Abstract
一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备及其应用,属于医药技术领域。由控制器、波长范围650nm‑780nm的LED矩阵和固定带构成,所述控制器底端的输出与波长范围650nm‑780nm的LED矩阵通过输出线连接,所述固定带固定安装在LED矩阵的两侧。本发明临床安全性更高,使用更简单方便,且费用低,通过近红外光刺激中枢神经束和外周血液,促进免疫系统加快免疫细胞的分裂和激活休眠T细胞,增加T细胞总量,并且刺激可以激发CD4+T和TM细胞线粒体信号表达上调后进行自我侦测,进而针对HIV致DNA变异的细胞发出凋亡指令。

技术领域
本发明属于医药技术领域,具体属于一种近红外LED设备提高HIV患者CD4+T细胞和有效降低未服用抗病毒药物患者外周血病毒载量的设备。
背景技术
人类免疫缺陷病毒(HIV),即艾滋病病毒,是造成人类免疫系统缺陷的一种病毒,它把人体免疫系统中最重要的CD4+T淋巴细胞作为主要攻击目标,大量破坏该细胞,使人体丧失免疫功能,这个时候,人体就会感染上机会性感染病,如肺炎、脑膜炎、肺结核等。一旦感染发生,则被确认为患了艾滋病。
当HIV病毒进入人体后,由于病毒可以识别出CD4+T淋巴细胞表面的特异蛋白质分子。因此也就可以识别并侵入CD4+T淋巴细胞,使CD4+T淋巴细胞遭受破坏而凋亡,而CD8+T淋巴细胞可以消灭受感染的细胞,也包括受HIV病毒感染的CD4+T淋巴细胞。未受HIV病毒感染的人体,CD4和CD8的比值通常大于1,而当HIV感染人体后,出现CD4+T淋巴细胞进行性减少,导致身体免疫系统对艾滋病毒的长期反应CD8+T一直保持高位,CD4/CD8细胞比值倒置,细胞免疫功能紊乱。
目前,对HIV患者的治疗普遍采用抗逆转录酶病毒药物联合疗法(鸡尾酒疗法),该方法同时作用于HIV-1的逆转录酶和蛋白酶,因此强有力地抑制体内HIV-1的复制,使得CD4细胞计数维持正常,CD4/CD8细胞增高,淋巴结构保持完整,但该方法存在严重的毒副作用,并且服用这些药物程序繁复,价格昂贵,常常会出现诸如乏力、头痛、恶心等不良反应。当代用这种方法还不能彻底根治HIV病毒,人们希望能有一种副作用更小,使用更简便、更安全且根治希望更大的HIV治疗方法。
发明内容
本发明的目的是针对上面所述疾病成因和治疗缺陷,提供一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备,该理疗设备使用方法简单、安全,能使CD4+T淋巴细胞大幅提升并趋于正常的中值,CD4/CD8的比值也有较大幅度提高。
本发明的另一目的是一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备的应用。
本发明的目的是通过以下技术方案予以实现的。
一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备,其特征在于:由控制器、波长范围650nm-780nm的LED矩阵和固定带构成,所述控制器底端的输出与波长范围650nm-780nm的LED矩阵通过输出线连接,所述固定带固定安装在LED矩阵的两侧。
所述固定带的两端安装有相互匹配的魔术贴。
所述LED矩阵内LED的波长为单一波长或两个波长的复合使用。
所述LED矩阵内的LED单一波长为730nm。
所述LED矩阵内的LED两个波长的复合使用为730nm+660nm。
所述LED矩阵内LED的数量为24-106粒。
所述单个LED的光空间分布要求严格控制在15°以内。
所述控制器上安装有显示屏、调节按钮、选择按钮和暂停按钮,顶端设有接口连接直流15V适配器或15V电池。
所述控制器内安装有1.2Khz脉冲发生器,发送准方波,15V电源提供9V稳压分别对准方波和驱动一供电,使准方波通过驱动一驱动730nm的矩阵进行工作。
