技术领域
本发明涉及包含信息素试剂组合的组合物,以及使用这种组合物改变动物行为的方法。更具体的,本公开涉及一种信息素组合物,其含有杂环芳族化合物与至少一种雄激素类固醇的组合,以及该组合物刺激或诱导母猪中的生殖行为的用途。
背景
信息素信号已知在哺乳动物生殖和行为中起到主要作用。在猪的情形中,当雌性发情会使用信息素以及明显行为(例如脊柱前凸或直立反射)时,它们会发送信号,招揽雄性接近。
早期的科学文献指出,唾液腺类固醇分子雄烯酮和雄甾烯醇是已被认识的公猪信息素,其负责诱导母猪中的生殖(见Melrose等,“雄激素类固醇与公猪气味结合,作为猪人工配种中检测发情的辅助”,The British Veterinary Journal,127,497-502(1971);另见Perry等,猪求偶行为:雄烯类固醇在雄性颌下腺分泌中的信息素性质,AnimalProduction,31,191-199(1980))。含有雄烯酮的BoarmateTM(ITSI,加拿大)在市场上已长期销售,用于检测发情中的母猪并在猪育种中节约时间和金钱。
然而,市场上存在的合成产品在复制公猪唾液以诱导母猪生殖行为和表现中并不总是成功。因此,决定母猪是否在发情变得越来越费力,费钱,和无效,因为可能需要圈养更多公猪,可能难以发现发情母猪,并且生殖成功率将可能下降。
因此,在业内需要一种合成的公猪唾液组合物,其在母猪中诱导生殖行为的可能性更大,提高生殖成功率和生产率上有更高的可能性。
发明内容
以下联合系统、工具和方法描述和说明的实施方式和方面是示例性和说明性的,不限制范围。在许多实施方式中,上述的一个或多个问题已被减轻或消除,而其他实施方式针对其他改进。
本发明的目的是提供在雌性猪属动物(suid)中诱导生殖行为的新方法和组合物,包含杂环芳族化合物和至少一种雄激素类固醇的组合。
本发明的组合物优选包含雄烯酮、雄甾烯醇和喹啉的组合,可任选地包含必需或所需的其他成分,这取决于终产物的形式和希望的用途。这样的任选的成分可包含但不限于,运载体(诸如水、醇类、溶剂等);芳香剂、着色剂、防腐剂和抗氧化剂等。所得产物的示例包括但不限于喷雾、扩散器或气雾剂。另外,所得的产品可以是泡沫、浸涂物、擦拭巾、乳膏、凝胶或乳液。

具体实施方式
本公开的组合物包含信息素和芳族化合物的组合,该组合在本文中统称为“信息素试剂”,其组合物已被发现能有用和有效地诱导或刺激雌性猪属动物中的生殖行为。

I.信息素组合物
本公开的组合物一般包含信息素试剂组合。本文公开的信息素试剂的独特组合已被发现能惊人有效地诱导雌性猪属动物中的生殖行为。
在本发明组合物中使用的信息素试剂包含至少一种类固醇激素。根据结合的受体,类固醇激素可分成五类:糖皮质激素、盐皮质激素、雄激素、雌激素和孕激素。用于本发明的类固醇激素可以是天然或合成的。优选地,类固醇激素包含至少一种雄激素类固醇,其可选自雄烯酮、雄甾烯醇、雄烯二酮、雌四烯醇及其组合。优选的,雄激素类固醇是雄烯酮和雄甾烯醇的组合。
通常,所述制剂中存在的类固醇激素的量是总组合物的至少0.0000001%(w/w)。在一个实施方式中,所述组合物中存在的类固醇激素浓度是约0.0000001%-约10%(w/w)。在另一个实施方式中,所述组合物中存在的类固醇激素浓度是约0.0001%-约5%(w/w)。优选地,所述组合物中存在的类固醇激素的浓度是约0.0001%-约1%(w/w),并且最优选地,所述组合物中存在的类固醇激素的浓度是约0.0008%(w/w)。
存在于组合物中的信息素试剂还包括芳族杂环化合物。用于本发明的优选的芳族杂环化合物是喹啉。存在于制剂中存在的芳族杂环化合物的量是总组合物的至少0.0000001%(w/w)。在一个实施方式中,所述组合物中的芳族杂环化合物浓度是约0.0000001%-约10%(w/w)。在另一个实施方式中,所述组合物中的芳族杂环化合物浓度是约0.0001%-约5%(w/w)。优选地,所述组合物中存在的芳族杂环化合物的浓度是约0.0001%-约1%(w/w),并且最优选地,所述组合物中的芳族杂环化合物的浓度是约0.0004%(w/w)。

