Abstract
一种胚胎干细胞,其是在GSK3抑制剂和MEK抑制剂存在下对小鼠MHC I类的H2‑D分子中的全部或部分结构域被置换为人MHC I类的HLA‑A分子的结构域的小鼠的胚胎进行培养而得到的;以及,一种使用该胚胎干细胞制作的小鼠。

技术领域
本发明涉及从小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为人MHC I类的HLA-A分子的结构域的小鼠中采集的胚胎干细胞(ES细胞)、和脏器被人源化的小鼠。
背景技术
以往,关于人脏器的模型小鼠的制作,例如就肝脏的模型小鼠而言,Heckel等报道了一种导入有构建体(Alb-Plau)的转基因小鼠(Tg(Alb-Plau),所述构建体在白蛋白(Alb)启动子上连接有尿激酶(urokinase)型的纤溶酶原激活物(Plau)基因(非专利文献1:Heckel et al.Cell 62:447-456,1990)),然而,该小鼠在出生后第四天因肠管等的出血而死亡,因此未能用于实验。与此相对地,在Tg(Alb-Plau)中,存活个体的系统建立是成功的,并且报道了利用肝细胞在分裂中发生Alb-Plau基因缺损的肝细胞再生出肝脏的例子(非专利文献2:Sandgren et al.Cell 66:245-256,1991)。另外有将Tg(MT-nLacZ)小鼠的成体肝细胞移植到Tg(Alb-Plau)并获得成功的例子,所述Tg(MT-nLacZ)小鼠是导入有在金属硫蛋白启动子上连接有lacZ基因的构建体的小鼠,即,用作为标记基因的lacZ标记作为供体的肝细胞的转基因小鼠(Tg(MT-nLacZ)小鼠)(非专利文献3:Rhim et al.Science 263:1149-1152,1994)。
进而,有对免疫缺陷小鼠进行人肝细胞的移植的例子。例如有如下例子:对Rag2-/-基因缺损免疫缺陷小鼠移植肝细胞而进行乙型肝炎病毒(HBV)的感染实验的例子(非专利文献4:Dandri et al.Hepatology 33:981-988,2001),或者对于使Tg(Alb-Plau)与作为免疫缺陷小鼠的SCID交配而变为免疫缺陷的SCID小鼠(Tg(Alb-Plau))移植人肝细胞(Tg(Alb-Plau);SCID)),并进行丙型肝炎病毒的感染实验(非专利文献5:Mercer etal.Nature Med.7:927-933,2001)等。
进而,Tateno等使肝脏具有损伤的白蛋白增强子/启动子尿激酶纤溶酶原激活物转基因小鼠(uPA小鼠)与SCID小鼠交配,制作任一者的性状均为同型接合体的uPA/SCID转基因小鼠(非专利文献6:Tateno et al.Amer.J.Pathol165:901-912,2004)。该报道中记载有人肝细胞向Tg(Alb-Plau;SCID)的移植法的改良,通过Futhan处理排除源自人肝细胞的补体的影响,即使为高度嵌合体也使死亡率降低。
进而,还报道了证实以Rag2基因缺损的免疫缺陷小鼠为模型进行基因治疗的可行性的研究(非专利文献7:整形/灾害外科“从分子水平观察的、整形外科疾病系列IV体细胞克隆技术和再生医疗(整形·災害外科「分子レベルからみた整形外科疾患シリーズIV体細胞クローン技術と再生医療」)Vol.45,NO.11,PAGE.1040-1041,2002)。
但是,这些模型小鼠中残留有作为宿主的小鼠的肝脏细胞,并不是100%的细胞被置换为源自人的细胞的肝细胞模型。另外,源自人的细胞未必进行再生,从而不得不移植源自人的细胞。进而,一旦残留源自小鼠的肝脏细胞,则人的肝功能的检验将变得不充分。
另一方面,为了建立进行种系传递的源自NOG小鼠的ES细胞株,还尝试了使用分化信号抑制剂(PD0325901、CHIR99021)来建立ES细胞(非专利文献8:日本实验动物学会总会讲演要旨集Vol.58th,Page 210,2011。)
然而,由于NOG小鼠难以繁殖这一点,难以得到实验用途所需的多个小鼠。
另外,本发明人此前已经成功地由采集自免疫缺陷小鼠的胚胎干细胞制作了肝脏被人源化的模型动物(专利文献1:WO2013/145331)。

发明内容
发明要解决的问题
本发明的目的在于,不是由源自免疫缺陷小鼠、而是由免疫应答正常的小鼠的胚胎来提供脏器被人源化的小鼠。更详细而言,目的在于提供:采集自小鼠主要组织相容性抗原(MHC)I类基因被破坏、取而代之具有人主要组织相容性抗原I类基因的小鼠的胚胎干细胞(ES细胞);以及脏器被人源化的小鼠。
用于解决问题的方案
本发明人为了解决上述课题进行了深入研究,结果成功地制作出通过使用胚胎干细胞制作脏器被人源化的小鼠,所述胚胎干细胞是制作小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为人MHC I类的HLA-A分子的结构域的小鼠的胚胎,在GSK3抑制剂和MEK抑制剂存在下对其进行培养而得到的。
发现通过向该小鼠的胎儿期的卵黄嚢血管中移植人肝细胞,即使不使用以往那样的免疫缺陷小鼠,人肝细胞也成活,至此完成了本发明。
即,本发明如下所述。
(1)一种胚胎干细胞,其是在GSK3抑制剂和MEK抑制剂存在下对小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为人MHC I类的HLA-A分子的结构域的小鼠的胚胎进行培养而得到的。
(2)根据(1)所述的胚胎干细胞,其中,H2-D分子的α1结构域、α2结构域和β2微球蛋白结构域分别被置换为人HLA-A分子的α1结构域、α2结构域和β2微球蛋白结构域。
(3)根据(1)或(2)所述的胚胎干细胞,其保藏号以NITE BP-02068表示。
(4)根据(1)~(3)中任一项所述的胚胎干细胞,其中,导入有雌激素受体基因和白喉毒素基因。
(5)根据(4)所述的胚胎干细胞,其中,细胞中内源性的生长激素基因被置换为源自人的该基因。
(6)根据(5)所述的胚胎干细胞,其中,细胞中内源性的药物代谢酶基因也被置换为源自人的该基因。
(7)根据(6)所述的胚胎干细胞,其中,细胞中内源性的药物代谢酶基因为选自由Cyp3a11、Cyp3a13、Cyp3a25和Cyp3a41组成的组中的至少1种。
(8)一种小鼠,其是使用上述(1)~(3)中任一项所述的胚胎干细胞制作的。
(9)一种小鼠,其是使用上述(4)~(7)中任一项所述的胚胎干细胞制作的。
(10)根据(9)所述的小鼠,其中,通过施予抗雌激素剂引起肝细胞损伤。
(11)一种肝脏被人源化的小鼠,其特征在于,向上述(9)所述的小鼠移植源自人的肝细胞,并且施予抗雌激素剂而除去源自该小鼠的肝细胞。
(12)根据(11)所述的小鼠,其中,源自人的肝细胞是源自具有肝脏疾病的患者的肝细胞。
(13)一种人肝脏疾病模型小鼠,其包括上述(12)所述的小鼠。
(14)一种源自小鼠的胚胎干细胞的制造方法,其特征在于,在GSK3抑制剂和MEK抑制剂存在下,对小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为人MHC I类的HLA-A分子的结构域的小鼠的胚胎进行培养。
