Abstract
The invention is prepared by adopting scientific methods such as volatile oil extraction, ethanol extraction, water extraction, alcohol precipitation and the like according to different properties of active ingredients in the six traditional Chinese medicines in the granules for tonifying deficiency and removing blood stasis in the national medicine standard (YBZ1142015) on the basis of component analysis of the six traditional Chinese medicines. The invention provides a traditional Chinese medicine composition with the functions of tonifying qi and deficiency, promoting blood circulation and removing obstruction in channels and a preparation method thereof, wherein the traditional Chinese medicine composition has the advantages of high content of active ingredients, small dosage and high curative effect.
Description
Technical Field
The invention belongs to the field of pharmaceutical technology development, and particularly relates to a traditional Chinese medicine composition with the effects of tonifying qi and deficiency, and activating blood and dredging collaterals and a preparation method thereof.
Background
Coronary heart disease and angina pectoris belong to common clinical diseases, can be classified into the categories of chest obstruction, chest pain and the like in traditional Chinese medicine, and have various traditional Chinese medicine syndrome phenotypes. In recent years, with the progress of aging of population and the change of dietary habits of people, the incidence of cardiovascular and cerebrovascular diseases is gradually increased year by year, and the cardiovascular and cerebrovascular diseases become one of the main diseases threatening human health.
The deficiency tonifying and stasis removing granules are a medicine with the functions of tonifying qi and deficiency, and activating blood circulation to remove meridian obstruction, are mainly used for treating arteriosclerosis and coronary heart disease caused by qi deficiency and blood stasis, are prepared by adding and subtracting a prescription based on 'yang tonifying and five decoction' in a classical name prescription in China and combining with the modern traditional Chinese medicine theory, are currently collected in the national medicine standard (YBZ1142015), are also unique production varieties of an applicant, and disclose the following information in the national medicine standard:

