮脂分泌过多、毛囊皮脂腺导管开口部位过度角化、痤疮丙酸杆菌的定植与侵袭繁殖,以及炎症反应。其中,痤疮丙酸杆菌在痤疮的发生中起重要作用。痤疮丙酸杆菌为毛囊内的正常共生菌,为革兰阳性厌氧杆菌,主要定植在毛囊管深部毛囊壁角质层最薄弱的部位。皮脂分泌的增加可引起亚油酸浓度的下降,进而导致毛囊壁的屏障功能受损,致使在此部位痤疮丙酸杆菌极易与角质形成细胞及朗格汉斯细胞直接接触,通过多种途径启动对痤疮丙酸杆菌天然及适应性免疫应答,导致痤疮炎症的发生。
除引起痤疮外,痤疮丙酸杆菌还在多个致病因素中起重要作用,例如,可引起感染性心内膜炎、血栓性静脉炎、骨炎、关节炎、急性化脓性心包炎及其他手术后感染。因此,选择性杀灭痤疮丙酸杆菌将极大的有利于痤疮及其他疾病的治疗,缓解因痤疮丙酸杆菌引起的炎症反应,降低耐药性细菌的产生。
抗菌肽作为一类新的抗生素具有抗菌活性高,抗菌谱广,对真核和原核生物有较好的选择性以及目标靶菌株不易产生抗性突变等特点,已经吸引了人们极大的兴趣和关注。在杀灭P.acnes方面,虽然有所报道,但相对较少,这些工作主要表现在以下几个方面:(1)研究表明,粒溶素及其衍生多肽可以在体外有效地杀灭痤疮丙酸杆菌。在其氨基酸序列中用色氨酸代替缬氨酸后,可以增加其与细菌表面的结合从而加强其抗菌活性。当用D型氨基酸取代原来的L型氨基酸后,得到的衍生物不易被蛋白酶降解,而且对杀死痤疮丙酸杆菌更有效。更重要的是,粒溶素衍生多肽还可抑制由痤疮丙酸杆菌引发的细胞因子的释放,有较好的抗炎作用,可以用于外用制剂治疗痤疮。(2)最近,Conlon等从青蛙的皮中分离得到五个抗P.acnes的多肽,MIC范围在3-12.5μM,研究同时发现这些多肽表现出较强的降低TNF-α和IFN-γ的作用。(3)2013年,我国昆明动物所的赖仞等通过设计的方法得到一个含15个氨基酸残基的小肽LZ1,VKRWKKWWRKWKKWV-NH2(SEQ ID NO.2),该肽不但显示了较好的杀灭P.acnes的活性(MIC=0.6μg/mL)而且能够降低TNF-α和IL-1β浓度,同时毒性较低,显示良好的应用前景。
在过去的五十年间主要采用抗生素来治疗痤疮。抗生素可直接作用于P.acnes。近年来,由于痤疮治疗过程中系统或局部广谱抗生素的普遍滥用,P.acnes的耐药现象明显增强,而且存在着交叉耐药,导致系统使用常规剂量的抗生素在毛囊皮脂腺局部难以达到有效的药物浓度,最终导致疗效降低。
后续不断出现一些替代药物,如BPO(过氧化苯甲酰)和维生素A酸类药物。BPO有显著抗菌活性,缺点不抗炎,使用时伴有皮肤局部红疹、刺痛、灼热等副作用。Retinoids抗炎效果好,缺点易引起胎儿畸形。
使用抗菌肽治疗痤疮成为新的思路,但是目前发现的这些抗痤疮杆菌多肽化合物存在一定的不足之处,例如含有较多的氨基酸残基(>15AA),或者虽然较短(12个氨基酸),但活性较低(>4μg/mL),并且在生理条件下易被酶分解,稳定性较差等。这些缺陷限制了它们的应用,因此有必要提高其生理条件下的稳定性,同时进一步减少它们的氨基酸残基的数目以降低成本。其次,这些肽多具有广谱的抗菌活性,专一性较差,使用时可能会把人体有益的菌群也一齐杀灭,从而造成二次感染。
发明内容
本发明的目的在于克服现有技术的不足,提供一种具有专一性抗痤疮丙酸杆菌作用及抗炎作用的短脂肽及添加或包含所述短脂肽的各种相关产品。所述短脂肽,其氨基酸序列长度比较短,不会产生免疫原性反应,并且易于合成,成本低。
本发明所采取的技术方案是:
一种短脂肽,所述短脂肽是由C6~C20脂肪酸与通式为KWKW(SEQ ID NO.1)的氨基酸序列组成,其中C6~C20表征碳链长度。
优选的,所述氨基酸为L-构型氨基酸或D-构型氨基酸。发明人研究发现D-构型氨基酸构建的短脂肽与L-构型氨基酸构建的短脂肽抗菌活性相差不大。
优选的,氨基酸序列的两端需进行修饰。
