Abstract
本发明公开了一种用于治疗咳嗽的中药组合物,是由以下重量比的组分制成:地骨皮:桑白皮:川贝母:鱼腥草:金荞麦:桔梗:橘红:当归:生白芍为15:15:9:30:30:9:9:15:15。本发明还公开了一种所述用于治疗咳嗽的中药组合物在制备用于治疗咳嗽的药物中的应用。本发明提供的用于治疗咳嗽的中药组合物是在几十年的临床经验的基础上总结出来的,其止咳祛痰药优点体现在:配方科学合理,既符合中医的理法、方、药,又有现代医学药理学为依据,适用范围广,对治疗肺热气喘、肺热咳嗽、肺热伴有阴虚咳嗽等的效果明显;无药物依赖性,配方简单,使用方便;疗效显著,见效快,安全可靠,无明显毒副作用。

技术领域
本发明属于医药技术领域,尤其涉及一种用于治疗咳嗽的中药组合物及其制备方法与应用。
背景技术
医者认为:咳嗽是人体清除呼吸道内的分泌物或异物的保护性呼吸反射动作。虽然有其有利的一面,但剧烈长期咳嗽可导致呼吸道出血。正确区分一般咳嗽和咳嗽变异性哮喘,防止误诊。治疗咳嗽应区分咳嗽类型,西药、中药皆可,但以食疗为最佳。
西医认为:咳嗽(Cough)是人体的一种保护性呼吸反射动作。通过咳嗽反射能有效清除呼吸道内的分泌物或进入气道的异物。但咳嗽也有不利的一面,剧烈咳嗽可导致呼吸道出血,如:长期频繁,剧烈咳嗽影响工作,休息,甚至引起喉痛,音哑和呼吸肌痛,则属病理现象。
中医认为:咳嗽是因外感六淫,脏腑内伤,影响于肺所致有声有痰之症。见《素问·五脏生成篇》。《素问病机气宜保命集》:“咳谓无痰而有声,肺气伤而不清也;嗽是无声而有痰,脾湿动而为痰也。咳嗽谓有痰而有声,盖因伤于肺气动于脾湿,咳而为嗽也”。因外邪犯肺,或脏腑内伤,累及于肺所致。
咳嗽是喉部或气管的粘膜受到刺激时迅速吸气,随即强烈的呼气,声带振动的现象,是人体清除呼吸道内的分泌物或异物的保护性呼吸反射动作,是呼吸系统疾病的主要症状,如急性咽喉炎、支气管炎、肺部感染、支气管内异物、肺结核等。目前,治疗咳嗽的药物有许多种,西医治疗咳嗽尽管能够治疗所叙述的病症,但是在实际使用过程中发现它们的效果并不是特别的理想;中医治疗咳嗽,不仅有很好的疗效,而且无副作用,是治疗咳嗽的最佳方案。
有鉴于此,有必要寻找一种可有效治疗咳嗽的中药组合物。
发明内容
本发明的目的是提供一种用于治疗咳嗽的中药组合物。
本发明另一个目的是提供一种所述用于治疗咳嗽的中药组合物的提取方法。
本发明的再一个目的是提供一种所述用于治疗咳嗽的中药组合物在制备用于治疗咳嗽的药物中的应用。
为了实现上述目的,本发明采用的技术方案如下:
本发明的第一个方面提供了一种用于治疗咳嗽的中药组合物,是由以下重量比的组分制成:
地骨皮:桑白皮:川贝母:鱼腥草:金荞麦:桔梗:橘红:当归:生白芍为15:15:9:30:30:9:9:15:15。
所述用于治疗咳嗽的中药组合物是由以下重量份的组分制成:地骨皮15份,桑白皮15份,川贝母9份,鱼腥草30份,金荞麦30份,桔梗9份,橘红9份,当归15份,生白芍15份。
本发明另一个方面提供了一种所述用于治疗咳嗽的中药组合物的提取方法,包括以下步骤:
按照所述配比将地骨皮、桑白皮、川贝母、鱼腥草、金荞麦、桔梗、橘红、当归、生白芍研磨成粗粉,取粗粉用水或乙醇水溶液浸泡回流提取1~3次,过滤合并,滤液减压浓缩至稀浸膏,真空干燥得粗提物,得率为7%~28%;
