Abstract
本发明提供了一种有益血糖健康的铁皮石斛组合物及其制备方法,包含铁皮石斛叶3~300份、线叶金雀花1~60份、大麦苗1~100份、金银花0.1~20份、杜仲叶1~50份、山药3~200份、桑叶提取物0.05~10份、库拉索芦荟全叶干粉0.1~30份。其经提取、干燥、粉碎、混合及成型等制成,具有降血糖、改善肾功能及光损伤视网膜的作用。

技术领域
本发明涉及医药、功能食品领域,具体涉及一种有益血糖健康的铁皮石斛组合物及其制备方法。
背景技术
人体一般空腹血糖值在6.1mmol/L以下,用餐后两小时血糖的正常值在7.8mmol/L以下,如高于这一范围则为高血糖。正常情况下,人体能够通过激素调节和神经调节确保血糖保持平衡。但是在遗传因素(如糖尿病家族史)与外部因素(如不合理的膳食、肥胖等)的作用下,两大调节功能紊乱,出现高血糖。短暂的高血糖对人体无严重损害,比如在情绪激动、高度紧张时,可出现短暂的高血糖;一次进食大量的甜食,也可出现短暂高血糖;一段时间后,血糖水平慢慢趋于正常。然而长期的高血糖会使全身各组织器官病变,导致各种急慢性并发症的发生。如营养缺乏、抵抗力下降、肾功能受损、神经病变、眼底病变、心脑血管疾病、糖尿病足等。因此,健康的血糖管理势在必行。目前国内常用口服降糖药物分为促胰岛素分泌剂类、二甲双胍类、α-糖苷酶抑制剂类、噻唑烷二酮衍生物、DPP-4酶抑制剂等;其中促胰岛素分泌剂类又分为磺脲类和非磺脲类(格列奈类)。这些药物的共同副作用为低血糖。同时,双胍类的降糖药对肠胃的伤害较大,容易导致消化不良,严重时可能会导致酮尿和乳酸酸中毒。磺脲类易造成白细胞减少、溶血性贫血。胰岛素增敏剂易引起水潴留,颜面和手脚浮肿。这些口服药物均不利于该类慢性疾病的长期健康管理。
本发明目的在于提供以一种将铁皮石斛叶等多种天然原料进行合理组方加工,能够有效控制高血糖、适宜长期保健使用的铁皮石斛组合物及其制备方法。
发明内容
本发明第一目的在于提供一种有益血糖健康的铁皮石斛组合物,第二目的在于提供一种有益血糖健康的铁皮石斛组合物的加工制备方法。
为实现上述目的,本发明提供的一种有益血糖健康的铁皮石斛组合物,其包铁皮石斛叶、线叶金雀花、大麦苗、金银花、杜仲叶、山药、桑叶提取物、库拉索芦荟全叶干粉。
其中,按重量份数计,所述有益血糖健康的铁皮石斛组合物包含铁皮石斛叶3~300份、线叶金雀花1~60份、大麦苗1~100份、金银花0.1~20份、杜仲叶1~50份、山药3~200份、桑叶提取物0.05~10份、库拉索芦荟全叶干粉0.1~30份。
本发明所述的有益血糖健康的铁皮石斛组合物,可加入可接受的辅料或食品添加剂,制成固体饮料、口服液。
本发明所述的有益血糖健康的铁皮石斛组合物,其特征在于,所述桑叶提取物以桑叶为主要原料,以生活饮用水、食用酒精、麦芽糊精为辅料,经物理提取、过滤、浓缩、干燥、包装等生产工艺制成;
优选地,所述桑叶提取物为安徽尚善生物科技股份有限公司生产的桑叶提取物;
优选地,所述桑叶提取物生产检验执行标准为Q/AS0010S-2015;更优选地,所述桑叶提取物中1-脱氧野尻霉素≥10.0%。
本发明所述的有益血糖健康的铁皮石斛组合物,其特征在于,所述库拉索芦荟全叶干粉以库拉索芦荟为原料,经清洗、榨汁、干燥、杀菌、粉碎、包装等生产工艺制成;
优选地,所述库拉索芦荟全叶干粉为云南万绿生物股份有限公司生产的库拉索芦荟全叶干粉;
优选地,所述库拉索芦荟全叶干粉生产检验执行标准为Q/YWL0012S-2017;更优选地,所述索芦荟全叶干粉中多糖≥30000mg/kg,芦荟苷≥2500mg/kg。
本发明还提供一种本发明所述有益血糖健康的铁皮石斛组合物的制备方法,所述制备方法包括如下步骤:
(1)将铁皮石斛叶加入10~53倍量的水煎煮提取3次,每次提取1~4小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将线叶金雀花、大麦苗、金银花、杜仲叶、山药加入6~15倍量的水煎煮提取3次,每次提取1~3小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入4~10倍量的40~85%乙醇煎煮提取2次,每次提取1~3小时,合并两次滤液,400目筛滤过,备用;
(4)将(1)、(2)、(3)中滤液合并,静置1~6小时,250目筛滤过,滤液于45~88℃减压浓缩至密度1.08~1.33g/ml,50~85℃减压干燥,控制水份<3.0%,粉碎过60~300目筛,备用;
(5)将(4)中物料加入桑叶提取物、库拉索芦荟全叶干粉及适宜辅料或食品添加剂,制成固体饮料、口服液。
本发明有益血糖健康的铁皮石斛组合物口感良好,服用方式容易被众多消费者接受,有助于其推广使用。本发明具有以下有益效果:本发明有益血糖健康的铁皮石斛组合物有降血糖、改善肾功能及光损伤视网膜的作用。
具体实施方式
下面通过具体实施方式进一步说明本发明,下述实施例、试验例中所用的原料、试剂材料等,如无特殊说明,均购买市售产品。以下实施例仅用于说明本发明,其中所述的实验方法,如无特殊定义,均为常规方法。