由于蛋白酶的水解作用,传统的肽类化合物半衰期较短。本发明实施例中采用的是Fmoc保护基团对KWKW的氨基酸序列的C端和N端进行修饰。修饰后的氨基酸序列具有较高的酶水解稳定性,其稳定性远远高于传统未加修饰的肽类化合物。此外,本领域技术人员也可以按照公知常识进行别的基团的修饰,以提高氨基酸的稳定性。
优选的,所述脂肪酸为C10、C12、C14、C16、C18脂肪酸。

根据2007年8月27日国家质检总局公布的《化妆品标识管理规定》,化妆品是指以涂抹、喷洒或者其他类似方法,散布于人体表面的任何部位,如皮肤、毛发、指趾甲、唇齿等,以达到清洁、保养、美容、修饰和改变外观,或者修正人体气味,保持良好状态为目的的化学工业品或精细化工产品。添加本发明的短脂肽后,可以给增加化妆品的祛痘功效,适合有这方面需求的人群。
本发明的有益效果是:
本发明的短脂肽能够专一性抑制痤疮丙酸杆菌生长,减少毛囊周围组织的炎症反应,现有体内、体外实验表明本发明短脂肽具有较高的抗菌、抗炎活性,较低毒性。本发明的短脂肽,其氨基酸序列长度比较短,易于合成,成本低。另外,本发明短脂肽具有与传统抗生素不同的作用机制,短脂肽极难产生耐药性,尤其是对现有抗生素已经产生耐药的菌株有效,因而能够达到防治痤疮,提高生活质量的根本目的。
本发明的短脂肽是含有脂质与氨基酸的化合物或复合体,可以添加于各种洗护用品或者化妆品中,安全环保,具有广阔的市场前景。

具体实施方式
如无特殊说明,实施例中所涉及的氨基酸均为L-构型氨基酸。此外,发明人也尝试过D-构型氨基酸,发现D-构型氨基酸构建的短脂肽与L-构型氨基酸构建的短脂肽抗菌活性相差不大。
实施例中所用的对比药物包括:
Melittin:蜂毒肽是蜂毒素(melittin,MLT)又叫蜂毒肽,是蜂毒的主要成分,约占蜂毒干重的50%,也是蜂毒中具药理作用和生物学活性的主要组分。MLT呈强碱性,易溶于水,相对分子质量为2 849,具有抗炎、降压、镇痛、抑制血小板凝集、抗辐射、抗菌、抗HIV、抗风湿性关节炎及抗肿瘤等多种药理活性。但是其临床应用却有很多局限,主要原因是现有技术难以将蜂毒中有致敏反应并与蜂毒肽分子量接近的磷脂酶A2完全去除。
Clindamycin:克林霉素是一种外观为白色结晶粉末的化学品,化学式为C18H33ClN2O5S,分子量为424.98300,熔点141~143℃。克林霉素为抗生素类药,临床上主要用于厌氧菌引起的腹腔和妇科感染,是金黄色葡萄球菌骨髓炎首选治疗药物。此外,克林霉素也用于痤疮的外用治疗。
下面结合具体实施例对本发明做进一步的说明,但不限于此。
实施例1
1.1实验方法
1.1.1多肽合成
通过使用酰胺MHBA树脂和标准的9-芴基甲氧基羰基(Fmoc)氨基酸的固相合成来合成肽。肽延伸反应条件如下:标准HBTU/HOBt用作偶联剂,N-二甲基甲酰胺作为溶剂,10倍过量的二异丙基乙胺,3倍过量的Fmoc保护基团氨基酸或5倍过量的游离脂肪酸。使用由87.5%三氟乙酸,2.5%乙二硫醇,5%茴香硫醚和5%去离子水组成的试剂从树脂上裂解肽,室温下反应3小时,之后将肽在甲基叔丁基醚-石油醚(体积比1:1)中沉淀。通过电喷雾电离质谱(ESI-MS,Waters,USA)确认肽的分子量。在C18柱(250×4.6mm;Shimadzu,Kyoto,Japan)上用Shimadazu 10AHPLC仪器分析肽的纯度,流动相由溶剂A(含有0.075%三氟乙酸的水)和溶剂B(含有0.075%三氟乙酸的甲醇)组成。梯度:15%至20%B 2分钟,20%至60%B 6分钟,60%至80%B 4分钟,80%至90%B 4分钟。所有短脂肽的纯度都在95%以上。
1.1.2菌株培养
痤疮丙酸杆菌(ATCC6919和ATCC11827)使用布氏肉汤培养基在厌氧条件下培养;表皮葡萄球菌(ATCC12228),铜绿假单胞菌(ATCC25853)和大肠杆菌(ATCC25922)使用LB培养基在需氧条件下中培养;变形链球菌(ATCC25175和UA159)使用BHI培养基在厌氧条件下中培养;金黄色葡萄球菌(ATCC12600和临床分离菌株)使用MH培养基在需氧条件下培养。所有菌株均在37℃下培养。热灭活痤疮丙酸杆菌的制作方法为将菌液置于80℃下孵育30分钟。
