Abstract

Description
技术领域
本发明涉及牙膏组合物,其特别涉及具有用于增强牙齿再矿化的氟化物源的牙膏组合物。
背景技术
龋齿(Dental caries)或牙龋洞(Dental cavity)是常见的口腔护理问题,更通常被称为蛀牙(tooth decay)。龋齿是由牙釉质的去矿化引起的。
口腔前庭中的细菌分解食物,并且在此过程中产生酸,酸侵蚀牙釉质,最终导致龋齿。构成牙釉质的钙和磷酸盐的化合物是羟基磷灰石的改性形式,其易受酸侵蚀。唾液通过中和这些酸和牙齿上的唾液沉积物中的矿物质以使牙釉质再矿化,在一定程度上防止牙釉质的腐蚀。只要去矿化速率和再矿化速率保持平衡,则牙齿保持强健。这一过程中的不平衡会导致形成龋齿,当牙齿失去的矿物质比可替代的矿物质更多时,就会发生龋齿。
氟化物是已知的抗龋齿剂。氟化物通过与羟基磷灰石相互作用以形成较不易受酸侵蚀的更强的化合物,有助于减缓去矿化。氟离子部分地对羟基磷灰石加氟(fluoridate),同时修复晶格不完整并改善再矿化过程。氟化物作用的有效性取决于可用于沉积在釉质上的氟离子的量。因此,期望配制牙膏组合物,其在使用牙膏组合物形成的刷洗溶液中提供最大的氟离子可用性。
含氟化物源的牙膏是本领域已知的。
US4455293 B(Harvey等,1984)公开了一种稳定的洁齿剂配制物,该洁齿剂配制物具有包含碱金属铝硅酸盐的抛光剂和稳定量的单氟磷酸根离子。
EP0102695 A2(Advanced Research&Technology Institute,1985)公开了含有煅烧高岭土的组合物具有更好的流变性质。该出版物中所公开的组合物含有煅烧高岭土、滑石和二氧化钛作为主要成分。所述组合物还含有氟化物源。
US4064231 A(Kao公司,1977)公开了包含水溶性氟化物和0.3-13重量%的具有特定组成的蒙脱石或锂蒙脱石的洁齿剂组合物导致洁齿剂中水溶性氟化物的浓度随着时间的推移而降低。
US4122163 A(Advanced Research&Technology Institute,1978)公开了新的且更有效的洁齿剂制备物,其含有用于磨耗的煅烧高岭土(1:1粘土)和氟化物源。示例性配制物含有作为2:1粘土的和作为研磨剂的柠檬酸三钠。
在牙膏组合物掺入氟化物源是有挑战性的。递送高水平氟化物的主要限制是由于氟化物离子会被牙膏组合物中的其它成分、特别是研磨组分失活并且变得不可用的倾向。虽然已知研磨剂与氟化物源在不同程度上相容,但在相容性上有很大差异。含钙研磨剂不是特别相容的。虽然不含钙研磨剂的相容性稍好些,但此类研磨剂抛光釉质的能力较差。
因此期望改善牙膏组合物中氟化物源的性能,并且这向牙齿递送最大益处,而不损害从牙膏组合物可获得的其它益处。
因此,本发明的第一目的是提供一种牙膏组合物,其中掺入的氟化物产生更高级别(higher order)的再矿化。
本发明的再一目的是提供一种牙膏组合物,其中掺入的氟化物呈容易获得的形式。
发明内容
本发明人惊讶地发现,当具有氟化物源的牙膏组合物包含研磨剂二氧化硅或基于钙的研磨剂、1:1层状硅酸盐粘土和2:1层状硅酸盐粘土时,所述组合物可提供改善的牙齿再矿化。
