一种牦牛骨含钙提取物的制备方法,本发明以120目骨粉为原料,采用稀盐酸溶解,静置后固液分离,经干燥后得到牦牛骨含钙提取物。经试验检测,本发明的骨粉收率70%以上,剩余的30%渣可用于家禽家畜饲料的添加剂;提取物中钙含量14.7%,总磷含量7.35%,并富含其他矿物质和营养有机物。本发明具有生产工艺简单、操作方便、生产周期短及生产成本低(仅为目前市场同类产品的1/3)的优点,并由于未使用强酸强碱、且提取液回收再用,使生产过程无废液废气排放,对生产设备的腐蚀和环境的污染降到最低;所得产品完全溶于水、无异味、容易被人体吸收,适用于各类饮品和各种冲剂及食品的补钙添加剂。

技术领域
本发明属于化学提取技术领域,具体涉及一种牦牛骨含钙提取物的制备方法。
背景技术
我国西部牦牛资源丰富,据藏药典籍《晶珠本草》记载,牦牛骨增热量、祛寒、生胃火、治胃寒。牦牛骨中含有丰富的钙、锌、磷、镁、铁、铜、锰、镍等十多种人体所需元素。传统藏医、蒙医和中医都将青藏高原特有的牦牛骨作为珍贵药材,因此牦牛骨是一种具有极高营养价值的食材。但由于牦牛骨的骨骼结构特殊,使其中所含的营养成分不易被人体消化吸收,须经过特殊加工才能被人体吸收利用。市场上主要的骨产品为骨胶、骨油、骨粉等,这些产品存在不易溶水、口感差,且营养成分难以被人体吸收等问题,不能满足市场对这类食品的需求,每年有大量的牦牛骨被废弃而不能利用。目前骨产品的加工技术主要有,酸解法、碱解法、酶解法和微生物发酵等,加工技术落后、成本高、有污染,且产品难以被人体吸收。
(1)酸解法:采用酸拆解胶原纤维中蛋白质的盐键、酯键,使与胶原纤维结合的羟基磷灰石裸露;再以酸作用于羟基磷灰石让骨钙转化为可溶性钙。该法可以更多地保留骨内的矿物质元素,所需生产设备少、生产过程易于操作,不足的是生产过程中强酸对设备的腐蚀及废液废气的排放。
(2)碱解法:采用碱处理的目的主要是除去骨粉内外包围缠绕的蛋白质和脂肪,进一步减小骨粉粒度,从而有利于人体对钙的吸收。但是碱解法会将蛋白质中的L-型氨基酸部分转变为D-型氨基酸,且水解过程中破坏氨基酸比较多,所以此法用的相对较少。
(3)酶解法:采用合适的酶分解骨粉中的残留蛋白质,使骨钙充分分离出来,并与氨基酸作用生成可溶性氨基酸钙促进钙的吸收。该法反应时间长,对生产设备和生产过程的控制要求较高,生产周期长、成本高。
(4)微生物发酵法:是利用微生物的降解作用,将骨粉中的蛋白质和多糖降解,使部分钙从结合态游离出来,再经分离提取获得。该方法受菌种品质和生产控制条件的影响很大,对生产设备和控制条件要求高,故生产工艺操作复杂、生产成本较高,并有废液废气的排放。
中国专利CN1483351A公开了一种牛骨提取铁钙蛋白食品及方法,是以牛骨为原料,通过清洗、酸浸泡、漂洗、加碱中和、晾晒除水、油锅固化、碱水泡发、清洗等步骤完成。该方法生产操作复杂且周期长,其中盐酸浸泡为3~20天,另外纯碱的浸泡再中和,容易破坏骨中的氨基酸。
总之,目前骨产品的加工方法操作工艺复杂、生产成本高、排放有污染,所得产品水溶性差、口感差、且难以被人体吸收。
发明内容