所述控制器内安装有1.2Khz脉冲发生器,发送锯齿波,15V电源提供14.5V稳压分别对锯齿波和驱动二供电,使锯齿波通过驱动二驱动730nm+660nm的矩阵进行工作。
一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备的应用,其特征在于:将730nm LED矩阵紧贴颈椎部位(施加300~500g压力),LED矩阵的四分之一压住发际线,第一次使用连续用15天,每天二次,每次90-180分钟;15天后每休息5天再连续使用5天,每天二次,每次60-120分钟,连续使用180天。
或者同时将730nm+660nm矩阵紧贴整个腹腔部位(施加300~500g压力),若不能覆盖整个腹腔,则间隙(可在腹部)移动LED矩阵至整个腹腔或使用多个LED矩阵覆盖(每个部位不得少于30分钟),每连续使用5天休息5天,连续使用180天,每天一次,每次30-60分钟。
经研究发现,HIV病毒除了可以在免疫系统中分布外还可以在肠道、生殖系统和神经系统分布。一旦艾滋病病毒藏身到记忆T细胞核的DNA中,它的命运就与记忆T细胞休戚相关,细胞活着,病毒也就活着,细胞死亡,病毒也就死亡。病毒增殖也完全依赖于细胞的分裂,但复制的是DNA改变后的记忆T细胞。前面提到的CD4和CD8T细胞都会分化形成各自的记忆T细胞。
本发明通过近红外光刺激中枢神经束和外周血液,促进免疫系统加快免疫细胞的分裂和激活休眠T细胞,增加T细胞总量;另外近红外光刺激可以激发CD4+T和TM细胞线粒体信号表达上调后进行自我侦测,进而针对HIV致DNA变异的细胞发出凋亡指令(线粒体是生物细胞中负责能量供给、调控细胞存亡的重要细胞器),HIV病毒的暴露很容易被免疫系统识别和消灭。
为大脑供血的动、静脉集中通过颈椎附近部位,给光照带来方便。近红外光直接作用于流经的外周血液,通过LED光子发射和接收团之间发生能量转移,刺激外周血中的免疫细胞(主要是T淋巴细胞)线粒体诱导各种细胞因子,趋化因子信号表达上调,使细胞达到自身最大的生物学潜力。
获得外周血病毒抑制是治疗成功的第一步。当本发明将近红外发光矩阵作用于颈椎部位时,人体中枢神经纤维束全部暴露在近红外光内,在光的刺激下会释放特定的神经递质,神经递质经扩散引发突触厚膜电位变化,产生一系列神经冲动,多极神经元将冲动从中枢传至肌肉(全身组织)或腺体等效应器,激活第二信使系统蛋白,协调诸如突触中神经递质的释放,干涉相应部位产生生理反应,完成对免疫系统的信号上调,激发免疫祖、原细胞加速分裂,生成更多的免疫细胞,同时也激活部分休眠的T细胞分化、成熟(激活T细胞应放在促生成T细胞前)。当这类神经递质传递到身体各组织细胞时,同样也会激发组织细胞线粒体的自我侦测,暴露感染了HIV病毒的记忆T细胞,引导免疫细胞对其攻击。
相较于目前的抗逆转录酶病毒药物治疗方法,本发明的临床安全性更高,使用更简单方便,且费用低,动物试验表明:经8天近红外LED光刺激颈椎,可有效降低外周血IL-4,提高γ-IFN/IL-4的比值,γ-IFNR的提高可提高记忆CD8+T细胞的形成。(γ-IFN:γ-干扰素是I型辅助T细胞(Th1细胞)的标志性的细胞因子,γ-干扰素具有抗病毒、免疫调节及抗肿瘤特性,可以结合到γ-干扰素受体IFNGR。
因此降低外周血IL-4,提高γ-IFN/IL-4的比值,是清除体内感染的HIV-1靶细胞有效方式。
经小范围HIV感染者试验,某种近红外LED光作用于颈椎部位20天左右可升高患者CD4+T细胞23%~140%,CD8+T细胞55%~98%,其在短时间提升患者白细胞(主要是T细胞)的能力显现。本次试验中有一例患者没有服用抗逆转录药物,三个月的试验期,患者病毒载量从5万多下降到3千左右,下降了94.04%。普通血常规试验也表明七天的颈椎近红外光刺激,淋巴细胞绝对值增加12.78%~14.80%;白细胞总量增加12.60%~26.0%。
本发明可以增加近红外LED与抗逆转录病毒药物共同使用以增强抑制病毒的复制和杀灭作用。

