Abstract

背景技术
抗原呈递细胞(antigen presenting cells,APC)在引发有效的免疫应答中是重要的。它们不仅将抗原呈递给具有抗原特异性T细胞受体的T细胞,而且还提供T细胞活化所必需的信号。APC既呈递抗原又传递T细胞活化信号的能力通常被称为辅助细胞功能。虽然单核细胞和B细胞已被证明是有能力的(competent)APC,但其体外抗原呈递能力似乎仅限于先前致敏过的T细胞的再激活。因此,单核细胞和B细胞不能直接激活功能上初始的或未接触过抗原的T细胞群体,它们也不能传递可以使诱导的免疫应答或者诱导时产生的免疫应答极化的信号。
树突细胞(dendritic cells,DC)是免疫系统中专门呈递抗原的细胞,其被认为能够激活初始和记忆T细胞。越来越多的树突细胞在体外被制备以用于免疫治疗,特别是癌症的免疫治疗。制备具有最佳免疫刺激性质的树突细胞需要理解和利用它们在体外培养的生物性质。已经描述了多种各自具有不同优点的用于培养这些细胞的方案。
树突细胞的活化启动将表型类似于皮肤朗格汉斯(Langerhans)细胞的未成熟DC转化为成熟的可以迁移到淋巴结的抗原呈递细胞的过程。这个过程导致了表征未成熟树突细胞的强大的抗原提呈(antigenuptake)能力循序渐进的丧失,以及共刺激细胞表面分子和各种细胞因子的表达的上调。多种刺激可以启动DC的成熟。这个过程是复杂的,并且,取决于所使用的树突细胞成熟剂,至少单核树突细胞(monocytic dendritic cell)在体外的完全成熟可能需要长达48小时才能完成。成熟的另一个后果是细胞体内迁移性质的改变。例如,诱导未成熟树突细胞成熟则诱导了几种趋化因子受体,包括CCR7,其将细胞引导到引流淋巴结的T细胞区域,在此区域中,在I类和II类MHC分子存在的情况下,成熟的DC针对呈递于DC表面上的抗原活化T细胞。术语“活化/激活(activation)”和“成熟/成熟化(maturation)”以及“经活化的/经激活的(activated)”和“成熟的(mature)”描述了诱导和完成从未成熟DC(部分特征为提呈抗原(take up antigen)的能力)到成熟DC(部分特征为有效刺激新生T细胞应答的能力)的过程。这些术语在本领域中通常可互换使用。
已知的成熟方案基于其中据信在暴露于抗原期间或之后遭遇DC的体内环境。这种方法的早期示例是使用单核细胞条件培养基(MCM)作为细胞培养基。在体外培养单核细胞以产生MCM,并将其用作成熟因子的来源。(参见例如US 2002/0160430,通过引用并入本文。)据报道,(促)炎性细胞因子白细胞介素1β(IL-1β)、白细胞介素6(IL-6)和肿瘤坏死因子α(TNFα)是MCM中负责成熟的主要成分。
因此,DC的成熟可以通过许多不同因子触发或启动,所述因子通过许多信号转导途径发挥作用。所以不存在单一的成熟途径或结果,而实际上DC的成熟有许多不同阶段,每个阶段都有其自己独特的功能特征。从概念上讲,这是有道理的,因为免疫系统需要不同的攻击策略来对身体面临的多方面的各种威胁做出应答。例如,尽管细菌感染最好通过补充有特异性抗体的活化巨噬细胞来清除,但病毒感染最好通过有效杀死病毒感染细胞的细胞毒性T细胞来攻击。杀死癌细胞通常涉及细胞毒性T细胞、自然杀伤细胞和抗体的组合。
因此可以设计DC的体外成熟以诱导免疫系统偏好一种类型的免疫应答超过另一种,即使得免疫应答极化。DC的定向成熟(directional maturation)描述了如下概念,即成熟过程的结果指示了经成熟的DC治疗所得的接踵而至的免疫应答的类型。在其最简单的形式中,定向成熟导致DC群体产生细胞因子,所述细胞因子引导了极化为Th1型或Th2型免疫应答的T细胞应答。DC表达多达9种不同的Toll样受体(TLR1至TLR9),其中每一种都可以用于触发成熟。毫不意外的,细菌产物与TLR2和TLR4的相互作用导致DC的定向成熟,导致对于处理细菌感染最为适宜的极化反应。相反,通过TLR7或TLR9触发的成熟似乎导致更多的抗病毒类型反应。作为另一个示例,在大多数成熟方案中加入干扰素γ(IFN-γ)导致成熟DC产生白细胞介素12,这指示了Th1型免疫应答。相反,包含前列腺素E2具有相反的效果。
完全成熟的树突细胞与未成熟的DC在定性和定量上不同。一旦完全成熟,DC表达更高水平的MHC I类和II类抗原,以及更高水平的T细胞共刺激分子,例如CD80和CD86。这些改变增加了树突细胞活化T细胞的能力,因为它们增加了细胞表面上的抗原密度,以及通过T细胞上的共刺激分子的对应物(例如CD28等)的T细胞激活信号的幅度。另外,成熟DC产生大量细胞因子,刺激和极化T细胞应答。这些细胞因子包括与Th1型免疫应答有关的白细胞介素12和与Th2型免疫应答有关的白细胞介素-10和白细胞介素-4。
通常用于离体产生DC的方法包括:由受试者获得富含DC前体细胞的细胞群,然后在体外将DC前体细胞分化成完全成熟的DC,再将完全成熟的DC回输至受试者。通常在该过程中,成熟中的DC与抗原接触以提呈并随着DC成熟而处理抗原。有些人认为DC必须终末分化(terminally differentiated),否则会分化回单核细胞/巨噬细胞,从而失去大部分免疫增强能力。用单核细胞产生的DC的离体成熟已经用本领域熟知的方法和试剂成功完成。
树突细胞(DC)被认为是癌症主动免疫治疗的选择载体。动物实验已经证实了基于DC的免疫疗法在保护小鼠免于肿瘤形成和消除已存在的肿瘤中的潜力。在小型临床试验于人类中至少部分地重复了这些成功。如上所述,DC制备的繁琐性质阻碍了其从小规模的安全性或概念证明实验转变为可以证明其活性或功效的大型实验。因此,尽管这些产品具有巨大的潜在治疗价值,但很少有公司对开发基于DC的癌症疫苗感兴趣。
