Abstract
本发明具体公开了一种保湿抗衰老中药组合物及其应用。该中药组合物由金钗石斛、银耳、麦冬、枸杞、仙人掌制成。其制备方法为:按配比称取中药原料金钗石斛、银耳、麦冬、枸杞、仙人掌,将上述中药组合物粉碎,混合,加入水或乙醇溶液作为提取溶剂,浸泡后,用常温高压差膨爆提取技术进行常温提取,提取液经离心分离、浓缩后,用大孔树脂吸附,用乙醇多次洗脱,收集洗脱液,浓缩后,再加入与提取浓缩液相同质量的丁二醇溶解,过滤,制得中药组合物。通过以上中药协同作用,达到滋润皮肤、增强皮肤弹性、增强皮肤细胞免疫力、清除自由基、抑制黑色素生成等综合保湿抗衰老功效。该中药组合物用于制备保湿抗衰老化妆品,具有即时保湿效果好,而且保湿效果持久,并兼具有抗衰老和美白的功效。
Description
技术领域
本发明涉及一种复配的中药组合物,该中药组合物具有保湿抗衰老功能,可以应用于保湿抗衰老化妆品的配制。
背景技术
人体的皮肤是覆盖于整个体表的一个重要的而且是最大的器官,具有特殊的独立的功能。皮肤分表皮、真皮及皮下组织三部分,其中表皮是皮肤最外面的一层组织,为皮肤直接与外界接触的部分,它由内而外包括基底层、薄细胞层、颗粒层、透明层、角质层。角质层为表皮最外面部分,由多层相叠的无生物活性的扁平细胞组成,它具有防止水分流失和外界异物内渗的作用。健康正常的角质层中一般含有10%-20%的水分,以使得皮肤维持丰满而富有弹性的状态。
然而随着季节、环境及年龄的变化,皮肤角质层中的水分也会出现波动。当角质层的水分降低到10%以下时就会出现皮肤干燥的症状,更为严重的出现皲裂的症状,长此以往会加速皮肤的衰老。因此,皮肤的保湿和抗衰老显得非常的重要,保湿抗衰老化妆品是最基本的护肤品,是最畅销的化妆品。
目前使用的保湿剂基本是单一功能的添加剂,仅能起到保湿效果,无法满足消费者增强皮肤弹性、增强皮肤屏障和清除自由基、抑制黑色素生成等诉求,所以开发具有多功能的保湿剂具有现实意义和良好的市场前景。
发明内容
本发明的目的是提供一种保湿抗衰老中药组合物,该中药组合物能使皮肤角质层水分长时间地保持充盈、稳定,并能增强皮肤弹性、增强皮肤屏障和清除自由基、抑制黑色素生成。
为达到本发明的目的,本发明采用了如下技术方案:
一种保湿抗衰老中药组合物,其特征在于由如下重量百分比的组分制成:金钗石斛5-50%,银耳 5-50%,麦冬 5-50%,枸杞 5-50%,仙人掌 5-50%。
优选的,一种保湿抗衰老中药组合物,其特征在于由如下重量百分比的组分制成:金钗石斛10-35%,银耳 10-35%,麦冬 10-35%,枸杞 10-35%,仙人掌 10-35%。
优选的,一种保湿抗衰老中药组合物,其特征在于由如下重量百分比的组分制成:金钗石斛20%,银耳 20%,麦冬 20%,枸杞 20%,仙人掌 20%。
优选的,一种保湿抗衰老中药组合物,其特征在于由如下重量百分比的组分制成:金钗石斛25%,银耳 20%,麦冬 15%,枸杞 20%,仙人掌 20%。
优选的,一种保湿抗衰老中药组合物,其特征在于由如下重量百分比的组分制成:金钗石斛15%,银耳 25%,麦冬 20%,枸杞 15%,仙人掌 25%。
优选的,一种保湿抗衰老中药组合物,其特征在于由如下重量百分比的组分制成:金钗石斛25%,银耳 20%,麦冬 25%,枸杞 15%,仙人掌 15%。
所述中药组合物制备方法:按配比称取中药原料金钗石斛、银耳、麦冬、枸杞、仙人掌,将上述中药组合物粉碎,混合,按液料质量比为5:1 -30:1加入水或乙醇溶液作为提取溶剂,浸泡1-5h后,在30-70MPa压力下用常温高压差膨爆提取技术进行常温提取,提取液经离心分离、浓缩后,用大孔树脂吸附,用10-95%的乙醇多次洗脱,收集洗脱液,浓缩至提取液与中药的质量比为1:0.1 -1:10,再加入与提取浓缩液相同质量的丁二醇溶解,过滤,制得中药组合物。
本发明另一目的是上述保湿护肤中药组合物在配制化妆品中的应用,其特征在于化妆品中含有上述保湿护肤中药组合物,其用量为0.01-50%,优选用量为1-30%。除了含有上述保湿护肤中药组合物以外,可以根据需要适当地添加适合化妆品使用的通常成分,如润肤剂、水、乳化剂、防腐剂、香精、色素等基质成分。
上述保湿护肤中药组合物的有益效果:本发明采用君臣佐使的中医理论进行保湿护肤中药组合物的组方。在本组方中,金钗石斛为君药,石斛具生津益胃、清热解毒功效,近代科学研究表明金钗石斛中的石斛糖蛋白能增加AQP3(水通道蛋白——真皮与表皮间水分交换的重要介质)的表达量,能畅通水通道而成为强劲水动力。麦冬和银耳为臣药,麦冬具有有生津解渴、润肺止咳之效,现代药理研究也表明,麦冬主要含甾体皂苷、生物碱、谷甾醇、氨基酸、维生素等,具有抗疲劳、清除自由基、提高细胞免疫功能等作用;银耳具有优异滋阴润肺功效,含有丰富的多糖类物质,具有非常好的保湿和保持皮肤弹性作用。枸杞为佐药,具有补肾益精、养肝明目、补血安神、生津止渴、润肺止咳之功效,现代研究也表明枸杞含有丰富的多糖类化合物,对于皮肤缺水引起的皮肤皲裂有良好的治疗效果,同时提升肌体免疫力。仙人掌为使药,调和诸药,具有行气活血、清热解毒之功效,现代研究结果表明仙人掌具有非常好的保湿和抑菌作用,对于炎症、冻伤具有非常号的治疗效果。通过以上中药协同作用,达到滋润皮肤、增强皮肤弹性、增强皮肤细胞免疫力、增强皮肤屏障、清除自由基和抑制黑色素生成等综合保湿和抗衰老功效。