(15)根据(14)所述的方法,其中,H2-D分子的α1结构域、α2结构域和β2微球蛋白结构域分别被置换为人HLA-A分子的α1结构域、α2结构域和β2微球蛋白结构域。
(16)一种肝脏损伤模型小鼠的制作方法,其特征在于,对上述(9)所述的小鼠施予抗雌激素剂。
(17)一种肝脏被人源化的小鼠的制作方法,其特征在于,向上述(9)所述的小鼠移植源自人的肝细胞,并且施予抗雌激素剂而除去源自小鼠的肝细胞。
(18)根据(17)所述的方法,其中,源自人的肝细胞为源自具有肝脏疾病的患者的肝细胞。
发明的效果
根据本发明,提供一种胚胎干细胞,其源自免疫应答正常的小鼠,用于确立最适合人的细胞移植的小鼠。就本发明的胚胎干细胞而言,在进行肝脏的人源化时能够导入与肝功能有关的各种人基因,能够建立人源化肝脏模型小鼠。因此,由本发明的胚胎干细胞建立的小鼠能够在人的各种脏器的细胞移植中利用,从能够100%人源化的观点出发是极为有用的。

具体实施方式
以下详细说明本发明。
1.概要
本发明为由正常小鼠中的、小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为源自人MHC I类的HLA-A分子的结构域的小鼠的胚胎建立的胚胎干细胞,进而,由该胚胎干细胞确立了脏器被人源化的小鼠。
通常,向小鼠的胎仔中移植人细胞时,该细胞在小鼠体内成活,因此所制作的小鼠被称为在细胞水平上人源化的小鼠。
但是,这样的人源化小鼠残留有源自作为宿主的小鼠的细胞,因此并不是脏器完全置换为源自人的脏器,在进行该脏器的功能分析、研究方面,未必是最适合的小鼠。另外,为了制作最适合的小鼠,需要进行各种基因修饰,但不能使用小鼠个体来进行。
因此,在本发明中,为了建立具有100%的细胞被人源化的肝脏的小鼠而建立了最适合人源化的基因修饰小鼠。对于该基因修饰小鼠,从个体产生的初期起进行以人源化为目标的深入研究,结果成功地由正常小鼠和HHB小鼠建立了胚胎干细胞(以下称为“ES细胞”)。还使用该ES细胞制作了嵌合小鼠,还成功地制作了进行种系传递的生殖嵌合小鼠。
在本发明中,成功地由HHB小鼠建立了目标ES细胞,如后所述,还成功地由正常小鼠建立了目标ES细胞。
本发明的小鼠是小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为源自人MHC I类的HLA-A分子的结构域的小鼠。
MHC I类分子是作为重链的α链与2个作为轻链的β2-微球蛋白链通过非共价结合而形成的二聚体,肽抗原与其结合而以三聚体形式在细胞表面表达。I类分子包含α1~α3这3个细胞外区域、跨膜区域和细胞内区域这些结构域。其中,成为将源自小鼠的分子置换为源自人的分子的对象的结构域(链)为α链(α1~α3链)和β2-微球蛋白链。在本发明中,可以将全部的结构域置换为人的,但优选使α3结构域为小鼠型的、将α3以外的结构域置换为人的(图1)。
在本发明的优选方式中,本发明的小鼠为破坏了小鼠的H2-D类基因和b2-微球蛋白基因、并且导入有人的HLA-A2.1基因中的α1和α2结构域的小鼠。在该方式中,仅a3结构域为源自小鼠的结构域。将该小鼠称为“HHB小鼠”。
另外,在本发明中,为了使肝脏功能在长期内维持并且确认安全性,而建立具有人正常肝脏的小鼠。进而,为了建立与具有肝脏疾病的人患者症状相同的疾病模型并且分析病态而建立具有人变异肝脏的小鼠。进而,为了开发通用性高的新的治疗方法而建立对人疾病最适合的模型小鼠。
2.小鼠的制作
本发明的小鼠是使用小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为源自人MHC I类的HLA-A分子的结构域的小鼠的胚胎而制作的小鼠。
该小鼠中,H2-D和b2-微球蛋白基因均被敲除、在该位置利用人HLA-A2.1基因进行了置换。优选α1和α2结构域由源自人的基因编码、α3结构域由源自小鼠的基因编码(图1)。将编码图1(左框)所示的分子的基因称为“HHD基因”,将具有HHD基因的小鼠称为“HHB小鼠”。HHB小鼠是已经确立的(Pascolo,S.,Bervas,N.,Ure,J.M.,Smith,A.G.,Lemonnier,F.A.and Perarnau,B.HLA-A2.1-restricted education and cytolytic activity ofCD8+T lymphocytes from b2microglobulin(b2m)HLA-A2.1monochain transgenic H-2Dbb2m double knockout mice.J.Exp.Med.185:2043-2051,1997)。
该敲除了H2-D和b2-微球蛋白基因的小鼠的制作方法和关于该基因的细节记载于Pacolo et al.J.Exp.Med.185:2043-2051,1997中,通常可以利用本领域中熟知的方法、例如使用靶向载体的方法来制作(Capecchi,M.R.,Science,(1989)244,1288-1292)。该方法利用了小鼠ES细胞中的H2-D和b2-微球蛋白基因与靶向载体上的基因的同源重组。
需要说明的是,HHB小鼠也可以从熊本大学生命资源研究支援中心获得。通过使这些小鼠与市售的C57BL/6小鼠回交,可以分别得到与C57BL/6小鼠具有同一品系的遗传背景的H2-D缺损(-/-)小鼠和b2-微球蛋白缺损(-/-)小鼠。
关于H2-D基因和b2-微球蛋白基因两者都缺损的双敲除小鼠的制作,首先使C57BL/6-H2-D缺损小鼠与C57BL/6-b2-微球蛋白缺损小鼠交配而得到F1,然后使F1之间交配而得到F2小鼠。然后,从其中选择H2-D缺损(-/-)和b2-微球蛋白缺损(-/-)的双缺损小鼠(C57BL/6-H2-D-/-:b2-微球蛋白-/-小鼠)即可。作为C57BL/6-H2-D-/-:b2-微球蛋白-/-小鼠的选择方法,例如可以通过PCR法或DNA印迹法(Southern blot)来确认H2-D和b2-微球蛋白这两个基因的缺损。
进而,将HHD基因注入到小鼠受精卵而得到转基因小鼠(Tg(HHD)小鼠)的方法记载于Pacolo et al.J.Exp.Med.185:2043-2051,1997。通过使该小鼠与C57BL/6-H2-D-/-:b2-微球蛋白-/-交配,能够得到C57BL/6-H2-D-/-:b2-微球蛋白-/-:Tg(HHD)(即HHB小鼠)(图1)。
除了HHD基因以外,还可以将小鼠H2-D分子中的全部结构域置换为人的,还可以将α3结构域以外的结构域保持小鼠型的,编码这样的结构域的基因能够通过常规基因工程方法得到。
3.ES细胞的建立
本发明的ES细胞可以通过在GSK3抑制剂及MEK抑制剂存在下对采集自如上得到的小鼠的胚胎进行培养来得到。
例如,在使用HHB小鼠的情况下,首先,通过由受精后的HHB雌小鼠对受精卵或2细胞期胚胎进行培养而得到、或者直接得到囊胚。受精通过自然交配、或体外受精法进行。体外受精通过对使雌小鼠过量排卵而得到的卵子和采集自雄小鼠的精子进行培养而进行。
