Abstract
本发明公开了促进植物破坏细胞快速生长的调节剂,将二甲基亚砜溶液加入到羧甲基壳聚糖溶液中反应制备水凝胶,另取布油菜素内酯、肽亚铁螯合物、细胞分裂素、6‑BA与尿素溶解于黄腐酸溶液中,然后再加入稀土化合物、2,5‑二甲基硼咾烷、复合氨基酸得生长调节剂,最后将生长调节剂加入到水凝胶中制备得到促进植物破坏细胞快速生长的调节剂。有益效果为:生长调节剂可以刺激植物细胞超氧化物歧化酶的基因表达,加速酶的分泌,提高酶的活性,保护细胞膜免受伤害,维持膜内外电解质平衡,促进破坏细胞的修复与生长,鱼皮胶原蛋白肽亚铁螯合物可以为破坏细胞提供稳定的亚铁离子,促进叶绿素的合成,促进细胞分裂与生长。

技术领域
本发明涉及植物生长调节领域,尤其是涉及促进植物破坏细胞快速生长的调节剂。
技术背景
温度是作物生长发育最为敏感的环境条件之一,温度是影响作物生产、限制其地域分布的主要因子。近年来全球性温室效应不断加剧,据预测我国各地的增温幅度将明显高于全球,整个种植业将面临严重挑战。20世纪80年代末,我国南方柑桔产区遭受的3次大范围高温热潮造成了巨大的经济损失。目前,不仅植物在干旱逆境下,各种生理生化的代谢变化的研究成为研究热点,而且在此基础上深入研究的逆境胁迫对细胞结构的影响也成为解决此类问题的关键。
干旱胁迫是因植物体的水分平衡遭到破坏所致。如果土壤缺水,根系吸收的水分少,而叶片蒸腾的水分多,植物体的水分收支失去平衡,就会发生水分亏缺,造成干旱。同时,如空气蒸发力大,蒸腾消耗的水分很多,而吸收的水分不足以补偿这种支出时,植物也会发生水分亏缺,植物水分亏缺,细胞膨压下降,细胞受损,如果细胞受损得不到有效抑制,植株叶片就会出现萎蔫。严重缺水会对植物体造成很大的伤害,对植物体的损害顺序为:(1)降低光合作用,加强呼吸作用;(2)有机物的运输减慢,积累减少;(3)有机物质合成与分解的正常比例遭到破坏;(4)原生质脱水、衰老、破坏;(5)细胞大量破坏甚至死亡,植株凋亡。
发明内容
本发明的目的在于提供促进植物破坏细胞快速生长的调节剂,生长调节剂可以刺激植物细胞超氧化物歧化酶的基因表达,加速酶的分泌,提高酶的活性,保护细胞膜免受伤害,维持膜内外电解质平衡,促进破坏细胞的修复与生长,鱼皮胶原蛋白肽亚铁螯合物的含量与纯度较高,可以为破坏细胞提供稳定的亚铁离子,促进叶绿素的合成,促进细胞分裂与生长。
本发明针对背景技术中提到的问题,采取的技术方案为:
促进植物破坏细胞快速生长的调节剂,其制备方法包括:制备水凝胶、制备生长调节剂、复配,具体包括以下步骤:
制备水凝胶:将15~18份羧甲基壳聚糖与850~980份去离子水混合,磁力搅拌至羧甲基壳聚糖完全溶解;另取1.65~1.78份1-乙基-(3-二甲基氨基丙基)碳二亚胺盐酸盐、1.26~1.32份N-羟基琥珀酰亚胺加入到560~580份二甲基亚砜中,磁力搅拌并完全溶解;在22~23℃温度下,将二甲基亚砜溶液逐滴加入到羧甲基壳聚糖水溶液中并持续反应4~6h,产物透析18~24h即得水凝胶;水凝胶均匀度良好,而且其水溶性、胶凝性、促渗透性、生物相容性和可降解性均较为优异,可以作为生长调节剂的载体,而且水凝胶能够吸收大量的水份,在植物表皮形成一层隔膜,大大降低植物叶片表面的蒸腾作用,有效缓解植物的缺水、蒸发而造成的失水现象;