the preparation method comprises the following steps: cutting the six medicines, adding water, decocting twice, adding 6 times of water for decocting for 4 hours for the first time, adding 5 times of water for decocting for 3 hours for the second time, mixing decoctions, filtering, concentrating the filtrate to obtain a clear paste with a relative density of 1.05-1.10(50 ℃), adding ethanol to make the ethanol content reach 70%, fully stirring, standing for 12 hours, filtering, concentrating the filtrate to obtain a thick paste with a relative density of 1.28-1.30(50 ℃), adding a proper amount of sucrose, mixing uniformly, preparing into granules, drying (60-70 ℃) and preparing into 1000 g.
The extraction method adopts a water extraction and alcohol precipitation method which is commonly used in traditional Chinese medicine preparations, and because the active ingredients of six traditional Chinese medicines such as red ginseng and the like are different, the single water extraction and alcohol precipitation method can cause that some active ingredients can not be extracted, or the content of some active ingredients is very low, so that the curative effect of the medicine is influenced, the curative effect can be achieved by taking 1 tablet or 1 granule, and the dosage can be multiplied as a result, and meanwhile, the toxic and side effects of the medicine are also increased.
Therefore, it is necessary to provide a Chinese medicinal composition and its preparation which can improve the content of active ingredients of the medicament, and has small dosage and high curative effect.
Disclosure of Invention
The invention aims to provide a traditional Chinese medicine composition with the functions of tonifying qi and deficiency, promoting blood circulation and removing obstruction in channels and a preparation method thereof, wherein the traditional Chinese medicine composition has the advantages of high content of active ingredients, small dosage and high curative effect.
The invention analyzes the components of six traditional Chinese medicines in the deficiency tonifying and stasis removing granules in the national medicine standard (YBZ1142015), and prepares the granules by adopting scientific methods such as volatile oil extraction, ethanol extraction, water extraction, alcohol precipitation and the like according to different properties of active ingredients in the medicines.
The red ginseng is a cooked product of ginseng and has the effects of greatly tonifying primordial qi, recovering pulse, relieving depletion, benefiting qi and controlling blood. The active ingredients mainly contain ginsenoside (ginsenoside Rg, ginsenoside Re and ginsenoside Rb) and polysaccharide, and the content of the active ingredients extracted by ethanol is high through research.
Radix astragali has effects of enhancing organism immunity, protecting liver, promoting urination, and resisting aging. The active ingredients mainly comprise astragaloside IV and Astragalus polysaccharides, and the active ingredients are extracted by water extraction method after research, and have high content of active ingredients.
Acanthopanax senticosus has the effects of strengthening body resistance, tonifying kidney, strengthening spleen, promoting intelligence, tranquilizing, etc., and its active ingredients mainly comprise alcohol-soluble syringin, eleutheroside B and eleutheroside E, and the content of active ingredients is high by ethanol extraction;
radix Paeoniae Rubra has effects of clearing heat and cooling blood, promoting blood circulation and removing blood stasis. The active ingredient contained in the extract is mainly paeoniflorin, and the active ingredient content is high by research and extraction with a water extraction method;
the salvia miltiorrhiza has the effects of activating blood and dissolving stasis, stimulating the menstrual flow and relieving pain, clearing away the heart fire and relieving restlessness, cooling blood and eliminating carbuncle. The active ingredients mainly comprise tanshinone, including tanshinone IIA, salvianolic acid B, cryptotanshinone and other fat-soluble non-quinone pigment compounds, and the content of the active ingredients is high by adopting an ethanol extraction method through research;
cassia twig has the functions of inducing sweat, expelling pathogenic factors from muscles, warming and activating meridian, invigorating Yang and regulating qi. The active ingredients mainly comprise volatile oil such as cassia twig bark aldehyde and cinnamic acid, and also comprise part of water-soluble ingredients such as phenols and organic acid. Through research, the effective active ingredient content of the compound is high by adopting a steam distillation extraction method and a water extraction method.
The invention provides a traditional Chinese medicine composition with the effects of tonifying qi and deficiency, and promoting blood circulation and removing obstruction in channels, which is prepared from the following components: 86.4g of red ginseng, 343.2g of astragalus, 343.2g of acanthopanax, 172.8g of red peony root, 86.4g of red sage root, and 50.4g of cassia twig, which are prepared by the following method: extracting volatile oil from ramulus Cinnamomi; mixing the residues after volatile oil extraction with radix astragali and radix Paeoniae Rubra, and extracting with water to obtain water extract; extracting Ginseng radix Rubri, radix Acanthopanacis Senticosi, and Saviae Miltiorrhizae radix with ethanol to obtain soft extract; mixing the water extract and ethanol extract, precipitating with ethanol, recovering ethanol, concentrating, drying, micronizing to obtain micropowder, mixing with different adjuvants, and making into oral solid preparation such as granule, tablet, capsule, etc.
The invention relates to a traditional Chinese medicine composition with the functions of tonifying qi and deficiency, and promoting blood circulation and removing obstruction in channels, which comprises the following components in part by weight:
1. firstly, the six medicinal materials such as red ginseng and the like are respectively cut into pieces for standby;
2. the method for extracting the volatile oil from the cassia twig comprises the following steps: extracting volatile oil from crushed ramulus Cinnamomi in a volatile oil extractor, and storing the extracted volatile oil in another extractor.
3. The water extraction of the dregs of a decoction after the volatile oil extraction, the astragalus and the red paeony root comprises the following steps: placing the crushed radix astragali and radix Paeoniae Rubra and the residue after volatile oil extraction in a multifunctional extraction tank, extracting with water twice, the first time adding amount of water 8-10 times of the material amount, extracting for 2-3 hr, the second time adding amount of water 6-8 times of the material amount, extracting for 1-2 hr, mixing the two water extractive solutions, filtering, and concentrating the filtrate to obtain soft extract with specific gravity of 1.20-1.30 (measured at 60 deg.C).
Preferably, the water extraction of the dregs of decoction, the astragalus and the red paeony root after the volatile oil extraction comprises the following steps: placing the crushed radix astragali and radix Paeoniae Rubra and the residue after volatile oil extraction in a multifunctional extraction tank, extracting with water twice, the first time adding amount of the residue 8 times the amount of the raw materials, extracting for 2-3 hr, the second time adding amount of the residue 6 times the amount of the raw materials, extracting for 2-3 hr, mixing the two water extractive solutions, filtering, and concentrating the filtrate to obtain soft extract with specific gravity of 1.20-1.30 (measured at 60 deg.C).