实施例1。
一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备,其特征在于:由控制器1、波长范围650nm-780nm的LED矩阵2和固定带3构成,所述控制器1底端的输出与波长范围650nm-780nm的LED矩阵2通过输出线连接,所述固定带3固定安装在LED矩阵2的两侧。
所述固定带3的两端安装有相互匹配的魔术贴4。
所述LED矩阵2内LED的波长为单一波长或两个波长的复合使用。
所述LED矩阵2内的LED单一波长为730nm。

下面是实施例1所述提高HIV患者CD4+T淋巴细胞的近红外LED理疗设备的实验报告,本次实验总共四位患者,二位年龄在35岁以下,二位年龄在65岁以上,其中三位患者正在服用抗逆转录病毒药物,一位65岁女性患者未服用抗逆转录病毒药物,详见表1。

注:在试验开始的CD4检验前,D、H、W 对LED已用了二、三天,只有L姓患者在27日检查后才开始使用。
D男:自2012年检查出HIV感染后CD4+T细胞一直在216~388之间波动,CD4+T细胞最高值388个/μl出现在2014年3月,使用抗逆转录药物效果不明显,在患者7月至8月使用近红外LED光疗后CD4+T细胞上升了23%,CD8+T细胞上升了42%,提高T细胞的作用还是比较明显。3个月后T细胞回调原因不明,需要进一步观察(事后该患者主治医师介绍该患者从事房屋装修工程,走动频繁,未按时使用)。
L男:自2015年检查出HIV感染后CD4+T细胞一直在214~296之间波动,CD4+T细胞最高值296个/μl出现在2016年3月,试验前仅有214,抗逆转录药物几乎没能提高患者的CD4+T细胞,在患者7月至8月使用近红外LED光疗后CD4+T细胞上升了140%,CD8+T细胞上升了55%,作用明显。
H男:自2015年检查出HIV感染后CD4+T细胞在抗逆转录药物支持下一直处于上升状况,药效明显,患者7月至8月在使用近红外LED光疗后CD4+T细胞略有下调,随后三个月又回复到前期高点附近,建议继续观察。猜测由于年龄原因,胸腺严重萎缩,短时间内小剂量近红外光疗作用难于体现。后期试验应使用更大的LED矩阵,光疗时间适当延长。
以上三位患者的CD3、CD8+T细胞均有明显提升。
W女:自2015年检查出HIV感染后CD4+T细胞一直处于缓慢下降通道,在LED支持下虽然CD4+T提高不见起色,但病载从5万下降到3千。

1.一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备,其特征在于:由控制器(1)、波长范围650nm-780nm的LED矩阵(2)和固定带(3)构成,所述控制器(1)底端的输出与波长范围650nm-780nm的LED矩阵(2)通过输出线连接,所述固定带(3)固定安装在LED矩阵(2)的两侧。
2.根据权利要求1所述的一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备,其特征在于:所述固定带(3)的两端安装有相互匹配的魔术贴(4)。

6.根据权利要求1所述的一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备,其特征在于:所述控制器(1)上安装有显示屏(9)、调节按钮(5)、选择按钮(6)和暂停按钮(7),顶端设有接口连接直流15V适配器或15V电池(8)。
7.根据权利要求3所述的一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备,其特征在于:所述控制器(1)内安装有1.2Khz脉冲发生器,发送准方波,15V电源提供9V稳压分别对准方波和驱动一供电,使准方波通过驱动一驱动730nm的矩阵进行工作。
8.根据权利要求3所述的一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备,其特征在于:所述控制器(1)内安装有1.2Khz脉冲发生器,发送锯齿波,15V电源提供14.5V稳压分别对锯齿波和驱动二供电,使锯齿波通过驱动二驱动730nm+660nm的矩阵进行工作。
9.一种提高HIV患者CD4+T淋巴细胞并降低病载的近红外LED理疗设备的应用,其特征在于:将730nmLED矩阵紧贴颈椎部位,施加300~500g压力,LED矩阵的四分之一压住发际线,第一次使用连续用15天,每天二次,每次90-180分钟;15天后每休息5天再连续使用5天,每天二次,每次60-120分钟,连续使用180天;
或者同时将730nm+660nm矩阵紧贴整个腹腔部位,施加300~500g压力,若不能覆盖整个腹腔,则间隙移动LED矩阵至整个腹腔或使用多个LED矩阵覆盖,每个部位不得少于30分钟,每连续使用5天休息5天,连续使用180天,每天一次,每次30-60分钟。