在一个实施方式中,该组合物可包含至少一种运载体溶剂。合适的运载体溶剂通常为本领域已知并且公认包括亲脂型有机稀释剂、醇类、乙二醇、丙二醇、双丙甘醇、醚、氯仿、苯、二硫化碳、油类(包括非挥发性和挥发性液体和油类)、水,及其组合。合适的醇类包括乙醇、丙醇、异丙醇、丁醇、戊醇、己醇、庚醇、辛醇和苯乙醇。在一个实施方式中,醇可包含乙醇、异丙醇、丁醇和苯乙醇。在示范性实施方式中,溶剂包含异丙醇。醇溶剂可以与水或亲脂性有机稀释剂或运载体(诸如乙二醇、丙二醇、双丙甘醇、双丙甘醇单乙基醚、双丙甘醇甲基醚或DowQ7-9180硅酮液体)合并。在一个示范性实施方式中,溶剂可以是水和选自乙醇或异丙醇的醇的组合。在一个实施方式中,所述组合物中存在的溶剂的量的范围可以是总组合物的约0.5%-99.99%(w/w)。存在于组合物中的水的量的范围可以是总组合物的约70%-99.99%(w/w),例如总组合物的约80%-98.5%(w/w)。存在于组合物中的醇的量范围可以是总组合物的约1%-20%(w/w),例如总组合物的1.5%-10%(w/w)。
所述组合物可另包含推进剂。合适的推进剂包括含氯氟烃(CFC)(诸如三氯一氟甲烷、二氯二氟甲烷和二氯四氟乙烷);氢氯氟烃(HCFC)或氢氟烃(HFC)(如氯代二氟甲烷、三氟一氟乙烷、氯代二氟乙烷、二氟乙烷和七氟丙烷);烃类(如丙烷、丁烷和异丁烯);以及其他压缩气体(如氮气、二氧化碳和一氧化二氮),以及上述推进剂的任何组合。在一个实施方式中,推进剂可以是丙烷。在另一个实施方式中,所述推进剂可以是1,1-二氟乙烷。所述推进剂可以是不包含致癌化合物的惰性气体,包括1,1,1,2-四氟乙烷、氯代二氟甲烷和二氯二氟甲烷。推进剂可具有小于约-50℃的闪点。通常当在组合物中包含推进剂时,其可以是总组合物的约75%-99.99%(w/w),例如是总组合物的约85%-99.99%(w/w),或总组合物的约95%-99.99%(w/w)。
组合物可任选包含一种或多种表面活性剂。表面活性剂通常用于制备配制成乳液的组合物。可配制成油包水或水包油乳液。合适的表面活性剂的示例包括:非离子型乙氧基化和非乙氧基化的表面活性剂、松香酸、杏仁油PEG、蜂蜡、丁基葡糖苷癸酸酯(butylglucoside caprate)、C18-C36酸二醇酯、C9-C15烷基磷酸酯、癸酸/辛酸甘油三酯PEG-4酯、鲸蜡硬脂醇-7、鲸蜡醇、磷酸鲸蜡醇酯、谷物油PEG酯、磷酸DEA-鲸蜡醇酯、糊精月桂酸酯、二月桂醇聚醚-7柠檬酸酯、磷酸二肉豆蔻酯、甘油聚醚-17椰酸酯、芥酸甘油酯、月桂酸甘油酯、氢化蓖麻油PEG酯、异硬脂醇聚醚-11羧酸、卵磷脂、溶血卵磷脂、壬苯醇醚-9、辛基十二醇聚醚-20、棕榈油酸甘油酯、PEG二异硬脂酸酯、PEG硬酯胺(stearamine)、泊洛沙胺(poloxamine)、聚甘油、亚油酸钾、PPG、棉子糖肉豆蔻酯、己酰乳酰乳酸钠、辛酸钠、椰酸钠、异硬脂酸钠、生育酚磷酸钠、硬脂醇聚醚、TEA-C12-C13链烷醇聚醚-3硫酸酯、三-C12-C15链烷醇聚醚-6磷酸酯和十三烷醇聚醚。
