本发明公开了一种具有高稳定性的薄荷精油乳状液,属于食品加工技术领域。本发明以薄荷精油、白藜芦醇、玉米醇溶蛋白以及果胶为原材料,利用反相溶剂法制备包埋了白藜芦醇的玉米醇溶蛋白‑果胶纳米粒子,再采用高压均质法制备稳定的薄荷精油乳状液。该乳状液对白藜芦醇包埋率可达80%以上,且白藜芦醇和薄荷精油具有协同抑菌效应,同时果胶的加入提高了白藜芦醇的稳定性。本发明提供了一种共包埋精油和两亲性功能因子的方法,同时提供一种增强白藜芦醇稳定性和制备含有白藜芦醇和薄荷精油抑菌剂的方法。该方法操作简便,易于工业化推广。

技术领域
本发明涉及一种具有高稳定性的薄荷精油乳状液,属于食品加工技术领域。
背景技术
由病原微生物引起的食源性疾病在世界范围内都是一个日益严重的卫生问题,天然抗菌剂由于其生物安全性和高效抑菌能力越来越引起人们的关注。
薄荷精油具有抗细菌、真菌、病毒的作用,可作为潜在的天然抑菌剂;白藜芦醇是一种多酚类化合物,具有广谱抗菌性,因此,以薄荷精油以及白藜芦醇作为原材料制备成的天然抗菌剂具有巨大的潜在市场。
然而薄荷精油水溶性差,具有强烈的气味且易挥发;白藜芦醇在水中和油中的溶解度均不高,且易于氧化而降解,大大限制了两者在实际工业生产及食品体系中的应用。
现有技术中,常采用包埋法,解决薄荷精油水溶性差、易挥发以及白藜芦醇溶解度低、易降解的问题。有研究报道利用水包油型乳状液对多种功能因子的包埋多集中在对疏水性功能因子的包埋和保护,但此方法对于两亲性的白藜芦醇包埋率并不高(约为50%);也有研究报道将乳清分离蛋白作预变性处理,并添加钙离子进行交联,虽然提高白藜芦醇的界面包埋率,却加速白藜芦醇的降解。
玉米醇溶蛋白是玉米中主要的贮藏蛋白,其廉价易得,结构中含有大量疏水性氨基酸使其具有独特的自组装特性。
果胶是一种天然的聚阴离子多糖,其主链由1→4糖苷键连接的α-D-半乳糖醛酸单元和以1→2糖苷键连接的α-L-鼠李糖残基构成。果胶分子中含有大量可电离的羧基,因此具有聚阴离子电解质特性。果胶能够与蛋白质在一定条件下形成亲水复合物,可改善蛋白质的环境敏感性。果胶还具有增稠、凝胶等多种功能性质,因此被广泛用于食品加工中。
本发明尝试以薄荷精油、白藜芦醇为原料,玉米醇溶蛋白以及果胶为载体,通过包埋法制备出一种包埋率高的同时稳定性也好的薄荷精油乳状液,但各成分的配比、具体的制备方法以及制备出的薄荷精油乳状液是否具有抗菌能力、其抗菌能力的大小,均需要进一步的研究。
发明内容
为解决上述问题,本发明以薄荷精油、白藜芦醇、玉米醇溶蛋白以及果胶为原材料,通过反相溶剂法制备玉米蛋白-果胶复合粒子,再采用高压均质法制备一种薄荷精油乳状液,此薄荷精油乳状液对白藜芦醇的包埋率可达80%以上且可以显著提高白藜芦醇的储藏稳定性;被包埋后薄荷精油和白藜芦醇显现出协同抑菌效果,此制备方法具有成本低、易于操作的优势。
本发明的技术方案如下:
本发明提供了一种具有高稳定性的薄荷精油乳状液,其包含溶剂、溶质以及乳化剂;所述溶质为薄荷精油;所述乳化剂由玉米醇溶蛋白、白藜芦醇和果胶组成。
在本发明的一种实施方式中,所述溶质薄荷精油在整个薄荷精油乳状液中的浓度为5-50mg/mL。
在本发明的一种实施方式中,所述溶质薄荷精油在整个薄荷精油乳状液中的浓度为50mg/mL。
在本发明的一种实施方式中,所述乳化剂中玉米醇溶蛋白、白藜芦醇和果胶的质量比为10:0.2-1:2.5-10。
在本发明的一种实施方式中,所述乳化剂中玉米醇溶蛋白、白藜芦醇和果胶的质量比为10:1:5。
在本发明的一种实施方式中,所述乳化剂中的玉米醇溶蛋白在整个薄荷精油乳状液中的浓度为2-5mg/mL。