将粗提物加水混悬,上样到大孔吸附树脂进行柱层析,先吸附后再分别用水及乙醇充分洗脱至洗脱液不浑浊,弃去洗脱液,再用浓度为20%~70%的乙醇洗脱,收集洗脱液减压浓缩干燥,得到所述用于治疗咳嗽的中药组合物的有效药效物质。
所述乙醇水溶液的浓度为20~95%,用量为粗粉的至少10倍量。
所述大孔吸附树脂为D1300大孔吸附树脂、D101大孔吸附树脂、AB-8型大孔吸附树脂中的至少一种。
所述分别用水及乙醇充分洗脱至洗脱液不浑浊的洗脱体积均不少于4倍的大孔吸附树脂用量。
所述用于治疗咳嗽的中药组合物的有效药效物质采用超高压液相色谱-飞行时间质谱仪进行化合物鉴别,鉴别出的化合物为102~140个。
所述超高压液相色谱-飞行时间质谱仪的色谱分析条件为:色谱柱xselect HSST3,2.1×100mm,2.5μm;流动相A为0.1%甲酸-乙腈,B为1%甲酸-水溶液,梯度洗脱程序为:0-2min,5%A;2-13min,5%~95%A;13-15min,95%A;流速0.4ml/min;柱温25℃,检测波长260nm。
所述用于治疗咳嗽的中药组合物的有效药效物质可用于制备治疗咳嗽的临床合适剂型,如:片剂、颗粒剂、丸剂、散剂、普通胶囊剂等。
本发明的再一个方面提供了一种所述用于治疗咳嗽的中药组合物在制备用于治疗咳嗽的药物中的应用。
所述用于治疗咳嗽的中药组合物具有止咳平喘、润肺化痰的功效,以达到有效治疗咳嗽的目的,所治疗的疾病主要涉及肺热气喘、肺热咳嗽、肺热伴有阴虚咳嗽等。
本发明的用于治疗咳嗽的中药组合物中,地骨皮、桑白皮为君药,桔梗、橘红、川贝母为臣药,鱼腥草、金荞麦为佐药,当归、生白芍为使药。
本发明的组方中各项药物的性味、归经和功用及常用剂量如下:
地骨皮:味甘、性寒,具有清肺降火之功效。主治肺热蕴结引起的咳嗽、气喘、咳血等症,现代药理研究本品具有解热、抗炎、抑菌等作用。
桑白皮:味甘、性寒,具有泻肺理气平喘之功效。主治热邪壅肺引起的胸闷、气急、咳嗽、气喘、咳血、痰黄粘稠、咯痰不爽等症。现代药理研究本品具有镇静、解痉、止咳、祛痰、抑菌等作用。
桔梗:味苦辛、性平,具有宣肺、利咽、祛痰、排脓之功效。本品性善上行,善于开宣肺气而利胸膈研究,并具有祛痰的作用,主治痰阻气滞引起的胸膈痞闷、咳嗽、气喘、咳痰不爽等症。现代药理研究本品具有祛痰、镇咳、平喘、抗炎、抑菌、解热镇痛及调节免疫的作用。
橘红:味辛、苦,性温,具有理气宽中,燥湿化痰之功效。主要用于脾虚生湿,湿痰聚肺引起的胸闷脘痞、咳吐白色黏痰,气喘等症。现代药理研究本品具有抗炎、止咳、祛痰的作用,本品富含的发挥油对胃肠道有温和的刺激作用,有利于胃肠积气的排出,并能促进胃液的分泌。
川贝母:味甘苦、性微寒,具有清热润肺,化痰止咳之功效。主治肺热引起的咳嗽、咳痰,痰黄粘稠,以及肺阴亏虚引起的干咳少痰、咯痰不爽、痰中带血等症。现代药理研究本品具有抗炎、抑菌、解痉、止咳、祛痰等作用。
鱼腥草:味苦微辛、性凉,具有清热解毒,消痈排脓之功效。主治肺痈咳吐脓血,以及肺热引起的咳嗽、痰黄粘稠,被誉为“治痰热壅肺,肺痈吐脓血之要药”。现代药理研究本品具有抑菌、抗病毒作用,能通过促进白细胞及巨噬细胞的吞噬能力发挥抗炎作用,镇咳、平喘作用明显。