所得牙膏组合物不仅显示出在产品首次配制时增加的氟化物可用性,而且还显示出在储存期间对氟化物可用性降低的显著耐受性。
为了本发明的目的,"牙膏"意指与牙刷一起使用的膏糊或凝胶组合物。尤其优选适于通过刷洗约两分钟来清洁的牙膏组合物。
如本文所用的术语1:1层状硅酸盐粘土是指两层型粘土,其中片状结构由一层二氧化硅四面体和一层氧化铝八面体的单元构成。
如本文所用的术语2:1层状硅酸盐粘土是指三层型粘土,其中片状结构由两层二氧化硅四面体和优选为二八面体层或三八面体层的一层中心八面体层构成。
根据第一方面,公开了一种牙膏组合物,其具有:
(i)2-70重量%的研磨剂;
(ii)氟化物源;
(iii)1:1层状硅酸盐粘土;和
(iv)2:1层状硅酸盐粘土,

发明详述
根据第一方面,公开了一种牙膏组合物,其具有研磨剂、氟化物源、2:1层状硅酸盐粘土和1:1层状硅酸盐粘土。
研磨剂
所公开的牙膏组合物包含2-70重量%的研磨剂,其中所述研磨剂是研磨剂二氧化硅或基于钙的研磨剂。
根据本发明使用的研磨剂是微粒研磨剂材料,如二氧化硅、碳酸钙、磷酸二钙、焦磷酸钙、羟基磷灰石或其混合物。还可使用团聚的微粒研磨剂材料。
透明或半透明的凝胶通常含有二氧化硅,而不透明的乳膏通常含有基于钙的研磨剂,尤其是白垩。研磨剂的量需要有一些控制,因为过量的研磨剂引起更大磨耗。此外,不受控量的研磨剂也会不利地影响牙膏组合物的粘度。
研磨剂优选以组合物的10-70重量%存在。优选地,研磨剂的量是所述组合物的20重量%、更优选25重量%、仍更优选30重量%、进一步优选40重量%且最优选45重量%。所公开的发明中所用的研磨剂进一步优选基于钙的。优选所公开的组合物包含40-60重量%、更优选45-55重量%的基于钙的研磨剂。
特别优选的基于钙的研磨剂是碳酸钙。精细研磨的天然白垩(FGNC)(一种白垩形式)是高度优选的。FGNC还可在研磨期间或在研磨之后通过热处理由涂层进行化学或物理改性。典型的涂层材料包括硬脂酸镁和油酸镁。还可通过使用各种研磨技术,例如球磨、空气分级器研磨或螺旋喷射研磨,在研磨加工期间改变FGNC的形态。FGNC可用作唯一的基于钙的研磨剂。然而,FGNC还可与其它基于钙的研磨剂一起使用以平衡磨耗。通常白垩的粒径为1至60微米,且优选的大小范围为1至15微米。其它优选的基于钙的研磨剂包括磷酸二钙(DCP)、焦磷酸钙和沉淀碳酸钙(PCC)。当使用基于钙的研磨剂的组合时,优选FGNC为全部基于钙的研磨剂的35-100%、更优选75-100%且尤其是95-100%。在此情况下,余量最优选为PCC(沉淀碳酸钙)。
取决于想要的磨耗程度,其它研磨剂也可任选地与基于钙的研磨剂一起使用。这些包括合成研磨剂抛光剂,如碳酸镁、偏磷酸钠、偏磷酸钾、硅酸锆、偏磷酸钾、正磷酸镁、正磷酸镁、磷酸三镁、硅酸铝、硅酸锆和珍珠岩。
氟化物源
所公开的牙膏组合物包含氟化物源。
优选地,氟化物源是提供氟化物离子的源的可溶性氟化物盐。可使用任何口腔可接受的氟化物盐或盐的组合。实例包括但不限于氟化亚锡、氟化钠、氟化钾、单氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺、氟化铵和其组合。优选地,氟化物源是氟化亚锡、氟化钠、单氟磷酸钠和其混合物。优选的是所述氟化物源是单氟磷酸钠。
