Abstract

本发明提供了一种用于发酵豆粕的复配酶制剂及其在豆粕发酵过程中的应用。该复配酶由中性蛋白酶,纤维素酶,木聚糖酶,β‑葡聚糖酶,甘露聚糖酶,葡萄糖氧化酶组成。其中含量按成分的重量百分比为:中性蛋白酶50~55%,纤维素酶10~15%,木聚糖酶5~10%,β‑葡聚糖酶10~15%,甘露聚糖酶1~5%,葡萄糖氧化酶1~5%。本发明可以有效降解饲料用豆粕中抗营养因子,降低发酵黏度,提高豆粕中总蛋白和小肽含量,促进乳酸菌增殖,减少发酵过程中大肠杆菌、沙门氏菌等有害菌的生长,改善发酵豆粕品质。本发明为生物活性物质,具有高效专一性、安全、环保、无毒、无副作用等优点。

Description

一种用于发酵豆粕的复配酶制剂及其应用
技术领域
本发明属于畜牧饲料技术领域,涉及饲料添加剂,具体涉及到一种用于发酵豆粕的复配酶制剂及其应用。
背景技术
豆粕作为一种质量稳定的植物蛋白原料,一直是饼粕消费的主体,加之动物性蛋白资源日益短缺,价格居高不下,使其使用量受限,豆粕因其蛋白质含量丰富、氨基酸较平衡,成为植物蛋白的首选原料,但豆粕中含有较多的抗营养因子,如大豆抗原、胰蛋白酶抑制因子、大豆凝集素、非淀粉多糖、脲酶、胀气因子等,阻碍营养物质的消化吸收。通过生物技术的使用以及酶制剂在饲料原料生产加工中的广泛应用等,使低质的蛋白原料转化为高质的蛋白原料,是目前提高植物性原料营养价值的主要途径。
通过生物发酵处理大豆粕,利用有益微生物和酶制剂协同作用,去除多种抗营养因子,丰富并平衡豆粕中的蛋白质营养水平,增加氨基酸和肽类物质的生成,从而改善豆粕的营养价值。利用微生物丰富的酶系,将植物大分子蛋白降解为寡肽,并将植物蛋白中的抗营养物质如胰蛋白酶抑制因子、脲酶、血凝素、抗原蛋白等彻底分解,改善产品的风味和适口性。由于发酵微生物分泌酶的种类和数量有限,因此,结合酶法处理大豆粕,可更有效降解豆粕细胞壁中非淀粉多糖,释放胞内物质,促进蛋白酶对蛋白质的水解作用,增加小肽的含量,其中小肽能直接由消化道吸收,促进矿物质微量元素在动物消化道中的消化吸收作用,金属矿物质元素在动物胃酸作用下溶解成离子,与水溶性的小肽、氨基酸结合,形成水溶性的、动物易吸收的金属、有机物的螯合物,供机体利用。目前,发酵豆粕已成为提供饲料优质蛋白质来源的研究热点。
发明内容
根据现有技术中大豆粕本身存在较多的抗营养因子,不能被动物吸收利用,甚至阻碍其他营养物质的消化吸收,微生物法处理后可去除一部分抗营养因子,其自身产酶量有限,对于降解非淀粉多糖,提高蛋白质的含量和小肽的生成量仍需加酶法降解。因此,本发明提供了一种用于发酵豆粕的复配酶制剂及其应用,利用本发明提供的复配酶制剂可以达到利用多酶处理降解大豆粕中的非淀粉多糖,提高大豆粕营养价值的目的。
为实现上述发明目的,本发明采用下述技术方案予以实现:
一种用于发酵豆粕的复配酶制剂按以下重量百分比组成:
本发明所述的发酵底物为饲料用大豆粕或其他饲料用饼粕类物质。
其中,所述复配酶制剂以固体粉末的形式直接使用。
本发明所述复配酶制剂的添加量为豆粕总量的0.1-0.3%,直接将其添加到饲料用大豆粕中发酵使用。
本发明的制备方法:将原酶分别测定酶活后,按比例进入自动混配机混匀,然后测定酶活,检验合格后包装为成品。