1.1.3最小抑菌浓度确定
96孔药敏板的制备:脂肪酸的最终测试浓度为125-3.9μg/mL,短脂肽浓度为31.2-0.5μg/mL。在对数生长期取细菌悬液,将痤疮丙酸杆菌稀释至1×106CFU/mL的最终浓度,并将其他菌株稀释至1×105CFU/mL。每孔中加入100μL细菌悬浮液,痤疮丙酸杆菌在96小时后读取结果,其他菌株在17-20小时读取结果,取没有细菌生长迹象的最低观察药物浓度为MIC值。每组测试结果独立重复4至6次。
1.1.4杀菌时效/量效关系测试
将P.acnes ATCC11827悬浮液稀释至1×106CFU/mL,接着分别加入终浓度为10μg/mL的C16-KWKW或0.5μg/mL的克林霉素在25℃下厌氧分别处理0,1,2,4,6,8,10小时,进行杀菌时效关系测定;或是加入终浓度为1,10,100μg/mL的C16-KWKW或0.01,0.1,1μg/mL克林霉素在25℃厌氧处理4小时,进行杀菌量效关系测定。之后将菌液分别按照1:10,1:100,1:1000比例稀释,并将20μL各稀释液置于布氏肉汤琼脂培养基平板上,在37℃无氧条件下培养5-7天,记录平板菌落数。
1.1.5细胞毒性测试
使用CCK-8法测定短脂肽对HaCaT和RAW264.7的细胞毒性。将细胞(2-5×104/孔)接种在96孔板中,生长过夜后,加入含有2-125μg/mL肽的新鲜培养基,HaCaT细胞37℃作用24小时,RAW264.7细胞37℃作用8小时。之后每孔加入10μL CCK-8(Dojindo,日本),37℃继续孵育2小时。使用酶标仪(Infinite M1000Pro,Tecan,瑞士)在450nm处测定吸光度。通过比较经过短脂肽处理后细胞的生长抑制率来评估短脂肽的细胞毒性,未经短脂肽处理的对照组细胞的抑制率设定为0%。
1.1.6酶联免疫吸附实验
将RAW264.7细胞(2-5×104/孔)接种在96孔板中并生长过夜,加入含有1,10,25μg/mL C16-KWKW或1μg/mL Melittin的新鲜培养基,2小时后,每孔中加入终浓度为100ng/mL的LPS或1×107CFU/mL热灭活的P.acnes。37℃,5%CO2条件下孵育6h,收集细胞培养液,根据试剂说明书(Neobioscience,中国)进行酶联免疫实验,测定TNF-α,IL-1β和IL-8水平。
1.1.7荧光实时定量PCR
使用Trizol试剂(Sigma,USA)提取总RNA。分光光度法测定RNA的产量和纯度。使用PrimeScript RT Master Mix(Takara,日本)按照说明合成cDNA。使用SYBR-green Ⅱ(Takara,Japan)荧光试剂通过ABI 7500序列检测系统(PE Applied Biosystems,USA)进行实时荧光定量PCR。反应条件如下:起始95℃30秒,接着95℃5秒,60℃34秒持续40个循环,之后95℃15秒,60℃1分钟,95℃15秒。每个样品测试三次。使用2-ΔΔCt方法比较基因相对含量表达。PCR引物序列如下:

1.1.8稳转NF-κB细胞系构建及荧光素酶报告基因检测
RAW264.7细胞以5×104/孔的密度接种在24孔板中孵育过夜,每孔按照140μL的PEI(100ng/mL)及112μL的pNF-κB-luc DNA(1μg/mL)的比例制作DNA-PEI混合物,室温下孵育45分钟。接着吸出细胞培养液,轻轻滴加DNA-PEI混合物并孵育4-6小时。之后将混合物吸出,更换含有10%胎牛血清的DMEM培养细胞。当细胞达到约80%浓度时,向培养基中加入1mg/mL选择性抗生素G418,筛选稳定转染的抗性细胞2周。
将稳定转染的RAW264.7细胞以5×104/孔接种在96孔板中,待细胞生长过夜后,分别加入含有1,10,25μg/mL C16-KWKW或10μg/mL PDTC。2小时后,将终浓度为100ng/mL的LPS或1×107CFU/mL的热灭活P.acnes加入各孔中,37℃孵育6小时。接着弃掉上清液,每孔加入100μL裂解液,避光孵育20分钟后,将细胞裂解液转移到白色不透明板上,各孔加入100μL化学发光底物,通过酶标仪(Infinite M1000Pro,Tecan,Swiss)检测化学发光。