以下通过具体实施例来对本发明作进一步的说明,但这些实施例只是为了说明,不是对本发明的限制。
实施例1 一种保湿抗衰老中药组合物(Ⅰ)
一种保湿抗衰老中药组合物(Ⅰ),其特征在于由如下重量百分比的组分制成:金钗石斛20%,银耳 20%,麦冬 20%,枸杞 20%,仙人掌 20%。
实施例2 一种保湿抗衰老中药组合物(Ⅱ)
一种保湿抗衰老中药组合物(Ⅱ),其特征在于由如下重量百分比的组分制成:金钗石斛25%,银耳 20%,麦冬 15%,枸杞 20%,仙人掌 20%。
实施例3 一种保湿抗衰老中药组合物(Ⅲ)
一种保湿抗衰老中药组合物(Ⅲ),其特征在于由如下重量百分比的组分制成:金钗石斛15%,银耳 25%,麦冬 20%,枸杞 15%,仙人掌 25%。
实施例4 一种保湿抗衰老中药组合物(Ⅳ)
一种保湿抗衰老中药组合物(Ⅳ),其特征在于由如下重量百分比的组分制成:金钗石斛25%,银耳 20%,麦冬 25%,枸杞 15%,仙人掌 15%。
实施例1-4所述中药组合物制备方法:按配比称取中药原料金钗石斛、银耳、麦冬、枸杞、仙人掌,将上述中药组合物粉碎,混合,按液料质量比为12:1加入水作为提取溶剂,浸泡2h后,在10MPa压力下用常温高压差膨爆提取技术进行常温提取,提取液经离心分离、浓缩后,用大孔树脂吸附,用10-95%的乙醇多次洗脱,收集洗脱液,浓缩至提取液与中药的质量比为1:1,再加入与提取浓缩液相同质量的丁二醇溶解,过滤,制得中药组合物。
对比例1-5 单种中药提取物
分别称取金钗石斛、银耳、麦冬、枸杞、仙人掌,粉碎,按实施例1-4所述中药组合物制备方法同样的方法进行提取分离浓缩,分别制得金钗石斛提取物、银耳提取物、麦冬提取物、枸杞提取物、仙人掌提取物。
试验例1 清除自由基和抑制黑色素生成试验
1、试样的配制
将实施例1的中药组合物和对比例1-5的单种中药提取物溶于水中,配成浓度为0.5%的样品溶液,进行下面试验。
2、抑制酪氨酸酶活力效果评价试验
按表1准确吸取样品溶液、pH值为 6.8的磷酸盐缓冲溶液(PBS)和质量分数为0.1 %的多巴溶液,充分混合,于30 ℃孵育5 min后,加入活度为110 U/mL的酪氨酸酶溶液,再在30℃下孵育10 min,迅速转移至比色皿中,在475 nm处测吸光度。使用曲酸、氢醌、抗坏血酸、熊果苷作为对照物。