本发明提供了一种具有高稳定性的薄荷精油乳状液的制备方法,包含如下步骤:
步骤1:将玉米醇溶蛋白和白藜芦醇按照一定的比例溶解于乙醇溶液,然后将溶有玉米醇溶蛋白和白藜芦醇的乙醇溶液加入水中,获得玉米醇溶蛋白-白藜芦醇纳米粒子;
步骤2:将步骤1所得的玉米醇溶蛋白-白藜芦醇纳米粒子加入果胶溶液中,获得玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子;
步骤3:在步骤2所得的玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子中加入薄荷精油,进行均质包埋处理,得到具有高稳定性的薄荷精油乳状液。
在本发明的一种实施方式中,所述步骤1中的玉米醇溶蛋白和白藜芦醇的质量比为10:0.2-1。

在本发明的一种实施方式中,所述步骤3为在步骤2所得的玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子中加入薄荷精油,然后在转速为8000rpm的条件下进行高速剪切2min,最后在压力为250Bar的条件下进行均质1min,得到具有高稳定性的薄荷精油乳状液。
本发明提供了上述一种具有高稳定性的薄荷精油乳状液的制备方法制备得到的薄荷精油乳状液。
本发明提供了上述一种具有高稳定性的薄荷精油乳状液或上述一种具有高稳定性的薄荷精油乳状液的制备方法制备得到的薄荷精油乳状液在制备食品、保健品、化妆品、医药配制品以及抗菌剂中的应用。

有益效果:
(1)采用本发明方法制备的薄荷精油乳状液能够同时包埋薄荷精油和白藜芦醇,薄荷精油包埋率可达87%,白藜芦醇的包埋率可达到80%以上;
(2)采用本发明方法制备的薄荷精油乳状液有显著的协同抑菌作用,该乳状液在稀释100倍后对金黄色葡萄球菌和鼠伤寒沙门氏菌的抑菌能力分别最高可达4.18和4.71个对数,相比于单独使用薄荷精油,抑菌能力分别提高了1.02和1.38个对数;
(3)采用本发明方法制备的薄荷精油乳状液贮藏稳定性显著提高,在25℃下储藏42天后,白藜芦醇残留率为81%,在未添加果胶的乳状液中白藜芦醇残留率为52%;在乳清分离蛋白稳定的乳状液中,在25℃下储藏25天后,白藜芦醇的残留率最高为34%;
(4)本发明采用的制备方法成本低,方法简便,易于工业推广应用。
(5)本发明采用的原材料薄荷精油和白藜芦醇具有抑菌作用,利用本发明的原材料制备得到的薄荷精油乳状液可同时具备上述优势。

具体实施方式
本发明实施例仅作为本发明内容的进一步说明,不能作为本发明的限定内容或范围。
粒径分布的检测方法:
采用粒径分析仪对纳米粒子的粒径分布进行检测。
ζ-电位检测方法:
采用ζ-电位分析仪对纳米粒子ζ-电位进行检测。
包埋率的检测方法:
采用超速离心法(离心速度为15,000g)和高效液相色谱仪分别测定包埋率。
抑菌能力的检测方法:
选用金黄色葡萄球菌作为革兰氏阳性菌模型,选用鼠伤寒沙门氏菌作为革兰氏阴性菌模型。鼠伤寒沙门氏菌在BPY培养基,pH7.0,37℃下培养10h,达到~108CFU/ml;金黄色葡萄球菌在肉汤培养基,pH7.4,37℃下培养24h,达到~108CFU/ml。将100μL乳状液加入到9.9mL含有细菌的0.85%NaCl溶液中,使得最终体系细菌浓度为104~106CFU/ml;同时分别设置阳性对照组和阴性对照组,乳状液与细菌混合后10min后,利用琼脂平板法进行菌落计数,每组至少设置3组平行,将平板在37℃下培养24h进行菌落计数。
稳定性的检测方法:
将制备乳状液放置不同的时间,采用高效液相色谱法测定白藜芦醇的含量,进而计算白藜芦醇的残留率。
实施例1:制备玉米醇溶蛋白-白藜芦醇-果胶纳米粒子
(1)采用反相溶剂法制备包埋白藜芦醇的玉米醇溶蛋白纳米粒子,将玉米醇溶蛋白溶于80%乙醇中制得浓度为40mg/mL的玉米醇溶蛋白溶液,然后加入白藜芦醇,获得玉米醇溶蛋白-白藜芦醇混合溶液,玉米醇溶蛋白和白藜芦醇的质量比为50-10:1;
(2)将上述(1)得到的玉米醇溶蛋白-白藜芦醇混合溶液逐滴或快速倒入正在搅拌的纯水中,玉米醇溶蛋白-白藜芦醇混合溶液与水的体积比例为1:9,得到玉米醇溶蛋白-白藜芦醇纳米粒子后经过旋蒸或氮吹去除体系中乙醇,并用纯水补足损失的乙醇量;
(3)将上述(2)中制备的玉米醇溶蛋白-白藜芦醇纳米粒子逐滴加入或者快速倒入正在搅拌的pH为4.0和浓度为0.5mg/mL-2.0mg/mL的果胶水溶液中,其中玉米醇溶蛋白-白藜芦醇纳米粒子溶液与果胶溶液的体积比为1:1,获得玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子;
(4)对获得的玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子的粒径分布、ζ-电位和包埋率进行检测。
检测结果如下:当玉米醇溶蛋白与白藜芦醇质量比在50:1-10:1范围内,纳米粒子粒径为-80nm,白藜芦醇不影响纳米粒子的粒径分布;在玉米醇溶蛋白-白藜芦醇纳米粒子基础上添加果胶,粒径进一步增大,并且随着果胶浓度增大而增大。(图1为不同配方制备玉米醇溶蛋白纳米粒子粒径分布图)
表1是不同条件下制备纳米粒子的ζ-电位和对白藜芦醇的包埋率。玉米醇溶蛋白纳米粒子在pH为4.0条件下的ζ-电位为32.8mV,白藜芦醇对ζ-电位没有显著影响,添加0.5mg/mL果胶后,ζ-电位变为-27.9mV,并且随着果胶浓度增加而增大,表明果胶与玉米醇溶蛋白纳米粒子作用生成玉米醇溶蛋白-果胶复合纳米粒子。玉米醇溶蛋白纳米粒子对白藜芦醇的包埋率约为70%,且不依赖玉米醇溶蛋白和白藜芦醇间的质量比;当果胶浓度范围在0.5-2mg/mL时,不会影响复合粒子对白藜芦醇的包埋率。

实施例2:制备玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子稳定的水包油型薄荷精油乳状液
(1)参照实施例1的方法制备玉米醇溶蛋白-白藜芦醇纳米粒子和玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子,其中玉米醇溶蛋白浓度为2mg/mL,白藜芦醇浓度为0.2mg/mL,果胶浓度0.5-2.0mg/mL;
(2)向(1)中得到的玉米醇溶蛋白-白藜芦醇纳米粒子和玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子中加入50mg/mL的薄荷精油,经过高速剪切(8000rpm,2min)后得到粗乳液,然后在250Bar条件下均质1min,获得最终薄荷精油乳状液;
(3)对获得的薄荷精油乳状液的粒径分布、ζ-电位进行检测。
检测结果如下:利用玉米蛋白纳米粒子和玉米蛋白-白藜芦醇纳米粒子稳定薄荷精油乳状液均呈121和434nm双峰分布且粒径分布无明显差异。当有果胶存在且浓度为0.5mg/mL时,乳状液粒径分布呈现单峰分布且粒径增大到583nm;进一步增大果胶浓度到2mg/mL时,乳状液粒径分布仍然呈现单峰分布且粒径先增大到709nm后有减小到528nm。(图2是不同配方制备的薄荷精油乳状液的粒径分布。)