Preferably, the red ginseng, acanthopanax senticosus and salvianic alcohol comprise the following steps: extracting the crushed red ginseng, acanthopanax and salvia miltiorrhiza twice with 70% ethanol under reflux in a reflux extractor, adding 8 times of the amount of the materials for the first time, extracting for 2-3 hours, adding 6 times of the amount of the materials for the second time, extracting for 1-2 hours, combining the extracting solutions, filtering, recovering ethanol from the filtrate under reduced pressure until no alcohol smell exists, and continuously concentrating to obtain a thick paste with the specific gravity of 1.20-1.30 (measured at 50-60 ℃) to obtain the alcohol extract.
Further preferably, the red ginseng, acanthopanax senticosus and salvia miltiorrhiza alcohol extraction method comprises the following steps: placing the cut red ginseng, acanthopanax and salvia miltiorrhiza into a multifunctional extraction tank, carrying out reflux extraction twice by using 75% ethanol, adding 10 times of the amount of the fed medicinal materials for the first time, extracting for 3 hours, adding 8 times of the amount of the fed medicinal materials for the second time, extracting for 2 hours, combining extracting solutions, filtering, decompressing filtrate, recovering ethanol until no alcohol smell exists, and continuously concentrating to obtain a thick paste with the specific gravity of 1.2-1.3 (measured by heat at 50-60 ℃), thus obtaining the alcohol extract.

Mixing the water extract and ethanol extract, precipitating with ethanol, recovering ethanol, concentrating, drying, and micronizing to obtain micropowder comprising the following steps: mixing the above soft extracts, adding ethanol to ethanol content of 50-90%, stirring, standing at 1-10 deg.C for 12-24 hr, collecting supernatant, filtering with 80-120 mesh sieve, recovering ethanol from filtrate under reduced pressure, concentrating to obtain soft extract with specific gravity of 1.20-1.30 (measured at 50-60 deg.C), drying, and micronizing.
Preferably, the water extraction thick paste and the alcohol extraction thick paste are combined for alcohol precipitation, ethanol is recovered, concentrated, dried and superfine ground to obtain micro powder, and the micro powder comprises the following steps: mixing the above soft extracts, adding ethanol to ethanol content of 80%, stirring, standing at 3-10 deg.C for 24 hr, collecting supernatant, filtering with 100 mesh screen, recovering ethanol from filtrate under reduced pressure, concentrating to obtain soft extract with specific gravity of 1.20-1.30 (measured at 50-60 deg.C), drying, and micronizing to obtain micropowder.
Further preferably, the step of mixing the water extract and the alcohol extract for alcohol precipitation, recovering ethanol, concentrating, drying and carrying out superfine grinding to obtain micro powder comprises the following steps: mixing the above soft extracts, adding ethanol to alcohol content of 75%, stirring, standing at 2-7 deg.C for 24 hr, collecting supernatant, filtering with 120 mesh screen, recovering ethanol from filtrate under reduced pressure, concentrating to obtain soft extract with specific gravity of 1.20-1.30 (measured at 50-60 deg.C), drying, and micronizing to obtain micropowder.
The invention also provides a preparation method of the traditional Chinese medicine composition, which comprises the following steps:
1. cutting the six medicinal materials such as red ginseng and the like into pieces for later use;
2. extracting volatile oil from crushed ramulus Cinnamomi in a volatile oil extractor, and storing the extracted volatile oil in another extractor;
3. placing crushed radix astragali and radix Paeoniae Rubra and the residue after volatile oil extraction in a multifunctional extraction tank, adding water for extraction twice, adding 8-10 times of the amount of the materials for the first time, extracting for 2-3 hr, adding 6-8 times of the amount of the materials for the second time, extracting for 1-2 hr, mixing the water extractive solutions, filtering, concentrating the filtrate to obtain soft extract with specific gravity of 1.20-1.30 (measured at 60 deg.C), and collecting the soft extract;