按式(1)计算样品对酪氨酸酶的相对抑制率 (I,简称抑制率,下同)。

3、抑制黑色素细胞中黑色素合成试验
将B16黑色素瘤细胞以1×105个/mL的密度接种于96孔板中,每孔90μL,CO2孵箱中孵育24小时后,每孔加入样品溶液。同时设立培养基和细胞的空白对照组。
将培养板置于孵箱中孵育72小时后,弃去上清液,PBS洗涤两遍,然后每孔加入0.5mL胰酶消化细胞3min,每孔加入2mL维持液终止消化。混匀后,每种浓度取出0.5mL做细胞计数。其余细胞悬液以2500r/min离心分离5min,弃去上清液,于沉淀中加入NaOH溶液,加热使黑色素溶解,选择490nm波长在酶联免疫检测仪下测定吸光度值。按式(2)计算样品对黑色素合成抑制率(I’)。

4、超氧阴离子自由基清除效果评价试验
按表2添加药液。取样品溶液2 mL,加入2.25 mL Tris-HCl缓冲溶液(pH 8.36),0.25 mL浓度为10 mmoL/L邻苯三酚溶液后,25 ℃下水浴,第4min时滴加10 mol/L盐酸1滴,终止反应。用紫外分光光度计在420 nm下测吸光度值。按照式(3)计算清除率(D)。

6、实验结果
美白护肤效果评价实验见表3所示。

实验结果表明,本发明的中药组合物具有良好清除自由基作用,同时能有效地抑制酪氨酸酶活性和抑制黑色素合成,这些作用均优于单一中药提取物的作用,说明经过科学组方后起到了协同增效作用。
试验例2 刺激性评价实验
采用兔红细胞溶血实验评价发酵液刺激性。具体操作如下:分别取实施例1的中药组合物和对比例1-5的单种中药提取物500μL置于2mL离心管中,再分别加入2%兔红细胞(市售)悬液500μL,轻摇混匀,置于37℃恒温培养箱中温育3h后,2000r/min离心3min,吸取上清液,以生理盐水为阴性对照,以蒸馏水为阳性对照,在波长414nm处测定吸光值,按公式计算溶血度。结果如表4所示。

实验结果对比表明,本发明的保湿抗衰老中药组合物溶血度比对比例1-5制得的单味中药的提取液低得多,说明经过科学组方后可显著降低提取物的刺激性。
实施例5保湿抗衰老霜
表5给出含有本发明的一种保湿抗衰老中药组合物的保湿抗衰老霜配方。