DC的瘤内(Intratumoral,IT)注射是基于DC的免疫疗法的特殊形式。在注射后,DC从体内(例如凋亡或濒临死亡的肿瘤细胞)提呈抗原,并在迁移到淋巴结后将抗原呈递(present)给T细胞。事实上,发现这种治疗在动物模型中的功效与肿瘤的凋亡程度相关(Candido等人,CancerRes.61:228-236,2001),这表明这种方法与在注射DC之前用化学治疗剂或辐射治疗肿瘤完全兼容。此外,几个研究小组已经证实,这样的联合疗法对于已存在的肿瘤是特别有效的(Nikitina等人,Int.J.Cancer 94:825-833,2001;Tanaka等人,Int.J.Cancer 101:265-269,2002;Tong等,CancerRes.61:7530-7535,2001)。
由于体内肿瘤细胞是抗原的来源,所以IT注射不需要肿瘤抗原的选择和制造,即使它们目前用于大多数基于体外DC的治疗方法中。肿瘤抗原的选择通常是由公司取得专利权地位(proprietary position)的需求所驱动,并且迄今为止鉴定的少数肿瘤抗原尚未被证明具有显著的临床益处。另外,使用这样的肿瘤抗原经常导致单价疫苗,如果肿瘤细胞下调免疫中使用的抗原的表达,则其会丧失效力。当然,在良好生产规范(GMP)要求的条件下生产肿瘤抗原的要求增加了传统的基于DC的免疫方法的附加成本。
DC的IT注射使树突细胞进入免疫抑制性肿瘤环境。已知肿瘤产生使DC失活的或具有使T细胞应答朝不太有效的Th2型免疫应答倾斜的能力的细胞因子。几个研究组已经使用DC的遗传修饰来尝试克服这些抑制效应,特别是通过产生细胞因子白细胞介素12(IL-12;Nishioka等,Cancer Res.59:4035-4041,1999;Melero等,Gene Therapy 6:1779-1784,1999)或CD40配体的表达(Kikuchi等,Blood 96:91-99,2000)。这些研究组描述的令人鼓舞的结果进一步证明了IT注射DC作为治疗方法的可行性。
Triozzi等人(Cancer 89:2647-2654,2000)描述了在患有转移性黑素瘤或乳腺癌的患者中IT注射DC。他们在4例黑素瘤患者和2例乳腺癌患者中获得肿瘤消退。退化病变的活组织检查显示浸润的T细胞,表明DC确实激活了针对肿瘤细胞的免疫应答。总体而言,这些数据表明,IT注射DC对人类是可行的,并且可以提供显著的临床益处。然而,观察到注射的DC上MHC II类抗原和B7-2共刺激分子的显著下调。预计这些关键分子的下调将降低DC的免疫刺激潜力。
在WO 2004/053072(通过引用并入本文)中已经公开了克服这种下调的一种方法,其中发现可以通过在给药之前使DC部分成熟来避免下调。在该方法中,在体外诱导树突细胞前体(红细胞溶解后的骨髓细胞或单核树突细胞前体)分化成未成熟的树突细胞,通过用树突细胞成熟剂,例如BCG和IFNγ、脂多糖(LPS)、肿瘤坏死因子α(TNFα)、咪唑并喹啉化合物、合成的双链多聚核糖核苷酸、Toll样受体(TLR)激动剂、已知会诱导树突细胞成熟的含有未甲基化的CpG基序的核酸序列,或其任何组合来培养细胞以诱导未成熟的树突细胞开始成熟。使未成熟的树突细胞继续成熟持续一段时间,该时间短于先前已经确定的未成熟的树突细胞完全成熟所需的时间。如果允许树突细胞在体外完全成熟,那么细胞在施用给患者后将不能提呈和处理抗原。发明人公开了在分离部分成熟的树突细胞和配制用于施用给患者之前,应允许树突细胞成熟1至约10小时以达到最佳活化。
出乎意料地,确定了与树突细胞成熟剂(例如BCG和IFNγ)接触约10至约19小时的未成熟的树状细胞被最佳活化以用于体内提呈和处理抗原,以及随后诱导在受试者体内的抗肿瘤应答。
发明内容
本公开提供了生产分离的且经活化的人树突细胞的方法,包括:i)从外周血分离包含人PBMC的细胞群;ii)使所述包含人PBMC的细胞群富含人单核树突细胞前体;iii)用补充有有效量的树突细胞分化剂的组织培养基培养富含人单核树突细胞前体的细胞群,培养的时间足以使人单核树突细胞前体分化成未成熟的人树突细胞;iv)用有效量的树突细胞成熟剂培养富含未成熟的人树突细胞的细胞群约10至约19小时,以活化所述未成熟的人树突细胞;和v)分离并清洗经活化的人树突细胞。
在另外的实施方案中,提供了产生分离的且经活化的人树突细胞的方法,其中所述方法包括以下步骤:
i)分离包含人单核树突细胞前体的细胞群;ii)用补充有有效量的树突细胞分化剂的组织培养基培养富含人单核树突细胞前体的细胞群,培养的时间足以使人单核树突细胞前体分化成未成熟的人树突细胞;iii)用有效量的树突细胞成熟剂培养富含未成熟的人树突细胞的细胞群约10至约19小时,以活化所述未成熟的人树突细胞;和iv)分离并清洗经活化的人树突细胞。
该方法可以使用获自皮肤、脾脏、骨髓、胸腺、淋巴结、脐带血或外周血的单核树突细胞前体。进一步的,所述单核树突细胞前体从与待治疗个体受试者HLA匹配的健康个体受试者获得。
在上述方法中,树突细胞分化剂可以是不含其他细胞因子的单独的GM-CSF,或者是与白细胞介素4(IL-4)、白细胞介素7(IL-7)、白细胞介素-13(IL-13)或白细胞介素15(IL-15)等组合的GM-CSF。在单独使用GM-CSF的典型实施方案中,使用未经活化(non-activated)的单核树突细胞前体,并且组织培养基还补充有至少1%的人或动物蛋白以防止未经活化的单核树突细胞前体粘附于组织培养基质。所述人或动物蛋白质可以是白蛋白、血清、血浆、明胶、聚氨基酸等。
