背景技术
侧柏叶(Cacluneu Platycladi )又名丛柏叶,是侧柏的干燥嫩枝梢及叶;中医认为侧柏叶气清香,味苦涩,性微寒.无毒入心`,肝、大肠经;具有凉血、止血、除内湿、散肿毒之功;可治吐血、鼻出血、尿血、血痢、崩漏、风湿痛咳嗽、烫伤等症;明代李时珍《本草纲目》中记载侧柏叶“乃多寿之木,所以可以入服食;道家以之点汤常饮;”侧柏叶作为传统的中药,其预防脱发和生发的功能一直被人忽略;而在诸多古代文献中记载其主要功能为乌发和生发,在《本草纲目》中谓其:浸油,生发,烧汁,黑发;和猪脂,沐发长黑;然而侧柏叶的生发物质基础却尚不明确,经检索关于侧柏叶中的生发活性物质未见报道。
发明内容
本发明公开了中药侧柏叶预防脱发和促进毛发生长作用的最佳提取溶媒和提取条件并制定了质量标准;公开了中药侧柏叶预防脱发和促进毛发生长作用的最佳提取溶媒和提取条件以及最新炮制加工工艺,此方法用于制备防脱发及生发药物、保健品、化妆品、食品等成本低廉,适用于工业化生产;
本发明所述的最佳提取溶剂和提取条件,其特征在于:
是以猪脂和亚麻籽油作为溶媒,提取最佳温度为110℃,提取液最佳浓度为0.25g/ml,提取最佳时间为30min的提取工艺;
本发明所述的所述从胶陀螺中提取的光敏活性物质的提取方法,包括以下步骤:
1)制备猪脂提取液
首先把猪脂放于锅内加热融化成液态,分别称取75ml、85ml、95 ml放于烧杯杯内;取蒸制后的侧柏叶粉碎过80目筛备用;分别制成猪脂混合物高剂量组(0.25g/ml),中剂量组(0.15g/ml),低剂量组(0.05g/ml)的受试液100ml;然后放于电磁炉上加热,温度始终控制在110℃,持续加热30min后,取出冷却;
2)制备亚麻籽油提取液
首先把亚麻籽油放于锅内加热后,分别称取75ml、85ml、95 ml放于烧杯杯内;取蒸制后的侧柏叶粉碎过80目筛备用;分别制成猪脂混合物高剂量组(0.25g/ml),中剂量组(0.15g/ml),低剂量组(0.05g/ml)的受试液100ml;然后放于电磁炉上加热,温度始终控制在110℃,持续加热30min后,取出冷却;
本发明的积极效果在于:本发明公开了中药侧柏叶预防脱发和促进毛发生长作用的最佳提取溶剂和提取条件以及最新炮制加工工艺,此方法用于制备防脱发及生发药物、保健品、化妆品、食品等成本低廉,具有明显的临床效果;
试验例:
通过以下试验证明本发明提取的物质是生发活性成分:
1.材料
1.1本发明提取的生发活性成分;
1.2受试动物:昆明种小鼠,体重 20-22g, 购自吉林大学实验动物中心;
2.试验方法:
2.1硫化钠脱毛模型的建立
健康成年昆明雄性SD小鼠80只,用基础饲料喂养3-5d,用剪刀剪去其背部的毛发,面积约2X3cm,然后涂上浓度为8%的硫化钠溶液,过5分钟后,用温的蒸馏水洗净,防止烫伤皮肤;此方法诱导可以高度同步的生长期毛囊的发育,该毛囊与自发形成的生长期毛囊在形态上无差别;
2.2实验动物分组及给药
2.2.1硫化钠脱毛模型动物分组及给药
对小鼠采用硫化钠脱毛模型,从而研究侧柏叶对促进正常毛发生长的作用;至于侧柏叶能否减弱因环磷酰胺引起的毛囊损伤导致的脱毛,从而起到预防脱毛的的作用,将由同时进行的预防脱毛作用试验进行研究;
选取脱毛区光滑且无皮肤破损的小鼠,随机分为8组,分别为:阴性对照组、猪脂高剂量组(Lard-G)、猪脂中剂量组(Lard-G)、猪脂低剂量组(Lard-G)、亚麻籽油高剂量组(Linseed-G) 、亚麻籽油中剂量组(Linseed-Z)、亚麻籽油低剂量组(Linseed-D)、阳性对照组;
实验小鼠正常饲养,从脱毛后第2天背部涂抹受试液及阳性药物0.3mL,用于促进毛发生长作用实验的小鼠背部连续涂抹给药12天;
2.2.2实验观察及评价方法
对于化疗模型组的小鼠逐日观察脱毛情况,并按照未脱毛记0分,头颈及背部稀疏脱毛记1分,头颈及背部中度脱毛记2分,头颈及背部明显脱毛记3分的标准对小鼠脱毛情况进行打分,结果进行秩和检验(脱毛积分)分析;对于促进毛发生长的小鼠无毛发生长记0分,有少量细小绒毛出现记1分,有较明显毛发出现记2分,毛发生长良好的记3分,毛发生长同正常小鼠无异记4分,所述状况显著记“*”;
结论
通过实验研究发现了中药侧柏叶预防脱发和促进毛发生长作用的最佳提取溶媒和提取条件,提取的两种最佳溶媒分别是猪脂和亚麻籽油,提取最佳温度为110℃,提取液最佳浓度为0.25g/ml,提取最佳时间为30min;侧柏叶作为药食两用的中药,具有非常明显的促进毛发生长、预防脱发、生发、乌发等作用;侧柏叶可以被开发为保健食品、药品、洗护用品等,使侧柏叶广泛受益于人类。
Claims (2)
1.中药侧柏叶以猪脂和亚麻籽油,提取最佳温度为110℃,提取液最佳浓度为0.25g/ml,提取最佳时间为30min的提取工艺方法。
2.根据权利要求1所述的应用,侧柏叶的猪脂和亚麻籽油提取物的制备过程如下:
猪脂组:首先把猪脂放于锅内加热融化成液态,分别称取75ml、85ml、95 ml放于烧杯杯内;取侧柏叶粉碎过80目筛备用;分别制成猪脂混合物高剂量组(0.25g/ml),中剂量组(0.15g/ml),低剂量组(0.05g/ml)的受试液100ml;然后放于电磁炉上加热,温度始终控制在110℃,持续加热30min后,取出冷却;
亚麻籽油组:首先把亚麻籽油放于锅内加热后,分别称取75ml、85ml、95 ml放于烧杯杯内;取侧柏叶粉碎过80目筛备用;分别制成猪脂混合物高剂量组(0.25g/ml),中剂量组(0.15g/ml),低剂量组(0.05g/ml)的受试液100ml;然后放于电磁炉上加热,温度始终控制在110℃,持续加热30min后,取出冷却;权利要求1所述的中药侧柏叶预防脱发和促进毛发生长作用的最佳提取溶剂和提取条件以及最新炮制加工工艺,此方法用于制备防脱发及生发药物、保健品、化妆品、食品等成本低廉,适用于工业化生产。