实施例1
原料:铁皮石斛叶3份、线叶金雀花1份、大麦苗1份、金银花0.1份、杜仲叶1份、山药3份、桑叶提取物0.05份、库拉索芦荟全叶干粉0.1份。
制备方法:
(1)将铁皮石斛叶加入10倍量的水煎煮提取3次,每次提取1小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将线叶金雀花、大麦苗、金银花、杜仲叶、山药加入6倍量的水煎煮提取3次,每次提取1小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入4倍量的40%乙醇煎煮提取2次,每次提取1小时,合并两次滤液,400目筛滤过,备用;
(4)将(1)、(2)、(3)中滤液合并,静置1小时,250目筛滤过,滤液于45℃减压浓缩至密度1.08g/ml,50℃减压干燥,控制水份<3.0%,粉碎过60目筛,备用;
(5)将(4)中物料加入桑叶提取物、库拉索芦荟全叶干粉及木糖醇12份、甜菊糖苷0.05份、赤藓糖醇5份、柠檬酸0.08份,制成口服液。
实施例2
原料:铁皮石斛叶150份、线叶金雀花30份、大麦苗50份、金银花10份、杜仲叶25份、山药100份、桑叶提取物5份、库拉索芦荟全叶干粉15份。
制备方法:
(1)将铁皮石斛叶加入30倍量的水煎煮提取3次,每次提取2小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将线叶金雀花、大麦苗、金银花、杜仲叶、山药加入10倍量的水煎煮提取3次,每次提取2小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入7倍量的65%乙醇煎煮提取2次,每次提取2小时,合并两次滤液,400目筛滤过,备用;
(4)将(1)、(2)、(3)中滤液合并,静置3.5小时,250目筛滤过,滤液于68℃减压浓缩至密度1.25g/ml,62℃减压干燥,控制水份<3.0%,粉碎过200目筛,备用;
(5)将(4)中物料加入桑叶提取物、库拉索芦荟全叶干粉及阿洛酮糖50份、罗汉果苷0.09份,制成固体饮料。
实施例3
原料:铁皮石斛叶300份、线叶金雀花60份、大麦苗100份、金银花20份、杜仲叶50份、山药200份、桑叶提取物10份、库拉索芦荟全叶干粉30份。
制备方法:
(1)将铁皮石斛叶加入53倍量的水煎煮提取3次,每次提取4小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将线叶金雀花、大麦苗、金银花、杜仲叶、山药加入15倍量的水煎煮提取3次,每次提取13小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入10倍量的85%乙醇煎煮提取2次,每次提取3小时,合并两次滤液,400目筛滤过,备用;
(4)将(1)、(2)、(3)中滤液合并,静置6小时,250目筛滤过,滤液于88℃减压浓缩至密度1.33g/ml,85℃减压干燥,控制水份<3.0%,粉碎过300目筛,备用;
(5)将(4)中物料加入桑叶提取物、库拉索芦荟全叶干粉及低聚异麦芽糖醇350份、三氯蔗糖0.06份,制成固体饮料。
实施例4
原料:铁皮石斛叶10份、线叶金雀花5份、大麦苗10份、金银花5份、杜仲叶10份、山药10份、桑叶提取物0.5份、库拉索芦荟全叶干粉0.3份。
制备方法:
(1)将铁皮石斛叶加入17倍量的水煎煮提取3次,每次提取2小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将线叶金雀花、大麦苗、金银花、杜仲叶、山药加入9倍量的水煎煮提取3次,每次提取1.5小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入6倍量的60%乙醇煎煮提取2次,每次提取1.5小时,合并两次滤液,400目筛滤过,备用;
(4)将(1)、(2)、(3)中滤液合并,静置2小时,250目筛滤过,滤液于60℃减压浓缩至密度1.12g/ml,65℃减压干燥,控制水份<3.0%,粉碎过200目筛,备用;
(5)将(4)中物料加入桑叶提取物、库拉索芦荟全叶干粉及阿洛酮糖200份、赤藓糖醇50份,制成固体饮料。
实施例5
原料:铁皮石斛叶10份、线叶金雀花5份、大麦苗10份、杜仲叶10份、山药10份、桑叶提取物0.5份、库拉索芦荟全叶干粉0.3份。
制备方法:
(1)将铁皮石斛叶加入17倍量的水煎煮提取3次,每次提取2小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将线叶金雀花、大麦苗、杜仲叶、山药加入9倍量的水煎煮提取3次,每次提取1.5小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入6倍量的60%乙醇煎煮提取2次,每次提取1.5小时,合并两次滤液,400目筛滤过,备用;