取决于应用,根据本发明的牙膏组合物可包含在以所述组合物的重量计1,000至20,000ppm范围内的游离氟离子。优选地,用于一般消费者使用的牙膏组合物中的游离氟离子浓度可以通常在以所述组合物的重量计1000至15000ppm的范围内。优选将氟化物源以组合物的约0.01-10重量%、更优选0.03-5重量%、仍更优选0.1-2重量%且最优选0.15-1重量%的水平添加至所公开发明的组合物中。氟化物源(其通常为氟化物盐,以提供适当水平的游离氟离子)的重量可以依赖于所述盐中抗衡离子的重量而变化。
1:1层状硅酸盐粘土
所公开的牙膏组合物包含1:1层状硅酸盐粘土。通常1:1层状硅酸盐粘土是非溶胀性粘土。
1:1层状硅酸盐粘土包括高岭石-蛇纹石族粘土。高岭石-蛇纹石族粘土包括高岭石和蛇纹石矿物质的亚族。蛇纹石是具有四面体片和八面体片的三八面体片状矿物质,具有镁与少量的铝,并且该亚族内的种类优选纤蛇纹石、利蛇纹石和叶蛇纹石。
优选地,1:1层状硅酸盐粘土是高岭石粘土。高岭石具有二八面体片,包括在高岭石亚族内的种类是高岭石、地开石、珍珠陶土和埃洛石粘土矿物质,但不限于此。在所公开的发明中特别优选高岭石。
高岭石,通常被称为高岭粘土,是天然存在的矿物质。高岭石可以是煅烧高岭土、高纯化的煅烧高岭土、胶体高岭土或水合高岭土,但不限于此。优选的高岭石是水合硅酸铝并且表示为Al2O3·2SiO2·2H2O。优选的高岭石的粒径分布应使得至少98重量%的粒子具有2μm或更小的粒径。优选地高岭石是精制高岭土,进一步优选地,所述精制高岭土包含38重量%的Al2O3和45重量%的SiO2及最大0.5重量%的Fe2O3。
优选地,1:1层状硅酸盐粘土的10%水溶液具有5-7的pH。
所公开的牙膏组合物包含0.1-20重量%、更优选0.1-5重量%且仍更优选0.1-3重量%且最优选0.1-2重量%的1:1层状硅酸盐粘土。
2:1层状硅酸盐粘土
所公开的牙膏组合物包含2:1层状硅酸盐粘土。通常2:1层状硅酸盐粘土是溶胀性粘土。
优选地,2:1层状硅酸盐粘土选自叶蜡石-滑石族粘土、蒙皂石族粘土、蛭石族粘土,更优选地,2:1层状硅酸盐粘土属于蒙皂石族。
叶蜡石-滑石族优选包括滑石,其为水合硅酸镁;叶蜡石,其为水合硅酸铝;或铁滑石,其为水合硅酸铁。
蛭石族粘土优选包括二八面体蛭石或三八面体蛭石。三八面体蛭石优选具有0.6至0.7的层电荷。
高度优选的是,2:1层状硅酸盐粘土属于蒙皂石族粘土,所述蒙皂石族粘土包括但不限于蒙脱石、蒙皂石、贝得石、绿脱石、皂石、锂蒙脱石、锌蒙脱石或合成锂皂石。蒙皂石族粘土优选二八面体蒙脱石或三八面体蒙脱石。
适合的二八面体蒙脱石的实例包括但不限于蒙脱石(经常被称为膨润土)、蒙皂石、贝得石、绿脱石。
通常蒙脱石和蒙皂石具有0.3至0.6的低总电荷含量,且大部分电荷源于八面体片中。贝得石是可膨胀的二八面体蒙皂石,其具有0.7或更高的总电荷含量和0.4的四面体电荷含量。当二八面体蒙脱石是所述蒙脱石时,其通常不含碳酸盐且优选是水洗的。
适合的三八面体蒙脱石的实例包括但不限于皂石、锂蒙脱石、锌蒙脱石和合成锂皂石。总电荷含量为0.3至0.5的三八面体蒙脱石包括八面体片中具有镁和锂的锂蒙脱石,以及八面体片中具有大量镁和四面体片中的一些铝取代物的皂石。