然后,将采集的囊胚或内部细胞块在动物细胞培养用培养基中且在GSK-3抑制剂及MEK抑制剂存在下培养约1~3周、优选14-18天。
GSK-3(糖原合成酶激酶3)为丝氨酸/苏氨酸蛋白激酶,为作用于涉及糖原的产生、凋亡、干细胞的维持等的许多信号通路的酶。作为GSK-3抑制剂,可以列举:CHIR99021(提供者:和光纯药)、6-溴靛玉红-3’-肟(6-Bromoindirubin-3’-oxime(BIO))(提供者:和光纯药)等。GSK-3抑制剂在培养基中的添加量为0.1~10μM(微摩尔),优选为0.3~3μM。GSK-3抑制剂在培养基中的添加时期没有特别限定,优选从囊胚的培养开始时起添加。
MEK抑制剂为抑制丝裂原活化蛋白激酶激酶(MAP Kinase Kinase(MEK))活性、抑制ERK1/ERK2的活化的蛋白激酶抑制剂。作为MEK抑制剂,可以列举例如:PD0325901(提供者:和光纯药)、U0126(提供者:Promega)等。PD0325901抑制剂在培养基中的添加量没有特别限定,例如为3μM。
培养条件没有特别限定,例如在约37℃、5%CO2的气氛中进行。传代培养可以以3~4天的间隔、在小鼠胚胎成纤维细胞(MEF)饲养层上或包被有胶原酶I的平板上进行。
作为培养基,可以列举例如GMEM培养基(Glasgow’s Minimal EssentialMedium)、DMEM(杜尔贝科改良伊格尔培养基)、RPMI1640培养基等。可以从KSR(KnockoutSerum Replacement,敲除血清替代物)、胎牛血清(FBS)、碱性成纤维细胞生长因子(bFGF)、β-巯基乙醇、非必需氨基酸、谷氨酸、丙酮酸钠及抗生素(例如青霉素、链霉素等)等中选择并适宜添加到培养用培养基中。
培养规定时间后,通过在含有EDTA或胶原酶IV的培养基中孵育来回收ES细胞。所回收的ES细胞也可以根据需要在饲养层细胞存在或非存在下进行培养来进行多次传代。需要说明的是,不含饲养层的条件下的内部细胞块的培养可以在用MEF驯化的培养基中进行。
所培养的ES细胞通常可以使用它们的标记基因进行鉴定。作为ES细胞的标记基因,可以列举例如Oct3/4、碱性磷酸酶、Sox2、Nanog、GDF3、REX1、FGF4等。标记基因或基因产物的存在可以利用PCR或蛋白质印迹等任意方法进行检测。
另外,也可以通过SNP标记的检测或者通过基于PCR或DNA印迹法的分析来确认本发明的ES细胞是否为目标细胞、是否为BALB/c。例如,小鼠的SNP的数据库发布于http://www.broadinstitute.org/snp/mouse,使用该数据库比照SNP信息则可以确认为BALB/c,判断为本发明的ES细胞。
如上得到的ES细胞称为“HHB10”,于2015年6月17日(保藏日)在国家高级工业科学技术学院,国际专利生物保藏中心(〒292-0818千叶县木更津市上总镰足2-5-8)基于布达佩斯条约进行了国际保藏。其保藏号为“NITE BP-02068”。
上述ES细胞的详细信息如下所述。
关于利用GSK抑制剂及MEK抑制剂的ES细胞建立方法,已有论文发表,所得到的ES细胞继承了原品系的遗传性质且含有各自特有的特征。
在预实验中,尝试使用以往所用的GMEM-KSR培养基来建立ES细胞,但仅能建立增殖非常差的细胞株,即使使用该细胞株制作嵌合小鼠,也只能得到50%左右的嵌合,而对种系(germ line)也没有助益。与此相对,在本发明中,通过向培养基加入对维持ES的未分化状态有效的GSK3的抑制剂和MEK的抑制剂,能够建立目标ES细胞。本发明的ES细胞的增殖力高,嵌合也高。其理由在于,与使用以往方法制作的ES细胞相比,良好地保持了未分化状态。在ES细胞的分化中起作用的重要的信号之一为从FGF4经由FGF受体的ERK/MEK通路。即,ERK作为分化的信号起作用。另外,GSK-3通过对b-连环素进行磷酸化来刺激Wnt信号,从而诱导分化。因此,通过使用作为强MEK抑制剂的PD0325901和GSK3抑制剂这2个抑制剂(2i),本发明的ES细胞能够抑制分化、维持多潜能。
4.基因修饰
为了建立最适合人源化的基因修饰小鼠,需要在ES细胞的阶段、而不是成为成体的小鼠阶段将内源基因置换为人的,或者将人基因导入ES细胞之后由该基因修饰和/或基因导入ES细胞制作小鼠。
因此,在本发明中,为了向ES细胞导入目标基因或将ES细胞中内源性的基因置换为人基因,利用基于如下系统的同源重组,所述系统为:作为源自噬菌体的重组系统的Cre-loxP系统、作为源自弧菌属菌的重组系统的VCre-Vlox系统、作为利用了Cre的同源体的重组系统的Dre/rox系统、或对这些重组系统进行了修饰的系统。
1oxP(locus of crossing(X-ring)over,P1)为由34个碱基(5’-ATAACTTCGTATAGCATACAT TATACGAAGTTAT-3’)构成的序列(序列号1),从5’末端起的13个碱基(称为反向重复序列1)及从3’末端起的13个碱基的序列(称为反向重复序列2)分别形成反向重复序列,由“GCATACAT”所示的8个碱基的被称为间隔区的序列夹在上述反向重复序列1和2间(图2)。“反向重复序列”是指以间隔区为边界、一侧的序列与另一侧的序列在彼此相对时互补的序列。
Cre(causes recombination)是指引起基因重组的重组酶(也称为recombinase),其识别上述重复序列并以形成粘末端的切断方式将间隔区部分的“cataca”切断。
其中,在细菌中,在2处1oxP间发生重组,发生插入或删除反应(图2)。如果能够在哺乳类细胞中引起插入反应,则此后能够插入任意的基因,因此,应用性极其广泛。由于哺乳类细胞的核较大,因此,具有暂时被删除的loxP的环状DNA会扩散,几乎观察不到插入反应。
因此,为了引起插入反应,本发明人想到了如下方式:在1oxP序列中导入变异而暂时将基因插入到基因组时,则所插入的基因不能删除(不从基因组中脱离),为此设计出多种变异型1oxP(lox66、lox71、lox511、lox2272)(图2)。这些变异型1oxP是公知的(WO01/005987号公报、日本特开2007-100号公报)。
此外,在本发明中,还可以利用被称为Vlox的系统。Vlox是作为源自弧菌属细菌的重组系统的VCre-Vlox(Suzuki,E.,Nakayama,M.VCre/VloxP and SCre/CloxP:new site-specific recombination systems for genome engineering.Nucleic Acid Res.2011,1-11),可以利用Vlox43L、Vlox43R、Vlox2272等(图2)。
将1oxP、变异型1oxP以及Vlox系统的碱基序列示于以下(图2)。

进而,在本发明中,可以采用Dre/rox系统。
Dre是指D6-位点特异性DNA重组酶,是识别下述rox位点的序列的酶(Sauer,B.andMcDermott,Nucic Acid.Res.32:6086-6095,2004)。将利用了该重组酶和rox识别序列的重组系统称为Dre/rox系统。该系统与Cre-lox系统密切相关,但所识别的DNA特异性不同。