(2)将大麦苗、金银花、杜仲叶、山药加入9倍量的水煎煮提取3次,每次提取1.5小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入6倍量的60%乙醇煎煮提取2次,每次提取1.5小时,合并两次滤液,400目筛滤过,备用;
(4)将(1)、(2)、(3)中滤液合并,静置2小时,250目筛滤过,滤液于60℃减压浓缩至密度1.12g/ml,65℃减压干燥,控制水份<3.0%,粉碎过200目筛,备用;
(5)将(4)中物料加入桑叶提取物、库拉索芦荟全叶干粉及阿洛酮糖200份、赤藓糖醇50份,制成固体饮料。

制备方法:
(1)将铁皮石斛叶加入17倍量的水煎煮提取3次,每次提取2小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将线叶金雀花、大麦苗、金银花加入9倍量的水煎煮提取3次,每次提取1.5小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入6倍量的60%乙醇煎煮提取2次,每次提取1.5小时,合并两次滤液,400目筛滤过,备用;

制备方法:
(1)将铁皮石斛叶加入17倍量的水煎煮提取3次,每次提取2小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将(1)中滤液静置2小时,250目筛滤过,滤液于60℃减压浓缩至密度1.12g/ml,65℃减压干燥,控制水份<3.0%,粉碎过200目筛,备用;
(3)将(2)中物料加入桑叶提取物、库拉索芦荟全叶干粉及阿洛酮糖200份、赤藓糖醇50份,制成固体饮料。
试验例1对高血糖大鼠的效果观察
1、试验动物及方法
选择健康雄性SD大鼠100只,随机分为10组,分为空白对照组、桑叶提取物组、实施例1-8组。空白组食用普通饲料,桑叶提取物组每天在饲料中添加500mg桑叶提取物,实施例1-8组大鼠在饲料中每天添加1.2g相应实施例样品,连续45天。末次给药前禁食不禁水12h,末次给药后1小时,各组均腹腔注射肾上腺素0.7mg/,同时灌服葡萄糖200mg。
2、测定方法
剪尾采血,用葡萄糖氧化酶法分别测定空腹,造模后1小时、2小时、4小时的血糖值。3、结果:本发明对肾上腺素高血糖大鼠有明显快速的降血糖作用。具体结果见表1。

试验例2对大鼠肾病综合征的效果观察
1、实验动物及方法
选择健康雄性SD大鼠100只,随机分为10组,分为假手术组、模型组、实施例1-8组。模型组和实施例1-8组大鼠静脉注射氨基核苷嘌呤霉素15mg,制作大鼠肾病综合征慢性,假手术组大鼠注射等体积的生理盐水。造模后假手术组、模型组大鼠每天食用普通饲料,实施例1-8组大鼠在饲料中每天添加1.0g相应实施例样品,连续90天。
2、测定方法
试验周期全部结束后,分别从各组大鼠眼眶取血,3000rpm离心10min,分离血清,采用全自动血生化分析仪测定血清肌酐指标。
3、结果:本发明表现出对肾功能的改善效果。具体结果见表2。

