技术领域
本发明提供了作为水包油型乳剂的包含海洋生物油(marine oil)和果蔬汁(juice)的组合物,其用于改善肌肉性能。
背景技术
在过去几年中,已经对营养组合物在每日补充或运动期间的作用进行了广泛研究,以增强耐力和运动后的运动表现。还旨在缩短肌肉恢复时间。
骨骼肌的底物可用性对于优化耐力表现至关重要。已显示,在耐力运动期间,碳水化合物(carbohydrate,CHO)氧化的提高有助于减轻疲劳和提高性能(Jeukendrup,2010)。有趣的是,在过去的二十年中,已经提出ω-3多不饱和脂肪酸(n-3PUFA)(特别是活性成分二十碳五烯酸(eicosapentaenoic acid,EPA)和二十二碳六烯酸(docosahexaenoic acid,DHA))通过其在骨骼肌膜上的不饱和作用来提高肌肉细胞利用葡萄糖的能力,从而有助于提高CHO氧化(Andersson等,2000,Vessby等,2002)。此外,补充n-3PUFA还已与氧(O2)消耗的降低相关,并且在动物模型(Pepe和McLennan,2002,2007)和人(Peoples等,2008)模型二者中均观察到。这种降低的氧耗以及因此提高的氧效率可归因于运动期间骨骼肌主要利用的燃料源的变化。
ω-3多不饱和脂肪酸(n-3PUFA)可介导多种生物过程。鉴于它们在改变骨骼肌膜完整性中的作用,n-3PUFA补充可提高葡萄糖可用性,从而影响休息时和/或运动期间的燃料代谢。

因此,期望改善肌肉性能的组合物。
发明概述
本发明提供了作为水包油型乳剂的包含海洋生物油和果蔬汁之组合的组合物,其用于改善肌肉性能,其中所述海洋生物油选自totox值低于20且基于鱼油的总重量ω-3含量高于按重量计10%的海洋生物油,并且其中使用合适的乳化剂来稳定所述乳剂。
本发明的组合物有益于改善的肌肉性能,其包括以下情况:延长的运动期间耐力;减轻的“迟发性肌肉酸痛(delayed onset of muscle soreness,DOMS)”;减轻的急性肌肉酸痛;缩短的肌肉恢复时间;降低的运动引起之肌肉损伤;以及提高的运动能力。

发明详述
本发明的一个目的是提供用于改善肌肉性能的本发明组合物。
本发明的另一个目的是提供将延长运动期间耐力的组合物。
本发明的另一个目的是提供将减轻“迟发性肌肉酸痛(DOMS)”的组合物。
本发明的另一个目的是提供将减轻急性肌肉酸痛的组合物。
本发明的另一个目的是提供将缩短肌肉恢复时间的组合物。
本发明的另一个目的是提供将降低运动引起之肌肉损伤的组合物。
本发明的另一个目的是提供将提高运动能力的组合物。
本发明的另一个目的是提供将提高碳水化合物(CHO)氧化以有助于减轻疲劳和提高性能的组合物。
现在出人意料地发现,本发明的组合物可用作营养补充剂,其显示出改善的肌肉性能。
已表明本发明中使用的组合物在化学和物理这两方面具有高稳定性。该组合物使得低氧化成为可能并使脆弱营养物保持完整和有效,这进而导致提高的吸收和高生物利用度。
该组合物包含:
a)海洋生物油,其具有低totox值,尤其是ω-3脂肪酸浓度足以实现营养作用,以及
b)含有天然存在的抗氧化剂的果蔬汁;
c)乳化剂。
WO 2007/064222(其是发明人自己的专利)中描述了该组合物的不同方面,但本发明中可使用该组合物的进一步改进。
组合物中的海洋生物油通常导致不需要的鱼的味道和余味。用于本发明的组合物具有例如为美味饮料的优点,这可以是经历味道改变和食欲不振的患者中的重要先决条件。