优选地,2:1层状硅酸盐粘土的10%水溶液具有8至10的pH。
所公开的牙膏组合物包含量为0.1-5重量%、更优选0.1-3重量%且仍更优选0.1-2重量%且最优选0.1-1重量%的2:1层状硅酸盐粘土。
磷酸盐源
优选可用于本发明中的磷酸盐源仅限于该磷酸盐源可用于适合用于口腔的组合物中的范围。适于本发明中的磷酸盐源类型的说明性实例包括磷酸一钠、磷酸二氢钠、磷酸氢二钠、焦磷酸钠、焦磷酸四钠、六偏磷酸钠、磷酸一钾、磷酸二氢钾、磷酸氢二钾、磷酸三钠、磷酸三钾、其混合物等。磷酸盐源优选水溶性的源。通常,磷酸盐源占洁齿剂组合物的0.5-15重量%,更优选2-12重量%,最优选4-9重量%,基于洁齿剂组合物总重量且包括其中所含的所有范围。在最优选的实施方案中,所用磷酸盐源是磷酸三钠和磷酸二氢钠,磷酸三钠与磷酸二氢钠的重量比为1:4-4:1,优选1:3-3:1,最优选1:2-2:1,包括其中所含所有比率。
另外的成分
湿润剂
根据本发明的牙膏优选包含湿润剂,所述湿润剂通常为多元醇,特别是选自甘油、山梨糖醇、丙二醇、木糖醇、麦芽糖醇、乳糖醇或其混合物的多元醇。湿润剂通常包含在牙膏中,用于获得柔软且柔顺的口感。湿润剂还降低牙膏组合物失去水分的倾向。尤其优选山梨糖醇,该材料还用作甜味剂,山梨糖醇通常以70%的水溶液获得。湿润剂的总量通常为总组合物的1-60重量%,特别是5-50重量%,且尤其是10-30重量%。
抗微生物剂
根据本发明的牙膏组合物优选包含抗微生物剂。实例包括三氯生、氯己定、铜、锌和亚锡的盐(如柠檬酸锌、硫酸锌、甘氨酸锌、柠檬酸钠锌、焦磷酸亚锡)、血根碱提取物、甲硝哒唑、季铵化合物(如西吡氯铵)、双胍(如氯己定)、二葡糖酸盐、海克替啶、奥替尼啶、阿来西定;和卤代双酚化合物(如2,2'-亚甲基双-(4-氯-6-溴苯酚))。
表面活性剂
根据本发明的牙膏组合物优选包含表面活性剂。
适合的表面活性剂是那些在较宽pH范围内相当稳定并提供泡沫的表面活性剂。所述表面活性剂是阴离子型的。
可用于本文中的阴离子型表面活性剂包括在烷基中具有8-20个碳原子的水溶性烷基硫酸盐(例如烷基硫酸钠)和具有8-20个碳原子的脂肪酸磺化甘油单酯的水溶性盐。月桂基硫酸钠和椰子基甘油单酯磺酸钠是该类型的阴离子型表面活性剂的实例。
优选的组合物含有0.25-12重量%、更优选0.5-8重量%且最优选1-约6重量%的阴离子型表面活性剂。
一些阴离子型表面活性剂、特别是月桂基硫酸钠本身具有抗细菌作用。此类作用提供一定程度的即时抗细菌效果。然而,这种使用通常是非常短暂的。因此,诸如三氯生的抗菌剂提供更持久的抗细菌作用。
还可包含其它表面活性剂,如非离子型表面活性剂、两性表面活性剂或两性离子型表面活性剂。非离子型表面活性剂可广义地定义为通过烯化氧基团(性质上为亲水性的)与有机疏水化合物缩合产生的化合物,该有机疏水化合物在性质上可以是脂族或烷基芳族的。

调味剂(flavouring agent)
