Abstract

技术领域
本发明涉及药品食品领域,具体而言,涉及一种用于改善男性不育症的组合物及其制备方法和用途。
背景技术
临床上把男性不育症分为性功能障碍和性功能正常两类,后者依据精液分析结果可进一步分为无精子症、少精子症、弱精子症、精子无力症和精子数正常性不育。
目前国内外利用西医治疗男性不育症的手段虽然丰富,但其价格昂贵,病人痛苦,或长期服用毒副作用大,又常易继发其他疾病。因此越来越多的人希望通过中医药的研究为该疾病的治疗带来更好的效果。但现有的多种治疗男性不育症的中药治疗效果尚不够理想,且不宜长期服用。因此,需要提出一种具有更好疗效、且可长期服用的治疗男性不育症的药品或保健品。
发明内容
本发明的目的在于提供一种用于改善男性不育症的组合物及其制备方法和应用,这种组合物能够促进男性精子活力和改善生殖能力,解决了现有植物蛋白粉类产品中营养配方单一、吸收利用率低的问题。
为了实现本发明的上述目的,特采用以下技术方案:

大豆蛋白粉1500~1550份、膳食纤维180~200份、蛹虫草15~30份、人参150~200份、黄精45~55份、玛咖90~105份、左旋肉碱1400~1600份、牡蛎95~105份。
一种用于改善男性不育症的组合物的制备方法,其包括:
按重量份数计,将所述大豆蛋白粉、所述膳食纤维、所述蛹虫草、所述人参、所述黄精、所述玛咖、所述左旋肉碱和所述牡蛎混合30~80min后,过40~80目筛。
一种上述组合物在制备改善或促进男性不育症的药品或者保健品中的用途。