制备生长调节剂:称取0.1~0.12份布油菜素内酯、0.3~2.5份肽亚铁螯合物、0.8~1.6份硼砂、0.02~0.5份细胞分裂素、0.04~0.06份赤霉素、0.02~0.05份6-BA与2~3份尿素,依次将上述物质溶解于200~230份8.8~9.2%的黄腐酸溶液中,然后再加入0.2~0.3份稀土化合物、0.02~0.025份2,5-二甲基硼咾烷、2~10份复合氨基酸,搅拌并混合均匀即得生长调节剂;生长调节剂能缩小气孔开张度,减少水分蒸腾,使植株和土壤保持较多的水分,通过物理化学作用与农药形成农药-黄腐酸复合物,既可降低农药毒性、减少农药用量、提高农药药效、提高对人畜的安全性,又可减少环境污染,络合微量元素,提高植物对微量元素的吸收与运转能力,同时还可有效促进细胞生长,加速细胞分裂,促进叶绿素合成,延长黄化时间,避免植物枯萎;
复配:在暗光、1~2℃温度条件下,将生长调节剂缓慢倒入羧甲基壳聚糖水凝胶中,边倒入边以80~100r/min的低转速搅拌羧甲基壳聚糖水凝胶溶液,混合均匀后即得促进植物破坏细胞快速生长的调节剂;将调节剂通过喷施、涂抹于植物表面后,会在植物表面形成一层调节剂水凝胶薄膜,薄膜不仅对植物表面具有隔水、防蒸作用,降低植株表面的水份蒸发,同时还可使调节剂水凝胶薄膜内的生长调节剂缓慢释放,使调节剂持久的发挥减蒸作用,同时还可以有效的加速破坏细胞生长的作用,加速细胞分裂,避免细胞坏死现象持续恶化,可以高效、持续的促进植物破坏细胞快速生长。
作为优选,2,5-二甲基硼咾烷中含有1/50~1/45%的(R,R)-2,5-二甲基硼咾烷;特殊配比的(R,R)-2,5-二甲基硼咾烷、(S,S)-2,5-二甲基硼咾烷具有协同作用,该协同作用可以刺激植物细胞超氧化物歧化酶的相关基因的表达,从而加速分泌超氧化物歧化酶,提高超氧化物歧化酶的活性,平衡了因高温胁迫造成的植物对活性氧自由基的清除能力和对细胞膜的保护作用,保护细胞膜免受伤害,维持膜内外电解质平衡,从而抑制内源乙烯和脱落酸的大量释放,降低植物果柄离层区因为纤维素酶和过氧化物酶活性,促进破坏细胞的修复与生长,降低落花落果量。
作为优选,肽亚铁螯合物的制备方法为:将经酶解后的鱼皮胶原蛋白肽溶液经低温干燥后得到固含量为16~18%的肽液,调节pH值至6.8~7.0,加入1.3~1.5倍的浓度为0.6~0.9M的FeCl2溶液,再加入胶原蛋白肽溶液干物重量0.1~0.3‰的复配助剂,在25~28℃温度下震荡螯合45~60min,将螯合液于1~2℃温度下以13000~13500r/min转速离心浓缩15~30min,取上清液真空浓缩,冷冻干燥后即得肽亚铁螯合物;鱼皮胶原蛋白肽螯合亚铁的螯合率较高,螯合物较稳定,不仅避免了无机铁盐稳定性差、生物利用率低等缺点,易于被植物体吸收,为植物体提供稳定的亚铁离子,可加速植物体叶绿素的合成,同时鱼皮胶原蛋白肽可以补充机体所需氨基酸,是一种高效的亚铁补剂与氨基酸补剂。