5. mixing the above soft extracts, adding ethanol to ethanol content of 50-90%, stirring, standing at 1-10 deg.C for 12-24 hr, collecting supernatant, filtering with 80-120 mesh sieve, recovering ethanol from filtrate under reduced pressure, concentrating to obtain soft extract with specific gravity of 1.20-1.30 (measured at 50-60 deg.C), drying, and micronizing to obtain micropowder.
The invention can further prepare the obtained micro powder into the following oral solid preparations:
1. pulverizing sucrose, sieving with 80-120 mesh sieve, adding the above micropowder, mixing, granulating with 30-90% ethanol as binder and 18-24 mesh sieve, drying at 50-70 deg.C, grading with 18-24 mesh sieve, spraying the above volatile oil, and mixing for 30-60 min to obtain sugar-containing granule;
2. sieving dextrin with 80-120 mesh sieve, adding the above micropowder and appropriate amount of correctant, mixing, granulating with 30-90% ethanol as binder and 18-24 mesh sieve, drying at 50-70 deg.C, grading with 18-24 mesh sieve, spraying the above volatile oil, and mixing for 30-60 min to obtain sugar-free granule.
The flavoring agent is one or more of stevioside, lactose, aspartame, sorbitol, xylitol, maltose, mannitol and sucralose.
3. The micro powder is taken, added with diluent, adhesive, disintegrating agent, lubricant and the like, and prepared into tablets according to the conventional preparation method of the tablets.
4. Mixing the above fine powders with diluent, binder, and lubricant, and making into capsule by conventional method.
Advantageous effects
The preparation method of the deficiency-tonifying and stasis-removing granules disclosed by the invention is further optimized, and the granules are prepared by adopting a plurality of technologies such as volatile oil extraction, ethanol extraction, water extraction and alcohol precipitation respectively, and the granules have the advantages of high content of active ingredients of the medicine, small dosage, high curative effect of the medicine, high bioavailability and the like.

Example 1
The preparation method comprises the steps of respectively cutting the six medicinal materials such as the red ginseng and the like in the prescription for later use;
(1) placing the crushed ramulus Cinnamomi in a volatile oil extractor, collecting the extracted volatile oil, and storing the residue in another extractor;
(2) placing the crushed radix astragali, radix Paeoniae Rubra and the above residue in a multifunctional extraction tank, adding water for extraction twice, adding 8 times of the amount of the materials for the first time, extracting for 2 hr, adding 6 times of the amount of the materials for the second time, extracting for 1 hr, mixing the two water extractive solutions, filtering, concentrating the filtrate to obtain soft extract with specific gravity of 1.20-1.30 (measured at 60 deg.C), and collecting the soft extract;
(3) placing the cut red ginseng, acanthopanax and salvia miltiorrhiza into a multifunctional extraction tank, performing reflux extraction twice by using 40% ethanol, adding 6 times of the amount of the fed medicinal materials for the first time, extracting for 2 hours, adding 6 times of the amount of the fed medicinal materials for the second time, extracting for 1 hour, combining extracting solutions, filtering, decompressing filtrate, recovering ethanol until no alcohol smell exists, and continuously concentrating to obtain a thick paste with the specific gravity of 1.20-1.30 (measured by heat at 50-60 ℃), so as to obtain an alcohol-extracted thick paste for later use;
(4) mixing the above soft extracts, adding ethanol to 55%, stirring, standing at 2-8 deg.C for 12 hr, collecting supernatant, filtering with 80 mesh screen, recovering ethanol from filtrate under reduced pressure, concentrating to obtain soft extract with specific gravity of 1.2-1.3 (measured at 50-60 deg.C), drying, and micronizing to obtain micropowder;

Taking a proper amount of sucrose, crushing, sieving with a 80-mesh sieve, adding the micro powder, uniformly mixing, granulating with a 18-mesh sieve by using 60% ethanol as a binder, drying at 60 ℃, grading with the 18-mesh sieve, spraying the volatile oil, and mixing for 30 minutes to obtain sugar-containing granules; (sugar-free) granule preparation

