Abstract
本发明公开了一种控制驴排卵时间段相对集中的药物组合物及使用方法,该药物组合物一份是由600国际单位绒毛膜促性腺激素和10国际单位的促卵泡素混合而成的粉剂。使用时,实时监控驴卵泡发育直径,当发现驴卵泡发育直径达到35mm时,一次性给予驴本发明50份混合粉剂的皮下剂量,要求50份混合粉剂用生理盐水稀释;在给予粉剂后的第32小时、42小时,分别使驴授精一次,达到提高情期受胎率的目的。本发明的药物组合物矿可以控制驴排卵时间和减少驴的流产率,保证母驴能足月分娩,且分娩后的驴仔体质发育良好存活率较高。
Description
技术领域
本发明属于马属动物繁殖领域,尤其是控制驴排卵时间段相对集中的药物组合物。
背景技术
由于马属动物的卵巢结构的独特性,在出现发情表现后发情持续期变化很大,一般会持续3~14天,驴的排卵时间也会因卵巢上卵泡生长位置的差异,排卵时间也会出现较大差异,排卵时间多在发情的第3~10天,在发情停止前的12~24小时内排卵不等,这就造成了驴输精时间难以确定。
曾经有报道采用促性腺激素释放激素或绒毛膜促性腺激素处理驴或马促进排卵,以提高受胎率。比如中国专利申请号2016108107595公开的一种驴的胚胎移植方法;再比如本申请人申请的申请号为201510554754.6公开的一种增加驴受胎率的鲜精人工授精方法。但实践中发现无论促性腺激素释放激素还是单纯绒毛膜促性腺激素处理,驴的排卵时间都难以满足提高受胎率的目的。
为了提高受胎率,中国专利申请号2014100851723公开了一种诱导马超数排卵的药物组合物,它是将单纯绒毛膜促性腺激素结合促卵泡素作为外源激素,从该文献中可以看出,外源激素中单纯绒毛膜促性腺激素与促卵泡素的量均是1000-2500单位,这种比例组合虽然达到马属动物超数排卵的目的,但是排卵数的增加,会引起怀双胎,驴怀双胎往往容易流产,即便能够足月分娩,出生驴驹体质发育不良的比例也很高,造成早期死亡,因此不能达到令人满意的效果。原因是,促性腺激素释放激素可以诱导垂体分泌促黄体素和促卵泡素,如果剂量使用量小,达不到促排卵的效果;如果使用剂量大使得促卵泡素的量也增加,促卵泡素量过大会造成卵泡选择过程中的从属卵泡能够发育至排卵大小,再在促黄体素的作用下发生排卵。因此无论大剂量促性腺激素释放激素处理还是绒毛膜促性腺激素与促卵泡素结合,只要促卵泡素的剂量过大就会造成排卵数增加。
发明内容:
本发明针对控制驴排卵时间,遵循激素在体内生理作用的原则,提供一种在可以防止驴流产基础上控制驴排卵时间段相对集中的药物组合物。
本发明提供利用上述组合物制备的方法。
为达到上述目的,本发明的药物组合物一份是由600国际单位绒毛膜促性腺激素和10国际单位的促卵泡素混合而成的粉剂。
本发明混合粉剂作用于驴的使用方法是:
第一步:首先实时监控驴卵泡发育直径,当发现驴卵泡发育直径达到35mm时,一次性给予驴本发明50份混合粉剂的皮下剂量,要求50份混合粉剂用生理盐水稀释;
第二步:在给予粉剂后的第32小时、42小时,分别使驴授精一次,达到提高情期受胎率的目的。
下面根据本发明原料功效以及作用机理说明本发明的优点。