根据本发明的牙膏组合物可有利地采用调味剂。用于本发明实践中的调味剂包括但不限于精油以及各种调味的醛、酯、醇和类似材料。精油的实例包括留兰香油、薄荷油、冬青油、丁香油、鼠尾草油、桉树油、肉桂油、柠檬油、酸橙油和橙油。还可使用的此类化学品如薄荷醇、香芹酮和茴香脑。优选地,调味剂是薄荷油、丁香油、柠檬油和留兰香油。
增稠剂
根据本发明的牙膏组合物可有利地采用增稠剂。所公开的组合物优选含有0-2重量%的增稠二氧化硅,这意味着组合物可含有或可不含有增稠二氧化硅。常规的凝胶牙膏组合物通常含有至多8.5重量%的增稠二氧化硅,而不透明牙膏组合物通常含有4-10重量%。牙膏组合物可包含最高达2重量%的增稠二氧化硅。优选的组合物具有1.5重量%或甚至小于1重量%的增稠二氧化硅。最佳的组合物具有小于0.5重量%的增稠二氧化硅。高度优选的组合物不含增稠二氧化硅。
当存在时,优选的增稠二氧化硅包括来自Degussa的T系列或来自Cabot公司的CAB-O-系列,硅胶,如来自W.R.Grace&Co的或系列,或沉淀二氧化硅,如来自J.M.Huber公司的265。可用的二氧化硅增稠剂还包括165、163和/或167以及153、177和/或265二氧化硅,全部得自J.M.Huber公司。其它优选的增稠二氧化硅包括-P(来自Madhu Silica)、22S和200(来自Evonik Industries)、来自WR Grace&Company的和增稠二氧化硅以及来自Rhodia的43和331、合成细碎的热解二氧化硅,如以商标244、266和D-200出售的那些。
还可添加至所公开的组合物中的其它增稠剂包括一种或多种其它增稠剂,如羧乙烯基聚合物,所述羧乙烯基聚合物包括可以系列(包括934、940、941和956)从B.F.Goodrich商购的卡波姆。其它优选的等级包括丙烯酸酯/丙烯酸C10-30烷基酯交联聚合物(crosspolymer),其可以21、TR-1和TR-2从Noveon公司商购。优选的组合物可包含0.05-10重量%、更优选0.1-5重量%且甚至更优选0.25-约4重量%的其它增稠剂。
脱敏剂(De-sensitising agent)
根据本发明的牙膏组合物可有利地采用脱敏剂,例如选自硝酸钾、氯化钾、柠檬酸钾、酒石酸钾和乙酸钾的钾盐,所述脱敏剂以优选0.5-3重量%、更优选1-2.5重量%且尤其是1.7-2.2重量%使用。
硅酸盐
根据本发明的组合物优选包含碱金属硅酸盐。所述碱金属是钠或钾,优选钠。硅酸钠通常可作为10%至40%水溶液获得,最通常为30%溶液。硅酸钠可以中性硅酸钠或碱性硅酸钠获得。优选的牙膏具有中性硅酸钠。硅酸钠可以不同比率的Na2O:SiO2获得。
Na2O:SiO2比率在3.0至3.8范围内的硅酸钠是优选的,更高度优选的范围是3.25至3.5。优选的牙膏组合物包含0.1-5重量%的硅酸盐(基于干重)。因此,将30%的硅酸钠溶液以0.3-16重量%范围内的量添加至所述组合物中。
粘结剂