为解决上述存在问题,本发明提出一种牦牛骨含钙提取物的制备方法,通过酸拆解将牦牛骨中丰富的营养物质释放,转化为易溶于水、人体易吸收的营养物质形态,从而用于补钙保健品生产,具有操作简便、生产成本低、无污染排放的优点。
本发明提出的一种牦牛骨含钙提取物的制备方法,其具体步骤包括:
步骤一:将检验检疫合格的牦牛骨原料清洗干净,在0.2~0.4Mpa和120~140℃条件下蒸煮2h后,再清洗干燥后粉碎成100~120目的骨粉;
步骤二:将骨粉按1:10的质量比加入到3.2%的稀盐酸溶液中,常温条件下搅拌1.5~2.5h后,静置至混合液分层;
步骤三:将下层混合液用清水洗涤2~3次后,得到的沉淀物可作为家禽家畜饲料的补钙添加剂,洗涤水回收用于配制稀盐酸溶液;
步骤四:将步骤二得到的上层悬浮液,通过真空冻干后粉碎,得到牦牛骨提取物冻干粉,可用于液体饮品或粉状冲剂的添加剂;或者将上层悬浮液通过喷雾干燥方式,得到提取物干粉,可用于液体饮品生产或粉状冲剂生产的添加剂;或者是将上层悬浮液在通风条件下自然干燥,得到牦牛骨胶状提取物,可用于液体饮品生产或粉状冲剂生产的添加剂;
步骤五:将步骤四得到的胶状提取物按3:90~120的质量比用热水溶解,所得溶液可用于液体饮品生产的原料;
步骤六:为解决沉淀问题,可以将步骤四得到的胶状提取物与柠檬酸按3:1的质量比进行混合,所得混合物按3:100~120的质量比用热水溶解,所得溶液可用于液体饮品生产的原料;
步骤七:将步骤六得到的混合物与淀粉按照1:2的质量比进行混合研磨后成粉,可用于粉状冲剂的生产。
本发明以牦牛骨为原料,利用稀盐酸拆解牦牛骨中的盐键、酯键等化学键,再分解骨骼晶体组织,从而使钙等有益元素分离,释放出可溶性物质和骨钙等有益元素,经提取、分离、干燥,所得牦牛骨含钙提取物可完全溶于水,实现了将牦牛骨中有益成分为人体所用的目的。由于本发明采用酸解法最大限度地保留了骨中的氨基酸、多糖及造血物质等不被破坏;同时,也使牦牛骨中钙等矿物质能以离子状态溶于水更易于人体吸收。经试验检测,本发明方法的骨粉收率70%以上,剩余的30%渣可用于家禽家畜饲料的添加剂;提取物中钙含量14.7%,总磷含量7.35%,并富含其他矿物质和营养有机物。本发明具有生产工艺简单、操作方便、生产周期短及生产成本低(仅为目前市场同类产品的1/3)的优点,并由于未使用强酸强碱、且提取液回收再用,使生产过程无废液废气排放,对生产设备的腐蚀和环境的污染降到最低;所得产品完全溶于水、无异味、容易被人体吸收,适用于各类饮品和各种冲剂及食品的补钙添加剂。

具体实施方式
以下结合实例对本发明技术方案的具体实施方式详细说明。实例中所选用试剂与用量以及条件仅用于说明本发明的实施方式,并不限定本发明所要求保护的内容。
实例中材料准备:1)取10kg检验检疫合格的牦牛骨(购于西宁)原料清洗干净,在0.2Mpa和120℃条件下蒸煮2h后,经清洗干燥、粉碎过120目筛,得到骨粉(含水量0.6%);2)采用食品级盐酸(含量31%)配制3.2%的稀盐酸溶液;3)所用柠檬酸采用食品级一水柠檬酸(含量99.9%)。实例1.