制备方法:A相80℃溶解混合均匀,B相80℃溶解混合均匀,环聚二甲基硅氧烷乳化前加入B相,乳化时B相加入A相并均质5分钟,降温至50℃时加入C相并均质2分钟,搅拌均匀,降温至35℃,复测pH后300目过滤出料,陈化24h后包装,即可。
对比例6-10 含有单味中药提取物的保湿霜
将实施例5中“一种保湿抗衰老中药组合物(Ⅰ)”用对比例1-5制备的“单种中药提取物”取代外,其余成分和制备方法与实施例5一致,分别值得对比例6-10含有单味中药提取物的保湿霜。
实施例6保湿抗衰老乳液
表6给出含有本发明的一种保湿抗衰老中药组合物的保湿抗衰老乳液配方,各物质的用量用质量分数表示:

制备方法:A相80℃溶解混合均匀,B相80℃溶解混合均匀,乳化前将C加入B相,乳化时A相加入B相并均质5分钟,降温至50℃时加入D并均质2分钟,搅拌均匀,降温至35℃,复测pH后300目过滤出料,陈化24h后包装,即可。
实施例7 保湿抗衰老面膜
一种保湿抗衰老中药组合物的保湿抗衰老面贴膜,该面贴膜的具体配方如表7所示:

制备方法如下:
1、将添加相原料对羟基苯乙酮和1,2-己二醇预先在1,3-丙二醇中加热溶解质澄清透明,备用;
2、将水相原料投入至去离子水中,加热至75-85℃,搅拌至溶解均匀;
3、水相降温至60℃,边搅拌边缓慢加入已预先在1,3-丙二醇中溶解好的羟苯甲酯和羟苯丙酯,搅拌至溶解均匀;
4、冷却至40℃,加入中药保湿组合物提取物(Ⅰ),再滴加氨甲基丙醇调节pH至5-6;
5、搅拌均匀,降温至35℃,复测pH后300目过滤出料,即可。
实施例8 保湿抗衰老精华液
一种保湿抗衰老中药组合物的精华液,该精华液的具体配方如表8所示:

上述精华液,其制备方法如下:
1、将添加相原料对羟基苯乙酮和1,2-己二醇预先在1,3-丁二醇中加热溶解质澄清透明,备用。
2、将水相投入到去离子水中,搅拌均匀,加热搅拌至75-85℃;
3、保温10分钟,然后开冷却水降温至60℃,边搅拌边缓慢加入已预先在1,3-丁二醇中溶解好的羟苯甲酯和羟苯丙酯,搅拌至溶解均匀;
4、冷却至40℃,加入中药保湿组合物提取物(Ⅰ),;
5、搅拌均匀,降温至35℃,复测pH后300目出料,即可。
试验例3 保湿效果评价
1、评价样品制备
取按照实施例5制备的含本发明组合物的保湿抗衰老霜和对比例6-10制备含有单味中药提取物的保湿霜。
2、测试方法
水分测试采用的是世界公认的CORNEOMETER®法——电容法,皮肤水分流失TEWL对评估皮肤水分保护层的功能是非常重要的参数,在国际上已经得到了非常广泛的认可。皮肤保护层越完好,水分的含量就会越高,皮肤水分流失TEWL的数值就越低。皮肤水分测试能及时发现皮肤的屏障保护功能是否已被破坏。
2.1测试仪器
德国Courage+Khazaka公司(德国CK公司)制造的皮肤水分含量Corneometer测试探头。德国德国Courage+Khazaka公司(德国CK公司)制造的皮肤水分流失(YEWL) TewameterTM300测试探头。
2.2皮肤水分含量及皮肤水分流失(TEWL)测试方法
将实施例5和对比例6-10进行下面实验。
选取干性肌肤人群(刺痛感累计≥3分者)150名,年龄在20-45岁,男女各半。随机分成5组,每组15名男性、15名女性。受试部位前15天不能使用任何产品(化妆品、外用药品、内服保健品)。试验前,要求受试者清洗颜面部并在清洗后2小时进入气候控制室(22±1℃,相对湿度50%)静坐30min以上,并保持放松状态。
实验中按随机表进行半边脸测试,一侧为实施例5的保湿抗衰老霜,另一侧为对比例6-10的保湿霜,选取志愿者的左右鼻唇沟部分进行皮肤水分含量及水分流失(TEWL)测试,取3个值的平均值作为左右鼻唇沟水分含量数据(d0)及鼻唇沟皮肤水分流失(TEWL)的数据(d0)。测试结束后,志愿者需按要求使用,一侧脸涂抹实施例产品,另一侧脸涂抹对比例产品。连续使用28天,并要求试验者在试验的第14天(d14)及第28天(d28)进行回访。
3、试验结果
结果见表9所示。