作为对本发明的进一步优化,复配助剂中含有下列含量的物质:60~78%的抗坏血酸、15~20%的苹果酸与2~10%的D-苯丙胺;螯合反应过程中,D-苯丙胺与抗坏血酸可以在极大程度上与螯合体系中的氧化剂发生反应,从而降低了氧化剂将二价铁氧化成褐色化三氧化二铁沉淀的占比,保持螯合体系中二价铁离子的充足含量;另外,在苹果酸存在的条件下,D-苯丙胺可以与螯合环境中的S2-离子产生配位络合并生成稳定的配合物,降低了螯合环境中的S2-含量,避免了S2-与稳定螯合物中的Fe2+发生反应,从而防止稳定螯合物的解离,有助于鱼皮胶原蛋白肽螯合亚铁复配物的生成,提高鱼皮胶原蛋白肽螯合亚铁复配物的含量与纯度,增大了可为植物体提供的稳定的亚铁离子的量,大大加快植物体叶绿素的合成,促进植物破坏细胞快速生长。
与现有技术相比,本发明的优点在于:
1)生长调节剂中特殊配比的(R,R)-2,5-二甲基硼咾烷、(S,S)-2,5-二甲基硼咾烷可以刺激植物细胞超氧化物歧化酶的相关基因的表达,加速酶的分泌,提高酶的活性,平衡因高温胁迫造成的植物对活性氧自由基的清除能力和对细胞膜的保护作用,保护细胞膜免受伤害,维持膜内外电解质平衡,从而抑制内源乙烯和脱落酸的大量释放,降低植物果柄离层区因为纤维素酶和过氧化物酶活性,促进破坏细胞的修复与生长,降低落花落果量;
2)鱼皮胶原蛋白肽与亚铁离子螯合反应过程中,D-苯丙胺与抗坏血酸可以在极大程度上与螯合体系中的氧化剂发生反应,从而降低了氧化剂将二价铁氧化成褐色化三氧化二铁沉淀的占比,保持螯合体系中二价铁离子的充足含量;
3)螯合反应时,在苹果酸存在的条件下,D-苯丙胺可以与螯合环境中的S2-反应生成稳定的配合物,降低了螯合环境中的S2-含量,避免了S2-与稳定螯合物中的Fe2+发生反应,从而防止稳定螯合物的解离,有助于鱼皮胶原蛋白肽螯合亚铁复配物的生成,提高鱼皮胶原蛋白肽螯合亚铁复配物的含量与纯度;
4)将生长调节剂通过喷施、涂抹于植物表面后,会在植物表面形成一层调节剂水凝胶薄膜,薄膜不仅对植物表面具有隔水、防蒸作用,降低植株表面的水份蒸发,同时还可使调节剂水凝胶薄膜内的生长调节剂缓慢释放,使调节剂持久的发挥减蒸作用,同时还可以有效的加速破坏细胞生长的作用,加速细胞分裂,避免细胞坏死现象持续恶化,可以高效、持续的促进植物破坏细胞快速生长。

实施例1:
促进植物破坏细胞快速生长的调节剂,其制备方法包括以下步骤:
1)将15份羧甲基壳聚糖与850份去离子水混合,磁力搅拌至羧甲基壳聚糖完全溶解;另取1.65份1-乙基-(3-二甲基氨基丙基)碳二亚胺盐酸盐、1.26份N-羟基琥珀酰亚胺加入到560份二甲基亚砜中,磁力搅拌并完全溶解;在22℃温度下,将二甲基亚砜溶液逐滴加入到羧甲基壳聚糖水溶液中并持续反应4h,产物透析18h即得水凝胶;2)称取0.1份布油菜素内酯、0.3份肽亚铁螯合物、0.8份硼砂、0.02份细胞分裂素、0.04份赤霉素、0.02份6-BA与2份尿素,依次将上述物质溶解于200份8.8%的黄腐酸溶液中,然后再加入0.2份稀土化合物、0.02份2,5-二甲基硼咾烷、2份复合氨基酸,搅拌并混合