Shows that: the sugar-containing granules prepared in examples 1 to 7 and the granules for tonifying deficiency and removing blood stasis prepared according to the preparation method of the national drug standard are subjected to component detection, and the active components of the sugar-containing granules prepared in examples 1 to 7 are higher than those of the granules for tonifying deficiency and removing blood stasis prepared according to the national drug standard, particularly the sugar-containing granules prepared in examples 3 and 4 are far higher than others, so that the beneficial effects of the invention are further shown.
Claims (12)
1. A Chinese medicinal composition with effects of invigorating qi, tonifying deficiency, promoting blood circulation and removing obstruction in collaterals is prepared from the following components: 86.4g of red ginseng, 343.2g of astragalus, 343.2g of acanthopanax, 172.8g of red peony root, 86.4g of red sage root, 86.4g of cassia twig and the like, and is characterized in that the traditional Chinese medicine composition is prepared by the following method: extracting volatile oil from ramulus Cinnamomi; mixing the residues after volatile oil extraction with radix astragali and radix Paeoniae Rubra, and extracting with water to obtain water extract; extracting Ginseng radix Rubri, radix Acanthopanacis Senticosi, and Saviae Miltiorrhizae radix with ethanol to obtain soft extract; mixing the water extract and ethanol extract, precipitating with ethanol, recovering ethanol, concentrating, drying, and micronizing to obtain micropowder;
the method comprises the following steps:
1.1, cutting the six medicinal materials such as red ginseng and the like into pieces for later use:
1.2, putting the crushed cassia twig into a volatile oil extractor to extract volatile oil, and storing the extracted volatile oil in another extractor for later use;
1.3, placing the broken astragalus and red paeony root and the cassia twig dregs after the volatile oil extraction into a multifunctional extraction tank, adding water for extraction twice, adding 8-10 times of the material dosage for the first time, extracting for 2-3 hours, adding 6-8 times of the material dosage for the second time, extracting for 1-2 hours, combining water extract solutions obtained in two times, filtering, and concentrating the filtrate into a thick paste with the specific gravity of 1.20-1.30 measured at 60 ℃ to obtain a water extract for later use;
1.4, placing the cut red ginseng, acanthopanax and salvia miltiorrhiza into a multifunctional extraction tank, performing reflux extraction twice by using 30-80% ethanol, adding 6-10 times of the amount of the fed medicinal materials for the first time, extracting for 2-3 hours, adding 6-8 times of the amount of the fed medicinal materials for the second time, extracting for 1-2 hours, combining extracting solutions, filtering, recovering ethanol from filtrate under reduced pressure until no alcohol smell exists, and continuously concentrating to obtain a thick paste with the specific gravity of 1.20-1.30 measured at 50-60 ℃ to obtain an alcohol extract for later use;
1.5, mixing the water extract and the alcohol extract, adding ethanol until the alcohol content is 50-90%, stirring uniformly, standing at 1-10 ℃ for 12-24 hours, taking supernatant, filtering with a 80-120 mesh screen, recovering ethanol from filtrate under reduced pressure, concentrating to obtain thick paste with the specific gravity of 1.20-1.30 at 50-60 ℃, drying, and micronizing to obtain micropowder.

The traditional Chinese medicine composition with the effects of tonifying qi and deficiency, and promoting blood circulation and removing obstruction in channels as claimed in claim 1, wherein the step of combining the cassia twig decoction dregs, the astragalus and the red paeony root after the volatile oil extraction and water extraction comprises the following steps: placing the crushed radix astragali and radix Paeoniae Rubra and the residue of ramulus Cinnamomi after extracting volatile oil in a multifunctional extraction tank, extracting with water twice, the first time adding amount of the extract 8 times the amount of the medicinal materials, extracting for 2-3 hr, the second time adding amount of the extract 6 times the amount of the medicinal materials, extracting for 2-3 hr, mixing the water extractive solutions, filtering, and concentrating the filtrate to obtain soft extract with specific gravity of 1.20-1.30 at 60 deg.C.