在某些应用中,可能需要使所述组合物增稠。合适的增稠或增粘剂包括:丙烯酰胺共聚物、琼脂糖、支链淀粉、膨润土、藻酸钙、羧甲基纤维素钙、卡波姆(carbomer)、羧甲基壳多糖、纤维素胶、糊精、明胶、氢化牛脂、羟乙基纤维素、羟丙基纤维素、羟丙基淀粉、棕榈酸异丙酯、藻酸镁、甲基纤维素、微晶纤维素、果胶、各种PEG、聚丙烯酸、聚甲基丙烯酸、聚乙烯醇、各种PPG、丙烯酸钠共聚物、角叉菜胶钠、黄原胶和酵母β-葡聚糖。当存在时,增稠剂的量可以是总组合物的约1%-30%(w/w),例如总组合物的约5%-20%(w/w),或总组合物的约10%-15%(w/w)之间。
组合物还可额外包含乳化剂或增稠剂,例如蒸馏的单和二甘油酯,丙二醇单酯、硬酯酰-2-乳酸钠、聚山梨醇酯60、卵磷脂、羟基化卵磷脂、羊毛脂、和任何其他工业上已知和使用的乳化剂。
所述组合物还可包含芳香剂。芳香剂可以是任何提供理想臭味掩蔽效果,而不显著影响组合物的主要性质的芳香剂。虽然可以采用多种芳香剂而不偏离本公开的范围,合适的芳香剂包括花香型香精、柑橘花、针叶树的油或提取物,或香料。花香型香精的示例包括玫瑰、丁香、薰衣草、栀子花和茉莉。合适的柑橘花包括橘和柠檬,而合适的针叶树的油或提取物包括松树和杜松。芳香剂通常可占所述总组合物的约0.25%-约1%(w/w)。
II.给予途径
哺乳动物(包括猪和其他猪属动物)有几个接收嗅觉信号的解剖学器官。两种主要的“嗅觉”器官是主要嗅觉上皮(MOE)和犁鼻器(VNO)。其他鼻腔内的感觉纤维可以感觉气味,但是主要嗅球和辅助嗅球(接收来自VNO的信号)是主要整合系统。
嗅球位于脑前部。其发出神经元投射穿过骨骼并使这些突起延伸进入嗅觉上皮。MOE是富含血液供给和粘膜的大片区域,其中使气味气雾剂分子透过进入肺部。气味或水滴会沉降在MOE上,并且如果存在气味受体,则该气味受体会被结合并且导致感觉神经元的激活。
通常通过使对象雌性猪属动物吸入有效量组合物来递送的任何方法来向对象动物给予组合物。这些给予方法包括例如,在猪属动物的环境内放置或分布组合物,通过在猪属动物生活环境的表面或直接在猪身上,例如其面部区域或头部施加(例如通过喷雾或擦拭)。例如,可以使用气雾剂、泵式喷雾、泡沫、项圈、擦拭巾、浸涂物、液体、凝胶、洗剂和/或乳膏向猪属动物局部给予信息素组合物。
术语“有效量”描述了存在于组合物中的信息素试剂的量,其足以产生如本文所述的行为观察所决定的可注意的效果,例如对象猪属动物从事生殖行为。有效量将取决于以下因素:如个体动物参数,包括年龄、身体情况、大小和体重;并行处理;处理频率;或给予模式。这些因素为本领域普通技术人员熟知。
在一个实施方式中,信息素试剂可以与溶剂合并形成液体溶液并且该液体溶液可以进一步制备成各种适于通过吸入递送给猪属动物的制剂。例如,液体溶液可以进一步按照本领域熟知的方法制备,如泵式喷雾、气雾剂、凝胶、泡沫、乳膏、洗剂、凝胶、或扩散体。
在示范性实施方式中,信息素试剂溶解或稀释于非水性有机溶剂或溶剂混合物中,形成掺入泵式喷雾的溶液。一种示范性的泵式喷雾溶液可含有约0.0012%(w/w)信息素试剂,约5%(w/w)异丙醇,约3.5%(w/w)羊毛脂油,约91%(w/w)水。另外,可以向溶液添加约0.25%(w/w)的防腐剂。
在另一个实施方式中,用溶剂和增稠剂溶解或稀释信息素试剂来形成用于扩散体的溶液。示范性的扩散体溶液可包含约0.0012%(w/w)的信息素试剂、约80%(w/w)-约85%(w/w)的溶剂、和约15%(w/w)的增稠剂。
III.在雌性猪属动物中使用信息素组合物以诱导生殖行为的方法
本公开还涉及用信息素组合物诱导或刺激雌性猪属动物中的生殖行为或生理学的方法,通过使猪属动物接触有效量的组合物,该组合物包含信息素试剂的组合,其中该组合物在总组合物中包含至少约0.00001%(w/w)信息素试剂。通常,组合物可包含占所述总组合物的约0.0001%-约1%(w/w)的信息素试剂。
可通过任何使猪属动物在一段根据行为学观察足以诱导生殖的时间内吸入组合物的方法,使猪属动物接触组合物。通常,根据选择的给予途径、特定猪属动物和情况,组合物对猪暴露的时间段将是至少1秒,但也可以是至少1小时、1小时-5小时、至少1天、至少1周、1周-4周、至少1个月或达到满意效应可能需要的任何时间段。
可通过例如,对对象猪属动物喷洒气溶胶喷雾(该喷雾可通过例如手动、机械或自动化喷雾装置分散),或将组合物掺入,或将组合物以液体扩散体形式等提供到猪属动物的物理区域,来测试组合物的效力。在任何情况下,该组合物具备足够的挥发性以供猪属动物吸入从而暴露至足够量的组合物以产生明显的行为效应。
在使用时,包含信息素试剂组合的组合物可以根据目标猪属动物来实现。例如,可在例如,猪属动物的鼻孔、面部、头部,其环境,和/或其饲料、给料器或水中喷射含有雄烯酮、雄甾烯醇和喹啉组合的示例性液体喷雾制剂(溶于合适的溶剂中),从而其可以通过嗅觉被感觉到,以所需频率进行所述处理以得到希望的效果。
应理解用于组合物的信息素试剂组合可以纯浓缩物(100%浓度)形式,或用其它赋形剂稀释的组合物形式的剂型提供(即所述组合物中活性成分的量低于或等于99.99%,剩余部分由无活性的赋形剂组成)。如果稀释,在各种剂型中分配的信息素试剂的量可以是约1.0pg/mL-约1.0g/mL,例如约1.0ng/mL-约1.0g/mL。本领域技术人员会理解,考虑到稀释,需调节添加到组合物的活性组分的体积并确保最终组合物包含适当终浓度的信息素试剂。可以各种剂型提供组合物的各种成分,包括但不限于,溶液或悬液,乳液,气溶胶等。
本公开的组合物可以多种方式施用,但是最好通过诸如以下的任何方式通过暴露嗅觉系统来施用,例如,直接在动物的面部区域上或环境中喷射薄雾。另外,本公开的方法最好通过让动物吸入组合物来进行,由于动物的鼻腔、鼻窦、肺和喉具有供芳族分子与嗅觉受体结合的大片面积。该组合物对动物或动物环境的施用可以根据所需频率重复以引起所需效果。