在所述方法中,所述树突细胞成熟剂可以是灭活卡介苗(BCG)、干扰素γ(IFNγ)、脂多糖(LPS)、肿瘤坏死因子α(TNFα)、咪唑并喹啉化合物、合成的双链多聚核糖核苷酸例如聚[I]:聚[C(12)U]、Toll样受体(TLR)激动剂、已知会诱导树突细胞成熟的含有未甲基化的CpG基序的核酸序列,或其任何组合。所述灭活的BCG可以包含完整的BCG、BCG的细胞壁组分(constituents)、BCG衍生的脂阿拉伯糖甘露聚糖(lipoarabidomannans)或BCG成分(component),所述灭活的BCG是热灭活的BCG、经福尔马林处理的BCG,或者热灭活且经福尔马林处理的BCG。
当在上述方法之一中使用时,BCG的有效量为每毫升组织培养基约105-107cfu,IFNγ的有效量为每毫升组织培养基约100-约1,000单位。当上述方法使用咪唑并喹啉化合物时,其中所述咪唑并喹啉化合物为咪唑并喹啉-4-胺化合物,例如4-氨基-2-乙氧基甲基-α,α-二甲基-1H-咪唑[4,5-c]喹啉-1-5乙醇或1-(2-甲基丙基)-1H-咪唑并[4,5-c]喹啉-4-胺或其衍生物。
包含部分成熟且经最佳活化的树突细胞的组合物可以直接施用于肿瘤;施用于手术移除或切除肿瘤之后的肿瘤床;施用于肿瘤周围的组织区域;施用于直接引流(draining)肿瘤区域的淋巴结;直接施用于将血液或淋巴输送到肿瘤或肿瘤相关器官的循环血管管道;或施用进入循环系统将所述细胞递送到肿瘤或肿瘤相关器官。
通过上述任何一种方法产生的部分成熟且经最佳活化的树突细胞可作为放射疗法、化学疗法或其组合的佐剂施用。例如,所述部分成熟且经最佳活化的树突细胞可以在放射疗法、化学疗法或其组合之前、同时或之后施用。
在另一个实施方案中,提供了用于产生抗肿瘤免疫应答和/或临床应答的方法,其包括施用包含富含人树突细胞的细胞群和药学可接受的载体的组合物,所述人树突细胞已经在体外用人树突细胞成熟剂部分成熟并最佳地活化约10至约19小时,其中所述组合物被施用于需要这种治疗的个体的肿瘤、肿瘤床或肿瘤周围的组织区域中。

树突细胞是在各种淋巴和非淋巴组织中发现的抗原呈递细胞的不同群体。(参见Liu,Cell 106:259-262(2001);Steinman,Ann.Rev.Immunol.9:271-296(1991))。树突细胞包括脾的淋巴树突细胞、表皮的朗格汉斯(Langerhans)细胞和血液循环中的隐蔽细胞(veiled cell)。总体而言,树突细胞根据其形态、高水平表面MHC-II类表达以及缺乏在T细胞、B细胞、单核细胞和自然杀伤细胞上表达的某些其他表面标记而被分类为一组。具体而言,来源于单核细胞的树突细胞(也称为单核树突细胞)通常表达CD11c、CD80、CD86,并且是HLA-DR+,但也是CD14-。
相反,单核树突细胞前体(通常是单核细胞)通常是CD14+。单核树突细胞前体可以从它们所在的任何组织获得,特别是淋巴组织,例如脾脏、骨髓、淋巴结和胸腺。单核树突细胞前体也可以从循环系统中分离出来。
外周血是容易获得单核树突细胞前体的来源。脐带血是单核树突细胞前体的另一来源。单核树突细胞前体可以从引发免疫应答的多种生物体中分离。所述生物体包括动物,例如包括人和非人类动物,如灵长类动物、哺乳动物(包括狗、猫、小鼠和大鼠),鸟类(包括鸡),以及其转基因物种。
在某些实施方案中,单核树突细胞前体和/或未成熟的树突细胞可从健康受试者或需要免疫刺激的受试者中获得,例如癌症患者或者对其而言细胞免疫刺激可为有益的或期望的其他受试者,(即,患有细菌或病毒感染或增生状况等的受试者)。树突细胞前体和/或未成熟的树突细胞也可以从HLA匹配的健康个体获得,用于部分激活并施用于需要免疫刺激的HLA匹配的受试者。
树突细胞前体和未成熟的树突细胞
由各种来源(包括血液和骨髓)分离富含树突细胞前体(例如未经活化的树突细胞前体)和未成熟的树突细胞的细胞群的方法是本领域已知的。例如,树突细胞前体和未成熟的树突细胞可以经以下获得:通过收集肝素化血液,通过单采或者白细胞分离法,通过制备血沉棕黄层、花结(rosetting)、离心、密度梯度离心(例如使用(如FICOLL-),(涂有不可透析的聚乙烯吡咯烷酮(PVP)的胶态二氧化硅颗粒(直径15-30nm))、蔗糖等)、细胞的差异裂解、过滤等。在某些实施方案中,可以通过例如通过从受试者收集血液,去纤维化去除血小板并裂解红细胞以制备白细胞群体。树突细胞前体和未成熟的树突细胞可以通过例如经梯度、抗体淘选(antibody panning)等等任选地富集单核树突细胞前体。如本文所使用的术语“富含/富集的(enriched)”意指细胞群中的单核树突细胞前体、未成熟的树突细胞或部分成熟的树突细胞占细胞群中细胞总数的至少25%、至少30%、至少50%、至少60%、至少70%、至少80%或至少90%。通常,富集的细胞将占群体中细胞总数的至少约50%。在某些实施方案中,富集的细胞将占细胞群中细胞总数的至少约90%并高达约100%。
树突细胞前体和未成熟的树突细胞可以任选地在封闭的无菌系统中制备。如本文所使用的,术语“封闭的无菌系统”或“封闭系统”是指其中暴露于非无菌的,外界的或循环的空气或其他非无菌条件被降低至最少或消除的系统。用于分离树突细胞前体和未成熟树突细胞的封闭系统通常不包括敞口试管(open top tube)中的密度梯度离心、细胞的敞口转移、组织培养板或未密封烧瓶中细胞的培养等。在一个典型的实施方案中,封闭系统允许树突细胞前体和未成熟树突细胞从初始收集器无菌转移至可密封组织培养器,而不暴露于非灭菌空气。