由结果可知,本发明实施例6制备的保湿抗衰老霜具有良好的保湿效果,而且实施例6的保湿抗衰老霜相较于对比例6-10保湿霜具有更好的保湿效果,说明本发明中药组合物经过科学复配后各组分之间具有协同保湿增效的作用。
试验例4皮肤弹性试验
1、评价样品制备
取按照实施例5制备的含本发明组合物的保湿抗衰老霜和对比例6-10制备含有单味中药提取物的保湿霜。
2、评价方法
采用德国CK公司皮肤天性测试仪 MPA580,通过测量使用产品前后测试皮肤表面产生一个负压将皮肤吸进一个特定测试探头内,皮肤被吸进测试探头内的深度是通过一个非接触式的光学测试系统测得的。测试探头包括光的发射器和接收器,光的比率同被吸入皮肤的深度成正比,然后通过MPA软件分析来确定皮肤的弹性性能。无负压时皮肤的回弹量与有负压时的最大拉伸量之比,比值越接近1,说明皮肤弹性越好。
试验方法:与试验例3同时进行,一致的志愿者和使用方法,也是持续28天,期间与D0,D14,D28进行回访,于恒温恒湿环境静坐30min后,进行VISIA图像拍摄及弹性的测试。再通过计算得出20名志愿者在不同时间间隔点的平均弹性数据,结果如表10所示:

由试验结果可以得出,使用了本发明实施例5保湿抗衰老霜和对比例6-10的保湿霜,皮肤弹性均有所提升,但本发明实施例5效果显著比对比例6-10效果更好,说明经过科学复配后中药间起到了良好的协同增效作用。
Claims (7)
1.一种保湿抗衰老中药组合物,其特征在于由如下重量百分比的组分制成:金钗石斛5-50%,银耳 5-50%,麦冬 5-50%,枸杞 5-50%,仙人掌 5-50%。
2.一种保湿抗衰老中药组合物,其特征在于由如下重量百分比的组分制成:金钗石斛10-35%,银耳10-35%,麦冬 10-35%,枸杞 10-35%,仙人掌 10-35%。
3.权利要求1-2任一项所述的中药组合物制备方法,其特征在于按如下步骤制备:按配比称取中药原料金钗石斛、银耳、麦冬、枸杞、仙人掌,将上述中药组合物粉碎,混合,按液料质量比为5:1 -30:1加入水或乙醇溶液作为提取溶剂,浸泡1-5h后,在30-70MPa压力下用常温高压差膨爆提取技术进行常温提取,提取液经离心分离、浓缩后,用大孔树脂吸附,用10-95%的乙醇多次洗脱,收集洗脱液,浓缩至提取液与中药的质量比为1:0.1 -1:10,再加入与提取浓缩液相同质量的丁二醇溶解,过滤,制得中药组合物。
4.权利要求1-2任一项所述的一种保湿抗衰老中药组合物在配制化妆品中的应用,其特征在于化妆品中含有权利要求1-2所述的一种保湿抗衰老中药组合物,其用量以质量分数计为0.01-50%。
5.一种化妆品,其特征在于:含有权利要求1~2任一项所述的一种保湿抗衰老中药组合物。
6.根据权利要求5所述的化妆品,其特征在于:所述化妆品含有以质量分数计为0.01~500%的权利要求1-2所述的一种保湿抗衰老中药组合物;所述的化妆品为:水剂、乳液类、膏霜类、面膜类、啫喱类、粉类、喷雾类、精华类、洗护类。
7.根据权利要求5或6所述的化妆品,其特征在于:
