Abstract

本发明涉及一种皮下注射用干细胞微球凝胶复合物及其制备方法和应用。具体步骤为:1、分离脐带间充质干细胞.2.制备干细胞‑藻酸盐微球。3.制备水凝胶。4.将上述干细胞‑藻酸盐微球与水凝胶混合,所述干细胞在水凝胶中的分散浓度按105‑106个/mL。5.将干细胞微球凝胶复合物皮下注射至组织内创面修复组织损伤。本发明将透明质酸等高分子材料与干细胞通过微球包埋结合起来,并将该微球分散至凝胶中,形成具有固定干细胞定向释放细胞因子功能的生物支架材料,可以显著提高干细胞的靶向修复和治疗作用,凝胶形成物理屏障规避干细胞成瘤风险,且微球结构使得干细胞所释放的细胞因子具有缓释可控特点,符合安全给药和精准给药的发展趋势,具有广泛的临床应用价值。

Description

一种皮下注射用干细胞微球凝胶复合物及其应用
技术领域
本发明涉及干细胞和生物材料领域,更特别地涉及到一种皮下注射用干细胞微球凝胶复合物及应用。
背景技术
干细胞是一种具有自我复制和分化潜能的细胞,多呈圆形或椭圆形,体积小,胞浆少,核/质比大。干细胞增殖速度较慢,自稳性好,利于自我复制和维持。由于增殖速度缓慢,使干细胞有充足的时间发现和纠正复制错误,避免自我突变,以便稳步进入特定的分化程序。一旦机体需要时,干细胞就可进入分化状态,此时增殖速度开始逐渐加快,以适应组织器官生长,发育和修复的需要。
由于干细胞具有自我复制和分化的潜能,因此对受损组织器官具有较好的修复效果。近年来,已有研究人员将干细胞制成液体制剂,将其应用于创口等组织修复的临床研究上,但采用的给药方式为静脉回注,这种方式会导致制剂中的干细胞随血液流动而流失,使得制剂中的干细胞只有极少量能够归巢到目的组织或器官,从而大大降低了干细胞对创口的修复效果。因此有研究人员改变了给药途径,直接给药于关节腔等受损组织内部,也取得了一定的效果,但同时出现了因干细胞失活带来疼痛或因分泌细胞因子浓度的不可控性而引起组织损伤等一系列的不安全的临床表现,同时存在着干细胞成瘤风险等一系列临床安全隐患。针对上述问题,研究人员将干细胞与生物材料结合成液体凝胶,大大提高了干细胞的成活率,且因凝胶层的存在而规避了成瘤风险。但仍存在一定的缺点,具体表现为分泌细胞因子浓度的不可控性和凝胶材料降解的不稳定性。因此开发一种既能够控制分泌细胞因子缓慢释放又能稳定降解的生物材料成为干细胞在修复受损组织应用上的待解决问题之一。
发明内容
为解决以上问题,本发明提供了一种皮下注射用干细胞微球凝胶复合物的制备方法,其利用所述凝胶、分散于凝胶中的可降解微球及包埋于该微球内的干细胞可稳定分泌细胞因子修复组织内部损伤。
可选地,所述的一种皮下注射用干细胞微球凝胶复合物及其制备方法和应用,其干细胞微球为采用静电注滴法制备的干细胞-藻酸盐微球。
可选地,所述的采用静电注滴法制备的干细胞-藻酸盐微球,所述微球的粒径为1~500μm,优选为50~200μm。
可选地,所述的一种皮下注射用干细胞微球凝胶复合物,其凝胶为透明质酸水凝胶。其具有良好的生物相容性和稳定的可降解性。
可选地,所述的一种皮下注射用干细胞微球凝胶复合物,其干细胞来源为间充质干细胞。
可选地,所述的一种皮下注射用干细胞微球凝胶复合物,其干细胞分散浓度为105-106个/mL。
可选地,所述的间充质干细胞为骨髓间充质干细胞,脐带间充质干细胞,脂肪间充质干细胞。
可选地,所述的干细胞微球凝胶复合物应用在关节炎、糖尿病足中。
本专利涉及到的一种皮下注射用干细胞微球凝胶复合物,能够最大程度地保持干细胞活性,稳定分泌细胞因子,微球结构的缓释作用能够使得细胞因子缓慢释放避免因细胞因子浓度过大引起过激反应而导致组织二次损伤,本专利涉及到的一种皮下注射用干细胞微球凝胶复合物有很好的生物相容性和稳定可降解性。
具体实施方式
以下结合实例对本发明的原理和特征进行描述,所举实例只用于解释本发明,并非用于限定本发明的范围。
干细胞-藻酸盐微球制备:
利用静电注滴法制备干细胞-藻酸盐微球,以2.5%(w/v)的CaCl2凝胶浴溶液为接收器。针头与凝胶浴液面为用微量注射器泵以5mL•h-1的速率推注出1.7%海藻酸钠(w/v)、105-106个/mL脐带间充质干细胞(取P2-P5代冻存脐带间充质干细胞复苏待用)的原料液。在电压为10.5kV的电场力的作用下原料液与CaCl2凝胶浴溶液交联固化形成不溶于水的海藻酸钙微球,微球的粒径为50~200μm。
水凝胶制备
水凝胶是透明质酸水凝胶。透明质酸水凝胶的具体过程为:透明质酸溶于水,得到质量浓度为1~10%的透明质酸水溶液,然后用0.6mol/L的盐酸将透明质酸水溶液的pH值调到0.5~2.5,再将透明质酸水溶液放入冰箱在-20℃的条件下冷冻4~6天,最后在25~35℃的水浴中解冻15-25h,并用无菌注射用水浸泡清洗去除多余的盐酸,得到透明质酸水凝胶,并在2-8℃的温度下保存待用;
干细胞微球凝胶复合物的制备
将水凝胶置于六孔板中,将干细胞-藻酸盐微球以105-106个/mL的浓度均匀移液于六孔板中,将6孔板放入37℃、5%CO2培养箱培养15min。

Claims (8)

1.一种皮下注射用干细胞微球凝胶复合物,其特征在于:所述凝胶、分散于凝胶中的可降解微球及包埋于该微球内的干细胞、可稳定分泌细胞因子修复组织内部损伤。
2.根据权利要求1所述的一种皮下注射用干细胞微球凝胶复合物,其特征在于:其干细胞微球为采用静电注滴法制备的干细胞-藻酸盐微球。
3.根据权利要求2所述的一种皮下注射用干细胞微球凝胶复合物,其特征在于:所述微球的粒径为1~500μm,优选为50~200μm。
4.根据权利要求1所述的一种皮下注射用干细胞微球凝胶复合物,其特征在于:其凝胶为透明质酸水凝胶。
5.根据权利要求1所述的一种皮下注射用干细胞微球凝胶复合物,其特征在于:其干细胞来源为间充质干细胞。
6.根据权利要求1所述的一种皮下注射用干细胞微球凝胶复合物,其特征在于:其干细胞分散浓度为105-106个/mL。
7.根据权利要求5所述的一种皮下注射用干细胞微球凝胶复合物,其特征在于:,所述的间充质干细胞为骨髓间充质干细胞,脐带间充质干细胞,脂肪间充质干细胞。
8.一种应用权利要求1所述的干细胞微球凝胶复合物的方法,其特征在于:在关节炎、糖尿病足中应用。