因此,本发明的一个方面涉及作为水包油型乳剂的包含海洋生物油和果蔬汁之组合的组合物,其用于改善肌肉性能,其中所述海洋生物油选自totox值低于20且基于所述海洋生物油的总重量ω-3含量高于按重量计10%的海洋生物油,并且其中使用合适的乳化剂来稳定所述乳剂。

改善的肌肉性能的所有方面都包括在本发明的范围内,特别是选自包含以下的组的情况:延长的运动期间耐力;减轻的“迟发性肌肉酸痛(DOMS)”;减轻的急性肌肉酸痛;缩短的肌肉恢复时间;降低的运动引起之肌肉损伤;以及提高的运动能力。
本发明的组合物通过促进肌肉从损伤性运动中恢复来提供改善的肌肉性能。本组合物的另一个有益作用是降低损伤和劳损的发生率,因为肌肉得到更好的保护,导致不太严重的肌肉损伤和酸痛。此外,本发明的组合物通过提高碳水化合物(CHO)氧化以有助于减轻疲劳和提高性能来提供改善的肌肉性能。此外,本发明的组合物通过爆发性地促进肌肉来提供改善的肌肉性能。
在这种情况下,组合物中使用的海洋生物油是从新鲜海洋生物物种制备的任何海洋生物油,其中所有处理步骤都是根据功能油标准仔细地并在严格的氧控制的情况下进行的,以防止油氧化。海洋生物油将具有低的氧化状态,显示为没有例如鱼的特征性气味或味道的无色油。作为totox值(2乘以过氧化值(peroxide value,PV)加上茴香胺值(anisidinevalue,AV))给出的氧化水平应低于20,优选低于15,更优选低于10,且最优选低于5。如今,许多食品补充剂中存在的海洋生物油包含具有高得多(通常为20至30或甚至更高)的totox值的油。
因此,在本发明的一个优选实施方案中,组合物的海洋生物油的totox值低于20,优选低于15,更优选低于10,且最优选低于5。
海洋生物油富含ω-3脂肪酸。但是,含量是不同的。在本发明的一个优选实施方案中,基于海洋生物油的重量,海洋生物油中ω-3脂肪酸的含量应为至少8%,优选至少12%,更优选至少16%,最优选约20%至35%。
根据本发明,海洋生物油可以是富含ω-3脂肪酸的任何油,例如鱼油、海豹油或磷虾油。海洋生物油可与植物来源的其他多不饱和油(例如藻油和草本油(例如月见草油和菜籽油))混合。
在本发明的一个优选实施方案中,基于海洋生物油制剂的总重量,该组合的组合物包含的海洋生物油的量为按重量计约0.5%至约15%,优选约0.5%至约10%,更优选约0.5%至约7%,最优选约1.5%至约6%,甚至更优选约5%至6%。
组合物包含作为水包油型乳剂的海洋生物油,其中水相包含含有天然抗氧化剂的果蔬汁。果蔬汁可选自水果汁、浆果汁或蔬菜汁,或其任意组合。优选地,果蔬汁获自具有合适的高水平抗氧化剂的水果、浆果或蔬菜。此外,优选的是,果蔬汁具有最低水平的发挥氧化剂作用的金属离子。优选的果蔬汁可选自以下的组:石榴、杏、葡萄柚、橙(orange)、蔓越莓(cranberry)、玫瑰果(rosehip)、菠萝、黑果腺肋花楸(black chokeberry)、野樱梅(aronia)、桑椹、云莓(cloudberry)、针叶樱桃(acerola)、树莓(raspberry)、西瓜、桃、葡萄、樱桃、胡萝卜、苹果、梨、柠檬、甜菜根(beetroot)、西番莲(passion fruit)、芒果、甜菜根(beet root)、胡萝卜和猕猴桃(kiwi)。