表2是不同配方制备薄荷精油乳状液的ζ-电位。在pH为4.0的条件下,利用玉米蛋白纳米粒子和玉米蛋白-白藜芦醇纳米粒子稳定薄荷精油乳状液的ζ-电位分别为+61.1mV和+64.1mV。添加果胶后,乳状液的ζ-电位由正值变为负值,且在果胶浓度为0.5-2mg/mL范围内,乳状液ζ-电位在-38mV。

实施例3:检测薄荷精油和白藜芦醇包埋率
(1)参照实施例2的方法制备不同配方的薄荷精油乳状液,其中薄荷精油含量为50mg/mL,玉米醇溶蛋白含量为2mg/mL,白藜芦醇含量为0.2mg/mL,果胶含量为0-2mg/mL;
(2)对制备得到的薄荷精油乳状液进行包埋率检测。
检测结果如下:在无果胶条件下,薄荷精油和白藜芦醇的包埋率分别为74%和66%;当添加了0.5mg/mL的果胶后,薄荷精油和白藜芦醇的包埋率分别增加到83%和87%。表明玉米醇溶蛋白-果胶复合纳米粒子具有更加包埋效果。随着果胶浓度进一步增加到2mg/mL时,薄荷精油包埋率增加到87%,但是白藜芦醇的包埋率反而降到78%。(图3所示是不同果胶浓度对薄荷精油和白藜芦醇包埋率的影响)
实施例4:检测薄荷精油乳状液的对两种致病菌的协同抑菌作用
选用实施例2中方法制备的不同配方的乳状液体系进行抑菌能力的检测,其中薄荷精油含量为50mg/mL,玉米醇溶蛋白含量为2mg/mL,白藜芦醇含量为0.2mg/mL,果胶含量为0-2mg/mL。
检测结果如下:采用玉米醇溶蛋白稳定的薄荷精油乳状液可使得金黄色葡萄球菌总数降低3.16个对数,当加入白藜芦醇后,抑菌能力提高达到3.74个对数,而玉米醇溶蛋白-白藜芦醇纳米粒子的抑菌能力仅有0.21个对数,这些结果表明白藜芦醇与薄荷精油具有协同抗菌作用。当添加0.5mg/mL果胶后,抑菌能力提高到4.18个对数;空白实验表明果胶在该实验条件下没有显著的抑菌能力,这些结果说明玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子增强了与薄荷精油的协同抗菌作用。(图4是不同配方乳状液对金黄色葡萄球菌的抑制效果,其中,Z代表玉米醇溶蛋白纳米粒子,Res代表白藜芦醇,ZP代表玉米醇溶蛋白-果胶复合纳米粒子,括号中数字代表果胶浓度(mg/mL)。)
利用玉米醇溶蛋白纳米粒子、玉米醇溶蛋白-白藜芦醇纳米粒子和玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子制备的薄荷精油乳状液的抑菌能力分别为3.33、3.87和4.71个对数,而三种纳米粒子单独使用的抑菌能力分别仅为0.05、0.21和0.51个对数,表明白藜芦醇和薄荷精油在该条件下制备的乳状液中具有协同抗菌作用。(图5是不同配方乳状液对鼠伤寒沙门氏菌的抑制效果,其中,Z代表玉米醇溶蛋白纳米粒子,Res代表白藜芦醇,ZP代表玉米醇溶蛋白-果胶复合纳米粒子,括号中数字代表果胶浓度(mg/mL)。)
实施例5:检测白藜芦醇的稳定性
参照实施例2中制备的不同配方的乳状液进行白藜芦醇的稳定性的检测,其中薄荷精油含量为50mg/mL,玉米醇溶蛋白含量为2mg/mL,白藜芦醇含量为0.2mg/mL,果胶含量为0-2mg/mL。
检测结果如下:在25℃下储藏的14天后,白藜芦醇未发生明显的降解,残留率约为95%;在储藏第28天后,在无果胶的体系中白藜芦醇的残留率为70%,在果胶浓度为0.5-2.0mg/mL的乳状液体系中,白藜芦醇的残留率仍保持在95%左右,表明添加果胶后能够显著提高白藜芦醇的储藏稳定性。当储藏了42天后,果胶浓度为0.5mg/mL的乳状液体系中白藜芦醇的残留率为70%,而在果胶浓度为1.0-2.0mg/mL的乳状液体系中白藜芦醇的残留率约为81%,表明白藜芦醇的稳定性随着果胶浓度的提高而增强。(图6不同果胶浓度对白藜芦醇在储藏期间残留率的影响)
虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书所界定的为准。

1.一种具有高稳定性的薄荷精油乳状液,其特征在于,所述薄荷精油乳状液包含溶剂、溶质以及乳化剂;所述溶质为薄荷精油;所述乳化剂由玉米醇溶蛋白、白藜芦醇和果胶组成。
2.如权利要求1所述的一种具有高稳定性的薄荷精油乳状液,其特征在于,所述溶质薄荷精油在整个薄荷精油乳状液中的浓度为5-50mg/mL。
3.如权利要求1或2所述的一种具有高稳定性的薄荷精油乳状液,其特征在于,所述乳化剂中玉米醇溶蛋白、白藜芦醇和果胶的质量比为10:0.2-1:2.5-10。
4.如权利要求1-3任一所述的一种具有高稳定性的薄荷精油乳状液的制备方法,其特征在于,包含如下步骤:
步骤1:将玉米醇溶蛋白和白藜芦醇按照一定的比例溶解于乙醇溶液,然后将溶有玉米醇溶蛋白和白藜芦醇的乙醇溶液加入水中,获得玉米醇溶蛋白-白藜芦醇纳米粒子;
步骤2:将步骤1所得的玉米醇溶蛋白-白藜芦醇纳米粒子加入果胶溶液中,获得玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子;
步骤3:在步骤2所得的玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子中加入薄荷精油,进行均质包埋处理,得到具有高稳定性的薄荷精油乳状液。
5.如权利要求4所述的一种具有高稳定性的薄荷精油乳状液的制备方法,其特征在于,所述步骤1为将溶有玉米醇溶蛋白和白藜芦醇的乙醇溶液快速加入水中或者逐滴加入正在搅拌的水中获得玉米醇溶蛋白-白藜芦醇纳米粒子;所述步骤1中溶有玉米醇溶蛋白和白藜芦醇的乙醇溶液与水的体积比为1:9。
6.如权利要求4或5所述的一种具有高稳定性的薄荷精油乳状液的制备方法,其特征在于,所述步骤2为将步骤1所得的玉米醇溶蛋白-白藜芦醇纳米粒子经过旋蒸或氮吹去除体系中的乙醇,并用纯水补足损失的乙醇的量后逐滴加入或快速倒入正在搅拌果胶溶液中,获得玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子;所述步骤2中玉米醇溶蛋白-白藜芦醇纳米粒子溶液与果胶溶液的体积比为1:1。
7.如权利要求4-6任一所述的一种具有高稳定性的薄荷精油乳状液的制备方法,其特征在于,所述步骤3为在步骤2所得的玉米醇溶蛋白-白藜芦醇-果胶复合纳米粒子中加入薄荷精油,然后在转速为8000-20000rpm的条件下进行高速剪切,最后在压力为50-500Bar的条件下进行均质,得到具有高稳定性的薄荷精油乳状液。
8.权利要求4-7任一所述的一种具有高稳定性的薄荷精油乳状液的制备方法制备得到的薄荷精油乳状液。
9.权利要求1-3任一所述的一种具有高稳定性的薄荷精油乳状液或权利要求4-7任一所述的一种具有高稳定性的薄荷精油乳状液的制备方法制备得到的薄荷精油乳状液在制备食品、保健品、化妆品、医药配制品以及抗菌剂中的应用。
10.含有权利要求1-3任一所述的一种具有高稳定性的薄荷精油乳状液或含有权利要求4-7任一所述的一种具有高稳定性的薄荷精油乳状液的制备方法制备得到的薄荷精油乳状液的食品、保健品、化妆品、医药配制品以及抗菌剂。