将lox和rox的碱基序列示于以下。

如上所述,本发明的目的在于建立具有人正常组织、例如人肝脏组织的小鼠,进而目的还在于建立组织疾病(例如肝脏疾病)模型小鼠。因此,在本发明中,对ES细胞进行基因操作,从而使毒素在小鼠肝细胞的细胞质中表达而能够诱导小鼠肝细胞死亡。另外,为了制作具有人正常肝脏的小鼠,需要移植人肝细胞并使其增殖,因此,在ES细胞中,将小鼠生长激素基因置换为人生长激素基因。另外,为了分析药物代谢等功能,将小鼠药物代谢酶基因置换为人药物代谢酶基因。
导入了肝细胞死亡的小鼠的肝功能消失,因此该小鼠除了可以作为肝脏损伤模型来利用外,还可以通过移植人正常肝细胞来得到肝脏人源化的小鼠。
图3为用于将小鼠生长激素(GH)基因置换为人GH基因的同源重组载体的构建图。
另外,图4为用于将作为药物代谢酶基因的Cyp基因置换为人Cyp基因的同源重组载体的构建图。
从小鼠基因向上述人基因的置换可以按照WO01/005987号公报中记载的基因捕获法进行。例如,使用如上所述制作的载体进行2阶段基因捕获。
第1阶段为常规基因捕获法。在该常规基因捕获中,向ES细胞导入上述捕获载体,捕获ES细胞内原本存在的内源性基因。由此,ES细胞中的内源性基因被破坏。然后,在质粒(置换载体)上将人基因连接在lox序列(例如1ox66等)的下游,进行第2阶段的基因捕获(图3、4)。
在第2阶段的基因捕获中,将连接在1ox66的下游的人基因(hGH、hCyp等)导入到ES细胞。由此,在通过第1阶段而导入的捕获载体的1ox71位点与通过第2阶段导入的载体的lox66之间发生重组,可以导入含有由“(lox71/66)-(人基因)-(loxP)”构成的盒的修饰基因。需要说明的是,可以在人基因和loxP之间连接嘌呤霉素抗性基因(puro)。
根据该方法,可以用人基因置换内源性小鼠基因。图3及4示出置换等位基因的图。
在此,在图3及图4中,Ex1、Ex2、Ex3及Ex4分别表示小鼠生长激素基因及小鼠Cyp3a13基因的外显子1~4,pA表示poly A序列,Frt表示FLP的识别位点,PGK-neo表示连接有PGK启动子的新霉素抗性基因,P-puro表示连接有PGK启动子的嘌呤霉素抗性基因。
在其它脏器的情况下,只要是能成为脏器移植对象的脏器则都可以与上述肝脏的情况同样进行。即,制作在脏器或组织特异性启动子上连接有Cre-ERT2的基因,将该基因与CAG-lox-EGFP-lox-DT-A等载体(后述的构建体1)一起导入HHB的ES细胞即可。
例如,在制作心脏被人源化的小鼠时,只要制作在作为心肌特异性启动子的αMHC(α-myosin heavy chain,肌球蛋白重链)启动子上连接有Cre-ERT2的基因(MC:α-myosinheavy chain-Cre-ERT2)并将该基因与CAG-loxP-EGFP-loxP-DT-A(CD)一起导入到HHB ES细胞,即可制作HHB:MCCD小鼠。如果向该小鼠的胎仔的心脏移植人心肌细胞,然后施予他莫昔芬,则可以制作小鼠心肌细胞消失、仅人心肌细胞存活的心脏人源化小鼠。如果在胎儿期移植人心肌细胞,则人心肌细胞可以避免排斥反应,因此也可以在形成成体后施予他莫昔芬或将冠状动脉结扎,由此制作心肌梗塞模型,然后移植人心肌细胞。
5.嵌合小鼠的制作
嵌合小鼠的制作可以用标准的方法进行。
首先,使上述建立的ES细胞、或者进行了基因的导入或置换的ES细胞与8细胞期胚胎进行聚集或将其注入囊胚。将如上制作的胚胎称为嵌合胚胎,通过将该嵌合胚胎移植到假孕代孕者的子宫内使其生产而制作嵌合小鼠。
例如,嵌合胚胎的制作中,首先,使利用激素剂实施了过排卵处理的雌小鼠与雄小鼠交配。然后,在规定天数后从输卵管或子宫中回收初期发生胚胎。使ES细胞与回收的胚胎聚集或向回收的胚胎中注入ES细胞,制作嵌合体胚胎。
在此,“胚胎”是指个体发生中的处于从受精到出生前的阶段的个体,包含2细胞期胚胎、4细胞期胚胎、8细胞期胚胎、桑椹期胚胎、囊胚等。在使用8细胞期胚胎的情况下,可以在从受精起的第2.5天从输卵管或子宫中回收初期发生胚胎,在使用囊胚的情况下,可以在从受精起的第3.5天从输卵管或子宫中回收初期发生胚胎。
作为使用ES细胞和胚胎而制作集合体的方法,可以使用显微注射法、聚集法等公知方法。“集合体”是指ES细胞及胚胎在相同空间内聚集而形成的集合体,也意味着将ES细胞注入于胚胎的形态、使胚胎分散成单个细胞并与ES细胞一同聚集的形态中的任一种。
在采用显微注射法的情况下,在回收的胚胎中注入ES细胞而制作细胞的集合体。另外,在采用聚集法的情况下,只要将ES细胞撒在除去了透明带的正常胚胎上使其聚集即可。
另一方面,用于作为代孕者的假孕雌小鼠可以通过使正常性周期的雌小鼠与通过输精管结扎等去势的雄小鼠进行交配来得到。对于所制作的假孕小鼠,将通过上述方法制作的嵌合胚胎移植到子宫内,然后使其生产,由此可以制作嵌合小鼠。
从这样的嵌合小鼠中选择源自ES细胞移植胚胎的雄小鼠。所选择的雄的嵌合小鼠成熟后,使该小鼠与纯系小鼠品系的雌小鼠交配。然后,在诞生的仔鼠中出现源自ES细胞的小鼠的被毛色,由此可以确认多能性干细胞被导入到了嵌合小鼠的种系。
6.人源化小鼠的制作
(1)最适合人源化的基因修饰小鼠的制作
如后所述,使用进行了基因的导入或置换的ES细胞建立的转基因小鼠、即基因修饰小鼠是成为建立具有100%人源化的脏器(例如肝脏)的小鼠的基础的小鼠。
由于已经明确了通过经由胎仔的卵黄嚢静脉进行人肝细胞的移植能够避免排斥反应,因此使用正常小鼠和HHB小鼠的ES细胞。
(i)正常小鼠:
只要是已经建立的近交系小鼠就都可以应用。本发明提供一种脏器被人源化的小鼠的制作方法,其特征在于,向正常小鼠的胎仔的卵黄嚢静脉移植源自人的该脏器的细胞。
(ii)HHB小鼠:
在本发明中,除了上述近交系小鼠以外还可以使用HHB小鼠。HHB小鼠是在C57BL/6小鼠的遗传背景中导入有H2-D和b2-微球蛋白基因的缺损、且导入有HHD基因的小鼠。
(2)肝脏损伤模型小鼠的制作
就肝脏损伤模型小鼠而言,通过施予抗雌激素剂而使毒素表达来除去小鼠肝细胞(使其消失),由此可以制作失去肝功能的损伤模型小鼠。
为了使小鼠肝细胞消失、还为了在小鼠肝细胞的细胞质中表达Cre-ERT2,制作以下的构建体1及构建体2。Cre-ERT2为连接有Cre重组酶基因和变异型雌激素受体基因的载体,所述变异型雌激素受体基因按照不与哺乳类体内产生的雌激素结合的方式进行了修饰。
构建体1:
CAG-ATG-lox-EGFP-lox-DT-A
构建体2:
SAP-Cre-ERT2
构建体1是在紧邻CAG启动子的下游连接有(i)ATG、(ii)被lox夹住的EGFP、和(iii)DT-A(白喉毒素片段A)的构建体。
EGFP的起始密码子与rox上游的ATG以框架相吻合的方式设计。另外,除去DT-A的起始密码子,并以与rox上游的ATG框架相吻合的方式设计。