果蔬汁可以是鲜榨果蔬汁(fresh pressed juice)或为果蔬汁浓缩物形式的果蔬汁。在配制之前将果蔬汁浓缩物稀释以获得正常的即用型果蔬汁。优选的果蔬汁或果蔬汁浓缩物是选自苹果、梨、石榴、花楸果(chokeberry)/野樱梅、西番莲及其组合的那些。
基于组合物的总重量,果蔬汁的含量为按重量计约20%至97%。当使用果蔬汁浓缩物时,20%至97%是指稀释以获得正常的即用型果蔬汁的果蔬汁量。在一个优选实施方案中,果蔬汁的含量为30%至97%,优选50%至95%,更优选75%至95%,且最优选80%至95%。如果适用,水相进一步用水平衡。
在组合物的制备中,可使用任何合适的乳化剂或乳化剂的组合。乳固体、乳清蛋白质和果胶、豌豆蛋白质、鱼蛋白质、大麻蛋白质、大豆蛋白质或其任意组合是优选的乳化剂。乳清蛋白质是优选的,特别是以乳清蛋白质粉或乳清蛋白质分离物的形式。
组合物还可包含作为营养来源添加的另外的蛋白质和氨基酸。这样的实例是瓜氨酸。
还涵盖添加肌酸。
组合物还可包含甜味剂、矫味剂和防腐剂。这样可给予组合物任何期望的味道。
合适的防腐剂是山梨酸钾。然而,可使用任何合适的防腐剂。合适的甜味剂是木糖醇。然而,可使用任何合适的甜味剂。
另一些合适的矫味剂是波萝蜜(jackfruit)和迷迭香(rosemary)提取物。另外,可添加pH调节剂,例如抗坏血酸。
组合物还可包含一种或更多种另外添加的抗氧化剂,包括但不限于生育酚、姜黄素、白藜芦醇、多酚、类胡萝卜素、辅酶Q10、番茄红素和叶黄素。
组合物还可包含一种或更多种维生素,例如维生素B、C和/或D。此外,组合物还可包含一种或更多种矿物质,例如镁、钾和/或钠。
组合物还可包含酸乳粉(yoghurt powder)。通过添加酸乳粉使制剂变稠,产生诱人的稠度。酸乳粉的量可以是组合物的按重量计5%至10%。在另一个实施方案中,组合物中可添加益生元(prebiotics)和/或益生菌(probiotics)。在另一个实施方案中,组合物可以是碳酸化的(carbonated)。
组合物的稠度可从可饮用的液体稠度至更粘稠的饮料(例如思慕雪(smoothie))变化。此外,组合物的稠度可以是像酸乳那样的奶油状。
优选地,组合物是饮料制剂,但还考虑组合物可以以其他公知的施用形式(例如片剂或胶囊剂或凝胶剂)存在。例如,可制备饮料制剂并随后干燥(例如冻干)成粉末或颗粒化,并随后以片剂或胶囊剂剂型存在。或者,可将饮料制剂浓缩以形成供患者使用的浓缩物或使用标准胶凝技术胶凝以提供可食用的凝胶。
优选地,组合物以每天为基础施用,且最优选地,其为以每天为基础施用的饮料制剂。如本文中所用的每天为基础意指每天施用至少一次,但也包括一天内多次施用,例如每天两次或三次。
优选地,组合物以剂量单位填充,优选气密瓶或小密封容器,但是可应用任何合适的容器。含有调整至成人、儿童和婴儿的推荐日剂量和每天施用次数的不同量脂肪酸的容器是可能的。
组合物中DHA和EPA各自的施用范围可以是每天约200mg至约5000mg,优选每天约3000mg,更优选每天约2000mg且最优选每天约1100mg或1000mg。在根据本发明的制剂作为饮料制剂提供的情况下,合适的单位体积为50至200ml。在本发明的组合物作为酸乳提供的情况下,合适的单位体积为25至100ml。
WO 2007/064222中描述了组合物的不同方面,但本发明中可使用组合物的进一步改进。
实施例
实施例1:
在每天补充本发明的组合物之后改善的运动恢复。
介绍:
足球运动员经常经历离心型运动(eccentric exercise)引起的肌肉损伤。已显示蛋白质补给(protein feeding)改善从离心型运动的肌肉功能恢复。由于来自鱼油的n-3多不饱和脂肪酸(n-3 PUFA)显示出抗炎特性,因此将鱼油与蛋白质组合可促进恢复。
目的:
该研究的目的是评估用含有n-3PUFA、乳清蛋白质和维生素D之组合的根据本发明的基于果蔬汁的含抗氧化剂的饮料进行6周补充对于运动恢复期间肌肉功能和酸痛、炎症和足球表现的影响。
方法:
将足球运动员(n=30)分配至三个条件之一中。实验条件确定的FO含有n-3PUFA(550mg DHA,550mg EPA)、乳清蛋白质(15g)、碳水化合物(14g)和维生素D(3μg),并且是可获自Smartfish AS标识了Recharge High Protein的根据本发明的饮料。蛋白质对照条件(PRO)含有乳清蛋白质(15g)、碳水化合物(14g)和维生素D(3.9μg)。适当热量(eucaloric)对照条件(CHO)含有碳水化合物。补充剂的营养组成列于下表1中。

研究设计示于图1中。
进行本领域中公知的足球专项测试,例如CMJ(反向跳(Counter MovementJump))、LSPT(拉夫堡足球传球测试(Loughborough Soccer Passing Test))和Yoyo测试。离心型运动由在两条腿上分别进行12组单侧膝关节伸展/屈曲收缩组成。统计分析包括常规p值和基于量级的推断方法。
结果:
如图2中所示,补充后,相比于PRO(25±1,p<0.05)或CHO(23±1,p<0.05),FO(36±2)中的血液%n-3PUFA/总PUFA更大。
如图3中所示,相比于PRO或CHO,FO中从EIMD(运动引起的肌肉损伤)的72小时运动恢复期间的肌肉酸痛(非优势腿)显著更低。表示为AUC的肌肉酸痛为FO中(1947±364mm×72小时),相比于PRO(4640±885mm×72小时,p<0.05)和CHO(4495±618mm×72小时,p<0.0.5)。

相比于PRO和CHO,FO对反向跳(CMJ)表现产生可能的(82%至85%)有益作用。
如图4中所示,72小时运动恢复期间的yoyo测试表现表明,在24小时和72小时时FO相对于PRO和CHO的改善的表现。
如图5中所示,相比于CHO,FO中的血液肌酸激酶浓度显著更低(p<0.05),并且相比于PRO,FO中趋向于更低(p=0.07)。相比于CHO(AUC:83±42mg/L×72小时,p<0.05),FO(AUC:42±11mg/L×72小时)中表示为AUC的血液肌酸激酶浓度更低,并且相比于PRO,FO中趋向于更低(p=0.07)。
如图6中所示,相比于CHO,FO中C-反应蛋白的血液浓度更低,并且相比于PRO,FO中也趋向于更低。
发现:
出人意料地发现,与不含鱼油的蛋白质对照和等热量的富含碳水化合物的对照相比,用根据本发明的基于果蔬汁的鱼油饮料进行6周补充在运动恢复期间改善了肌肉功能和疼痛、降低了炎症标志物并提高了足球表现。
实施例2:
每天补充本发明的组合物后,碳水化合物(CHO)氧化提高。
介绍:
ω-3多不饱和脂肪酸(n-3PUFA)可介导多种生物过程,包括改变骨骼肌膜组成。考虑到骨骼肌的代谢作用,n-3PUFA补充可影响休息时和/或运动期间的燃料代谢。
目的
该研究的目的是评估用含有n-3PUFA、乳清蛋白质和维生素D的组合的根据本发明的基于果蔬汁的饮料进行4周补充对于耐力训练个体在休息时和稳态运动期间二者的影响。
方法