1.1.9等温量热滴定法
将细菌脂质溶解在5%DMSO中,浓度为4mg/mL。脱气处理10分钟后,通过ITC法测定C16-KWKW(150μg/mL)与脂质相互作用的微热量的变化(MicroCal PEAQ-ITC,马尔文,英国)。200μg/mL Melittin作为对照组。ITC反应条件如下:环境温度:25℃;滴定数量:19;滴定体积:2μL;滴定间隔:100秒;参比功率:10μcal/sec;搅拌速度:750转/分钟。使用MicroCal PEAQ-ITC分析软件对实验数据进行分析,计算各热力学参数。细菌脂质平均分子量设定为3000。
1.1.10色氨酸荧光光谱法
将制备的细菌脂质调整至浓度为4mg/mL,加入终浓度为150μg/mL的C16-KWKW或C10-KWKW处理2小时。通过荧光光谱法(FLS 980,爱丁堡,英国)检测色氨酸荧光发射光谱,激发波长设定为280nm,扫描范围300-400nm。以150μg/mL Melittin与脂质的作用结果作为对照。
1.1.11透射电镜
P.acnes调整浓度为5×106CFU/mL,用10μg/mL C16-KWKW处理2小时。离心(1000r/min,5分钟)弃掉上清液,PBS缓冲液洗涤两次,之后用2.5%戊二醛处理过夜。用0.1M磷酸盐冲洗至少三次,每次15分钟,用1%四氧化锇固定2-3小时。脱水条件:50%乙醇15-20分钟,70%乙醇15-20分钟,90%乙醇15-20分钟,90%乙醇和90%丙酮15-20分钟(1:1,v/v)。孵育条件:丙酮和包埋液(2:1,v/v)室温下孵育3-4小时,丙酮和包埋液(1:2,v/v)37℃孵育2-3小时。固化条件:37℃过夜,45℃12小时,60℃24小时。固化后切片,用3%醋酸双氧铀和柠檬酸铅作为染料,用透射电子显微镜观察样品。
1.1.12紫外吸收物检测
紫外吸收物的检测方法如文献所述并稍作修改。P.acnes培养至对数生长期,用PBS缓冲液稀释至1×106CFU/mL。加入终浓度为10μg/mL的C16-KWKW,25℃分别作用0,2,4,6,8,10小时。10μg/mL Melittin为阳性对照组,0.1g/mL克林霉素为阴性对照组。用0.229μm纤维素酯微孔膜将菌液过滤,使用紫外分光光度计(UV757CRT,中国)在260nm处测量滤液的吸光度。
1.1.13PI摄取实验
将P.acnes菌悬液稀释至1×106CFU/mL,分别加入终浓度为1,10,100μg/mL的C16-KWKW,25℃处理4小时。1000r/min离心10分钟,弃掉上清液,PBS缓冲液洗涤两次,加入终浓度为30μM的PI染色10分钟后,1000r/min离心10分钟,弃掉过量的PI并用PBS缓冲液洗涤两次。之后将细菌用1mL PBS缓冲液重悬浮,通过流式细胞仪(FACSCanto II,BD,USA)检测荧光强度。剩余的细菌悬液以每孔100μL加入黑色96孔板中,通过荧光酶标仪(InfiniteM1000Pro,Tecan,Swiss)检测荧光强度。其中激发波长和发射波长分别设定为535nm和615nm。
1.1.14体内实验
20只6周龄雌性昆明小鼠购自南方医科大学实验动物中心。小鼠随机分为4组,每组5只。一组为空白对照,其余三组小鼠双耳内皮内注射浓度为3.0×107CFU/mL的P.acnes菌悬液40μL。之后将100,200μg C16-KWKW或10μg分别与0.05g凡士林混合后涂抹于右耳表面。同时,左耳涂抹等量凡士林作为对照。24小时后,颈椎脱臼法处死小鼠并迅速切除耳朵。使用微量厚度测量厚度后,将耳组织加入液氮中研磨成粉末状,加入500μL PBS匀浆。3000r/min离心15min后,收集上清液,在布氏肉汤琼脂平板上37℃厌氧培养96小时后,记录菌落数量;同时通过qRT-PCR(7500,PE Applied Biosystems,USA)测量匀浆液中TNF-α,IL-1β和iNOS含量。剩余的耳组织制作石蜡包埋切片,HE染色后进行组织学检查。