步骤一:将100g骨粉加至1L的3.2%稀盐酸溶液中,常温下搅拌1.5h后,静置2h至混合液分层,得到下层混合液和上层悬浮液;
步骤二:将下层混合液以离心方式再进行固液分离,所得上清液与上述的上层悬浮液混合,沉淀物用清水洗涤2次并固液分离;将洗涤后的沉淀物干燥后粉碎作为饲料的添加剂,将分离的洗涤水回收用于稀盐酸溶液的配制;
步骤三:将步骤一的上层悬浮液在通风条件下自然干燥,得到含水的胶状提取物98.4g,可作为液体饮品生产或粉状冲剂生产的添加剂。
如:将3g胶状提取物溶于90mL的80℃热水中,得到无色、无味的含钙提取物的水溶液(有少许白色沉淀)。另将3g胶状提取物中加入1g柠檬酸混合,再加入100mL的70℃热水,得到无色、无味的含有钙提取物的水溶液(无沉淀);经检测,pH为2,含钙量14.9%。
实例2.
步骤一:将500g骨粉加至5L的3.2%稀盐酸溶液中,常温下搅拌2h后,静置2.5h至混合液分层,得到下层混合液和上层悬浮液;
步骤二:将下层混合液用清水洗涤3次并固液分离;将洗涤后的沉淀物和洗涤水按上述实例1方式处理;
步骤三:将步骤一的上层悬浮液在通风条件下自然干燥,得到含水的胶状提取物481.4g,可作为液体饮品生产或粉状冲剂生产的添加剂。
如:将6g胶状提取物溶于200mL的76℃热水中,得到无色、无味的含有钙提取物的水溶液(有少许白色沉淀),pH为2,含钙量15.2%。另将6g胶状提取物中加入柠檬酸2g混合后,再加入16g的淀粉研磨,得到白色、无味的含钙提取物的粉状物,可用于粉状冲剂生产的添加。
实例3.
步骤一:将1000g骨粉加至10L的3.2%稀盐酸溶液中,常温下搅拌2.5h后,静置3h至混合液分层,得到下层混合液和上层悬浮液;
步骤二:将下层混合液用清水洗涤3次并固液分离;将洗涤后的沉淀物和洗涤水按实例1方式处理;
步骤三:将步骤一得到的上层悬浮液,通过真空冻干后粉碎,得到白色、无味的含有钙提取物的冻干粉705.3g。
或将步骤一得到的上层悬浮液,通过喷雾干燥后,得到白色、无味的含钙提取物干粉702.6g。
经检测,所得含钙提取物中钙含量14.7%,总磷含量7.35%,并富含其他矿物质和营养有机物,适用于液体饮品或粉状冲剂的添加剂,或与其他食品复配。
Claims (4)
1.一种牦牛骨含钙提取物的制备方法,其特征在于:具体步骤包括:
步骤一:将检验检疫合格的牦牛骨原料清洗干净,在0.2~0.4Mpa和120~140℃条件下蒸煮2h后,再清洗干燥后粉碎成100~120目的骨粉;
步骤二:将骨粉按1:10的质量比加入到3.2%的稀盐酸溶液中,常温条件下搅拌1.5~2.5h后,静置至混合液分层;
步骤三:将下层混合液用清水洗涤2~3次后,得到的沉淀物作为饲料的补钙添加剂,洗涤水回收用于配制稀盐酸溶液;
步骤四:将步骤二得到的上层悬浮液,通过真空冻干后粉碎,得到牦牛骨提取物冻干粉,适用于液体饮品或粉状冲剂的添加剂;或者将上层悬浮液通过喷雾干燥方式,得到提取物干粉,适用于液体饮品生产或粉状冲剂生产的添加剂;或者是将上层悬浮液在通风条件下自然干燥,得到牦牛骨胶状提取物,适用于液体饮品生产或粉状冲剂生产的添加剂。
2.根据权利要求1所述的制备方法,其特征在于:将步骤四得到的胶状提取物按3︰90~120的质量比用热水溶解,所得溶液可用于液体饮品生产的原料。
3.根据权利要求1所述的制备方法,其特征在于:将步骤四得到的胶状提取物与柠檬酸按3:1的质量比进行混合,所得混合物再按3:100~120的质量比用热水溶解,所得溶液可用于液体饮品生产用原料。
4.根据权利要求3所述的制备方法,其特征在于:将得到的混合物与淀粉按1:2的质量比进行混合研磨后成粉,可用于粉状冲剂生产。