二十一位男性自行车运动员和铁人三项运动员接受了间隔四周的两次实验性试验。在间隔期间,参与者每天两次(2×200ml)补充含有鱼油的根据本发明的基于果蔬汁的饮料(FO)或不含鱼油的味道匹配的对照饮料(CON)。一个单位(200ml)FO含有1g EPA和1gDHA,并且是可获自SmartfishAS标识了Recharge Lipid+的根据本发明的饮料。
在实验性试验期间,在休息时和在85%的个体乳酸阈(lactate threshold)下骑行60分钟期间收集呼出的气体。
结果
下表3和4示出了在4周补充之前和之后,FO和CON组二者中参与者的特征。

发现:
出人意料地发现,用根据本发明的基于果蔬汁的饮料进行4周补充可增强耐力训练自行车运动员在稳态运动期间燃料利用向碳水化合物氧化的转变。
实施例3:
包含瓜氨酸的根据本发明的饮料

*每200ml的单位体积含有500IU的维生素D3
所有制剂中使用的海洋生物油都是来自Orkla的Denomga arctic oil。其totox值低于8,并且根据功能油标准进行制备。
实施例3和4的饮料如下制备:
1.油相
将油与Toco 50(有利于油稳定的抗氧化剂制剂)混合。将任何其他油溶性成分与油混合。重要的是处理期间保护油不被氧化。
2.水相
在罐中填充入纯化去离子水。添加任何水溶性成分。然后将油相引入到水相中,之后添加乳化剂以获得乳剂。
或者,可对所得乳剂进行快速巴氏消毒(pasteurization)(约90℃持续8秒),随后均质化并冷却至温度为4至8℃。
最后,将饮料填充入气密的无菌容器(优选单剂量容器,例如约200ml的TetraBrick)中,并在使用前储存于6至8℃。
在所有步骤中必须进行严格的氧气控制以避免海洋生物油的氧化。
Claims (12)
1.作为水包油型乳剂的包含海洋生物油和果蔬汁之组合的组合物,其用于改善肌肉性能,
其中所述海洋生物油选自totox值低于20且基于所述海洋生物油的总重量ω-3含量高于按重量计8%的海洋生物油,并且其中使用合适的乳化剂来稳定所述乳剂。
2.权利要求1所述的组合物,其中所述改善的肌肉性能选自包含以下的组:
延长的运动期间耐力;
减轻的“迟发性肌肉酸痛(DOMS)”;
减轻的急性肌肉酸痛;
缩短的肌肉恢复时间;
降低的运动引起之肌肉损伤;以及
提高的运动能力。
3.根据权利要求1所述的组合物,其中所述海洋生物油的totox值低于10。
4.根据权利要求1所述的组合物,其中所述海洋生物油是鱼油。
5.根据权利要求1中任一项所述的组合物,其中基于所述组合物的总重量,所述海洋生物油含量为按重量计约0.5%至15%。
6.根据权利要求1中任一项所述的组合物,其中基于所述组合物的总重量,所述果蔬汁的含量为按重量计约20%至97%。
7.根据权利要求6所述的组合物,其中所述果蔬汁选自具有合适的高水平抗氧化剂的水果和浆果。
8.根据权利要求6或7所述的组合物,其中所述果蔬汁选自以下的组:石榴、杏、葡萄柚、橙、蔓越莓、玫瑰果、菠萝、黑果腺肋花楸、桑椹、云莓、针叶樱桃、树莓、西瓜、桃、葡萄、樱桃、蒲桃、苹果、芒果、梨、野樱梅、西番莲、甜菜根、胡萝卜和猕猴桃。
9.根据权利要求1所述的组合物,其中所述乳化剂选自以下的组:乳固体、乳清蛋白质、燕麦蛋白质和豌豆蛋白质。
10.根据权利要求1所述的组合物,其中所述组合物还包含果胶。
11.根据权利要求1所述的组合物,其中所述组合物还包含甜味剂、矫味剂、抗氧化剂和防腐剂。