1.1.15统计学分析
每组数据重复至少三次,用平均值±标准偏差(SD)表示。数据通过SPSS13.0软件进行单向方差分析或配对样本T检验。统计学显着性差异定义为*P<0.05,**P<0.01,***P<0.001。
1.2实验结果
1.2.1确认了目标化合物KWKW,并通过连接不同长度脂肪酸的方法进行结构修饰改造前期研究中我们对几种天然存在的抗菌肽进行结构分析,发现氨基酸R,W和K是抗菌肽序列中发挥抗菌活性的关键残基。因此,我们以K,W和R作为基本骨架,通过与不同长度的脂肪酸相连接来设计抗菌肽库。在目前的研究中,考虑到脂肪酸对于P.acnes的定殖起关键作用,我们使用痤疮丙酸杆菌作为靶向细菌对肽库进行筛选,结果见表1。

表1结果显示,C10-KWKW表现出良好的抗P.acnes活性,最小抑制浓度(MIC)达到3.9μg/mL。
接下来我们通过测定从C6-C20不同脂肪酸长度对肽类化合物KWKW的抗菌活性,对C10-KWKW进行结构修饰改造,如表2中所列。

1.2.2短脂肽C16-KWKW具有高效专一抗P.acnes活性
通过测定MIC来确认所设计的短脂肽对P.acnes的抗菌活性。从表1中的数据可以看出,这些短脂肽的抗菌活性受脂肪链长度的影响,其中C16-KWKW表现出最高的抗P.acnes活性,其MIC值为2.0μg/mL,低于其同系物C10-KWKW(3.9μg/mL)C12-KWKW(3.9μg/mL)、C14-KWKW(3.9μg/mL)和C18-KWKW(7.8μg/mL)。此外我们发现,C16-KWKW对P.acnes的MIC值明显低于S.mutans和E.coli等其他测试菌株,这表明C16-KWKW具有专一性抑制P.acnes的特性,该结论通过实时定量PCR法得到进一步证实。我们分别用0.5,1,2μg/mL浓度C16-KWKW处理P.acnes,S.mutans,E.coli菌株4小时,对照组菌液未经C16-KWKW处理,比较两者16S rRNA基因的表达量,结果如图1a所示,经C16-KWKW处理后,16S rRNA基因在P.acnes中的表达量显著降低,而在S.mutans,E.coli菌株中几乎没有影响。
为了确认短脂肽的抗P.acnes活性是否来自单独的脂肪酸本身,我们测试了不同长度脂肪酸的抗P.acnes活性。结果如表3所示。

表3结果显示:在相同的条件下,只有C12和C14脂肪酸展示出微弱的抗P.acnes活性,IC为62.5μg/mL,而其他长度脂肪酸在我们的测试范围内均未表现出抗P.acnes活性。这说明对于P.acnes的抗性主要来源于氨基酸,而不是脂肪酸。然而如前所述,不同的脂肪酸也会影响短脂肽的抗痤疮丙酸杆菌活性。
虽然说,抗菌肽对于细菌的主要抗菌机制有一定共性,然后针对不同的细菌,如痤疮丙酸杆菌,C16-KWKW可能还有其他作用机制共同作用。目前所述作用机理还在研究中。
由于抗菌肽易受到盐离子,尤其是二价阳离子的影响而降低活性,对其实际应用产生重要影响。我们评估了C16-KWKW在不同浓度盐离子溶液中的抗P.acnes活性。结果如表4所示。

表4结果显示:在10mM浓度的CaCl2,NaCl和MgCl2溶液中,C16-KWKW的杀菌效果并未受到明显影响。
1.2.3短脂肽C16-KWKW的杀菌速度高于克林霉素
在确认了短脂肽C16-KWKW的抗P.acnes活性后,我们进一步测试了C16-KWKW抗菌时效(图1b)和量效(图1c)关系。时效关系结果如图1b所示,用10×MIC浓度的C16-KWKW处理P.acnes(1×106CFU/mL)4小时后,约有5×104个菌落存活,而相同条件下用克林霉素处理后,剩余菌落数约9×105个。6小时后,C16-KWKW处理组仅观察到3×103个菌落,而克林霉素处理组中仍有约2×105个菌落剩余。