试验例3对光损伤大鼠视网膜的影响
1、实验动物及方法
取SD大鼠100只,重约200g,分为空白组、模型组、实施例1-8组。空白组、模型组大鼠每天食用普通饲料,实施例1-8组大鼠在饲料中每天添加1.5g相应实施例样品,连续20天。模型组和实施例1-8组大鼠每天置(6000±100)Lx光强度下持续照射12小时,再置于暗箱12小时。空白组每天置(100±10)Lx光强度下持续照射12小时,再置于暗箱12小时。末次给药后置于暗处4小时处死大鼠,摘除眼球待用。
2、测定方法
将大鼠眼球完整分离出晶状体,晶状体同生理盐水冰浴研磨,制成质量浓度为9%的冰冷匀浆液,用试剂盒测定晶状体中MDA(丙二醛)、GSH(谷胱甘肽)含量。
3、结果:本发明表现出对视网膜光损伤的改善效果。具体结果见表3。

1.一种有益血糖健康的铁皮石斛组合物,其包含铁皮石斛叶、线叶金雀花、大麦苗、金银花、杜仲叶、山药、桑叶提取物、库拉索芦荟全叶干粉。
2.根据权利要求1所述的有益血糖健康的铁皮石斛组合物,其特征在于,按重量份数计,所述有益血糖健康的铁皮石斛组合物包含铁皮石斛叶3~300份、线叶金雀花1~60份、大麦苗1~100份、金银花0.1~20份、杜仲叶1~50份、山药3~200份、桑叶提取物0.05~10份、库拉索芦荟全叶干粉0.1~30份。
3.根据权利要求1至2任一所述的有益血糖健康的铁皮石斛组合物,其特征在于加入可接受的辅料或食品添加剂,制成固体饮料、口服液。
4.根据权利要求1至3任一所述的有益血糖健康的铁皮石斛组合物,其特征在于,所述桑叶提取物以桑叶为主要原料,以生活饮用水、食用酒精、麦芽糊精为辅料,经物理提取、过滤、浓缩、干燥、包装等生产工艺制成;
优选地,所述桑叶提取物为安徽尚善生物科技股份有限公司生产的桑叶提取物;
优选地,所述桑叶提取物生产检验执行标准为Q/AS0010S-2015;更优选地,所述桑叶提取物中1-脱氧野尻霉素≥10.0%。
5.根据权利要求1至4任一所述的有益血糖健康的铁皮石斛组合物,其特征在于,所述库拉索芦荟全叶干粉以库拉索芦荟为原料,经清洗、榨汁、干燥、杀菌、粉碎、包装等生产工艺制成;
优选地,所述库拉索芦荟全叶干粉为云南万绿生物股份有限公司生产的库拉索芦荟全叶干粉;
优选地,所述库拉索芦荟全叶干粉生产检验执行标准为Q/YWL0012S-2017;更优选地,所述索芦荟全叶干粉中多糖≥30000mg/kg,芦荟苷≥2500mg/kg。
6.一种根据权利要求1至5任一所述的有益血糖健康的铁皮石斛组合物的制备方法,所述制备方法包括如下步骤:
(1)将铁皮石斛叶加入10~53倍量的水煎煮提取3次,每次提取1~4小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(2)将线叶金雀花、大麦苗、金银花、杜仲叶、山药加入6~15倍量的水煎煮提取3次,每次提取1~3小时,收集滤液,合并三次滤液,200目筛滤过,备用;
(3)将(2)中滤渣加入4~10倍量的40~85%乙醇煎煮提取2次,每次提取1~3小时,合并两次滤液,400目筛滤过,备用;
(4)将(1)、(2)、(3)中滤液合并,静置1~6小时,250目筛滤过,滤液于45~88℃减压浓缩至密度1.08~1.33g/ml,50~85℃减压干燥,控制水份<3.0%,粉碎过60~300目筛,备用;
(5)将(4)中物料加入桑叶提取物、库拉索芦荟全叶干粉及适宜辅料或食品添加剂,制成固体饮料、口服液。
