In conclusion, the preparation method of the alcohol-soluble beta-glucan extract provided by the embodiment of the invention has the advantages of simple process, convenience in operation, short production period and high efficiency, and is suitable for industrial mass production; the prepared alcohol-soluble beta-glucan extract has good alcohol solubility, can be dissolved in ethanol with different concentrations, and is suitable for producing highland barley beta-glucan products; the alcohol-soluble beta-glucan extract provided by the embodiment of the invention is used for preparing health care wine; the health care wine provided by the embodiment of the invention contains a large amount of highland barley beta-glucan.

Claims (5)

1. A method for preparing an alcohol-soluble beta-glucan extract, characterized by comprising the steps of:
taking highland barley, adding an acid solution, and performing acid extraction at 40-60 ℃ to obtain an acid extracting solution;
adding alpha-amylase accounting for 0.05-0.2% of the mass of the acid extracting solution and saccharifying enzyme accounting for 0.2-0.6% of the mass of the acid extracting solution into the acid extracting solution, carrying out enzymolysis at 40-60 ℃, and carrying out enzyme deactivation treatment after enzymolysis is finished to obtain an enzymolysis extracting solution;
centrifuging and concentrating the enzymolysis extracting solution to obtain a concentrated solution;
adding an ethanol solution into the concentrated solution, and carrying out ethanol precipitation to obtain an ethanol precipitation solution;
filtering and drying the alcohol precipitation solution;
the method for adding the acid solution comprises the following steps: adding water in an amount which is 15-25 times the weight of the highland barley medicinal material into the highland barley medicinal material, and adding citric acid to adjust the pH value to 4-6;
the acid extraction method comprises the following steps: extracting for 2-3 times at 40-60 ℃ for 1-2 hours each time;
the enzymolysis time is 1-2 hours;
the enzyme deactivation method comprises the following steps: heating to 80-90 ℃ and preserving the heat for 40-60 minutes;
the method for alcohol precipitation comprises the following steps: adding 90-95% ethanol by mass concentration into the concentrated solution, and controlling the ethanol concentration in the ethanol precipitation solution to be 50-70%.
2. The method for preparing an alcohol-soluble β -glucan extract as claimed in claim 1, wherein the concentration is performed by concentrating the supernatant obtained by centrifugation to 1-3 times the mass of the highland barley herb.
3. An alcohol-soluble β -glucan extract, which is produced by the method for producing an alcohol-soluble β -glucan extract according to any one of claims 1 to 2.
4. Use of the alcohol-soluble β -glucan extract according to claim 3 for the preparation of a health wine.
5. A health wine is characterized by comprising white spirit with an alcoholic strength of 40-60% vol and the alcohol-soluble beta-glucan extract as claimed in claim 3, wherein the addition amount of the alcohol-soluble beta-glucan extract is 1-8% of the mass of the white spirit.