根据本发明的组合物优选包含使膏糊具有良好结构的粘结剂。纤维素粘结剂是尤其优选的。优选的纤维素粘结剂包括纤维素醚,其包括羟乙基纤维素(HEC)、羟丙基纤维素(HPC)、乙基羟乙基纤维素(EHEC)、羧甲基纤维素(CMC)、羧甲基羟乙基纤维素(CMHEC)、羟丙基羟乙基纤维素(HPHEC)、甲基纤维素(MC)、甲基羟丙基纤维素(MHPC)、甲基羟乙基纤维素(MHEC)、羧甲基甲基纤维素(CMMC)、疏水改性的羧甲基纤维素(HMCMC)、疏水改性的羟乙基纤维素(HMHEC)、疏水改性的羟丙基纤维素(HMHPC)、疏水改性的乙基羟乙基纤维素(HMEHEC)、疏水改性的羧甲基羟乙基纤维素(HMCMHEC)、疏水改性的羟丙基羟乙基纤维素(HMHPHEC)、疏水改性的甲基纤维素(HMMC)、疏水改性的甲基羟乙基纤维素(HMMHPC)、疏水改性的甲基羟乙基纤维素(HMMHEC)和疏水改性的羧甲基甲基纤维素(HMCMMC)。
其它纤维素粘结剂包括阳离子型羟乙基纤维素(阳离子型HEC)、阳离子型疏水改性的羟乙基纤维素(阳离子型HMHEC)和微晶纤维素。
高度优选的粘结剂是羧甲基纤维素钠(SCMC)。特别优选的羧甲基纤维素钠包括取代度为0.6至0.99、优选0.7至0.95的那些。
除了纤维素粘结剂之外,或作为纤维素粘结剂的替代物,可使用瓜尔胶或其衍生物,然而,此类粘结剂不如纤维素粘结剂优选。此类衍生物包括羧甲基瓜尔胶(CM瓜尔胶)、羟乙基瓜尔胶(HE瓜尔胶)、羟丙基瓜尔胶(HP瓜尔胶)、羧甲基羟丙基瓜尔胶(CMHP瓜尔胶)、阳离子型瓜尔胶、疏水改性的瓜尔胶(HM瓜尔胶)、疏水改性的羧甲基瓜尔胶(HMCM瓜尔胶)、疏水改性的羟乙基瓜尔胶(HMHE瓜尔胶)、疏水改性的羟丙基瓜尔胶(HMHP瓜尔胶)、阳离子型疏水改性的羟丙基瓜尔胶(阳离子型HMHP瓜尔胶)、疏水改性的羧甲基羟丙基瓜尔胶(HMCMHP瓜尔胶)和疏水改性的阳离子型瓜尔胶(HM阳离子型瓜尔胶)。
其它不太优选的胶包括黄原胶、角叉菜胶和衍生物(如爱尔兰苔藓和viscarin)、结冷胶、菌类植物胶和衍生物、短梗霉聚糖、鼠李糖胶(rhamsan gum)、威兰胶、魔芋胶、凝胶多糖、褐藻胶、海藻酸、海藻酸盐和衍生物、磷酸淀粉衍生物、琼脂和衍生物、阿拉伯胶和衍生物、果胶和衍生物、壳聚糖和衍生物、刺梧桐树胶、槐豆胶、纳豆胶、黄蓍胶、几丁质衍生物、明胶、β葡聚糖、糊精、右旋糖酐、环糊精和聚季铵盐、呋喃泡沫树脂胶(furcellarengum)、印度树胶、车前籽胶、温柏树胶、罗望子胶、落叶松胶和塔拉胶。
防腐剂
具有含钙的研磨剂(尤其是白垩)的牙膏组合物易发生细菌生长。某些防腐剂,例如对羟基苯甲酸甲酯、对羟基苯甲酸乙酯、对羟基苯甲酸丁酯、对羟基苯甲酸丙酯和对羟基苯甲酸异丙酯可能对于抵抗细菌生长特别可用。对羟基苯甲酸甲酯、对羟基苯甲酸乙酯、对羟基苯甲酸丁酯和对羟基苯甲酸丙酯的混合物是特别优选的。可通过添加苯氧基乙醇来增强该混合物的活性。甲醛和二甲基乙内酰脲是其它优选的防腐剂。防腐剂通常以0.05-0.8重量%的水平包含在所述组合物中。
甜味剂
根据本发明的组合物还可含有甜味剂。优选的甜味剂包括糖精钠、阿斯巴甜、三氯蔗糖、索马甜、乙酰舒泛钾(acesulfame potassium)、甜菊苷、甜菊提取物、对甲氧基肉桂醛、新橙皮苷二氢查尔酮和紫苏糖。典型的水平是0.005-5重量%、更优选0.01-1重量%。
牙膏组合物
所公开的牙膏组合物可以是透明的、不透明的或半透明的,这取决于所用研磨剂体系的类型。