另一种经报道的分离树突细胞前体的方法是使用商业处理的塑料底物(例如珠子或磁珠)来选择性去除粘附的单核细胞和其他“非树状细胞前体”。(参见例如美国专利号5,994,126和5,851,756)。丢弃粘附的单核细胞和非树突细胞前体,而保留非粘附细胞进行离体培养和成熟。在另一种方法中,将单采血液成分细胞培养在塑料培养袋中,向其中加入塑料(即聚苯乙烯或苯乙烯)微载体珠以增加袋的表面积。
将细胞培养足够的时间以使某些细胞粘附到珠上,并从袋中清洗掉非粘附细胞。(Maffei等,Transfusion 40:1419-1420(2000);WO 02/44338,在此引入作为参考)。在某些其他实施方案中,单核细胞树状细胞前体通过松散粘附于单核细胞结合底物而分离,如WO03/010292中所公开的,其公开内容通过引用并入本文。例如,可以使(例如,通过白细胞分离法分离的)白细胞群体与单核细胞树突细胞前体粘附基质(substrate)接触。当白细胞群体与基质接触时,白细胞群体中的单核细胞树突细胞前体优先粘附于基质,但不被激活分化或成熟。其他白细胞(包括其他潜在的树突细胞前体)表现出对基质的降低的结合亲和力,由此允许单核细胞树突细胞前体优先富集在基质的表面上。
合适的基质包括例如具有大的表面积与体积比的基质。基质可以是例如颗粒状或纤维状基质。合适的颗粒状基质包括例如玻璃颗粒、塑料颗粒、玻璃涂覆的塑料颗粒、玻璃涂覆的聚苯乙烯颗粒和其它适用于蛋白质吸收的珠粒。适用于本发明方法的纤维状基质包括微毛细管和微绒毛膜等。颗粒状或纤维状基质典型地允许粘附的单核细胞树突细胞前体被洗脱而基本上不降低粘附细胞的生存力(viability)。颗粒状或纤维状基质可以是基本上无孔的,以便于从基质上洗脱单核树状细胞前体或树突细胞。“基本上无孔的”基质是指存在于其中的至少大部分孔小于细胞的基质,以最少可能在基质中捕获细胞。
可以任选地通过添加结合培养基来增强单核树突细胞前体对基质的粘附。合适的结合培养基包括,例如,单独地或以任何组合地补充了例如细胞因子(例如粒细胞/巨噬细胞集落刺激因子(GM-CSF)或GM-CSF联合白细胞介素4(IL-4)、白细胞介素15(IL-15)或白细胞介素13(IL-13))、血浆、血清(例如人血清,如自体或同种异体血清)、纯化的蛋白质如血清白蛋白、二价阳离子(如钙离子和/或镁离子),以及其他帮助单核树突细胞前体特异性粘附于基质或者防止非单核树突细胞前体粘附于基质的分子的单核树突细胞前体培养基(例如AIM-RPMI 1640,DMEM,等)。在某些实施方案中,血浆或血清可被加热灭活。热失活的血浆对于白细胞可以是自体的或异源的。
在将单核树突细胞前体粘附到基质上之后,将未粘附的白细胞从单细胞树突细胞前体/基质复合物中分离出来。可以使用任何合适的手段将未粘附的细胞与复合物分离。例如,可使未粘附的白细胞和复合物的混合物沉降,并将未粘附的白细胞和培养基倒出或排出。或者,可将混合物离心,由沉淀的复合物中倒出或排出含有未粘附的白细胞的上清液。需注意的是,如果没有另外的刺激,单核树突细胞前体与基质的粘附不会诱导单核树突细胞前体活化并分化或成熟为未成熟的树突细胞、成熟的树突细胞或巨噬细胞。
在另一种方法中,未经活化的单核树突细胞前体可以从富含白细胞的细胞群中分离,所述细胞群以切向流过滤装置制备,如国际专利申请公开号WO 2004/000444,申请日2003年6月19日,现美国专利号7,695,627中所描述,两者都通过引用并入本文。用于分离富含单核树突细胞前体的细胞群的切向流过滤装置可以包括具有错流室(cross-flowchamber)、滤液室和置于其间的过滤器的去除器单元。过滤器在一侧即滞留物(retentate)表面与错流室流体连通,在另一侧即滤液(filtrate)表面与滤液室流体连通。错流室具有适于将包含白细胞的血液成分样品引入错流室并且平行于过滤器的滞留物表面的入口。错流室中还设有出口,该出口居中设置在与过滤器的滞留物表面相对的该室的一部分中。适用于切向流过滤装置的过滤器通常具有约1至约10微米的平均孔径。过滤器可具有约3至约7微米的平均孔径。还可以包括提供样品进入错流室入口的预定输入速率的装置以及用于控制滤液通过过滤器并进入滤液室的过滤速率的装置。过滤速度控制装置将过滤速度限制为小于过滤器的无对抗过滤速率。包含血液成分的样品可以由来源设备例如白细胞分离设备提供或包含从白细胞分离设备收集的样品的容器提供。
可以离体(ex vivo)或体外(in vitro)培养单核树突细胞前体和富含前体的细胞群以进行分化和部分成熟和/或扩增。如本文所用,“分离的未成熟的树突细胞”、“树突细胞前体”和其他“细胞”是指以人工培养的方式生存于其天然生存环境之外的细胞,因此并非天然产物。分离的细胞可以以纯化形式、以半纯化形式和/或在非天然环境中存在。简而言之,体外和/或离体树突细胞分化通常包括在一种或多种树突细胞分化剂存在下培养单核树突细胞前体或具有树突细胞前体的细胞群。合适的分化剂可包括例如细胞生长因子(例如细胞因子如(GM-CSF)或GM-CSF与白细胞介素4(IL-4)、白细胞介素13(IL-13)、白细胞介素15(IL-15)或白细胞介素7(IL-7)的组合)。在某些实施方案中,单核树突细胞前体分化形成单核细胞衍生的未成熟的树突细胞。