构建体2是在紧邻肝细胞特异性血清淀粉样蛋白P成分(serum amyloid Pcomponent:SAP)的启动子的下游连接有Cre-ERT2的构建体。
通过将这些构建体1及2共导入本发明的ES细胞,可以在施予他莫昔芬(Tamoxifen)后发生位点特异性重组,肝细胞特异性表达白喉毒素而诱导细胞死亡。
即,非类固醇性的抗雌激素剂、例如他莫昔芬(Tamoxifen)为通过与雌激素竞争性地结合于雌激素受体而显示抗雌激素作用、从而具有抗肿瘤活性的物质。若对表达了Dre-ERT2的人源化小鼠施予他莫昔芬,则Dre-ERT2通过他莫昔芬转移至核。在2个rox间发生重组,白喉毒素基因的启动子发挥作用。由此,毒素DT-A表达,小鼠的肝细胞消失(图5)。
他莫昔芬的施予次数及施予时期只要可以使肝细胞消失就没有特别限定,例如如下进行。
从妊娠第18.5天起,将他莫昔芬以0.1g/200g饲料的比例与粉末饲料混合。2天后产仔,此时给予普通饲料3天。然后,再次给予同浓度的饲料1周,然后给予普通饲料3天。然后再次持续给予同浓度的饲料。
(3)肝细胞被置换为人肝细胞的人源化小鼠的制作
如上所述,肝细胞被置换为人肝细胞的小鼠通过施予抗雌激素剂而除去小鼠肝细胞、并且将人肝细胞移植到小鼠中,由此可以得到肝细胞被置换为人肝细胞的人源化小鼠。
为了长期维持肝脏功能及确认安全性,需要建立具有人正常肝脏的小鼠。
(i)小鼠生长激素基因被置换为人基因的ES细胞的制作
为了使移植后的人肝细胞能够增殖,在ES细胞阶段将小鼠生长激素基因置换为人基因。
具体而言,如上所述以2个步骤来进行ES细胞的基因置换。
在第1个步骤中,使用导入有SAP-Cre-ERT2和CAG-lox-EGFP-lox-DT-A的正常细胞(称为ES:SAP-Cre-ERT2;CAG-lox-EGFP-lox-DT-A(ES:SCCD))和HHB ES细胞(称为HHB ES:SAP-Cre-ERT2;CAG-lox-EGFP-lox-DT-A(HHB ES:SCCD))进行同源重组,建立在起始密码子处破坏小鼠生长激素基因、且在该区域重组有lox71-PGK-neo-loxP的ES细胞(ES:SCCD;Ghneo)或HHB ES细胞(HHB ES:SCCD;Ghneo)。
在第2个步骤中,使用该ES细胞和置换载体,可以建立代替neo基因而重组有人生长激素基因cDNA的ES细胞(ES:SCCD;GhhGH或HHB ES:SCCD;GhhGH)。
使用如上建立的ES细胞,可以得到产生人生长激素的小鼠。
(ii)小鼠肝细胞和未分化肝细胞的除去
他莫昔芬的施予次数和施予时期与上述同样。
(iii)要移植的人肝细胞的制作
要移植的人肝细胞可以由iPS细胞进行诱导。
关于人肝细胞,可以由人iPS细胞使用支持细胞、细胞外基质确立高效的内胚层和肝脏分化诱导方法。
iPS细胞可以通过将编码包括Oct、Sox、Klf、Myc、Nanog、Lin等家族成员在内的3~6个转录因子(核初始化因子)的基因导入到体细胞中来进行诱导(Takahashi,K.,etal.Induction of pluripotent stem cells from fibroblast cultures.Nat.Protoc.2,3081-9(2007);Fusaki N,Ban H,Nishiyama A,Saeki K,Hasegawa M.Efficientinduction of transgene-free human pluripotent stem cells using a vector basedon Sendai virus,an RNA virus that does not integrate into the hostgenome.Proc Jpn Acad Ser B Phys Biol Sci.2009;85(8):348-62.)。
作为Oct家族成员,可以列举例如:Oct3/4、Oct1A、Oct6等,优选Oct3/4。
作为Sox(SRY相关HMG盒)家族成员,可以列举例如:Sox1、Sox2、Sox3、Sox7、Sox15等,优选Sox2。
作为Klf(Kruppel样因子)家族成员,可以列举例如:Klf1、Klf2、Klf4、Klf5等,优选Klf4。
作为Myc家族成员,可以列举c-Myc、N-Myc、L-Myc等,优选c-Myc。
Nanog为在囊胚的内部细胞块中表达最多、在分化细胞中不表达的同源异形盒蛋白。
作为Lin家族成员,可以列举例如作为未分化人ES细胞的标记的Lin28。
更具体而言,作为转录因子,优选Oct3/4、Sox2、Klf4和c-Myc的组合(Takahashi,K.and Yamanaka,S.,Cell 126,663-676(2006)),此外还可以使用Oct3/4、Sox2和Klf4的组合或Oct3/4、Sox2、Klf4和L-Myc的组合。
作为体细胞,可以列举例如皮肤细胞、肝脏细胞、成纤维细胞、淋巴细胞等。
对体细胞的基因导入法可以列举例如脂质转染、电穿孔、显微注射、利用病毒载体的导入等,没有特别限定。作为病毒载体,可以列举例如逆转录病毒载体、慢病毒载体、腺病毒载体、腺伴随病毒载体、仙台病毒等。也可以利用市售的载体、例如仙台病毒载体(DNAVEC)。
在使用载体的情况下,为了使所导入的基因能够表达,也可以以能够起作用的形式与启动子及增强子等调节序列进行连接。作为启动子,有例如CMV启动子、RSV启动子、SV40启动子等。这些载体可以进一步含有抗药性基因(例如嘌呤霉素抗性基因、新霉素抗性基因、氨苄青霉素抗性基因、潮霉素抗性基因等)等正向选择标记、负向选择标记(例如白喉毒素A片段基因或胸苷激酶基因等)、核糖体内部进入位点(IRES(internal ribosomeentry site))、终止子、复制起点等。
若将体细胞(例如0.5×104~5×106细胞/100mm平皿)在约37℃下、在MEF饲养层上或不含饲养层的条件下转染含有上述核初始化因子的载体并进行培养,则在约1~4周后诱导出iPS细胞。
作为培养基,可以列举例如GMEM培养基(Glasgow’s Minimal EssentialMedium)、DMEM(杜尔贝科改良伊格尔培养基)、RPMI1640培养基、OPTI-MEMI培养基等。可以从KSR(Knockout血清替代品)、胎牛血清(FBS)、激活素-A(activin-A)、碱性成纤维细胞生长因子(bFGF)、视黄酸、地塞米松、β-巯基乙醇、非必需氨基酸、谷氨酸、丙酮酸钠及抗生素(例如青霉素、链霉素等)等中选择并适宜添加到培养用培养基。
培养规定时间后,与培养ES细胞时同样地,通过在含有EDTA或胶原酶IV的培养基中孵育来回收细胞。在不含饲养层的条件下,可以将细胞在包被有基质胶的平板上、在用MEF驯化后的培养基中进行。
通常,经过3个阶段来进行从iPS细胞向人肝细胞的分化诱导。作为原则,如下所述:
(a)从多能性干细胞向内胚层系的诱导;
(b)从内胚层系向未成熟肝细胞的诱导;及
(c)从未成熟肝细胞向成熟肝细胞的诱导。