“猪属动物”指任何偶蹄目,有蹄类猪科(Suidae)的成员,包括野生和家养的猪、鹿豚、公猪、灌木猪、猪(swine)和疣猪。猪属动物是具有小眼睛和粗砺(有时稀疏)毛发的肥硕动物。它们都具有止于圆形软骨的鼻口部分,用于挖掘食物。有些物种具有獠牙。猪属动物是杂食性的,通常是群居动物。

实施例
实施例1-包含信息素试剂的喷雾组合物的制备
根据表1列出的配方制备含有信息素试剂雄烯酮、雄甾烯醇和喹啉组合的泵式喷雾制剂。

实施例2-确定包含雄烯酮、雄甾烯醇、和喹啉的喷雾组合物在母猪中诱导生殖,从而提高生殖成功率和生产率的效果
该研究评估了雄烯酮、雄甾烯醇和喹啉单独和组合的效力,以确定其在母猪中诱导生殖的效力。本研究的目的是确定是否雄烯酮、雄甾烯醇和喹啉作为信息素试剂的组合比每种信息素试剂单独或与另一种信息素试剂联合(如表2所示)可更有效诱导生殖行为,从而提高母猪中的生殖成功率和生产率。

用相同比例的雄烯酮、雄甾烯醇和喹啉以各种组合如表2所示制备了七(7)种不同的喷雾组合物,以及仅用异丙醇制备了对照溶液。全部成分以4.0μg/ml混合,并置于异丙醇载剂溶液中。
在大型商业养猪场测试含雄烯酮、雄甾烯醇和喹啉的喷雾组合物的效力。监督养猪场工人给母猪断奶,并每周配种2-4次。喂食时间后,或在上午7.00-8:30之间,通过使1-2头公猪走过(使用公猪机器人(Boar Bot,一种推或拉活公猪的机器人装置,以提供母猪“公猪体验”)),测试动物发情,并让工人对每只动物进行背压测试。背压测试用于测定动物是否处于授精的最佳状态。测试包括在母猪背部施加重压和摩擦以模仿公猪在交配时的压力。发情的母猪将通过锁紧和拱起它们的背部,整个身体下沉和头部运动,垂直竖起耳朵和咕哝来响应。已确定通过了背压测试的动物在上午9-10点之间被移到单独的猪圈内,用于人工授精。
根据观察,断奶后的母猪通常在断奶后第四天进入发情,用杀虫剂喷雾器对每个猪圈内的母猪喷洒信息素组合物或对照,并用背压测试检测。总共测试了947只母猪。在养猪场工人测试发情迹象前1-2个小时对母猪进行喷雾。对母猪喷雾和检查后,让一只公猪跑过猪圈,标记发情的动物并将其移到人工授精猪圈。喷雾过程对剩余的动物在第5日继续。在断奶后第4和第5天观察总共947只母猪的生殖行为。
组合三种最佳的确定母猪对公猪授精的接受度的测量值(直立、脊柱前凸和竖耳)来建立总体行为评分。表3和图1描述了脊柱前凸行为的单独行为发现。表4和图2描述了直立行为的发现。表5和图3代表了竖耳表达的母猪的数据。表6和图4代表了生殖行为的总体行为评分

加入行为和天数间的排名以后,确定了含有雄烯酮、雄甾烯醇和喹啉组合的制剂在母猪中显示了最强的整体反应。因此,这些结果说明本发明有效刺激母猪中的生殖行为,其相应提高生殖成功率和生产率。

Claims (16)
1.一种用于在雌性猪属动物中刺激生殖行为的组合物,所述组合物包含信息素试剂组合,其中所述信息素试剂组合是雄烯酮、雄甾烯醇、和喹啉,其中所述雄烯酮和雄甾烯醇分别以所述组合物的0.0001%-5% (w/w)的量存在,所述喹啉以所述组合物0.0001%-5% (w/w)的量存在。
2.如权利要求1所述的组合物,其特征在于,所述雄烯酮和雄甾烯醇分别以所述组合物的0.0001%-1% (w/w)的量存在。
3.如权利要求1所述的组合物,其特征在于,喹啉以所述组合物0.0001%-1% (w/w)的量存在。
4.如权利要求1所述的组合物,其特征在于,所述组合物还包含运载体溶剂。
5.如权利要求4所述的组合物,其特征在于,所述运载体溶剂选自下组:亲脂性有机稀释剂、水,及其组合。
6.如权利要求4所述的组合物,其特征在于,所述运载体溶剂选自醇类、醚、氯仿、苯、二硫化碳、油,及其组合。
7.如权利要求6所述的组合物,所述油是非挥发性或挥发性液体。
8.如权利要求6所述的组合物,其特征在于,所述醇选自下组:乙醇、丙醇、异丙醇、丁醇、戊醇、己醇、庚醇、辛醇、苯乙醇、 乙二醇、丙二醇、双丙甘醇,及其组合。
9.如权利要求1所述的组合物,其特征在于,所述组合物还包含选自下组的其它成分:表面活性剂、增稠剂、推进剂、芳香剂及其组合。
10.一种在雌性猪属动物中刺激生殖行为和提高生殖成功率和生产率的方法,包括:对所述猪属动物给予信息素组合物,所述组合物包含信息素试剂组合,其中所述信息素试剂组合是雄烯酮、雄甾烯醇、和喹啉,其中给予动物的所述信息素试剂的量在1.0ng/ml到1.0g/ml之间;其中所述雄烯酮和雄甾烯醇分别以所述组合物的0.0001%-5% (w/w)的量存在,所述喹啉以所述组合物0.0001%-5% (w/w)的量存在。
11.如权利要求10所述的方法,其特征在于,所述猪属动物是母猪。
12.如权利要求10所述的方法,其特征在于,所述给予方法是吸入给予。
13.如权利要求10所述的方法,其中,所述组合物被配制成喷雾。
14.如权利要求10所述的方法,其特征在于,所述喷雾组合物的给予包括对所述猪属动物喷射或对所述猪属动物的环境喷射所述组合物。
15.一种在雌性猪属动物中刺激生殖行为和提高生殖成功率和生产率的方法,包括:对所述猪属动物给予有效量的信息素喷雾组合物,所述喷雾组合物包含0.0004%(w/w)雄烯酮、0.0004%(w/w)雄甾烯醇、0.0004%(w/w)喹啉、5%(w/w)异丙醇、和91%(w/w)水。
16.如权利要求15所述的方法,其特征在于,所述猪属动物是母猪。
