一种早产诊断用生物标志物及其应用
技术领域
本发明涉及生物检测技术领域,特别涉及一种早产诊断用生物标志物及其应用。
背景技术
早产(Preterm Birth,PTB)是指妊娠满28周至不足37周间分娩者。胎膜早破(Premature rupture of membrane,PROM)是指37周后但临产前发生胎膜破裂。如果PROM发生在孕28周至37周之间,该PROM被定义为早产胎膜早破(preterm premature rupture ofmembrane,PPROM)又称为未足月胎膜早破,发生率约2.1%-3.5%。WHO在2012年发布了全球有1500万早产儿。早产的发生率在美国为12.1%,法国:5.3%(单胎),47.8%(双胎),中国:5~15%,而约1/3的早产来自于PPROM。早产是围生医学中的一个重要、复杂而又常见的妊娠并发症,早产儿后遗症多、病死率高,约1/4的早产儿遗留有神经或智力方面的后遗症(尤其是孕龄小于35周的早产儿)。西方发达国家早产儿死亡率为5%~9%,发展中国家为12.7%~20.8%,死亡原因主要是围生期窒息、颅内出血、畸形。目前数据显示早产是新生儿死亡率及并发症发生的首位原因。所以预防早产是降低围生儿死亡率和提高新生儿素质的主要措施之一。目前对早产及早产胎膜早破的确切发病机制仍不明确,临床上尚缺乏有效的手段对其进行早期预测。因此,发现新的高特异性和敏感性的生物标志物(Biomarker)对于早产及早产胎膜早破的早期筛查和早期预测显得尤为重要。
发明内容
为了解决现有技术的问题,本发明实施例提供了一种早产诊断用生物标志物及其应用。所述技术方案如下:
一方面,一种早产诊断用生物标志物,所述生物标志物为:lncDesmin。
另一方面,上述早产诊断用生物标志物在制备早产检测试剂盒中的应用。
具体实施方式
为使本发明的目的、技术方案和优点更加清楚,下面将对本发明实施方式作进一步地详细描述。
实施例
对下述A、B、C、D四组研究对象的母体外周血lncDesmin、mRNA及Desmin蛋白的表达情况,运用实时定量荧光PCR、Wsetern Blot及ELISA等实验方法和技术,通过分析中孕各组外周血中lncDesmin的表达,并探讨其与早产、早产胎膜早破的相关性,期望为早产、早产合并胎膜早破的早期预测、早期干预提供新的理论基础,为进一步临床检测提供方法及技术支撑。
(a)取材:将在中孕早期(孕14-18周)及临产后收集母体外周血,根据她们的妊娠结局选取四组研究对象,分别是A:早产胎膜早破(PPROM),B:足月无胎膜早破(full-termbirth,FTB),C:早产无胎膜早破(preterm birth,PTB),D:足月胎膜早破(prematurerupture of membrane,PROM)。
(b)研究对象:选择在Desmin基因位点上的lncRNA,即lncDesmin,为研究对象,探索lncDesmin在母体外周血的差异性表达,并且研究lncDesmin对Desmin基因的转录及转录后的调控。
具体步骤:
(1)不同孕妇组外周血中lncDesmin的表达变化研究
针对在孕中期和临产后采集的四组孕妇(FTB,PTB,PROM和PPROM组)的外周血,利用Trizol法提取总RNA,并通过反转录PCR合成cDNA模板;根据lncDesmin的序列设计引物,通过Real-time PCR法(RT-PCR)检测各组孕妇的孕中期和临产后外周血中lncDesmin的表达量。通过比较各组间同一时期以及各组内不同时期lncDesmin的表达变化,探讨孕妇外周血中lncDesmin的表达变化是否与早产的发生存在相关性。
(2)不同孕妇组外周血中Desmin基因mRNA的表达变化研究
根据Desmin基因的mRNA序列设计引物,以外周血cDNA为模板,通过RT-PCR法检测各组孕妇的孕中期和临产后外周血中mRNA的表达量。通过比较各组间同一时期以及各组内不同时期的mRNA表达变化,探讨孕妇外周血中Desmin的mRNA的表达变化是否与早产的发生存在相关性。
(3)不同孕妇组外周血中Desmin蛋白含量变化研究
购置抗Desmin的单克隆抗体,利用ELISA法测定各组孕妇在孕中期和临产后外周血中Desmin蛋白的含量,通过比较各组间同一时期以及各组内不同时期Desmin蛋白的含量变化,探讨孕妇外周血中Desmin蛋白的含量变化是否与早产的发生存在相关性。