本实施方式提供一种用于改善男性不育症的组合物,其按重量份数计包括:
大豆蛋白粉1500~1550份、膳食纤维180~200份、蛹虫草15~30份、人参150~200份、黄精45~55份、玛咖90~105份、左旋肉碱1400~1600份、牡蛎95~105份。
进一步的,按重量份数计,大豆蛋白粉为1510~1540份、膳食纤维为180~190份、蛹虫草为18~25份、人参为160~190份、黄精为47~53份、玛咖为95~103份、左旋肉碱为1450~1550份、牡蛎为97~103份。
进一步的,按重量份数计,大豆蛋白粉为1520~1530份、膳食纤维为182~187份、蛹虫草为18~22份、人参为170~180份、黄精为48~51份、玛咖为97~101份、左旋肉碱为1480~1520份、牡蛎为99~102份。
其中,膳食纤维是一种复杂的混合物,其含有独特的生物活性物质混合物,包括抗性淀粉、维生素、矿物质、植物化学物质和抗氧化成分。膳食纤维具有降血糖、降血脂、抗癌、改善肠道菌群、治疗便秘、利于减肥以及替代脂肪等的作用。水溶性膳食纤维的有效功能成分在保障人体正常生理功能方面作用显著,它可降低胆固醇水平和血压,防治胃肠问题的产生。
蛋白质是生命的物质基础,是身体不可或缺的物质,大豆蛋白属于优质蛋白。蛋白质作为小分子物质调节生理功能,维持新陈代谢,在男性生殖过程中起重要作用,男性一旦缺失蛋白质,会导致精子质量下降,精子活力降低以及精子不液化造成男性不育。蛋白质含有生物活性物质,能够预防前列腺癌。
蛹虫草,又名北虫草,属子囊菌亚门、核菌纲、球壳目、麦角菌科、虫草属真菌。蛹虫草中含有虫草素、虫草酸、核苷类、多糖类、糖醇、甾醇类各种氨基酸和微量元素以及SOD酶在内的多种有效成分,在免疫调节、抗肿瘤、抑菌抗炎、抗氧化衰老等方面有很强的活性,具有重要的研究和开发价值。尤为重要的是,蛹虫草可以增加附睾重量、精子数量和提高精子能动性、精子运动的连续性,改善精子的质量,增强性欲,促进生殖器官的蛋白质合成,对机体多个系统均有广泛影响,能够促进代谢,改善生殖功能。
人参,为五加科人参属植物,自古誉为“百草之王”“滋阴补生,扶正固本”之极品,《神农本草经》记载,人参性温、味甘、微苦,能“补五脏,安精神,定魂魄,止惊悸......”。人参药材中的主要有效成分为人参皂苷和人参多糖。人参在中药里,一般用作强壮剂,可以补养元气。且人参具有增强性腺机能的作用。人参酊对于麻痹型、早泄型阳痿有显著的疗效,对因神经衰弱所引起的皮层性和脊髓性阳痿也有一定治疗效果。此外,人参还具有提高免疫力、增强性行为等作用,能够用于改善性生活和治疗性功能障碍(尤其是勃起功能障碍),且无副作用。同时,人参作用于脑部,可增强性兴奋,激活控制勃起的行为神经;人参作用于阴茎,增加NO(一氧化氮是阴茎勃起的关键物质,在阴茎勃起过程中起重要调控作用)释放,引起平滑肌松弛,提高血流量而发生勃起,提高阴茎硬度和延长勃起持续时间。
黄精,为百合科植物滇黄精、黄精或多花黄精的燥根茎。黄精味甘、性平,具有补气养阴,健脾,润肺,益肾的功效。《本草纲目》中记载:“黄精宽中意气,使五脏调良,肌肉充实,骨髓坚强,其力增倍,多年不老。颜色鲜明,发白更黑,齿落更生。”黄精主要有黄精多糖、甾体皂苷等多种功效成分。黄精具有抗病原微生物、抗疲劳等功效,有抗氧化、抗衰老等功能作用,与蛹虫草和人参共同使用,可协同性地改善精子质量,治疗少精子症和弱精子症,增加精液量,提高精子密度、精子活动力和精子存活率。
玛咖Lepidium meyenii Walp.为十字花科独行菜属植物,药用地下块根,为一年生或两年生草本。其传统上被用做性欲激活剂,具有“植物伟哥”的美名,与治疗性功能的其他药物相比具有安全无副作用的优势。玛咖含有甾醇、生物碱、玛咖酰胺、玛咖烯、芥子油苷及其衍生物等功能性成分,其具有促进性欲、改善性功能、提高生育力、增强免疫力、抗疲劳、抑制前列腺增大等作用。玛卡富含高单位营养素,对人体有滋补强身的功用,食用过的人会有体力充沛、精神旺盛不会疲劳的感觉。当玛咖与黄精、蛹虫草和人参共同使用时,可进一步增强其改善男性生殖功能的能力。
左旋肉碱(L-carnitine),又称左卡尼汀、肉毒素或维生素BT,是一种促使脂肪转化为能量的类氨基酸。左旋肉碱作为转运脂肪酸进入线粒体的重要载体,具有参与精子能量代谢、降低精浆中活性氧物质、稳定精子细胞膜、抗精子凋亡等作用。左旋肉碱在男性生殖中具有重要作用,如启动精子运动、促进精子成熟和提高精子受精能力,调节Sertoli细胞功能,保护精子对抗活性氧(ROS)诱导的氧化损伤,减少生精细胞凋亡,抑制子聚集,而ROS增多引起精子膜脂质过氧化,导致精子运动和形态学改变,甚至细胞死亡。同时,左旋肉碱与精子密度、活动率、正常形态精子百分率等精液参数之间存在相关性。左旋肉碱可显著提高弱精子症患者前向精子活动率、总活动率、曲线运动速度、直线运动速度以及平均轨迹速度等主要精液参数指标,明显改善精子活力、活率及正常形态精子百分率。
牡蛎中,含有锌,其是人体必需的微量元素之一,在人体生长发育、生殖遗传、免疫、内分泌等重要生理过程中起着极其重要的作用。锌在男性生殖器官中的浓度非常高,特别是在前列腺和精液中的含量明显高于其他器官或体液,正常前列腺液或精液中的锌含量大约相当于血浆中的100倍。锌是铜/锌-SOD的辅助因子。锌直接参与精子的生成、成熟或获能过程,对精子活力、代谢及其稳定性等起重要作用,有利于受精。另外,锌对精子头尾连接起着非常重要的作用。锌离子是钙镁依赖性核酸内切酶的抑制因子,缺锌时可通过激活此酶而诱发细胞凋亡,并影响睾丸的发育和功能。
进一步的,该组合物按重量份数计还包括:
紫菜粉450~550份、番茄粉45~55份、菠菜粉145~155份、芝麻粉3~5份、白砂糖640~650份。
进一步的,按重量份数计,紫菜粉为470~530份,番茄粉为47~53份,菠菜粉为147~153份,芝麻粉为4~5份,白砂糖642~647份。
其中,紫菜中含有牛磺酸,牛磺酸是机体生殖腺和生殖细胞中含量最丰富的游离氨基酸之一,对机体生殖机能具有重要的作用。牛磺酸是精子活力激动剂,在精液中添加牛磺酸可增加精子体外存活时间,保持精子的正常运动;在精液稀释液中添加牛磺酸,可提高冷冻精液中精子的成活率,增加精子活力。睾丸组织可生物合成牛磺酸,机体补充牛磺酸可促进睾酮的分泌,显著提高精液品质;并可提高机体内NO的含量,显著增强性功能。
番茄中含有番茄红素,其是植物中所含的一种天然亲脂性类胡萝卜素。主要存在于茄科植物西红柿的成熟果实中。它是目前在自然界的植物中被发现的最强抗氧化剂之一。人体内的单线态氧和氧自由基是侵害人体自身免疫系统的罪魁祸首。番茄红素清除自由基的功效远胜于其他类胡萝卜素和维生素E,其淬灭单线态氧速率常数是维生素E的100倍。其可以有效的防治因衰老,免疫力下降引起的各种疾病。番茄红素在睾丸中的含量高于其它器官。番茄红素能增加精子的数量和改善精子的质量,提高男性生育能力,降低机体的脂质过氧化和DNA损伤。血浆番茄红素浓度与精浆番茄红素浓度正相关,血浆、精浆番茄红素浓度可以影响精子活动力。
菠菜中含有辅酶Q10(泛醌),其是电子传递链的重要组成部分,参与细胞有氧呼吸,产生三磷酸腺苷(ATP)。心脏、肝脏和肾脏等高耗能器官的组织细胞中具有高浓度的辅酶Q10。精子细胞和精浆中都有一定量的辅酶Q10存在,其浓度水平与精子某些参数存在一定相关性。辅酶Q10能促进精子线粒体中能量产生,中和产生的活性氧,精液中辅酶Q10水平的降低可能会引起精子能动性下降。辅酶Q10能有效改善精浆的抗氧化能力,降低氧化应激有关的DNA损伤,增强人体免疫力,提高精子质量和整体受孕率。
芝麻粉中含有硒,硒是人体必需的微量元素,是谷光甘肽过氧化物酶(GSH-Px)的活性中心元素,具有抗氧化、增强免疫力、改善精子质量等作用。硒在男性不育患者精液中的浓度明显低于正常生育能力的男性。硒在男性生殖发育和改善精子活力、维持精子正常形态和功能中有重要作用。随着男性性腺的发育成熟,性腺中硒含量明显上升,对维持精子鞭毛结构和功能的完整性有重要作用。另外,硒能够参与清除体内活性氧、保护DNA免受活性氧引起的氧化损伤。因此,精液缺硒引起的精子质量下降可能与缺硒引起的抗氧化作用下降有关。在不育者饮食中添加适量的硒,可明显改善精子活力和活动精子百分率,并降低精子畸形率。精液硒浓度与精子总数、精子活力、a级精子百分率之间呈明显的正相关。

