Abstract

本发明公开了一种去除红细胞的层析缓冲液,所述层析缓冲液为0.2M的PB溶液、pH=6.0‑8.0。本发明的层析缓冲液配合荧光免疫层析检测卡使用,在使用时,将其与全血样本充分混合后加入检测卡的加样孔,样本在经过检测卡的样品垫时红细胞会留在检测卡的样品垫上,从而避免红细胞出现在层析膜上对检测结果造成干扰,克服了现有技术中的不足。

一种去除红细胞的层析缓冲液
技术领域
本发明涉及及细胞生物技术领域,特别涉及一种去除红细胞的层析缓冲液。
背景技术
免疫荧光双抗体夹心法常用于定量检测N-端脑利钠肽前体(NT-proBNP)、肌酸激酶同工酶(CK-MB)、肌钙蛋白I(cTnI)或肌红蛋白(Myo)等生化标志物,其原理如下:将待测样本(全血、血清或血浆)加入检测卡的加样孔,如果样本中存在待测生化标志物,则包被有相应抗体的荧光微球与生化标志物结合形成复合物。该复合物再沿着检测卡的试纸条侧向移动,结合了荧光微球的复合物就会在检测区被捕捉到,而多余的荧光微球则在质控区被捕捉。当检测卡插入荧光免疫分析仪后,分析仪自动扫描、检测检测区和质控区上的荧光强度,用两个荧光强度的比值即可计算出所测生化标志物的含量。
在实际操作中发现,全血样本中的红细胞可对生化标志物的检验造成干扰,导致检验结果产生假阴性或假阳性的情况;有许多现有技术可用于分离样品中的红细胞,除了惯用的离心法,公告号为CN101750244B的中国发明专利还公开了一种分离血液样本中红细胞的方法。这些方法用物理和化学方法来分离样品中的红细胞,缺陷在于需要购置额外的试剂盒或者设备、需要增加操作步骤,在实际应用中存在一定的局限性。
发明内容
本发明所要解决的技术问题是,提供一种去除红细胞的层析缓冲液,与之混合后的全血样本中的红细胞会留在荧光免疫层析检测卡的样品垫上,从而避免红细胞出现在层析膜上对检测结果造成干扰,以克服现有技术中的不足。
本发明的技术方案:一种去除红细胞的层析缓冲液,所述层析缓冲液为0.2M的PB溶液、pH=6.0-8.0
优选地,所述PB溶液的pH=7.4。
优选地,所述层析缓冲液还含有0.1wt%procline 300和0.5wt%Tween-20。
与现有技术相比,本发明的层析缓冲液配合荧光免疫层析检测卡使用,在使用时,将其与全血样本充分混合后加入检测卡的加样孔,样本在经过检测卡的样品垫时红细胞会留在检测卡的样品垫上(即去除了全血样本中的红细胞),从而避免红细胞出现在层析膜上对检测结果造成干扰,层析缓冲液中的PB浓度至关重要,只有特定浓度的PB溶液才具备上述效果。本发明的层析缓冲液可以代替荧光免疫层析检测盒本身配备的层析缓冲液,不增加检测操作步骤,不需要额外的设备,克服了现有技术中的不足。

具体实施方式
下面结合附图和实施例对本发明作进一步详述,以下实施例只是描述性的,不是限定性的,不能以此限定本发明的保护范围。
实施例1:本发明的去除红细胞的层析缓冲液为0.2M的PB溶液、pH=6.0。
实施例2:本发明的去除红细胞的层析缓冲液为0.2M的PB溶液、pH=7.4,所述层析缓冲液还含有0.1wt%procline 300(防腐剂)和0.5wt%Tween-20(吐温20、表面活性剂)。
实施例3:本发明的去除红细胞的层析缓冲液为0.2M的PB溶液、pH=8.0。
实施例4:测试层析缓冲液对于红细胞的去除效果
1、配置如下浓度的PB溶液(pH=6.0-8.0):0.01M、0.1M、0.15M、0.2M。
2、将配置好的PB溶液分别与全血样本充分混合后加入检测卡的加样孔;
3、从检测卡的观察窗观察层析膜的颜色,参见附图1,0.2M的PB溶液测试后的层析膜完全看不出红色,即去除了全血样本中的红细胞。

1.一种去除红细胞的层析缓冲液,其特征在于:所述层析缓冲液为0.2M的PB溶液、pH=6.0-8.0。
2.根据权利要求1所述的层析缓冲液,其特征在于:所述PB溶液的pH=7.4。
3.根据权利要求1或2所述的层析缓冲液,其特征在于:所述层析缓冲液还含有0.1wt%procline 300和0.5wt%Tween-20。