上述(a)中,激活素A、Wnt信号较为重要,(b)中,FGF、BMP较为重要,以及,(c)中,肝细胞生长因子(hepatocyte growth factor)、制瘤素(Oncostatin)、地塞米松(Dexamethasone)较为重要。
但是,上述(b)和(c)的步骤可以适宜替代为DMSO、视黄酸、FGF4、氢化可的松。
人肝细胞的移植时期为妊娠第15.5天、或出生8周前后的成体小鼠。
人肝细胞的移植细胞数优选为105~106个。
关于人肝细胞的移植途径,在胚胎的情况下从卵黄嚢静脉(yolk sac vessel)注入而进行移植(图6)。在成体的情况下,注入到脾脏内。
(iv)人肝细胞的增殖
使用小鼠生长激素基因被置换为人生长激素基因的ES细胞建立的小鼠能够产生人生长激素。该人生长激素作用于所移植的人肝细胞,促进其生长,能够建立具有正常尺寸的人肝脏的人源化肝脏小鼠。
关于小鼠肝细胞全部(100%)被置换为人肝细胞这一点的确认、即不存在小鼠肝细胞这一点的确认,可以利用RT-PCR法等分析在小鼠肝脏中表达的基因的表达来进行。
(4)人源化肝脏小鼠的评价
肝脏被人源化这一点可以通过单独检查以下事项或将以下事项适宜组合并检查来进行确认。
(i)肝功能的检验
作为用于检验肝功能的检查项目,可以列举例如以下的项目。检查期没有特别限定,优选进行1年以上。
蛋白相关:总蛋白、ALB、TTT、ZTT、CRP、触珠蛋白、C3、C4
非蛋白性氮成分:总胆红素,直接胆红素
糖质:葡萄糖
脂质:甘油三酸酯、总胆固醇、HDL-胆固醇、LDL-胆固醇、ApoAI、ApoCII
酶:乳酸脱氢酶(LDH)、天冬氨酸氨基转移酶(AST(GOT))、丙氨酸氨基转移酶(ALT(GPT))、γ-谷氨酰转移酶(GGT)、肌酸激酶(CK)、碱性磷酸酶(AP)、淀粉酶(AML)
其它:钙、Fe、无机磷酸
ICG检查:经静脉施予吲哚菁绿(ICG),经时测定血中的ICG浓度,检查肝脏的色素排泄功能。ICG与血中的脂蛋白结合而被输送至肝脏,在通过血窦期间被肝细胞摄取,不接受抱合而排泄至胆汁,因此,可以分析肝脏整体的作为脏器的功能、而不是分析肝细胞的功能。
CT检查:检查肝脏的形态变化。
(ii)药物代谢
使用PCR阵列法分析以下的药物代谢相关酶。
·细胞色素P450:CYP11A1、CYP11B1、CYP11B2、CYP17A1、CYP19A1、CYP1A1、CYP1A2、CYP1B1、CYP21A2、CYP24A1、CYP26A1、CYP26B1、CYP26C1、CYP27A1、CYP27B1、CYP2A13、CYP2R1、CYP2S1、CYP2B6、CYP2C18、CYP2C19、CYP2C8、CYP2C9、CYP2D6、CYP2E1、CYP2F1、CYP2W1、CYP3A4、CYP3A11、CYP3A13、CYP3A43、CYP3A5、CYP3A7、CYP3A25、CYP3A41、CYP4A11、CYP4A22、CYP4B1、CYP4F11、CYP4F12、CYP4F2、CYP4F3、CYP4F8、CYP7A1、CYP7B1、CYP8B1。
在本发明中,优选选自由CYP3A11、CYP3A13、CYP3A25和CYP3A41中的至少1种。
需要说明的是,小鼠细胞中的内源性的药物代谢相关酶基因除了首字母以外用小写来表示。例如,人的“CYP11A1”基因在小鼠的情况下记作“Cyp11a1”基因,人的“CYP3A11”基因在小鼠的情况下记作“Cyp3a11”基因。
·醇脱氢酶:ADH1A、ADH1B、ADH1C、ADH4、ADH5、ADH6、ADH7、DHRS2、HSD17B10(HADH2)。
·酯酶:AADAC、CEL、ESD、GZMA、GZMB、UCHL1、UCHL3。

(iii)肝细胞功能的体外检验
由于肝细胞源自内胚层,因此,通过调查内胚层系及肝细胞内表达基因的经时表达、糖原的蓄积、细胞色素酶的表达等,能够检验是否具有人肝脏的功能。
在内胚层系及肝细胞中表达的基因的经时表达可以用Oct3/4、T、Gsc、Mixl1、Foxa2、Hex、Hnf4a、Hnf6、Afp、Alb、Ttr、αAT等检验。其检验方法例如为一般的RNA印迹法、RT-PCR法、蛋白质印迹法。
肝细胞的分泌能力可以通过培养液中的ALB、转铁蛋白(transferring)、alpha1抗胰蛋白酶(alpha1-antitrypsin)、纤维蛋白原(fibrinogen)的浓度测定来检验。其检验方法例如为一般的蛋白质印迹法或EIA(酶联免疫分析;enzyme-immuno assay)法。
糖原的蓄积可以通过PAS(过碘酸-希夫染色反应;periodic acid-Schiff)染色来检验。过碘酸选择性地氧化葡萄糖残基而生成醛,在希夫试剂作用下变色为红紫色。
细胞色素酶的表达可以通过5个主要的细胞色素、即CYP3A4、CYP1A2、CYP2C9、CYP2C19和CYP2D6的分析来检验。其检验方法例如为一般的RNA印迹法、RT-PCR法、蛋白质印迹法。
(5)利用源自人患者的肝细胞进行了置换的肝脏疾病模型小鼠的制作
对本发明的小鼠移植源自人患者的肝细胞,并且施予抗雌激素剂而除去源自小鼠的肝细胞,由此可以得到人肝脏疾病模型小鼠。
为了建立与人患者症状相同的疾病模型及分析病态,需要建立具有人变异肝脏的小鼠。然后可以在建立最适合人疾病的模型、开发通用性高的新的治疗法中利用。
以下,通过实施例对本发明进一步具体地进行说明。但是,本发明并不受这些实施例限定。需要说明的是,关于由iPS细胞起的肝细胞诱导、由来自人家族性淀粉样变多发性神经病的患者或人丙酸血症的患者起的iPS细胞建立、使用诱导出的人肝细胞的向小鼠的移植实验等,已向伦理委员会、动物实验委员会、第2种重组DNA实验安全委员会申请并全部得到许可。
实施例1
ES细胞的建立
在本实施例中,为了确立最适合人肝细胞移植的人源化最适小鼠,由HHB小鼠胚胎进行ES细胞株的建立,也确立了小鼠品系。
(1)HHB小鼠的建立及其ES细胞株的建立
使用HHB小鼠,进行体外受精而得到33个囊胚胚胎,将对于维持ES的未分化状态有效的作为GSK3的抑制剂的CHIR99021、和作为MEK的抑制剂的PD0325901加入到培养基中(GMEM-KSR-2i培养基),尝试建立ES。
具体而言,通过体外受精采集HHB的胚胎。将33个囊胚在KSOM培养基上培养4天直到形成胚囊,在48个孔(仅包覆明胶)中各放入1个胚胎。使用的培养基为KSR-GMEM-2i培养基。作为该培养基组成,在G-MEM(Glasgow minimum essential medium)中含有1X MEM非必需氨基酸、0.1mMβ-巯基乙醇、1mM丙酮酸钠、1%胎牛血清(FBS)(Hyclone)、14%KnockoutTMSR(KSR)、1100单位/ml白血病抑制因子(LIF)、2μM PD0325901和3μM CHIR99021。培养期为14天,中途进行2次培养基更换。14天后开始至18天后,从ICM逐渐增加的孔向带有饲养层细胞的24孔进行传代。进而,依次向12孔板、6孔板、6cm培养皿中进行传代,最终成功建立了21个增殖速度和形态都没有问题的ES株。