均匀即得生长调节剂;3)在暗光、1℃温度条件下,将生长调节剂缓慢倒入羧甲基壳聚糖水凝胶中,边倒入边以80r/min的低转速搅拌羧甲基壳聚糖水凝胶溶液,混合均匀后即得促进植物破坏细胞快速生长的调节剂;将调节剂通过喷施、涂抹于植物表面后,会在植物表面形成一层调节剂水凝胶薄膜,薄膜不仅对植物表面具有隔水、防蒸作用,降低植株表面的水份蒸发,同时还可使调节剂水凝胶薄膜内的生长调节剂缓慢释放,使调节剂持久的发挥减蒸作用,同时还可以有效的加速破坏细胞生长的作用,加速细胞分裂,避免细胞坏死现象持续恶化,可以高效、持续的促进植物破坏细胞快速生长。
实施例2:
促进植物破坏细胞快速生长的调节剂,其制备方法包括包括以下步骤:
1)制备水凝胶:将18份羧甲基壳聚糖与980份去离子水混合,磁力搅拌至羧甲基壳聚糖完全溶解;另取1.78份1-乙基-(3-二甲基氨基丙基)碳二亚胺盐酸盐、1.32份N-羟基琥珀酰亚胺加入到580份二甲基亚砜中,磁力搅拌并完全溶解;在23℃温度下,将二甲基亚砜溶液逐滴加入到羧甲基壳聚糖水溶液中并持续反应6h,产物透析24h即得水凝胶;水凝胶均匀度良好,而且其水溶性、胶凝性、促渗透性、生物相容性和可降解性均较为优异,可以作为生长调节剂的载体,而且水凝胶能够吸收大量的水份,在植物表皮形成一层隔膜,大大降低植物叶片表面的蒸腾作用,有效缓解植物的缺水、蒸发而造成的失水现象;
2)制备生长调节剂:称取0.12份布油菜素内酯、2.5份肽亚铁螯合物、1.6份硼砂、0.5份细胞分裂素、0.06份赤霉素、0.05份6-BA与3份尿素,依次将上述物质溶解于230份9.2%的黄腐酸溶液中,然后再加入0.3份稀土化合物、0.025份2,5-二甲基硼咾烷、10份复合氨基酸,搅拌并混合均匀即得生长调节剂;生长调节剂能缩小气孔开张度,减少水分蒸腾,使植株和土壤保持较多的水分,通过物理化学作用与农药形成农药-黄腐酸复合物,既可降低农药毒性、减少农药用量、提高农药药效、提高对人畜的安全性,又可减少环境污染,络合微量元素,提高植物对微量元素的吸收与运转能力,同时还可有效促进细胞生长,加速细胞分裂,促进叶绿素合成,延长黄化时间,避免植物枯萎;

复配助剂中含有下列含量的物质:78%的抗坏血酸、15%的苹果酸与7%的D-苯丙胺;螯合反应过程中,D-苯丙胺与抗坏血酸可以在极大程度上与螯合体系中的氧化剂发生反应,从而降低了氧化剂将二价铁氧化成褐色化三氧化二铁沉淀的占比,保持螯合体系中二价铁离子的充足含量;另外,在苹果酸存在的条件下,D-苯丙胺可以与螯合环境中的S2-离子产生配位络合并生成稳定的配合物,降低了螯合环境中的S2-含量,避免了S2-与稳定螯合物中的Fe2+发生反应,从而防止稳定螯合物的解离,有助于鱼皮胶原蛋白肽螯合亚铁复配物的生成,提高鱼皮胶原蛋白肽螯合亚铁复配物的含量与纯度,增大了可为植物体提供的稳定的亚铁离子的量,大大加快植物体叶绿素的合成,促进植物破坏细胞快速生长。
本发明操作步骤中的常规操作为本领域技术人员所熟知,在此不进行赘述。

Claims (9)
1.促进植物破坏细胞快速生长的调节剂,其特征在于:所述调节剂的制备方法包括:
制备水凝胶:将羧甲基壳聚糖完全溶解于去离子水中,将1-乙基-(3-二甲基氨基丙基)碳二亚胺盐酸盐、N-羟基琥珀酰亚胺完全溶解于二甲基亚砜中,将二甲基亚砜溶液逐滴加入到羧甲基壳聚糖溶液中并持续反应,产物透析即得水凝胶;
制备生长调节剂:称取布油菜素内酯、肽亚铁螯合物、硼砂、细胞分裂素、赤霉素、6-BA与尿素,依次将上述物质溶解于黄腐酸溶液中,然后再加入稀土化合物、2,5-二甲基硼咾烷、复合氨基酸,搅拌并混合均匀即得生长调节剂;
复配:将生长调节剂缓慢倒入羧甲基壳聚糖水凝胶中,边倒入边低转速搅拌羧甲基壳聚糖水凝胶溶液,混合均匀后即得促进植物破坏细胞快速生长的调节剂。
2.根据权利要求1所述的促进植物破坏细胞快速生长的调节剂,其特征在于:所述制备水凝胶步骤中,羧甲基壳聚糖的重量份为15~18份,去离子水为850~980份,1-乙基-(3-二甲基氨基丙基)碳二亚胺盐酸盐为1.65~1.78份,N-羟基琥珀酰亚胺为1.26~1.32份,二甲基亚砜为560~580份。
3.根据权利要求1所述的促进植物破坏细胞快速生长的调节剂,其特征在于:所述制备水凝胶步骤中,二甲基亚砜溶液与羧甲基壳聚糖水溶液的反应温度是22~23℃,反应时间为4~6h。
4.根据权利要求1所述的促进植物破坏细胞快速生长的调节剂,其特征在于:所述制备生长调节剂步骤中,布油菜素内酯的重量份为0.1~0.12份,肽亚铁螯合物为0.3~2.5份,硼砂为0.8~1.6份,细胞分裂素为0.02~0.5份,赤霉素为0.04~0.06份,6-BA 为0.02~0.05份,尿素为2~3份,黄腐酸溶液的质量分数为8.8~9.2%、重量份为200~230份,稀土化合物重量份为0.2~0.3份,2,5-二甲基硼咾烷为0.02~0.025份,复合氨基酸为2~10份。
5.根据权利要求1或4所述的促进植物破坏细胞快速生长的调节剂,其特征在于:所述制备生长调节剂步骤中,2,5-二甲基硼咾烷中含有1/50~1/45的(R,R)-2,5-二甲基硼咾烷。
6.根据权利要求1或4所述的促进植物破坏细胞快速生长的调节剂,其特征在于:所述制备生长调节剂步骤中,肽亚铁螯合物的制备方法为:将经酶解后的鱼皮胶原蛋白肽溶液经低温干燥后得到肽液,加入1.3~1.5倍的浓度为0.6~0.9M的FeCl2溶液,再加入胶原蛋白肽溶液干物重量0.1~0.3‰的复配助剂,震荡螯合,将螯合液离心浓缩取上清液真空浓缩,冷冻干燥后即得肽亚铁螯合物。
7.根据权利要求6所述的促进植物破坏细胞快速生长的调节剂,其特征在于:所述肽亚铁螯合物的制备方法中,复配助剂中含有下列含量的物质:60~78%的抗坏血酸、15~20%的苹果酸与2~10%的D-苯丙胺。
8.根据权利要求6所述的促进植物破坏细胞快速生长的调节剂,其特征在于:所述肽亚铁螯合物的制备方法中,震荡螯合的条件为:pH值6.8~7.0,温度25~28℃,震荡螯合时间45~60min。
9.根据权利要求1所述的促进植物破坏细胞快速生长的调节剂,其特征在于:所述复配步骤中,生长调节剂倒入羧甲基壳聚糖水凝胶中的环境条件为:暗光、1~2℃温度条件下。