可以在合适的体外培养条件下培养和分化树突细胞前体。合适的树突细胞组织培养基包括但不限于AIM-RPMI 1640、DMEM、X-VIVO等。组织培养基可补充有血清、血浆、氨基酸、维生素、细胞因子如GM-CSF和/或IL-4、IL-7、IL-13、IL-15、二价阳离子等,以促进细胞的分化。在某些实施方案中,树突细胞前体可以在无血清培养基中培养。培养条件可以任选地排除任何动物衍生产品。与树突细胞培养基一起使用的典型的细胞因子组合包含每一种约500单位/ml的GM-CSF和IL-4、IL-7、IL-15或IL-13。在使用未经活化的树状细胞前体的典型实施方案中,典型的树状细胞组织培养基可以在一定条件下补充有GM-CSF而不含任何其他细胞因子。例如,当单独使用GM-CSF时,组织培养基通常还补充有高浓度的人或动物蛋白以防止未经活化的单核树突细胞前体粘附到组织培养基质上从而激活树突细胞前体的成熟。通常人或动物蛋白以大于1%的浓度加入,并且通常以10%或更少的浓度使用。人或动物蛋白质可以是白蛋白,如人血清白蛋白、血清、血浆、明胶、聚氨基酸等。
当分化形成未成熟的树突细胞时,树突细胞前体在表型上类似于皮肤朗格汉斯细胞。未成熟的树突细胞通常是CD14-和CD11c+,表达低水平的CD86和CD83,并且能够通过特化内吞捕获可溶性抗原。
树突细胞成熟剂可以包括例如但不限于:BCG、IFNγ、LPS、TNFα、咪唑并喹啉化合物,例如咪唑并喹啉-4-胺化合物,如4-氨基-2-乙氧基甲基-α,α-二甲基-1H-咪唑[4,5-c]喹啉-1-5乙醇(命名为R848)或1-(2-甲基丙基)-1H-咪唑并[4,5-c]喹啉-4-胺和其衍生物(参见例如WO2000/47719,通过引用将其全部内容并入本文),合成的双链多聚核糖核苷酸,如聚[I]:聚[C(12)U]等,Toll样受体(TLR)激动剂,如TLR-3、TLR-4、TLR-7和/或TLR-9,已知会诱导DC成熟的含有未甲基化的CpG基序的核酸序列等,或其任何组合。在灭活之前,BCG的有效量通常在相当于每毫升组织培养基约105-107cfu的范围内。IFNγ的有效量的范围为每毫升组织培养基约100-约1,000单位。
卡介苗(BCG)是牛分枝杆菌的无毒菌株。如本文所使用的,BCG可以包含完整的BCG以及BCG的细胞壁组分、BCG衍生的脂阿拉伯糖甘露聚糖或其他BCG成分。BCG可任选的是灭活的,例如热灭活的BCG、经福尔马林处理的BCG,或者热灭活且经其他灭活方法处理的BCG等。有效量的咪唑并喹啉化合物,例如,其中所述咪唑并喹啉化合物为咪唑并喹啉-4-胺化合物,例如,4-氨基-2-乙氧基甲基-α,α-二甲基-1H-咪唑[4,5-c]喹啉-1-5乙醇(命名为R848)可以为约1至约50μg/ml培养基,更通常为5至约10μg/ml培养基。咪唑并喹啉化合物可以单独使用或者可以与例如BCG和/或IFN,或者另外的TLR激动剂组合。
通常使未成熟的DC与有效量的树突细胞成熟剂如BCG和IFNγ接触约10小时至约19小时以诱导其成熟,并使所述树突细胞最佳活化但不完全成熟。当使用BCG和IFNγ成熟树突细胞时,通常需要至少24小时的温育时间才能完全成熟,并且取决于使用的树突细胞成熟剂,通常需要约48至约72小时的温育时间成熟。在某些实施方案中,时间段可以是约10小时、11小时、12小时、13小时、14小时、15小时、16小时、17小时、18小时和至多约19小时。在更典型的实施方案中,树突细胞的部分成熟且最佳活化的时间段可以是约15至约18小时、约15至约17小时,或在特定实施方案中约16小时。未成熟的树突细胞可以在合适的成熟培养条件下培养并部分成熟且最佳活化。合适的组织培养基包括但不限于AIM-RPMI1640、DMEM、X-VIVO等等。组织培养基可以补充有氨基酸;维生素;细胞因子如GM-CSF(参见例如美国专利号8,389,278,其全部内容通过引用并入本文)或GM-CSF与IL-4、IL-7、IL-13或IL-15的组合;二价阳离子等,以促进诱导细胞成熟。典型的细胞因子可以是单独的GM-CSF和高浓度的人或动物蛋白,或者当组合使用时,GM-CSF以约500单位/ml至约1000单位/ml使用,并使用100ng/ml的IL-4、IL-13或IL-15。
未成熟的树突细胞的部分成熟和最佳活化可以通过本领域已知的用于树突细胞的方法来监测。细胞表面标记可以在本领域熟悉的分析中检测,例如流式细胞术、免疫组织化学等。还可监测细胞的细胞因子产生(例如,通过ELISA,另一种免疫测定或通过使用寡核苷酸阵列)。在树突细胞成熟剂(例如但不限于BCG和INFγ)的存在下,根据本说明书进行培养并部分成熟且经最佳活化的DC中,可以通过本领域公知的方法测量与未成熟的树突细胞相比而言增高水平的磷酸化的JAK2(janus activated kinase 2),以指示成熟的开始。细胞表面标记和细胞因子表达的诱导以及信号分子例如jak2的磷酸化也被认为是指示,其标志着一旦树突细胞被施用给个体,树突细胞经调整以适应在体内提呈抗原和诱导免疫应答。
本发明的方法包括使未成熟的树突细胞处于成熟条件中仅持续启动未成熟的树突细胞的成熟化(maturation),使树突细胞部分成熟并最佳活化而所必需的一段时间。已经发现,当与药学上可接受的载体组合施用于受试者时,与有效量的BCG和有效量的IFNγ作为组合物一起温育约10至19小时的时间可使树突细胞部分成熟且最佳活化。完全成熟的DC丧失提呈抗原并呈递共刺激细胞表面分子和各种细胞因子的上调表达的能力。具体而言,成熟DC比未成熟的树突细胞表达更高水平的MHC I类和II类抗原,并且成熟树突细胞一般被鉴定为CD80+、CD83+、CD86+和CD14-。更高的MHC表达导致DC表面上的抗原密度增加,而共刺激分子CD80和CD86的上调通过共刺激分子的对应物例如T细胞上的CD28而增强T细胞激活信号。如本公开中使用的部分成熟且经最佳活化的树突细胞通常包含那些一旦暴露于树突细胞成熟剂则表现出与未成熟的树突细胞相比而言上调的细胞表面上共刺激分子表达的树突细胞。这些共刺激分子包括但不限于CD80、CD86和/或CD54。这些细胞能或者不能表达CD83,但是细胞确实保持提呈和处理抗原以在树突细胞表面呈递的能力。在本申请方法的一个实施方案中,部分成熟的树突细胞在施用后暴露于抗原。此外,部分成熟且经最佳活化的树突细胞可以产生一种或多种通常不被未成熟的树突细胞大量产生的TNF-α、IL-6、IL-8、IL-10和/或IL-12。在一个具体的实施方案中,部分成熟且经最佳活化的树突细胞与有效浓度的BCG和IFNγ接触约16小时,在24小时内产生149ng/100万个树突细胞。与相同浓度的BCG和IFNγ接触的未成熟的树突细胞在24小时内仅产生50ng/100万个细胞。