其中,维生素C又称抗坏血酸,是存在于精液中的一种抗氧化剂,能减轻抗坏血酸过氧化物酶基底的氧化应力,可以提高精子质量,在人体内参与许多重要的代谢。不育男性精液中维生素C水平显著降低,通过在食物中补充维生素C能提高25岁以上男性精子的存活力和运动力,增加成熟精子数,减少精子畸形率。
维生素E是一种脂溶性维生素,又名生育酚,它分为生育酚(tocopherols)和生育三烯酚(tocotrienols)两类。维生素E作为脂溶性抗氧化剂和自由基清除剂,主要对抗生物膜上脂质过氧化所产生的自由基,保护生物膜的结构和功能。维生素E是生育过程中一个必需营养成分,也是生殖系统最基本的抗氧化物质。补充维生素E可以提高男性精子的浓度、活力及形态正常精子百分率,显著减少畸形精子的比率,从而提高自然怀孕率。
维生素B12与叶酸作为合成DNA的重要辅酶,在组织的合成与修复,以及精子的形成都起了重要的作用。生育男性精浆中低浓度的叶酸、维生素B12与多数DNA损伤之间有密切的关系。维生素B12和叶酸共同参与同型半胱氨酸的甲基化过程,维生素B12或叶酸缺乏都将使叶酸依赖性合成产物(胸苷酸和嘌呤)的生成受阻,从而导致DNA合成障碍。人睾丸中的精子细胞是以极高的速率合成DNA的,每天大约产生数千万精子。维生素B12或叶酸缺乏有可能导致精子DNA合成下降,从而影响精子质量,并进而影响男性生殖功能。
本实施方式还提供一种用于改善男性不育症的组合物的制备方法,其包括:
按重量份数计,将大豆蛋白粉、膳食纤维、蛹虫草、人参、黄精、玛咖、左旋肉碱和牡蛎混合30~80min后,过40~80目筛。
进一步的,其包括:
先将按重量份数计的大豆蛋白粉和左旋肉碱混合25~35min后,过40~80目筛,得到第一物料;
再将膳食纤维、蛹虫草、人参、黄精、玛咖、和牡蛎混合,过40~80目筛,得到第二物料;
将第一物料与第二物料混合40~60min。
以下结合实施例对本发明的特征和性能作进一步的详细描述:
实施例1
一种用于改善男性不育症的组合物,其包括:
大豆蛋白粉1500mg、膳食纤维200mg、蛹虫草15mg、人参150mg、黄精55mg、玛咖90mg、左旋肉碱1600mg、牡蛎105mg。
该组合物的制备方法包括:
a.称取大豆蛋白粉和左旋肉碱后,在混合机内混合25min,过80目筛,得到第一物料;
b.称取膳食纤维、蛹虫草、人参、黄精、玛咖、和牡蛎混合,过80目筛,得到第二物料;
c.将第一物料与第二物料混合40min;
d.检验合格后用三封(背封)包装机进行自动计量充填,再装塑胶篮(送整合室),检验合格入箱。
实施例2
一种用于改善男性不育症的组合物,其包括:
大豆蛋白粉1520mg、膳食纤维187mg、蛹虫草24mg、人参170mg、黄精49mg、玛咖97mg、左旋肉碱1520mg、牡蛎99mg。