根据本发明的牙膏组合物优选包含重量比率在1.5至5、更优选1.5至3、仍更优选1.6至3、仍更优选1.6至2且最优选1.8至2范围内的1:1层状硅酸盐粘土与2:1层状硅酸盐粘土。
根据本发明的牙膏组合物优选包含重量比率在1.5至15、更优选1.5至5、仍更优选1.6至5、仍更优选1.6至2且最优选1.8至2范围内的1:1层状硅酸盐粘土与氟化物源。
根据本发明的牙膏组合物优选包含重量比率在1至10、更优选1至6、仍更优选1.05至5、仍更优选1.05至4且最优选1.05至2范围内的2:1层状硅酸盐粘土与氟化物源。
使用牙膏组合物的方法
根据本发明的第二方面,公开了促进牙齿的再矿化的方法,其包括以下步骤:(i)使牙齿与根据第一方面的牙膏组合物接触;和(ii)任选地,用适合的溶剂冲洗口腔。优选所述溶剂为水。
所述方法本质上可以是治疗性的。或者且更优选地,所述方法本质上是非治疗性的或化妆用的。
根据第三方面,公开了1:1层状硅酸盐粘土与2:1层状硅酸盐粘土一起在具有研磨剂二氧化硅或基于钙的研磨剂和氟化物源的牙膏组合物中用于促进牙齿再矿化的用途。
优选所述用途是针对非治疗性目的或化妆目的的。
根据第四方面,公开了1:1层状硅酸盐粘土与2:1层状硅酸盐粘土一起在具有研磨剂二氧化硅或基于钙的研磨剂和氟化物源的牙膏组合物中用于降低牙齿去矿化的用途。
优选该用途是针对非治疗性目的或化妆目的的。
具体实施方式
将在以下非限制性实施例中进一步说明本发明。
实施例
制备具有如表1中所示成分的4种不同牙膏组合物。4种牙膏组合物中,组合物A、B和C是比较组合物,组合物1根据本发明。将表1中所提供的每种牙膏组合物用于如下文所提供的去矿化和再矿化研究。
a)去矿化研究
将完好的牛门齿切牙的唇表面磨平并用氧化铝浆料抛光。从每个牙齿上切下四个区段,通过用耐酸指甲油涂抹其余牙齿区段,在抛光表面上形成测试区域,以提供用于研究的牙齿样品。分析并记录所制备的牙齿样品的微硬度。随后用大量超纯水(Milli-Q water)洗涤牙齿样品,然后进行如下pH循环方案:
●首先,将牙齿样品浸渍在牙膏浆料中5分钟。所述牙膏浆料通过将1份牙膏组合物与3份具有10UI/mL碱性磷酸酶的去离子水混合来制备。
●接下来,将牙齿样品浸渍在酸性缓冲液中1小时。所述酸性缓冲液通过混合50mmol/L乙酸和1.5mmol/L的KH2PO4来制备,该缓冲液pH为5.0。
●此后,将牙齿样品浸渍在中性缓冲液中1分钟。所述中性缓冲液通过混合20mmol/L HEPES和1.5mmol/L KH2PO4来制备,该缓冲液pH为7.0。
●重复该循环12次。
●在每一步骤之后洗涤牙齿样品,以彻底冲洗掉来自在先步骤的活性剂。
●通过在pH-循环之前和之后对牙齿样品进行微硬度分析来评估功效。
●对每个牙齿样品进行4次读数,并将去矿化表示为硬度降低的百分比(HK降低%)。
●每种牙膏组合物的HK降低%结果提供于表1中。
b)再矿化研究
将完好的牛门齿的唇表面磨平并用氧化铝浆料抛光。从每个牙齿上切下四个区段,通过用耐酸指甲油涂抹其余牙齿区段,在抛光表面上形成区域,以提供用于研究的牙齿样品。
将牙齿样品在由等份的8%甲基纤维素和乳酸制备的凝胶体系中在37℃和pH 4.6下去矿化10天。在去矿化后,对每个牙齿样品进行压痕,以获得牙齿样品的硬度努普(Knoop)值。