本发明的绒毛膜促性腺素,简写HCG,它是一种糖蛋白,分子质量为39000,具有诱导排卵前的卵泡生长发育,促进卵泡成熟、排卵功效。促卵泡素,简写FSH,也是一种糖蛋白,分子量为30000左右,主要具有刺激卵泡生长和发育,能提高卵泡壁细胞的摄氧量,增加蛋白质合成,并对卵泡内膜细胞分化、颗粒细胞增生和卵泡液的分泌具有促进作用。从表面上看,两种激素都对卵泡的发育和生长有促进作用,但他们在作用的时间段和顺序上存在差异,生理上,体内的促卵泡素FSH要在卵泡发育到一定阶段后才能正常发挥作用,利用下丘脑分泌的促性腺激素释放激素在体内先促使垂体释放促卵泡素和促黄体素,再由促卵泡素和促黄体素引起卵泡发育的变化。本发明的绒毛膜促性腺素是促黄体素的类似物,是从孕妇尿中提取的药物,用绒毛膜促性腺素结合促卵泡素可以代替下丘脑分泌的促性腺激素释放激素的作用,而且作用直接。这样就容易控制驴的排卵时间。虽然垂体分泌的促卵泡素FSH可以促进卵泡发育,但是外源的促卵泡素FSH在组合药物中比例很小,不能够引起排卵数的增加,但可以内膜细胞分化、颗粒细胞增生和卵泡液的分泌有助于优势卵泡的成熟,基于这一点,本发明将两种药物的比例控制在60∶1,更适合促进已经发育接近完成的卵泡的发育和排卵,避免了现有技术利用大剂量外援促卵泡素FSH处理造成的排卵数增加,出现怀双驹流产的现象。
本发明的使用方法在于控制驴排卵时间和减少驴的流产率,保证母驴能足月分娩,且分娩后的驴仔体质发育良好存活率较高。在实施过程中对驴没有伤害,不是以治疗为目的,也不是以非治疗为目的的外科手术方法,更不属于所谓的介入性治疗或处置的方法,属于专利法保护的客体。
具体实施方式
下面结合一具体实施例进一步说明本发明的技术方案和优点。
本发明混合粉剂作用于驴的使用方法是:
第一步:首先实时监控驴卵泡发育直径,当发现驴卵泡发育直径达到35mm时,一次性给予驴本发明50份混合粉剂的皮下剂量,要求50份混合粉剂用生理盐水稀释;所述的一份混合粉剂是由600国际单位绒毛膜促性腺激素和10国际单位的促卵泡素混合而成的。
第二步:在给予粉剂后的第32小时、42小时,分别使驴授精一次,达到提高情期受胎率的目的。
为了验证本发明的效果,随机选取同批正常繁殖的3岁驴30头,其中11头驴采取自然排卵,剩余的19头驴用本发明混合粉剂进行处理,在处理后的时间段内对该30头驴的排卵情况进行监控,监控结果如表一。
从表一看出,19头驴用本发明混合粉剂处理后,排卵时间集中在24-48小时内,有14头驴排卵,占总数的73.7%,最集中时间在处理后的32-40小时,有8头驴排卵。采用自然排卵的11头驴,排卵时间比较分散,持续时间长,在24-48小时内,只有1头驴排卵,占总数9.09%。这说明本发明的混合粉剂确实有促进驴集中排卵的效果很好。
对用本发明混合粉剂处理后的驴采用鲜精在第32小时、42小时分别授精一次,受胎率达到45%以上。而采用自然排卵的受胎率不到30%。

1.一种控制驴排卵时间段相对集中的药物组合物,其特征在于,该药物组合物一份是由600国际单位绒毛膜促性腺激素和10国际单位的促卵泡素混合而成的粉剂。
2.一种如权利要求1所述的的药物组合物的使用方法,其特征在于,
第一步:首先实时监控驴卵泡发育直径,当发现驴卵泡发育直径达到35mm时,一次性给予驴本发明50份混合粉剂的皮下剂量,要求50份混合粉剂用生理盐水稀释;
第二步:在给予粉剂后的第32小时、42小时,分别使驴授精一次,达到提高情期受胎率的目的。