大豆蛋白粉1550mg、膳食纤维180mg、蛹虫草30mg、人参200mg、黄精45mg、玛咖105mg、左旋肉碱1400mg、牡蛎95mg。

一种用于改善男性不育症的组合物,其包括:
大豆蛋白粉1510mg、膳食纤维190mg、蛹虫草为18mg、人参190mg、黄精53mg、玛咖95mg、左旋肉碱1550mg、牡蛎97mg、紫菜粉450mg、番茄粉55mg、菠菜粉155mg、芝麻粉5mg、白砂糖650mg。
该组合物的制备方法包括:
a.称取大豆蛋白粉和左旋肉碱后,在混合机内混合25min,过40目筛,得到第一物料;
b.称取膳食纤维、蛹虫草、人参、黄精、玛咖、牡蛎、紫菜粉、番茄粉、菠菜粉、芝麻粉和白砂糖混合,过40目筛,得到第二物料;
c.将第一物料与第二物料混合40min;

该组合物的制备方法包括:
a.称取大豆蛋白粉和左旋肉碱后,在混合机内混合25min,过60目筛,得到第一物料;
b.称取膳食纤维、蛹虫草、人参、黄精、玛咖、牡蛎、紫菜粉、番茄粉、菠菜粉、芝麻粉和白砂糖混合,过60目筛,得到第二物料;
c.称取维生素C、维生素E、叶酸、维生素B12,混合后,得到第三物料;
c.将第一物料、第二物料与所述第三物料混合45min;
d.检验合格后用三封(背封)包装机进行自动计量充填,再装塑胶篮(送整合室),检验合格入箱。