量效关系结果进一步确认了短脂肽C16-KWKW相比克林霉素具有更快的杀菌效果。分别用0.5×,5×,50×浓度的C16-KWKW或克林霉素处理P.acnes 4小时,记录平板菌落数,结果如图1c所示,C16-KWKW和克林霉素的杀菌效果具有剂量依赖关系,其中5×,50×浓度C16-KWKW的杀菌速度明显快于相同条件下的克林霉素。C16-KWKW和克林霉素在0.5×浓度时均未发现杀菌效果,与MIC结果一致。
1.2.4C16-KWKW的细胞毒性较低
使用CCK-8法测评估C16-KWKW对HaCaT和RAW264.7两种细胞的毒性。结果如图2a所示,C16-KWKW在浓度为31.25μg/mL范围内时没有观察到显着的毒性,而对照药物Melittin在3.9μg/mL浓度没有显著毒性。
1.2.5C16-KWKW可显著抑制由LPS或热灭活P.acnes诱导的炎症因子的表达
通过测定C16-KWKW对小鼠巨噬细胞RAW264.7产生的促炎细胞因子表达的抑制作用来探究其抗炎活性。据报道,痤疮炎症可能由皮脂脂质或P.acnes引起。因此本发明研究中分别用脂多糖(LPS)和热灭活的P.acnes诱导细胞炎症因子的产生。基于细胞毒性测定结果,使用用31.25μg/mL浓度范围内的C16-KWKW不会对RAW264.7细胞产生抑制作用(图2b),因此将C16-KWKW浓度设定为1,10,25μg/mL。ELISA结果显示,C16-KWKW可显著抑制由LPS(图2c)和P.acnes诱导的(图2d)TNF-α,IL-8和IL-1β的产生。QPCR结果进一步证实,C16-KWKW可显著抑制IL-1β和TNF-α的基因表达。除此之外,C16-KWKW还可有效抑制参与炎症应答的其他关键酶,如诱导型一氧化氮合酶(iNOS)和前列腺素内源性过氧化物合酶-2(PTGS2),结果如图2e和2f所示。
此外,考虑到核转录因子κB(NF-κB)在炎症发展以及多种下游炎症因子的信号转导中起到重要作用,我们测试了在与诱导炎症因子相同条件下C16-KWKW对NF-κB表达的抑制作用。先用C16-KWKW预处理编码NF-κB和荧光素酶报告基因稳转质粒的RAW264.7细胞,2小时后,分别用LPS和热灭活的P.acne诱导细胞6小时。最后通过比较加药组与对照组萤光素酶的活性来评估C16-KWKW对NF-κB的抑制作用。结果显示,在浓度高于10μg/mL时观察到C16-KWKW对NF-κB表达的显著抑制(图2g)。
1.2.6C16-KWKW通过与P.acne细菌细胞膜的相互作用从而发挥抗菌效果
通常抗菌肽的作用靶点涉及与细胞膜之间相互作用,并诱导细胞内含物的渗漏。为了确定C16-KWKW的抗菌机制并验证其与细菌细胞膜的相互作用,我们使用等温滴定量热法(ITC)来比较C16-KWKW和几种细菌细胞膜脂质之间相互作用的热力学参数。分别从P.acnes,S.aureus和E.coli中提取脂质,真空干燥,并用5%DMSO调整至浓度为4mg/mL。几种细菌脂质与150μg/mL C16-KWKW之间相互作用引起的热变化如图3a所示,相同条件下C16-KWKW与P.acnes脂质的结合程度高于S.aureus和E.coli。分通过析软件计算出的结果见表5。

表5结果显示:C16-KWKW与P.acnes脂质之间的结合系数为3.484μM-1,而在相同测试条件下无法计算出C16-KWKW与其他两种细菌脂质的Ka值。此外,温谱图显示ITC峰值为负值,ΔH和TΔS值均为负,(表5),表明主要相互作用是放热反应。阳性对照Melittin在200μg/mL浓度与几种脂质之间相互作用的ITC等温线没有表现出显着差异(图3a和表5)。
为了进一步证实这一结果,我们使用色氨酸荧光光谱分析法进一步研究了细菌细胞膜和C16-KWKW之间的相互作用。如表6和图3b所示。