在损伤(lesion)准备好后,对牙齿样品进行如下pH循环方案。
●首先,将牙齿样品浸渍在牙膏浆料中5分钟。牙膏浆料通过将1份牙膏组合物与3份具有10UI/mL碱性磷酸酶的去离子水混合来制备。
●接下来,将牙齿样品浸渍在酸性缓冲液中30分钟。酸性缓冲液通过混合50mmol/L乙酸、1.5mmol/L二水氯化钙、0.9mmol/L正磷酸二氢钾和130mmol/L氯化钾来制备,该缓冲液pH为5.0。
●此后,将牙齿样品浸渍在中性缓冲液中10分钟。中性缓冲液通过混合20mmol/LHEPES、1.5mmol/L二水氯化钙、0.9mmol/L正磷酸二氢钾和130mmol/L氯化钾来制备,该缓冲液pH为7.0。
●如上文所述,通过在pH-循环方案之前和之后对牙齿样品进行微硬度分析来评估功效。
●对每个牙齿样品进行4次读数,并将再矿化如下表示为恢复的硬度(硬度努普=HK)的%:
HK恢复%=(ΔHK/HK基线)*100
其中ΔHK计算如下:处理后的HK-处理前的HK。

表1中的结果清楚地显示,与比较组合物(A、B、C)相比,同时具有1:1层状硅酸盐粘土和2:1层状硅酸盐粘土的根据本发明的组合物1提供了对牙齿样品的改善的再矿化。根据本发明的组合物1还降低牙齿样品的去矿化水平。
Claims (14)
1.一种牙膏组合物,其包含:
(i)2-70重量%的研磨剂;
(ii)氟化物源;
(iii)1:1层状硅酸盐粘土;和
(iv)2:1层状硅酸盐粘土;
其中所述研磨剂是研磨剂二氧化硅或基于钙的研磨剂。
2.根据权利要求1所述的组合物,其中所述1:1层状硅酸盐是高岭石粘土。
3.根据权利要求2所述的组合物,其中所述高岭石粘土选自高岭石、埃洛石、珍珠陶土和地开石。
4.根据权利要求3所述的组合物,其中所述高岭石是水合硅酸铝。
5.根据权利要求1所述的组合物,其中所述2:1层状硅酸盐粘土来自蒙皂石族粘土。
6.根据权利要求5所述的组合物,其中来自所述蒙皂石族粘土的粘土是蒙脱石、蒙皂石、贝得石、绿脱石、皂石、锂蒙脱石、锌蒙脱石或合成锂皂石。
7.根据权利要求5或6所述的组合物,其中来自所述蒙皂石族粘土的粘土是硅酸镁铝。
8.根据前述权利要求1至7中任一项所述的组合物,其中所述1:1层状硅酸盐粘土以按所述组合物的重量计0.1%至20%的量存在。
9.根据前述权利要求1至8中任一项所述的组合物,其中所述2:1层状硅酸盐粘土以按所述组合物的重量计0.1%至5%的量存在。
10.根据前述权利要求1至9中任一项所述的组合物,其中所述氟化物源选自单氟磷酸钠、氟化钠或氟化亚锡。
11.根据前述权利要求1至10中任一项所述的组合物,其中所述组合物包含钙源。
12.一种促进牙齿再矿化的方法,该方法包括以下步骤:
(i)使牙齿与根据前述权利要求1至12中任一项所述的组合物接触;和
(ii)任选地,用适合的溶剂冲洗口腔。
13.1:1层状硅酸盐粘土与2:1层状硅酸盐粘土一起在包含研磨剂二氧化硅或基于钙的研磨剂以及氟化物源的牙膏组合物中用于促进牙齿再矿化的用途。
14.1:1层状硅酸盐粘土与2:1层状硅酸盐粘土一起在包含研磨剂二氧化硅或基于钙的研磨剂以及氟化物源的牙膏组合物中用于降低牙齿去矿化的用途。