(2)使用了HHB ES细胞株的嵌合小鼠制作和HHB小鼠品系的建立
使用成功建立的ES株中的12个细胞系,与通过B6雌和BDF1雄的交配而得到的桑葚胚胎进行聚集,由此进行嵌合小鼠的制作(表1)。
由通过3个ES系(HHB-3,HHB-9及HHB-10)得到的100%嵌合确认了向种系的传递。
需要说明的是,将得到的ES细胞中的第10个细胞系称为“HHB10”,于2015年6月17日(保藏日)保藏在国家高级工业科学技术学院,国际专利生物保藏中心(〒292-0818千叶县木更津市上总镰足2-5-8)基于布达佩斯条约进行了国际保藏。其保藏号为“NITE BP-02068”。

实施例2
小鼠肝细胞死亡的诱导
(1)用于诱导小鼠肝细胞死亡的构建体的制作
为了制作能够使肝细胞特异性消失的基因修饰小鼠,制作了两种构建体。
构建体1(CAG-ATG-lox-EGFP-lox-DT-A)在紧邻CAG启动子的下游连接有ATG、被lox夹持的EGFP、和DT-A(白喉毒素片段A)。
EGFP的起始密码子和rox上游的ATG以框架吻合的方式进行设计。另外,除去DT-A的起始密码子,并以与rox上游的ATG框架相吻合的方式进行设计。
构建体2(SAP-CreERT2)在紧邻肝细胞特异性血清淀粉样蛋白P成分(serumamyloid P component:SAP)的启动子的下游连接有Cre-ERT2。而且,在SAP启动子的上游连接有嘌呤霉素抗性基因。
具体的方法如下所述。
(1-1)构建体1
构建体1如下来制作。
(i)将p6SEAZ用PstI进行限制性内切酶处理,将pSP-rox2用KpnI进行限制性内切酶处理,用T4连接酶(TaKaRa)制成平末端。然后,用EcoRI进行限制性内切酶处理并连接,由此制作pSP-lox-EGFP-lox。
(ii)将pSP-lox-EGFP-lox和pBSK-atg-rox2(合成DNA,Biomatik公司)用EcoRI和SmaI进行限制性内切酶处理并连接,由此制作pBSK-atg-lox-EGFP-lox。
(iii)将pBSK-atg-lox-EGFP-lox和P71hAXC-DT用BamHI和PstI进行限制性内切酶处理并连接,由此制作pBSK-atg-lox-EGFP-lox-DT-A。
(iv)将pCAGGS-EGFP用KpnI进行限制性内切酶处理,将pBSK-atg-lox-EGFP-lox-DT-A用SpeI进行限制性内切酶处理,用T4聚合酶(TaKaRa)制成平末端。然后,用Hind III进行限制性内切酶处理并连接,由此制作CAG-atg-lox-EGFP-lox-DT-A。
(1-2)构建体2
构建体2如下来制作。
(i)以pkSAP-CrePP为模板,用PCR对从起始密码子至终止密码子为止进行扩增。反向引物上附加有BamHI位点。
PCR kit TaKaRa Ex Taq
正向引物CCATGGCCCCCAAGAAGAAAA(序列号14)
反向引物CGGGATCCATGAGCCTGCTGTT(序列号15)
将pGEM-T Easy Vector和上述PCR产物连接,由此制作T easy-Dre。
(ii)将pkSAP-CrePP和T easy-Cre用SalI和EcoRI进行限制性内切酶处理并进行连接,由此制作T Easy SAP。
(iii)将上述T Easy Cre和T easy-SAP用SacII和NotI进行限制性内切酶处理并进行连接,由此制作T easy-SAP-Cre。
(iv)使用T Easy-SAP-Cre和pkSA-CremERT2PP,用BamHI和NotI进行限制性内切酶处理并进行连接,由此制作T easy-SAP-CremERT2。
(v)将pkSAP-CrePP和T easy-SAP-CremERT2用SalI和NotI进行限制性内切酶处理并进行连接,由此制作pKSAP-CreERT2。
(vi)将pKSAP-CreERT2用SpeI进行限制性内切酶处理,将pFPacpaF2用KpnI进行限制性内切酶处理,用T4聚合酶(TaKaRa)制成平末端。然后,将pKSAP-CreERT2用SalI进行限制性内切酶处理,将pFPacpaF2用XhoI进行限制性内切酶处理并进行连接,由此制作Puro-SAP-CreERT2。
(2)雌激素受体基因及白喉毒素基因向ES细胞的导入
对插入人基因时使其高效表达的条件进行了研究
(Li,Z.et al.,Transgenic Res.20:191-200,2011.DOI10.1007/s11248-010-9389-22)。
通过有无PGK-嘌呤霉素盒和IRES来分析怎样组合时表达效率最好。
使用同源重组载体,利用常规方法(Zhao,G.,Li,Z.,Araki,K.,Haruna,K.,Yamaguchi,K.,Araki,M.,Takeya,M.,Ando,Y.and Yamamura,K.Inconsistency betweenhepatic expression and serum concentration of transthyretin in mice humanizedat the transthyretin locus.Genes Cells 13:1257-1268,2008.)预先破坏了小鼠甲状腺素运载蛋白(Ttr)基因的第1外显子。此时得到第1外显子的ATG被破坏、且在该部分嵌入有lox71-PGK-beta-geo-loxP-poly A-lox2272的靶重组克隆。
然后制作2种置换载体。置换载体1含有lox66-hTTR cDNA-polyA-Frt-PGK-puro-Frt-loxP。置换载体2含有lox66-IRES-hTTR cDNA-polyA-Frt-PGK-puro-Frt-loxP。将这些置换载体分别与Cre表达载体一起通过电穿孔法导入到靶重组克隆。
其结果是,得到lox71/66-hTTR cDNA-polyA-Frt-PGK-puro-Frt-loxP克隆(简称为I(-)P(+))和lox71/66-IRES-hTTR cDNA-polyA-Frt-PGK-puro-Frt-loxP克隆(简称I(+)P(+))。这2个克隆均具有PGK-puro,但I(-)P(+)不具有IRES。
通过电穿孔法向这2个克隆中导入CAG-FLP,删除Frt间的PGK-puro,由此制作I(-)P(-)和I(+)P(-)克隆。
由上述4个ES克隆制作小鼠并进行表达分析,结果获知:I(-)P(+)中的表达最高,且表达按照I(-)P(-)、I(+)P(+)、I(+)P(-)的顺序下降。另外获知:I(-)P(+)的情况下,肝脏中的人TTR(甲状腺素运载蛋白)的表达与对照小鼠中的小鼠Ttr(甲状腺素运载蛋白)的表达水平基本相同。
该结果表明:存在PGK-嘌呤霉素且不存在IRES的组合的情况下,所插入的人基因的表达效率最好。
实施例3
利用人生长激素基因的置换