完全成熟的树突细胞对于本发明的方法和组合物来说不是优选的,因为一旦它们完全成熟,细胞就不再有效地处理抗原。另外,并不期望在现有方法中使用的尚未诱导开始成熟的未成熟树突细胞,因为通常在肿瘤内或在肿瘤周围的组织中发现的免疫抑制环境包括已知会阻止未成熟的树突细胞对抗原进行处理的大量浓度的细胞因子。在本公开中,未成熟的树突细胞的部分成熟和最佳活化下调了细胞表面上的细胞因子受体,使得它们对瘤内空间或周围组织中存在的细胞因子的任何免疫抑制作用较不敏感或响应,并提供给细胞能够有效摄取和处理瘤内空间或周围组织内存在的抗原。树突细胞提呈和处理瘤内空间或周围组织内发现的凋亡和濒临死亡的肿瘤细胞的大量肿瘤抗原。一旦施用的部分成熟且经最佳活化的树突细胞在瘤内空间内通过例如趋化因子受体CCR7的表达而测定为有效成熟,树突细胞迁移到淋巴结,其中现在呈递抗原的树突细胞将接触T细胞,特别是初始T细胞以上调对树突细胞呈递的任何肿瘤抗原的免疫应答。
根据说明书的又一个方面,本发明的各种DC可以与药学上可接受的载体组合后或不与药学上可接受的载体组合以通过例如冷冻保存为成熟前或部分成熟后的单核树突细胞前体、未成熟的树突细胞。可以使用的冷冻保存剂包括但不限于二甲基亚砜(DMSO)、甘油、聚乙烯吡咯烷酮、聚乙二醇、白蛋白、葡聚糖、蔗糖、乙二醇、i-赤藓糖醇(i-erythritol)、D-核糖醇、D-甘露糖醇、D-山梨糖醇、肌醇、D-乳糖、氯化胆碱、氨基酸、甲醇、乙酰胺、单乙酸甘油酯和无机盐。受控的缓慢冷却速度可能是至关重要的。不同的冷冻保护剂和不同的细胞类型通常具有不同的最佳冷却速率。
水转变成冰的融溶(fusion)阶段的热量通常应该是最小的。冷却过程可以通过使用例如可编程冷冻设备或甲醇浴过程来进行。可编程的冷冻设备允许确定最佳的冷却速率并有助于标准的可重复冷却。可编程控制速率冷冻机,如或允许将冷冻方案调整为所需的冷却速率曲线。
在彻底冷冻后,可以将具有或不具有药学上可接受的载体的单核前体细胞、未成熟的DC或部分成熟的DC迅速转移到长期低温存储容器中。在典型的实施方案中,样品可以低温储存在液氮(-196℃)或其蒸气(-165℃)中。造血干细胞特别是来自于骨髓或外周血的造血干细胞的操作、冷冻保存和长期储存的考虑(consideration)和程序大部分适用于本说明书中的细胞。这样的讨论可以在例如以下参考文献中找到,其通过引用并入本文:Taylor等人,Cryobiology 27:269-78(1990);Gorin,Clinics in Hematology 15:19-48(1986);“骨髓保存、培养和移植(Bone-Marrow Conservation,Culture andTransplantation)”,会议记录,莫斯科,1968年7月22-26日,国际原子能机构,维也纳,第107-186页。
优选的,将冷冻细胞快速解冻(例如在保持在37℃-41℃的水浴中)并在解冻后立即降温。可能需要处理细胞以便防止细胞在融化时结块。为防止结块可以使用各种方法,包括但不限于冷冻之前和/或之后加入DNase(Spitzer等人,Cancer45:3075-85(1980))低分子量葡聚糖和柠檬酸盐、羟乙基淀粉(Stiff等,Cryobiology 20:17-24(1983))等。如果冷冻保存剂对人体有毒,则应在治疗性使用融化的部分成熟的DC之前除去。去除冷冻保护剂的一种方法是稀释到不显著的浓度。一旦冷冻单核树突细胞前体、未成熟的树突细胞或部分成熟的DC已经解冻并回收,则可以将它们用于进一步的方法中以制备配制的药物产品。配制的部分成熟且经最佳活化的树突细胞可以如本文关于未冷冻的部分成熟且经最佳活化的DC所述的那样施用。
部分成熟的树突细胞的体内施用
提供了用于向具有例如癌症或肿瘤的受试者施用部分成熟且经最佳活化的树突细胞,或经富集并含有这些细胞的细胞群的方法和组合物。在某些实施方案中,通过获得树突细胞前体或未成熟的树突细胞,在树突细胞成熟剂如BCG和IFNγ或如上所例举之外的任何其他树突细胞成熟剂的存在下使这些细胞分化并部分成熟。可使用本领域技术人员熟知的方法和组合物,用生理上可接受的载体、赋形剂、缓冲剂和/或稀释剂来配制部分成熟且经最佳活化的树突细胞。部分成熟且经最佳活化的树突细胞可以直接施用于需要免疫刺激的受试者。通常,将约102至约1010个细胞悬浮于药学上可接受的载体中。将细胞直接注射到肿瘤中或者注射到与肿瘤或肿瘤床接近、邻近、或循环接触或淋巴接触的区域中,以确保细胞能够接近癌症或肿瘤抗原。