本发明的优点和积极效果是:本发明根据发酵豆粕的加工工艺,豆粕中抗营养因子的存在,提供了一种复配酶制剂,选用适用于发酵豆粕使用的酶制剂品种,确定不同酶制剂的添加比例进行复配,在发酵豆粕的生产过程中可有效降解饲料用大豆粕的抗营养因子,促进有益微生物的生长,抑制大肠杆菌、沙门氏菌等的繁殖,提高发酵豆粕总蛋白和小肽含量,改善发酵豆粕的产品品质。利用本发明提供的复配酶制剂可以达到提高发酵豆粕总蛋白和小肽含量,去除大豆粕中抗营养因子的目的,增加有益微生物的增殖。
从使用效果看,本发明所述的复配酶制剂处理发酵豆粕的优点为:1)提高豆粕中的营养成分,破解植物细胞壁,释放细胞内营养物质,使营养成分更易被动物消化吸收;2)可降解多种抗营养因子,降低发酵粘度;3)增加豆粕中总蛋白和小肽含量,促进大豆蛋白水解为氨基酸、寡肽等小分子物质;4)促进有益微生物的增殖,5)改善发酵环境,降低发酵pH值。6)经济、成本低,不需要加热或添加化学试剂,无化学残留。本发明具有高效专一性、安全、环保、无毒、无副作用等优点。
结合数据图表观察本发明的具体实施方式后,本发明使用特点和优点将变得更加清楚。
具体实施方式
下面结合试验数据和具体实施方式对本发明的技术方案做进一步详细的说明。
本发明所述的复配酶制剂的具体组分为:中性蛋白酶,纤维素酶,木聚糖酶,β-葡聚糖酶,甘露聚糖酶,葡萄糖氧化酶。
本发明所述的复配酶发酵豆粕的应用方法为:所述复配酶制剂的添加量为豆粕总量的0.2%,直接将其添加到饲料用大豆粕中发酵使用。
检测指标:
1)常规成分
粗蛋白:采用GB/T 6432-1994测定饲料中粗蛋白质含量
水分:采用GB/T 6435-1986测定饲料水分的含量
粗灰分:采用GB/T 6438-1992测定饲料中粗灰分的含量
粗脂肪:采用GB/T 6433-1994测定饲料中粗脂肪的含量
粗纤维:采用GB/T 6434-2006测定饲料中粗纤维的含量
钙:采用GB/T 6436-2002测定饲料中钙的含量
磷:采用GB/T 6437-2002测定饲料中总磷的含量
2)有益活菌数
采用平板计数法。
3)pH值
称取过60目筛的样品5g,加蒸馏水45mL,浸泡过夜,离心取上清,用酸度计检测pH值。
4)酸溶蛋白含量
酸溶蛋白是指能够溶解于15%TCA(三氯乙酸)溶液的可溶性蛋白,本试验采用QB/T 2653-2004(大豆肽粉)的处理方法,利用15%TCA(三氯乙酸)处理试样,除去了未水解的较大分子量的蛋白质,而分子量在10000Dalton以下的肽类、氨基酸经离心后留在上清液中,然后采用Folin一酚法测定上清液中的酸溶蛋白含量。
5)乳酸
首先对0.1mol/L的NaOH溶液进行标定,然后称取发酵后样品5g,加新煮沸冷却后的蒸馏水45ml,置摇床120r/min摇床30min。离心,取10ml上清,再加入同上蒸馏水15mL。滴2滴1%酚酞指示剂,用标准NaOH溶液进行滴定,滴定至淡粉红色,到终点记录消耗标准NaOH的体积,重复3次,取平均值。计算公式:

结果分析:
从数据结果可以看出,使用酶处理发酵豆粕后,其营养物质得到了改善,增加酸溶蛋白的含量,降低pH值,增加乳酸含量及发酵活菌数,从而整体改善发酵豆粕品质。