如表6和图3b所示:相同条件下观察到C16-KWKW与P.acnes脂质之间相互作用导致发射光谱产生23nm的蓝移,与来自S.aureus和E.coli脂质的相互作用之间存在显著差异。然而,Melittin与三种细菌脂质之间的相互作用中没有观察到光谱蓝移的差异。
1.2.7C16-KWKW诱导细菌细胞膜损伤,从而导致细菌内容物泄露
接下来我们需要探究C16-KWKW与细菌膜的相互作用是否导致细胞内容物释放。为此,我们使用透射电子显微镜(TEM)(图3a),测定核酸泄露(图3b)和PI摄取实验(图3c和3d)等方法来揭示C16-KWKW可能的作用机制。如图3b所示,在用10μg/mL C16-KWKW或Melittin处理P.acnes后,均显示出对细菌细胞膜的破坏作用并检测出明显的核酸泄漏,其中Melittin的作用远快于C16-KWKW。相反,在相同的处理条件下,克林霉素对细胞膜的完整性无明显影响。该结果通过PI摄取实验进一步确证,PI荧光强度通过流式细胞术分析或荧光酶标仪进行检测,结果如图3c和3d所示,对细菌细胞的处理显示出明显的量效关系,在C16-KWKW在10μg/mL浓度下PI的摄取量最高。
1.2.8C16-KWKW阻止P.acnes在体内的定植
测试C16-KWKW在体内抗P.acnes的作用。将P.acnes以对小鼠耳部皮内注射3.0×107CFU/40μL P.acnes,之后分别用C16-KWKW(100/200μg)和克林霉素(10μg)与0.05g凡士林混合涂抹,PBS处理组作为对照。结果如图4a所示,24小时后,PBS处理组耳部P.acnes菌落数为1.36×106CFU/mL,而C16-KWKW组100和200μg C16-KWKW处理后菌落数分别降至67%和45%。在对照组中,用10μg克林霉素治疗后剩余菌落数仅有23%。
1.2.9C16-KWKW有效降低体内诱发的炎症反应
为了研究C16-KWKW在体内的抗炎作用,我们采用P.acnes诱导的炎性动物模型来检测耳厚度及与炎症相关细胞因子的变化。结果如图4b所示,在注射P.acnes 24小时后观察到耳部有明显的红斑和肿胀,其耳厚度是没有注射P.acnes的空白对照组的1.5倍。仅用等量PBS注射的对照小鼠的耳朵中未观察到肿胀。以0.2%和0.4%浓度C16-KWKW处理后,耳厚度分别为对照组的124%和109%,克林霉素处理组耳厚度为对照组的103%(图4c)。
通过qRT-PCR进一步评估C16-KWKW的抗炎作用,检测经C16-KWKW处理后炎症动物模型的细胞炎症因子水平。结果如图4d所示,注射P.acnes 24小时后,PBS处理组(阴性对照)中TNF-α,IL-1β和iNOS的表达水明显增加。而在C16-KWKW和克林霉素处理组其表达水平均受到显著抑制。经0.2%C16-KWKW处理后,TNF-α,IL-1β和iNOS的表达水平分别为PBS处理组的32%,42%和16%;而在0.4%C16-KWKW处理组中达到27%,24%和8%。同时,克林霉素处理组的TNF-α,IL-1β和iNOS水平也有明显降低,其中TNF-α,IL-1β和iNOS表达水平分别为PBS处理组15%,33%和10%。
接下来采用组织学染色观察C16-KWKW在小鼠炎症模型中的抗炎作用。如图4e所示,注射P.acnes 24小时后,组织切片部位有明显肿胀,浸润的炎症细胞数量显着增加(图4e中的ii,其中i小图为对照)。而在C16-KWKW及克林霉素处理后的组织切片中,炎症肿胀程度及炎症细胞数目大大降低(组织学检测的图片见图4e中的iii-v小图)。总体来说,0.4%C16-KWKW的抗炎效果与克林霉素接近,0.2%C16-KWKW稍差一些。但与对照组相比,都能明显看出药物的治疗效果。
1.3实验讨论