金荞麦:味苦微辛、性凉,具有清热解毒、排脓祛瘀之功效。主治肺热引起的咳嗽、咳痰黄稠,咽喉肿痛,还可用于治疗热毒蕴肺所致的肺痈,主要表现为咳吐脓血,气味腥臭。现代药理研究本品具有抗炎、解热、祛痰的作用,对金黄色葡萄球菌凝固酶、溶血素及绿脓杆菌内毒素均有对抗作用。
当归:味甘辛、性温,具有补血活血、调经止痛、润肠通便的功效。本品既能补血,又可活血,兼行气止痛,主治一切血症,为血病之要品。《神农本草经》记载本品有治疗“咳逆上气”之功效,可用于久咳、难治性咳嗽。现代药理研究本品对气管平滑肌有松弛作用,能抗炎、平喘、抑制肺纤维化,能对抗组胺-乙酰胆碱引起的支气管痉挛吗,能改善肺部血液循环。
生白芍:味苦酸、性微寒,具有养血调经、敛阴止汗、柔肝止痛、平抑肝阳之功效。主治阴血亏虚引起的头痛、眩晕、血虚萎黄、月经不调、盗汗等症。早在《伤寒杂病论》中就有记载运用生白芍治疗咳嗽的记载,现代药理研究本品通过直接作用或抑制副交感神经末梢释放的乙酰胆碱,发挥缓解支气管平滑肌痉挛的作用,临床常用语治疗变态反应引起的咽痒、干咳。
由于采用上述技术方案,本发明具有以下优点和有益效果:
本发明提供的用于治疗咳嗽的中药组合物,以地骨皮、桑白皮、川贝母、鱼腥草、金荞麦、桔梗、橘红、当归、生白芍9味药材按一定比例组成治疗咳嗽的药方,并通过不同比例的醇回流提取后再上大孔吸附树脂,以确定处方中各药材的提取和纯化工艺,获得药效混合物可制备成任一可药用的剂型,可将其制成胶囊、颗粒剂、片剂、丸剂、散剂。
本发明提供的用于治疗咳嗽的中药组合物具有止咳平喘、润肺化痰的功效,无不良反应、无明显毒副作用,安全有效。

实施例1
一种用于治疗咳嗽的中药组合物,是由以下重量份的组分制成:
称取地骨皮15g、桑白皮15g、川贝母9g、鱼腥草30g、金荞麦30g、桔梗9g、橘红9g、当归15g、生白芍15g备用。
一种用于治疗咳嗽的中药组合物的提取方法包括以下步骤:
制备粗提物:将上述药材磨成粗粉,按照上述比例取上述药材粗粉147g,用10倍量浓度为80%的乙醇回流提取2次,每次2h,过滤,合并滤液,温度为60℃的条件下减压浓缩至稀浸膏,真空干燥得粗提物35.50g,得率为24.15%。
纯化:取上述粗提物加水混悬后总体积为294ml,加入到装有735ml经预处理过(90%乙醇浸泡24小时)的D101大孔吸附树脂层析柱上,上样完毕后吸附2小时,用2940ml水洗脱(弃去),再用2940ml浓度为10%的乙醇洗脱(弃去),最后用2940ml浓度为60%的乙醇洗脱,浓度为60%的乙醇洗脱后的流份减压浓缩,真空干燥(60℃),得粉末6.62g,得率为4.5%。
采用超高压液相色谱-飞行时间质谱仪对该粉末进行化合物鉴别,鉴别出的化合物为140个。色谱分析条件为色谱柱xselect HSS T3(2.1×100mm,2.5μm);流动相为0.1%甲酸-乙腈(A),1%甲酸-水溶液(B),梯度洗脱程序为:0-2min,5%A;2-13min,5%~95%A;13-15min,95%A;流速0.4ml/min;柱温25℃,检测波长260nm。
实施例2
一种用于治疗咳嗽的中药组合物的提取方法包括以下步骤:
制备粗提物:按照实施例1中的比例取药材粗粉147g,用10倍量浓度为20%的乙醇回流提取2次,每次2h,过滤,合并滤液,温度为60℃的条件下减压浓缩至稀浸膏,真空干燥得粗提物26.88g,得率为18.29%。
纯化:取上述粗提物加水混悬后总体积为294ml,加入到装有735ml经预处理过的D101大孔吸附树脂层析柱上,上样完毕后吸附2小时。用2940ml水洗脱(弃去),再用2940ml浓度为10%的乙醇洗脱(弃去),最后用2940ml浓度为20%的乙醇洗脱,浓度为20%的乙醇洗脱后的流份减压浓缩,真空干燥(60℃),得粉末3.09g,得率为2.1%。
采用超高压液相色谱-飞行时间质谱仪对该粉末进行化合物鉴别,鉴别出的化合物为112个。
实施例3
一种用于治疗咳嗽的中药组合物的提取方法包括以下步骤:
制备粗提物:按照实施例1中的比例取药材粗粉147g,用10倍量浓度为50%的乙醇回流提取2次,每次2h,过滤,合并滤液,温度为60℃的条件下减压浓缩至稀浸膏,真空干燥得粗提物29.10g,得率为19.80%。
纯化:取上述粗提物加水混悬后总体积为294ml,加入到装有735ml经预处理过的D1300大孔吸附树脂层析柱上,上样完毕后吸附2小时。用2940ml水洗脱(弃去),再用2940ml浓度为10%的乙醇洗脱(弃去),最后用2940ml浓度为60%的乙醇洗脱,洗脱液减压浓缩,真空干燥(60℃),得粉末4.70g,得率为3.2%。
采用超高压液相色谱-飞行时间质谱仪对该粉末进行化合物鉴别,鉴别出的化合物为122个。
实施例4
一种用于治疗咳嗽的中药组合物的提取方法包括以下步骤:
制备粗提物:按照实施例1中的比例取药材粗粉147g,用10倍量浓度为95%的乙醇回流提取2次,每次2h,过滤,合并滤液,温度为60℃的条件下减压浓缩至稀浸膏,真空干燥得粗提物21.32g,得率为14.5%。
纯化:取上述粗提物加水混悬后总体积为294ml,加入到装有735ml经预处理过的D1300大孔吸附树脂层析柱上,上样完毕后吸附2小时。用2940ml水洗脱(弃去),再用2940ml浓度为10%的乙醇洗脱(弃去),最后用2940ml浓度为50%的乙醇洗脱,洗脱液减压浓缩,真空干燥(60℃),得粉末3.68g,得率为2.5%。
采用超高压液相色谱-飞行时间质谱仪对该粉末进行化合物鉴别,鉴别出的化合物为102个。
实施例5
本发明的临床观察
一、临床疗效
1.临床基本资料:选择病程1周内,临床辨证诊断为肺热咳喘的患者245例,男性125例,女性120例,主要症状为反复咳嗽,咯黄黏痰,气喘,同时伴有发热(或无)、出汗(或无)、口干、咽干(或无)、咽痛(或无)、舌质红、苔薄黄、脉滑数;听诊肺部呼吸音增粗或可闻及中细湿啰音。
2.治疗方法:按照体重、年龄、病情酌情给予本发明中药组合物,各中药按重量地骨皮10-30份、桑白皮10-30份、川贝母3-9份、鱼腥草10-30份、金荞麦10-30份、桔梗3-9份、橘红3-9份、当归10-30份、生白芍10-30份加减,并告知煎煮及服用方法,即每剂以水煎煮两次,合并煎液,分早晚两次服用。
3.治疗效果评价标准:
治愈:咳嗽、咯痰及肺部体征消失,或偶有咳嗽,体温升高者恢复正常,其他临床症状消失;显著有效:咳嗽、咯痰及肺部体征明显好转,体温升高者恢复正常,其他临床症状明显缓解;好转有效:咳嗽、咯痰及肺部体征有所好转,体温升高者体温下降或降至正常,其他临床症状部分缓解;无效:咳嗽、咯痰及肺部体征无改善,体温升高者为下降,其他临床症状无改善。
4.治疗效果:临床治疗效果见表1。

二、典型病例
吴某,男,25岁,2015年10月12日初诊。患者3天前因洗冷水澡后调摄不慎,出现发热(37.8℃),伴恶寒,鼻塞流清涕,咽喉痛,自行服用感冒胶囊2天后无明显效果,体温升高,伴胸闷咳嗽而求治于中医。诊见:咳嗽,胸闷气喘,咯黄黏痰,咯痰不畅,发热(38.8℃),不恶寒,口干,咽喉疼痛,大便偏干,舌质红,脉滑数,听诊右肺部可闻及湿啰音,诊断为肺热咳嗽之证。药用:以重量份数地骨皮30、桑白皮30、川贝母9、鱼腥草30、金荞麦30、桔梗9、橘红9、当归15、生白芍15,服用七剂后热退喘平,咳止痰无,其他病症皆消失,听诊呼吸音恢复正常。

2.根据权利要求1所述的用于治疗咳嗽的中药组合物,其特征在于:所述用于治疗咳嗽的中药组合物是由以下重量份的组分制成:地骨皮15份,桑白皮15份,川贝母9份,鱼腥草30份,金荞麦30份,桔梗9份,橘红9份,当归15份,生白芍15份。
3.一种权利要求1或2所述的用于治疗咳嗽的中药组合物的提取方法,其特征在于:包括以下步骤:按照所述配比将地骨皮、桑白皮、川贝母、鱼腥草、金荞麦、桔梗、橘红、当归、生白芍研磨成粗粉,取粗粉用水或乙醇水溶液浸泡回流提取1~3次,过滤合并,滤液减压浓缩至稀浸膏,真空干燥得粗提物,得率为7%~28%;
将粗提物加水混悬,上样到大孔吸附树脂进行柱层析,先吸附后再分别用水及乙醇充分洗脱至洗脱液不浑浊,弃去洗脱液,再用浓度为20%~70%的乙醇洗脱,收集洗脱液减压浓缩干燥,得到所述用于治疗咳嗽的中药组合物的有效药效物质。
4.根据权利要求3所述的用于治疗咳嗽的中药组合物的提取方法,其特征在于:所述乙醇水溶液的浓度为20~95%,用量为粗粉的至少10倍量。
5.根据权利要求3所述的用于治疗咳嗽的中药组合物的提取方法,其特征在于:所述大孔吸附树脂为D1300大孔吸附树脂、D101大孔吸附树脂、AB-8型大孔吸附树脂中的至少一种。
6.根据权利要求3所述的用于治疗咳嗽的中药组合物的提取方法,其特征在于:所述分别用水及乙醇充分洗脱至洗脱液不浑浊的洗脱体积均不少于4倍的大孔吸附树脂用量。

8.根据权利要求3所述的用于治疗咳嗽的中药组合物的提取方法,其特征在于:所述超高压液相色谱-飞行时间质谱仪的色谱分析条件为:色谱柱xselect HSS T3,2.1×100mm,2.5μm;流动相A为0.1%甲酸-乙腈,B为1%甲酸-水溶液,梯度洗脱程序为:0-2min,5%A;2-13min,5%~95%A;13-15min,95%A;流速0.4ml/min;柱温25℃,检测波长260nm。
9.根据权利要求3所述的用于治疗咳嗽的中药组合物的提取方法,其特征在于:所述用于治疗咳嗽的中药组合物可制成片剂、颗粒剂、丸剂、散剂、普通胶囊剂。
10.一种权利要求1或2所述的用于治疗咳嗽的中药组合物在制备用于治疗咳嗽的药物中的应用。