The traditional Chinese medicine composition with effects of benefiting qi, tonifying deficiency, promoting blood circulation and removing obstruction in channels as claimed in claim 1, wherein the red ginseng, acanthopanax senticosus and salvianic acid extract comprises the following steps: placing the cut red ginseng, acanthopanax and salvia miltiorrhiza into a multifunctional extraction tank, performing reflux extraction twice by using 30-80% ethanol, adding 6-10 times of the amount of the fed medicinal materials for the first time, extracting for 2-3 hours, adding 6-8 times of the amount of the fed medicinal materials for the second time, extracting for 2 hours, combining extracting solutions, filtering, recovering ethanol from filtrate under reduced pressure until no alcohol smell exists, and continuously concentrating to obtain a thick paste with the specific gravity of 1.20-1.30 measured at 50-60 ℃ to obtain the alcohol extract.

The traditional Chinese medicine composition with effects of benefiting qi, tonifying deficiency, promoting blood circulation and removing obstruction in channels as claimed in claim 1, wherein the red ginseng, acanthopanax senticosus and salvianic acid extract comprises the following steps: placing the crushed red ginseng, acanthopanax and salvia miltiorrhiza into a reflux extractor, carrying out reflux extraction twice by using 70% ethanol, adding 8 times of the amount of the fed medicinal materials for the first time, extracting for 2-3 hours, adding 6 times of the amount of the fed medicinal materials for the second time, extracting for 1-2 hours, combining extracting solutions, filtering, decompressing the filtrate, recovering ethanol until no alcohol smell exists, and continuously concentrating to a thick paste with the specific gravity of 1.20-1.30 measured at 50-60 ℃ to obtain the alcohol extract.

The traditional Chinese medicine composition with effects of benefiting qi, tonifying deficiency, promoting blood circulation and removing obstruction in channels as claimed in claim 1, wherein the red ginseng, acanthopanax senticosus and salvianic acid extract comprises the following steps: placing the broken red ginseng, acanthopanax and salvia miltiorrhiza in a multifunctional extraction tank, carrying out reflux extraction twice by using 75% ethanol, adding 10 times of the amount of the fed medicinal materials for the first time, extracting for 3 hours, adding 8 times of the amount of the fed medicinal materials for the second time, extracting for 2 hours, combining extracting solutions, filtering, decompressing filtrate, recovering ethanol until no alcohol smell exists, and continuously concentrating to obtain a thick paste with the specific gravity of 1.2-1.3 measured at 50-60 ℃ to obtain the alcohol extract.

The traditional Chinese medicine composition with the effects of tonifying qi and deficiency, and activating blood and dredging collaterals as claimed in claim 1, wherein the steps of combining the water extract thick paste and the alcohol extract thick paste for alcohol precipitation, recovering ethanol, concentrating, drying and carrying out superfine grinding to obtain the micro powder comprise the following steps: mixing the above soft extracts, adding ethanol to ethanol content of 80%, stirring, standing at 3-10 deg.C for 24 hr, collecting supernatant, filtering with 100 mesh screen, recovering ethanol from filtrate under reduced pressure, concentrating to obtain soft extract with specific gravity of 1.20-1.30 at 50-60 deg.C, drying, and micronizing to obtain micropowder.

1) pulverizing sucrose, sieving with 80-120 mesh sieve, adding the above micropowder, mixing, granulating with 30-90% ethanol as binder, sieving with 18-24 mesh sieve, drying at 50-70 deg.C, grading with 18-24 mesh sieve, spraying the above volatile oil, and mixing for 30-60 min to obtain sugar-containing granule;
2) sieving dextrin with 80-120 mesh sieve, adding the above micropowder and appropriate amount of correctant, mixing, granulating with 30-90% ethanol as binder and 18-24 mesh sieve, drying at 50-70 deg.C, grading with 18-24 mesh sieve, spraying the above volatile oil, and mixing for 30-60 min to obtain sugar-free granule.

The traditional Chinese medicine composition with the effects of tonifying qi and deficiency, and activating blood and dredging collaterals according to claim 9, which is characterized in that the micro powder is taken, added with a diluent, a binder, a disintegrating agent and a lubricant and prepared into tablets according to a conventional preparation method of the tablets.