作为示例但并非进行限制,细胞可以直接施用于肿瘤,施用于手术移除或切除肿瘤之后的肿瘤床,施用到瘤周间隙,施用于与肿瘤直接接触的引流淋巴结,施用于引向肿瘤或受肿瘤折磨(afflict)的器官等的或为其提供营养的血管或淋巴管,例如门静脉或肺静脉或动脉等。本公开的部分成熟且经最佳活化的树突细胞的施用可以与肿瘤的其它治疗,例如化学疗法或放射疗法同时进行或在其之后进行。此外,本公开内容的部分成熟的树突细胞可以与另一种药剂共同施用,所述药剂作为肿瘤或肿瘤附近或邻近肿瘤内的树突细胞成熟化和/或抗原处理的佐剂。此外,还可以将树状细胞配制或复合成缓释基质以植入肿瘤或肿瘤床内或周围的区域,使得细胞缓慢释放到肿瘤或肿瘤床中以接触肿瘤抗原。
如本公开中使用的肿瘤包括实体瘤,例如但不限于肉瘤、胰腺肿瘤、结直肠肿瘤、黑色素瘤、肺肿瘤、乳房肿瘤、卵巢肿瘤、头部或颈部肿瘤、胃肿瘤、前列腺肿瘤、食管肿瘤、宫颈或阴道肿瘤、脑肿瘤例如成胶质细胞瘤、星形细胞瘤、脑膜瘤或成神经管细胞瘤等等。其他的实体瘤也使用本文所公开的组合物或方法来进行治疗。
本公开的部分成熟且经最佳活化的树突细胞可以通过适合于制剂和施用模式的任何方式施用。例如,所述细胞可以与药学上可接受的载体组合,并与注射器、导管、套管等一起施用。如上所述,细胞可以配制成缓释基质。当以这种方式施用时,可以通过适合所用基质的方式施用制剂。适用于本说明书的其他方法和模式对于本领域技术人员来说是公知的。
本说明书的组合物本身可用于治疗个体。另外,所述组合物可以与任何其他方法联合使用来治疗癌症或肿瘤。例如,本说明书的方法可以与肿瘤的手术切除、化学治疗(细胞毒性药物、细胞凋亡剂、抗体等)、放射疗法、冷冻疗法、近距离放射疗法、免疫疗法(施用抗原特异性成熟的且经活化的树突细胞、NK细胞、对癌细胞或肿瘤抗原具有特异性的抗体等)等。这些方法中的任何一种和所有也可以任何组合形式使用。联合治疗可以是同时的或依序的,并且可以按治疗医师确定的任何顺序施用。
在另一个实施方案中,树突细胞和受体受试者具有相同的MHC(HLA)单体型(haplotype)。确定受试者的HLA单体型的方法是本领域已知的。在相关的实施方案中,部分成熟的树突细胞与受体受试者是同种异体的(allogeneic)。同种异体细胞通常匹配至少一个MHC等位基因(allele)(例如共享至少一个但不是全部MHC等位基因)。在一个较不典型的实施方案中,树突细胞和受体受试者彼此都是同种异体的,但都具有至少一个共同的MHC等位基因。
抗肿瘤免疫应答可以通过任何一种或多种公知的方法来测量。例如,可以通过肿瘤尺寸的减小、肿瘤细胞死亡或肿瘤细胞坏死的诱导、肿瘤细胞增殖的减少,或者通过肿瘤抗原特异性T细胞(TIL)的浸润等等来测量抗肿瘤应答。
实施例
提供以下实施例仅仅是为了说明本说明书的各个方面,而不应被解释为以任何方式限制本文公开的方法和组合物。虽然已经说明和描述了该方法和/或方法的优选实施例,但是将能理解的是,在不脱离本说明书的精神和范围的情况下,可以在其中进行各种改变。
在本实施例中,从分离自患有癌症(例如但不限于作为实体组织肿瘤的肿瘤,例如肉瘤、胰腺肿瘤、结肠直肠肿瘤、黑色素瘤、乳房肿瘤、肺肿瘤和卵巢肿瘤等)的患者的血液样品中获得未成熟的树突细胞,并将未成熟的树突细胞在补充有GM-CSF的树突细胞培养基中单独培养,用灭活的BCG和IFNγ诱导成熟16或20小时,然后收集并制备以施用给病人。发现部分成熟且经活化约16小时的树突细胞相比与BCG和IFNγ接触约20小时的树突细胞在患者体内诱导显著更好的临床反应。
对患者进行白细胞分离术,在采集过程中处理大约10升或两个血量。调整血浆速率以获得白细胞分离产物的约2%的目标血细胞比容。通过切向流过滤(Tangential FlowFiltration,TFF)处理白细胞分离产物以分离和纯化单核细胞。将白细胞分离样品通常为血袋连接至TFF装置并处理该样品,并在组织培养袋中获得悬浮于培养基(不含酚红的RPMI-1640)中的富集的单核树突细胞前体产物。
将补充有重组人GM-CSF(500U/ml)的培养基添加到经富集的未经活化的单核树突细胞前体产物中,并将细胞置于培养箱中培养5天。在5天培养期间,单核树突细胞前体转化为未成熟的树突细胞。在此温育之后,经热灭活的BCG(1:300v/v稀释或每个活树突细胞大约一(1)个BCG颗粒)和干扰素γ(500至1,000U/ml)被加入到未成熟的树突细胞中,所述袋被放回到培养箱中温育16到20小时以活化树突细胞。这段时间不足以将未成熟的树突细胞完全转化为成熟的树突细胞。
在树突细胞活化之后,搅动组织培养袋并将内容物排入250ml锥形离心管中。将冷dPBS加入袋中,并通过手动按压袋将所有剩余的粘附细胞移除。将该洗涤物与初始细胞悬液合并,并在550x g下离心使细胞沉淀。将经活化的树突细胞重悬于小体积的RPMI-1640、40%人血清白蛋白和10%DMSO中。经活化的树突细胞以3x 106、8x 106、17x 106活树突细胞/小瓶的等份冷冻在1.0ml/小瓶中。必要时,每批注射量调整到每次注射总计2、6或15万个活树突细胞。为了制备注射用等分试样,将小瓶从气相液氮贮存器中取出并置于20℃至25℃下5-10分钟。这是完全解冻内容物的充足时间。将小瓶打开并使用注射器无菌移取1ml产物。空气被吸入注射器以确保针头是空的,取出针头,然后将合适的注射针连接到注射器上。产品被推回入注射针。然后在实时超声或间歇导引(intermittent guidance)下(MRI或CT,或使用多模式组合)触及肿瘤。在产品给药期间,将单个针在成像下导引至肿瘤周围。一旦放置确定,则将预定体积的第一次注射,例如如果使用四次注射则每次注射0.2ml,注射到肿瘤内。
40名患者参加了本研究。大多数患者因其他疗法失败而参加,例如,患有正在进展的癌症。观察患者在每次给药后2小时的急性毒性,以及最后一次免疫后30天的临床和实验室毒性以及自身免疫迹象。进行体外免疫学评估以评估免疫应答。通过身体检查测量、放射照相手段和肿瘤活检来测量疾病进展,以评估肿瘤反应和复发。因此,肿瘤生长的稳定被认为是积极的结果。