痤疮丙酸杆菌是一种革兰氏阳性菌,能够产生许多致病因子,例如释放促炎细胞因子,因此在早诱导痤疮病变过程中起到关键作用。一些肽类化合物具有抗菌及免疫调节双重功能,与目前临床使用的抗生素作用位点不同,细菌难以形成耐药性,并且通常具有更快的杀菌速度。因此,我们选择抗菌肽作为对抗痤疮丙酸杆菌的有力武器。此外,痤疮丙酸杆菌的生长主要依靠毛囊皮脂腺部位皮脂分泌的脂肪酸,这提示我们,若将脂肪酸作为靶点,可能得到具有专一抗P.acnes的抗痤疮药物。因此,我们对设计的短脂肽库进行筛选,从中得到了具有强效抗P.acnes的化合物C16-KWKW。合成方法是将软脂酸结合到KWKW的N端。结构修饰研究表明,用C16-脂肪酸取代的KWKW是同系衍生物物中抗菌活性最高的短脂肽,并且具有比阳性对照药物克林霉素更快的杀菌速度。因此C16-KWKW被选做进一步的体内外研究。
MIC实验结果(表1)表明,C16-KWKW对P.acnes表现出比其他细菌菌株如S.mutans、E.coli等更高的抗菌活性,qRT-PCR实验进一步验证了该结果(图1a)。分别用C16-KWKW处理P.acnes、S.mutans、E.coli后,16S rRNA的相对表达含量差异清楚地表明C16-KWKW具有专一抗P.acnes的特点。
为了探究C16-KWKW可能的作用靶点,使用ITC和色氨酸荧光光谱法研究细菌脂质和C16-KWKW之间的相互作用。ITC是用于确定小分子和较大大分子(例如蛋白质和DNA)之间相互作用的热力学参数的物理技术。结果如图3a和表4所示,该反应为放热反应,在滴定开始时出现负的ITC峰,C16-KWKW与P.acnes脂质之间的结合常数为3.484μM-1,表明两者之间存在强烈的相互作用。此外,经过C16-KWKW作用后,在色氨酸荧光光谱分析中观察到的更明显的蓝移现象。该结果表明,与其他细菌脂质的作用相比,KWKW残基能够更深入地埋在P.acnes脂质膜中,进一步证实了ITC实验的结论。
我们通过TEM,核酸泄漏物检测和PI摄取实验(图3c-3d)进一步深入探究了C16-KWKW和P.acnes之间的相互作用,结构表明C16-KWKW能够破坏细胞膜的完整性并诱导核酸等内容物泄漏,从而导致细菌死亡。
考虑到炎症反应在痤疮发展中的重要性,我们进一步探究C16-KWKW是否能够通过对宿主免疫系统的调节从而在抗炎过程中发挥关键作用。痤疮的炎症反应受到多种炎症细胞因子的调节,包括由单核巨噬细胞分泌的TNF-α和IL-1β等。因此,炎症因子的表达水平代表炎症反应程度。我们通过ELISA和qRT-PCR的方法分别检测由LPS和热灭活P.acnes诱导的炎症因子表达水平(图2d),结果表明,C16-KWKW可显著抑制TNF-α,IL-8和IL-1β的分泌,表现出抗炎应用方面的巨大潜力。
在此基础上,需要深入了解C16-KWKW的抗炎作用位点,确定其抗炎作用机制。我们知道,NF-κB转录因子在炎症反应的调控中起关键作用。多种促炎细胞因子的表达受到NF-κB的调控作用,包括TNF-α,IL-8和IL-1β。因此我们将经C16-KWKW处理后NF-κB的表达情况与对照组进行比较,结果如图2g所示,C16-KWKW可有效抑制NF-κB的表达,提示其可能是通过对NF-κB的抑制从而发挥抗炎作用。深入的抗炎机制仍需要进一步研究,包括对参与调控NF-κB活性的几种关键蛋白表达的检测。
为了评估C16-KWKW的治疗效果,我们建立了P.acnes诱导的小鼠体内炎症模型,以确定C16-KWKW在体内的抗P.acnes及抗炎活性。首先进行了初步毒性实验,对正常及有划痕的小鼠皮肤予以10倍浓度C16-KWKW处理,未观察到任何毒副作用。体内动物实验表明,C16-KWKW在体内能有效抑制P.acnes的生长,减少耳肿胀,降低炎症部位致炎细胞因子的分泌水平,在体内展示出良好的抗菌和抗炎作用。
总的来说,我们的发现基于“食物样”的设计策略,研究结果表明,C16-KWKW在体内外展示出高效专一抗P.acnes及抗炎活性。现有用于治疗痤疮的抗菌肽一般由15个以上氨基酸残基构成,缺乏对P.acnes的专一性,且对哺乳动物细胞具有一定毒性。而C16-KWKW与之相比具有更快的杀菌速度,对哺乳动物细胞毒性相对较低。C16-KWKW仅由四个氨基酸残基组成,表明该短脂肽类化合物具有开发成为新一代抗P.acnes药物的潜力。