使用同源重组载体,与实施例2同样地通过常规方法预先破坏小鼠生长激素(growth hormone,Gh)基因的第1外显子和第2外显子。此时得到第1外显子的ATG被破坏、且在该部分嵌入有lox71-PGK-beta-geo-loxP-poly A-lox2272的靶重组克隆。然后制作置换载体。置换载体含有lox66-基因组hGH基因-polyA-Frt-PGK-puro-Frt-loxP。将该置换载体与Cre表达载体一起通过电穿孔法导入到靶重组克隆。
其结果是,得到小鼠的Gh基因被置换为人GH基因的ES克隆。
实施例4
利用人药物代谢酶基因的置换
利用常规方法预先用同源重组载体破坏小鼠Cyp3a13基因的第1外显子。此时得到第1外显子的ATG被破坏、且在该部分嵌入有lox71-PGK-beta-geo-loxP-poly A-lox2272的靶重组克隆。然后制作置换载体。置换载体含有lox66-hCYP3A4cDNA-polyA-Frt-PGK-puro-Frt-loxP。将该置换载体与Cre表达载体一起通过电穿孔法导入到靶重组克隆。
其结果是,得到小鼠的Cyp3a13基因被置换为人CYP3A4基因的ES克隆。
实施例5
肝脏人源化小鼠的制作
基本确立了由人iPS细胞起的人肝细胞分化诱导方法,也进行了用于诱导小鼠肝细胞死亡的构建体的制作。
(1)由人iPS细胞起的肝细胞分化诱导
构建了由人iPS细胞起的高效的内胚层和肝脏分化诱导方法。
为了由iPS细胞分化为人肝细胞,从首日到第2天用含有Rock抑制剂的培养基进行培养。然后,从第3天到第4天用DMEM培养基进行培养。该DMEM培养基含有以下成分:4500mg/l葡萄糖、激活素A(100ng/ml)、CHIR99021(3mm)、bFGF(50ng/ml)。
然后,从第4天到第13天加入4500mg/l葡萄糖、地塞米松1mm、肝细胞生长因子10mm进行培养。
最后,从第15天到第30天用含有以下成分的DMEM培养基进行培养:10%KSR、地塞米松1mm、肝细胞生长因子10mm、制瘤素M(30ng/mL)。
(2)源自iPS的人肝细胞的移植方法的研究
为了确立使肝脏人源化所必需的、向小鼠肝脏高效地导入源自iPS的肝细胞的方法,开发出将源自iPS的人肝细胞经由妊娠期第16.5天或第17.5天的小鼠胎仔的羊膜上所存在的卵黄嚢血管(yolk sac vessel)进行导入的方法(图6)。
将上述(1)中所制作的肝细胞用于移植。
利用该培养方法,培养第4天时分化诱导为Sox17阳性的内胚层,培养第7天时分化诱导为AFP阳性的未成熟的肝脏细胞,培养第16天时分化诱导为ALBUMIN阳性的成熟肝细胞。
另外,通过使用小鼠特异性引物的RT-PCR分析,肝脏细胞中确认不到小鼠基因的表达,因此可知其100%源自人。
移植肝细胞,在第14天时将肝脏取出体外,用抗人细胞角蛋白8/18抗体进行免疫染色,结果确认人肝细胞已成活。另外,4周后进行同样的分析,结果获知人肝细胞的集落尺寸扩大、且人肝细胞已嵌入肝小叶结构中。
实施例6
变异人源化肝脏小鼠的建立
本实施例中进行FAP和PA的模型小鼠的繁殖。
(1)源自人患者的变异肝细胞的诱导
(i)家族性淀粉样变多发性神经病的患者(FAP):已建立
FAP是由甲状腺素运载蛋白(transthyretin:TTR)基因的点突变引起的常染色体显性遗传病。例如,FAP中,甲状腺素运载蛋白的氨基酸序列中的第30位的氨基酸、即缬氨酸被置换为甲硫氨酸(Val30Met)。使用采集自具有该Val30Met变异的患者的成纤维细胞建立了iPS细胞。
并且可知:通过与此前所记载的方法相同的方法,可以由该iPS细胞分化诱导为肝细胞。
(ii)由人丙酸血症(PA)患者建立iPS细胞
PA是由丙酰辅酶A羧化酶(propionyl CoA carboxylase:PCCA)基因的异常引起的常染色体隐性遗传病。例如,在PA中,PCCA的氨基酸序列中的第52位的精氨酸被置换为色氨酸(Arg52Trp)。使用采集自具有该变异的患者的成纤维细胞建立了iPS细胞。并且可知:通过与此前所记载的方法相同的方法,可以由该iPS细胞分化诱导为肝细胞。
(2)变异人源化肝脏小鼠(FAP和PA的模型小鼠)的建立
关于变异人源化肝脏小鼠的建立,可以与人源化肝脏小鼠(将由源自正常人的iPS诱导的肝细胞移植而制作的小鼠)的制作方法同样地,将由源自FAP和PA患者的iPS细胞分化诱导得到的肝细胞移植到本发明的小鼠中,从而建立。
产业上的可利用性
根据本发明,提供源自HHB小鼠的ES细胞。通过向使用本发明的ES细胞制作的胚胎中移植人细胞,由此可以制作目标脏器(例如肝脏)被人源化的小鼠,可以研究人的脏器功能。

Claims (18)
1.一种胚胎干细胞,其是在GSK3抑制剂和MEK抑制剂存在下对小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为人MHC I类的HLA-A分子的结构域的小鼠的胚胎进行培养而得到的。
2.根据权利要求1所述的胚胎干细胞,其中,H2-D分子的α1结构域、α2结构域和β2微球蛋白结构域分别被置换为人HLA-A分子的α1结构域、α2结构域和β2微球蛋白结构域。
3.根据权利要求1或2所述的胚胎干细胞,其保藏号以NITE BP-02068表示。
4.根据权利要求1~3中任一项所述的胚胎干细胞,其中,导入有雌激素受体基因和白喉毒素基因。
5.根据权利要求4所述的胚胎干细胞,其中,细胞中内源性的生长激素基因被置换为源自人的该基因。
6.根据权利要求5所述的胚胎干细胞,其中,细胞中内源性的药物代谢酶基因也被置换为源自人的该基因。
7.根据权利要求6所述的胚胎干细胞,其中,细胞中内源性的药物代谢酶基因为选自由Cyp3a11、Cyp3a13、Cyp3a25和Cyp3a41组成的组中的至少1种。
8.一种小鼠,其是使用权利要求1~3中任一项所述的胚胎干细胞制作的。
9.一种小鼠,其是使用权利要求4~7中任一项所述的胚胎干细胞制作的。
10.根据权利要求9所述的小鼠,其中,通过施予抗雌激素剂引起肝细胞损伤。
11.一种肝脏被人源化的小鼠,其特征在于,向权利要求9所述的小鼠移植源自人的肝细胞,并且施予抗雌激素剂而除去源自该小鼠的肝细胞。
12.根据权利要求11所述的小鼠,其中,源自人的肝细胞是源自具有肝脏疾病的患者的肝细胞。
13.一种人肝脏疾病模型小鼠,其包括权利要求12所述的小鼠。
14.一种源自小鼠的胚胎干细胞的制造方法,其特征在于,在GSK3抑制剂和MEK抑制剂存在下,对小鼠MHC I类的H2-D分子中的全部或部分结构域被置换为人MHC I类的HLA-A分子的结构域的小鼠的胚胎进行培养。
15.根据权利要求14所述的方法,其中,H2-D分子的α1结构域、α2结构域和β2微球蛋白结构域分别被置换为人HLA-A分子的α1结构域、α2结构域和β2微球蛋白结构域。
16.一种肝脏损伤模型小鼠的制作方法,其特征在于,对权利要求9所述的小鼠施予抗雌激素剂。
17.一种肝脏被人源化的小鼠的制作方法,其特征在于,向权利要求9所述的小鼠移植源自人的肝细胞,并且施予抗雌激素剂而除去源自小鼠的肝细胞。
18.根据权利要求17所述的方法,其中,源自人的肝细胞为源自具有肝脏疾病的患者的肝细胞。