白细胞分离后约3至4周给予第一次免疫。前三次注射分别间隔1周给予(即第0天、第1周、第2周),然后在第8、16和32周给予,或者只要产品可用并且只要可注射肿瘤块存在于同一个位置。如果原始注射部位的肿瘤质量不足,则使用不同位置的肿瘤块。向患者提供已被活化16小时(方法B)或20小时(方法A)的树突细胞。在每次就诊时注射单一肿瘤,并监测患者的肿瘤生长、存活、肿瘤细胞死亡(坏死)和免疫细胞浸润。图中提供了每个时间点的个体存活的比例。使用标准成像技术(CT或MRI)测量肿瘤生长。

方法B(16小时处理)制备的树突细胞比用欠佳的方法制备的树突细胞产生更多的TNFα:平均149ng/100万DC/24小时,相比于50ng/100万DC/24小时。在第8周,TNFα又与稳定的疾病(SD)相关联(参见上表1),这是更好生存率的预测因子(参见上表2)。
数据表明16小时处理(方法B)对用于癌症免疫疗法中肿瘤内注射的部分成熟的DC的活化而言是最佳的。
Claims (20)
1.用于生产分离的部分成熟且经最佳活化的人树突细胞的方法,包括:
i)从外周血分离包含人PBMC的细胞群;
ii)使所述包含人PBMC的细胞群富含人单核树突细胞前体;
iii)用补充有有效量的树突细胞分化剂的组织培养基培养富含人单核树突细胞前体的细胞群,培养的时间足以使人单核树突细胞前体分化成未成熟的人树突细胞;
iv)用有效量的树突细胞成熟剂培养富含未成熟的人树突细胞的细胞群约10至约19小时,以活化所述未成熟的人树突细胞;和
v)分离并清洗经活化的人树突细胞。
2.用于生产分离的部分成熟且经活化的人树突细胞的方法,包括:
i)分离包含人单核树突细胞前体的细胞群;
ii)用补充有有效量的树突细胞分化剂的组织培养基培养富含人单核树突细胞前体的细胞群,培养的时间足以使人单核树突细胞前体分化成未成熟的人树突细胞;
iii)用有效量的树突细胞成熟剂培养富含未成熟的人树突细胞的细胞群约10至约19小时,以活化所述未成熟的人树突细胞;和
iv)分离并清洗经活化的人树突细胞。
3.根据权利要求2所述的方法,其中所述单核树突细胞前体获自皮肤、脾脏、骨髓、胸腺、淋巴结、脐带血或外周血。
4.根据权利要求1-3中任一项所述的方法,其中所述单核树突细胞前体细胞是未经活化的单核树突细胞前体。
5.根据权利要求1-4中任一项所述的方法,其中所述单核树状细胞前体从待治疗的个体受试者获得。
6.根据权利要求1-4中任一项所述的方法,其中所述单核树突细胞前体从与待治疗个体受试者HLA匹配的健康个体受试者获得。
7.根据权利要求1和2中任一项所述的方法,其中所述树突细胞分化剂是不含任何其他细胞因子的GM-CSF或者是与IL-4、IL-7、IL-13或IL-15组合的GM-CSF。
8.根据权利要求1和2中任一项所述的方法,其中所述树突细胞成熟剂是灭活卡介苗(BCG)、干扰素γ(IFNγ)、脂多糖(LPS)、肿瘤坏死因子α(TNFα)、咪唑并喹啉化合物、合成的双链多聚核糖核苷酸、Toll样受体(TLR)激动剂、已知会诱导树突细胞成熟的含有未甲基化的CpG基序的核酸序列,或其任何组合。
9.根据权利要求8所述的方法,其中所述灭活的BCG包含完整的BCG、BCG的细胞壁组分、BCG衍生的脂阿拉伯糖甘露聚糖或BCG成分。
10.根据权利要求9所述的方法,其中所述灭活的BCG是热灭活的BCG、经福尔马林处理的BCG,或热灭活且经福尔马林处理的BCG。
11.根据权利要求8-10中任一项的方法,其中BCG的有效量为每毫升组织培养基约105-107cfu,IFNγ的有效量为每毫升组织培养基约100-约1,000单位。
12.根据权利要求8所述的方法,其中所述咪唑并喹啉化合物为咪唑并喹啉-4-胺化合物。
13.根据权利要求12所述的方法,其中所述咪唑并喹啉-4-胺化合物是4-氨基-2-乙氧基甲基-α,α-二甲基-1H-咪唑[4,5-c]喹啉-1-5乙醇或1-(2-甲基丙基)-1H-咪唑并[4,5-c]喹啉-4-胺,或其衍生物。
14.根据权利要求8所述的方法,其中所述合成的双链多聚核糖核苷酸是聚[I]:聚[C(12)U]。
15.根据权利要求1-14中任一项所述的方法,其中将部分成熟且经最佳活化的树突细胞直接施用于肿瘤,施用于手术移除或切除肿瘤之后的肿瘤床,施用于肿瘤周围的组织区域,施用于直接引流肿瘤区域的淋巴结,直接施用于将血液或淋巴输送到肿瘤或肿瘤相关器官的循环血管管道,或施用进入循环系统将所述细胞递送到肿瘤或肿瘤相关器官。
16.根据权利要求1-15中任一项所述的方法,其中部分成熟和且经最佳活化的树突细胞作为放射疗法、化学疗法或其组合的佐剂施用。
17.根据权利要求1-15中任一项所述的方法,其中所述经活化的树突细胞在放射疗法、化学疗法或其组合之前、同时或之后施用。
18.用于产生抗肿瘤免疫应答的方法,其包括施用包含富含人树突细胞的细胞群和药学可接受的载体的组合物,所述人树突细胞已经在体外用人树突细胞成熟剂部分成熟并最佳地活化约10至约19小时,其中所述组合物被施用于需要这种治疗的个体的肿瘤、肿瘤床或肿瘤周围的组织区域中。
19.包含部分成熟且经最佳活化的人树突细胞的组合物,其中所述人树突细胞是通过用人树突细胞成熟剂培养未成熟的树突细胞约10至约19小时而诱导成熟的,其中所述部分成熟且经最佳活化的树突细胞产生与炎症反应相关的细胞因子。
20.根据权利要求19所述的组合物,其中所述细胞因子是肿瘤坏死因子α(TNFα)、白细胞介素6(IL-6)和/或白细胞介素8(IL-8)。
